Abstract

Triglyceride (TG) is a simple and the main lipid of the daily diet. Nowadays, TG is emerging as an independent risk factor for cardiovascular disease, and increasing clinical data that indicate a high level of this simple lipid in serum may alert and play a role in liver impairment. Red yeast rice (RYR) reduces serum TG in human and animals. The rice is widely used as a natural inhibitor of HMG-CoA reductase to correct serum cholesterol level. Bioactive components of the rice are well known to have antioxidant properties. This study examined whether RYR protects hepatocytes by evaluating the serum AST-ALT, HDL-Cholesterol (HDL-C), TG levels, and the number of liver foam cells in hypertriglyceridemic rats. Twenty male Wistar rats were grouped into 5. Four groups received a high-fat diet (HFD), 40 g/animal/day for 60 days to induce hypertriglyceridemic condition. Along with the HFD treatment, three groups received 108, 54, and 27 mg/kgBW/day of RYR, respectively. Two other groups received standard and only HFD diet, respectively. Intracardiac blood was collected for measuring AST and ALT using AST or ALT activity assay Kit, respectively, serum TG and HDL-C by Enzymatic Caloric Test.  Hematoxylin-Eosin–stained 4µmm thick slices of liver tissues were prepared to count foam cell number by a light microscope with 400x magnificence. Data were analyzed using the Kruskal Wallis continued by the Mann Whitney U test. The p values of < 0.05 were considered to be significant. The present study found that 108, 54, and 27 mg/kgBW/day of RYR significantly decreased serum TG, HDL-C, AST, and ALT compared with those of hypertriglyceridemic rats receiving no RYR. The decreasing levels of the parameters were in relation to the doses of RYR. The doses of 108 and 54 mg/kgBW/day resulted in complete recovery of the liver tissues suffered from steatosis (p < 0.05). The RYR dose of 108 mg/KgBW/day completely corrects the serum HDL-C level. In conclusion, red yeast rice may have a potency to protect hepatocytes injuries due to hypertriglyceridemia.

Abstract

Introduction: The essential oil of soursop (Annona muricata Linn.) leaves contains terpenoid, alkaloid, and phenolic compounds. Terpenoid, alkaloid and phenolic function as antioxidants and antiinflammatory agents appropriately. When used in high concentrations and in long period, it may become toxic, which mayincreasein LDL and decrease in HDL. The study is to determine the safety of soursopleaf essential oil on LDL and HDL levels in wistar rats.Methods: This was an invivo experimental study with a control group post test only design using 20 male and 20 female wistar rats divided into 8 groups, consisting of KN (tween 80), P1 (1,25 mg/kgBW), P2 (2,5 mg/kgBW), and P3 (5mg/kgBW) both male and female rats. This study was conducted for 28 days then the LDL and HDL levels were checked after the treatment. Results were analized using One Way ANOVA test followed by Least Significant Difference (LSD). Results are assumed significant if p<0.05.Results and discussion: Administration of soursop leaf essential oil in P1 group showed the highest LDL levels and a higher dose of soursop leaf essential oil will cause a lower LDL levels in both male and female groups. The P2 group showed the highest HDL levels and P3 group had decreased HDL levels compared to P2 group in both male and female groups. The increasing of LDL levels and the decreasing of HDL levels in all treatment groups, is suspected to be caused by active compounds in soursop leaf essential oil, body weight differences and food consumption differences on rats.Conclusion: The doses between 1,25 to 5 mg/kgBW is considered to be safe, in which no increase in LDL and decrease in HDL in normal male and female rats was observed.Keywords: Essential oil of soursop leaves,Annona muricata Linn., Subchronic,LDL,and HDL.

Abstract

AbstractBackground. Hyperglycemia was reported to harm liver and induce metabolic inbalance. The tree bark of Cinnamomum burmannii (C. burmanii) was potential as anti-hiperglycemic agent. The bark was rich with secondary metabolites that may lead to possess other advantages for diabetes.Objectives. To study the effect of aqueos extract of C.burmannii (AEC) on liver and metabolic condition by measuring necrotic hepatocytes, serum urea and uric acid levels in alloxan-induced diabetic rats (AIDRs).Methods. Three groups of fasted rats (free allowed to water) were intraperitoneally injected with 150 mg/Kg BW in 1 ml normal saline 6 hours prior to intraperitoneal injection of 2 ml 10% glucose to induce AIDRs, the animals were allowed to 5% glucose solution for 24 hours afterward. The AIDRs were treated with 0.5 ml, 1ml, and 2 ml of AEC respectively for 2 weeks. Hematoxyline-Eosine-stained liver slices were observed for necrotic hepatocytes by light microscope. Serum urea and uric acid levels were measured by Glutamate dehydrogenase and Uricase-PAP methods respectively. Data were analyzed by ANOVA followed by LSD test, p values of < 0.05 were considered to be significant.Results.The given doses of AEC lowered urea and uric acid serum levels of AIDRs (p = 0.000) which approximated to the non diabetic ratss serum levels (p > 0.05). Except the smallest dose, the remaining doses of AEC reduced necrotic hepatocytes number in AIDRs (p = 0.000).Conclusion.Aqueos extract of C.burmannii protected hepatocytes and corrected serum urea and uric acid levels of alloxan-induced diabetic rats.Keywords: Cinnamomum burmanii; diabetes; necrotic hepatocytes; serum urea: serum uric acid.

Abstract

Desa Sukodono Kecamatan Dampit Kabupaten Malang merupakan salah satu daerah tujuan KKN Universitas Islam pada periode Pebruari 2020. Tujuan Kegiatan pengabdian kepada masyarakat (PPM) dalam rangka kegiatan KKN yang dilakukan oleh kelompok 63 adalah (1) Meningkatkan perekonomian masyarakat di Desa Sukodono dengan cara pembuatan produk olahan selai kopi, (2) Meningkatkan movitasi belajar siswa di SDN 1 Sukodono, dan (3) Meningkatkan tindakan preventif dan promotif stunting di Desa Sukodono. Kegiatan dilakukan melalui metode penyuluhan dan pelatihan, diskusi dan tanya jawab, serta demonstrasi secara langsung. Hasil akhir dari kegiatan menunjukkan bahwa kegiatan pengabdian kepada masyarakat (PPM) mampu memberikan motivasi dan perubahan pola fikir kepada masyarakat di Desa Sukodono terutama tentang bagaimana cara membuat produk olahan dari serbuk kopi, meningkatkan motivasi belajar siswa dan meningkatkan tindakan preventif dan promotif stunting. Kendala-kendala yang muncul dapat diatasi dengan metode pelatihan secara bertahap. Adanya MOU kerjasama dengan pemerintah daerah dan pemerintah desa perlu dilakukan untuk keberlajutan program secara kontinue dan dievaluasi keberhasilannya dalam meningkatkan ekonomi, kesejahteraan dan penurunan angka stunting di Kabupaten Malang.

Abstract

ABSTRACTInflammatory reactions give rise to a series of processes and symptoms that can cause pain and damage to nearby cells. Inflammatory reactions are mediated by inflammatory and proinflammatory cytokines. NSAID class (non-steroidal anti-inflammatory drug) drugs have been developed to inhibit the inflammatory process by suppressing COX-1 and / or COX-2 enzyme activity. However, this drug can cause gastric mucosal irritation. the ethanol extract of Annona muricata leaf has an analgesic and anti-inflammatory effect[3].The purpose of this research is to know analgesic and antiinflamatory activity from ethanol extract and essential oil of Annona muricata leaf.The method used was the study of in silico by molecular docking of active compounds in ethanol extracts and essential oils to the COX-1, COX-2, phospolipase-A, and TNF-alpha receptor molecules as target molecules. The molecular docking results show that Annona muricata ethanol extract can act as an anti-inflammatory and analgesic, especially through strong bonds between "rutine" compounds with COX-2 receptors with affinity -10 kcal / mold and "quercetin" with affinity bindings -9.8 kkal.mol, better than dexametason (binding affinity -9.6 kcal / mol). Keywords:Inflamation, COX-1, COX-2, phospolipase-A, TNF-alfa, docking

Abstract

The aim of the study was to prove the potential of Soursop Leaves Water Extract (SLWE) content as DPP4 inhibitor an in silico method. The extraction process of soursop leaves uses water solvent with the infusion method. Withdrawal of the active compounds using acetone solvent. Identification using the GCMS method. Molecular docking of SLWE active compounds, Linagliptin as a control and DPP4 protein target  using  Autodoc Vina application. 2D visualization using LigPlot. Validation of affinity of active compounds of Annona muricata Leaves  using the measurement results of free binding energy, the number  similarity of bond position to active site ot  the protein target, and the number of hydrogen bond compared to controls. To determine the potential effect as a drug and toxicity using the 5 rules of Lipinski and ADME. The results of molecular docking found that the active SLWE compounds that have affinity close to control is butenedioic acid. It has the ∆G of  -7 kcal / mol, binds 53% of amino acid residues of DPP4 and has one hydrogen bond. Based on 5th rules of Lipinski and ADMET, Buetenoic acid has the potential to be developed as DPP4 inhibitor drugs which is administered orally and had a non-toxic effect. The the Conclusion Butenedioic acid, the active compound found from SLWE is potential as DPP4 inhibitors.

Abstract

 AbstractIntroduction: Rifampicin is one of anti-tuberculosis drugs are reported to have hepatotoxic effects through increasing ROS (Reactive Oxygen Species) so thattriggering necrosis of liver cells and stimulates the formation of TNF-α. Essential oil of Annona muricata Linn. contains linalool, β-caryophyllen, trans-caryophyllen, germacrene A, germacrene B, germacrene D, sesquisterpene, β-canidine, γ-canidine. The objective of this study is to investigate the effect of essential oil of Annona muricata Linn. on levels of Tumor Necrosis Alpha(TNF-α) liver in male wistar rats which induced by rifampicin.Methods: This was an invivo experimental study with control group post test only design and use 25 male wistar rats and divided into 5 groups, they are KN (corn oil), KP (Rifampicin), P1 (Rifampicin + Essential oil 1,5%), P2 (Rifampisin + Essential oil 3%), P3 (Rifampisin + Essential oil 6%) for 6 weeks and then levels of TNF-α liver will be checked. Analysis of the data using One Way (ANOVA) test and then tested by post hoc test using Least Significant Difference (LSD). The result significant if p<0,05.Result: Induced rifampicin can increased levels of TNF-α liver than group KN (p<0,05). Treatment of essential oil of Annona muricata Linn. on P1, P2, and P3 can reduced levels of TNF-α liver than group KP (p<0,05). Allegedly this is due to the active ingredient in the essential oils act as antioxidants and antiinflammatory.Conclusion: Essential oil of Annona muricata Linn. at all doses can reduced levels of TNF-α liver in male wistar rats which induced by rifampicin.Keywords:rifampicin,essential oil of Annona muricata Linn., TNF-α liver

Abstract

A high-fat and high-fructose diet can lead to obesity which contributes to an increased risk of heart failure complications. Soursop leaves are known to have the potential of antioxidant, but research about the effects of soursop leaf on complications of heart failure has never been studied. This study evaluated whether soursop leaf aqueous extract can inhibit the total of cardiomyocyte necrosis, cardiomyocyte diameter, and density of cardiac collagen in obese rats. The heart organs of obese rats that have been paraffin, which randomly divided into normal group, obesity group, and soursop leaf aqueous extract (SLAE) group with dose I (100 mg/kgbw), II (200 mg/kgbw), and III (400 mg/kgbw) (n=6 rats). The rats were induced by high-fat high-fructose and SLI diets for 10 weeks. The number of necrosis and diameter cardiomyocytes were measured by using Hematoxylin Eosin staining, while the density of cardiac collagen was measured by Masson's Trichrome staining and then observed with a trinocular and dot side microscope at 200x and 400x magnification. Statistical analysis using One Way ANOVA and continued by LSD test. We found that group SLAE with doses I, II, and III significantly reduced the number of cardiomyocyte necrosis and cardiac collagen density compared to the obesity group. The SLI group with doses II and III significantly reduced the diameter of cardiomyocytes compared to the obesity group, while the SLAE group with the dose I was unable to reduce the diameter of cardiomyocytes. In conclusion, Soursop leaf aqueous extract can inhibit the increase of cardiac collagen density, number necrosis and diameter cardiomyocyte.

Abstract

ABSTRAK  Pendahuluan: Tempe merupakan makanan produk fermentasi yang memiliki kandungan probiotik. Kacang merah memiliki efek antihiperkolesterol dikarenakan dalam kacang merah mengandung proantosianidin dan isoflavon yaitu genestein, daidzein, biochanin A, glycitein, dan formononetin. Tujuan penelitian untuk memprediksi afinitas yang terbentuk antara senyawa aktif tempe kacang merah dengan enzim target serta memprediksi fisikokimia,  ADME, toksisitas senyawa aktif tempe kacang merah.Metode: Penelitian secara in silico dilakukan pada 10 senyawa aktif tempe kacang merah dengan metode molecular docking pada http://www.dockingserver.com. Struktur 3D senyawa aktif  tempe kacang merah dan orlistat diambil dari Pubchem, enzim Lipase pankreas dan Fatty Acid Synthase (FAS)  diambil dari RCSB dan Uniprot. Konversi format pdb menggunakan http://swissmodel.expasy.org. Analisa afinitas senyawa ligan terhadap enzim dengan membandingkan energi bebas, konstanta inhibisi, interaksi permukaan, dan residu asam amino dengan orlistat. Analisa fisikokimia, ADME, dan toksisitas senyawa aktif tempe kacang merah menggunakan pKCSMHasil: Hasil pKSCM online tool didapatkan seluruh senyawa aktif tempe kacang merah memiliki efektifitas yang baik untuk bekerja di intestinal dan tidak mempunyai  efek hepapatotoksik serta efek toksisitas lainnya, sementara catechin diprediksi bersifat karsinogenik. Hasil molecular docking didapatkan Kaempferol-O, Quercetin-O-acylhexoside dan Glisitein memiliki afinitas tinggi dalam menghambat enzim FAS. Genistein mempunyai afinitas rendah dalam menghambat lipase pankreas,namun  diprediksi memiliki potensi yang sama dengan kontrol.Kesimpulan: Seluruh senyawa aktif tempe kacang merah kecuali catechin, efektif bekerja di interstinal dan tidak bersifat toksik. Senyawa aktif tempe kacang merah yang memiliki afinitas terhadap FAS adalah  Kaempferol-O, Quercetin-O-acylhexoside dan Glisitein. Adapun yang memiliki afinitas terhadap lipase pankreas adalah Genistein.Kata Kunci: antikolesterol, tempe, senyawa aktif tempe kacang merah, in silico, pkCSM.

Abstract

Pendahuluan: Biji kacang merah dalam bentuk produk fermentasi mengandung senyawa aktif yang diketahui dapat menurunkan kadar glukosa darah diduga bekerja pada enzim α-amylase dan maltase-glucoamylase (MGAM). Tujuan penelitian ini memprediksi afinitas yang terbentuk antara senyawa aktif produk fermentasi biji kacang merah dengan enzim target dan memprediksi fisikokimia, farmakokinetik, toksisitas senyawa produk fermentasi biji kacang merah.Metode: Penelitian secara in silico dilakukan pada 10 senyawa aktif yang dominan pada produk fermentasi biji kacang merah. Proses docking menggunakan metode molecular docking di web http://www.dockingserver.com. Menggunakan kontrol acarbose dan enzim α-amylase dan MGAM. Struktur 3D senyawa dan kontrol diambil dari Pubchem. Konversi format pdb menggunakan http://swissmodel.expasy.org. Analisa afinitas senyawa ligan terhadap enzim membandingkan energi bebas, konstanta inhibisi, interaksi permukaan, dan residu asam amino dengan acarbose. Analisa fisikokimia, farmakokinetik, dan toksisitas menggunakan pKCSM pada senyawa aktif produk fermentasi biji kacang merah dan acarbose.Hasil: Berdasarkan afinitasnya, senyawa hesperetin-glucoronide-hexoside dan kaempferol 3-0-glucoside diprediksi memiliki afinitas terhadap enzim enzim α-amylase dan MGAM. Analisa pkCSM menunjukkan senyawa hesperetinglucoronide-hexoside dan kaempferol-3-O-glucoside berpotensi bekerja optimal pada lumen usus dan tidak toksik.Kesimpulan: Senyawa produk fermentasi biji kacang merah hesperetin-glucoronide-hexoside dan kaempferol 3-0- glucoside diprediksi menghambat enzim α-amylase dan MGAM. Pada pkCSM, hesperetin-glucoronide-hexoside dan kaempferol-3-O-glucoside diprediksi bekerja optimal di lumen intestinal.Kata Kunci : antidiabetes; fermentasi biji Phaseolus vulgaris L.; in silico; pkCSM
