Abstract

Moringa oleifera tergolong mudah dijumpai di Indonesia dan mengandung senyawa quercetin yang termasuk dalam jenis flavonoid, dimana senyawa ini berperan sebagai agen hipoglikemik. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian ekstrak Moringa oleifera terhadap kadar glukosa dan histopatologi pankreas mencit hiperglikemia. Metode yang digunakan pada penelitian ini yaitu ekstraksi daun Moringa oleifera dengan konsentrasi 42 mg/kgBB, 70 mg/kgBB, dan 98 mg/kgBB, lalu penginduksian aloksan secara intraperitoneal, pengaplikasian ekstrak Moringa oleifera, pengambilan organ pankreas dan pembuatan histologi organ pankreas. Hasil penelitian menunjukkan bahwa Pemberian ekstrak Moringa oleifera berpengaruh pada penurunan kadar glukosa dan penambahan diameter pulau Langerhans pankreas mencit diabetes. Konsentrasi ekstrak yang paling berpengaruh adalah 98 mg/kgBB. Maka dapat diambil simpulan bahwa ekstrak Moringa oleifera pada konsentrasi berpotensi sebagai antihiperglikemia.

Abstract

Aspartame has a lower calorie rating than other sweeteners. for example, cyclamate, lactose, saccharin, fructose and maltosa. Although it has low calorie, aspartame have 60 times sweeter than sugar (sucrose). Induction of aspartame can cause elevated levels of blood glucose and damage to the pancreatic β cells of mice. The purpose of this study was to analyze the dosing of aspartame levels; 13 mg/Kg body weight, 39 mg/Kg and 78 mg/Kg and those effect to the levels of glucose, cholesterol and haemoglobin of female mice (Mus musculus). The methode of research laboratory experiments using a complete randomized design (RAL). This study was using 24 mice with 4 treatment groups. Mice were given aspartame during surgical treatment, mice were made to perform measurement of Hb levels, glucose levels and cholesterol levels of neonatal mice. Data analysis using the Kruskal Wallis because Gaussian data but not homogeneous. The results showed an increase in the levels of glucose and cholesterol levels, but statistically there is no difference in glucose levels (p = 0,087), Hb (p = 0,899) and cholesterol (p = 0,168) in mice (Mus musculus) who were given variations of the dose of aspartame.

Abstract

Daun jambu air (Syzygium aqueum) dikenal sebagai salah satu obat tradisional, untuk menyembuhkan beberapa infeksi akibat antigen berupa bakteri. Penelitian ini bertujuan untuk mengetahui kandungan senyawa aktif yang terdapat dalam daun jambu air dengan beberapa pelarut pada proses maserasi. Metode ekstraksi yang digunakan dalam penelitian ini adalah ekstraksi dingin berupa maserasi dengan pemilihan pelarut yakni metanol (CH3OH), etil asetat dan n-heksana. Uji fitokimia yang dilakukan adalah uji saponin, flavonoid, alkaloid dan triterpenoid/steroid. Dari uji yang dilakukan diketahui sampel ekstrak daun jambu air dengan pelarut metanol positif mengandung beberapa senyawa aktif berupa flavonoid, saponin, alkaloid dan triterpenoid. Ekstrak daun jambu air dengan pelarut etil asetat dan n-heksana positif mengandung beberapa senyawa aktif berupa flavonoid, alkaloid dan triterpenoid. Pelarut yang paling bagus untuk mendapatkan banyak senyawa aktif adalah metanol. Kata kunci : Jambu air (Syzygium aqueum), maserasi, senyawa aktif

Abstract

A Virus is an individual that cannot be described as an animal or a plant. If animals and plants contain two nucleic acids (DNA and RNA), on the contrary, the virus only has one of them. These nucleic acids can stimulate a complete virus replication cycle. The virus can only replicate and live on a living host if the host is finally dead, then the virus will move on the cells that are still alive. The virus has genetic material which is a protective protein coat called a capsid. Viruses can infect various varieties of organisms, both eukaryotes (animals, plants, protists, and fungi) and prokaryotes (bacteria and archaea). The Virus infects bacteria known as bacteriophage (phage). The Virus can cause serious diseases for humans such as AIDS, HIV, rabies, etc. Dogs belonging to Canidae family are the sibling of wolves, foxes and raccoon dogs. Among all members of Canidae, dogs have the most closely related to wolves which are the ancestors of dogs. The Canidae family generally has a small elongated body, sharp ear and muzzle, sharp smelling, can run fast and can swim. Dogs are human best friends. While taking care of the dogs, they can be attacked by various diseases. The closeness of the relationship between humans and dogs raises the potential for disease transmission, especially zoonosis and pandemics viruses. Abstract

C.capitulata and C.latofilia are recognized as a medical plants in Asia, especially for ancient India and China. In Indonesia, i.e Kebonharjo (a village in Samigaluh District, Kulon Progo, Yogyakarta), people use it as a sweetener replacing sugar. Phenol, neonorlignan, and protein inside these plants had been examined by some researchers. However, the nutrient content inside the fruit are rare or slight recorded. This study aims to reveal the composition of minerals (Ca, Fe, Mg, K), carbohydrate compound (glucose, fructose, sucrose) and Vitamin C in their fruit, therefore, the development to preserve these species can be concerned due to the plan of “Bedah Menoreh” program, a program of road construction connecting between New Yogyakarta International Airport (NYIA) and Borobudur Temple. The method for finding out the nutrient content is by laboratorium analysis. Result showed that both fruit contained potentially high of mineral, especially Ca (calcium), but C.capitulata and C.latifolia contained no sucrose and glucose respectively.

Abstract

Salah satu komponen perekonomian penting yang ada di Indonesia adalah perindustrian. Perkembangan perindustrian yang ada di Indonesia mengalami peningkatan yang begitu cepat. Percepatan pertumbuhan industri dapat mengakibatkan masalah bagi lingkungan. Limbah yang berasal dari Industri mengandung logam berat merupakan salah satu masalah yang bisa berdampak pada kesehatan manusia. Beberapa mikroorganisme diketahui mampu meremediasi logam berat seperti Bacillus sp dan Pseudomonas sp. Penelitian ini bertujuan untuk mengetahui jenis bakteri pengakumulasi logamberat timbal (Pb) pada saluran pembuangan limbah industry kabupaten gowa. Sampel tanah berasal dari saluran pembuangan limbah pabrik tali rafia PT. Intra Polyplast Palangga, pabrik beton PT. Cipta Beton Sinar Perkasa. Sebanyak 6 isolat bakteri berhasil diisolasi dari limbah industri dan di uji kemampuan pengakumulasi logam berat timbal (Pb) menggunakan AAS (Atomic Absorpsion Spectrophotometer), dan isolat (1mTR) bergenus Bacillus yang mampu mengakumulasi logam sebesar 7 mg/l, isolat (5mTR) bergenus Pseudomonas yang mampu mengakumulasi logam sebesar 7 mg/l, isolat (10mTR) bergenus Bacillus yang mampu mengakumulasi logam sebesar 8 mg/l, isolat (1mPB) bergenus Bacillus yang mampu mengakumulasi logam sebesar 9 mg/l, isolat (5mPB) bergenus Bacillus yang mampu mengakumulasi logam sebesar 11 mg/l, dan isolat (10mPB) bergenus Bacillus yang mampu mengakumulasi logam sebesar 9 mg/l.

Abstract

DNA isolation is one of a series of methods that must be carried out on the basic techniques of Molecular Biology Analysis. Especially PCR-based molecular marking techniques. Many ways are done in DNA isolation. This study discusses the comparison of the results of DNA isolation using two methods. Simple DNA isolation methods and using Kit. Samples of Psidium guajava leaves used were taken from 15 different locations used. In general, DNA isolation methods include three steps, namely destruction, precipitation, and purification. Simple DNA isolation is done with detergents, alcohol groups, which are commonly available in the laboratory. Methods of DNA isolation with KIT are carried out according to the Promega Universal Wizard KIT protocol. The comparison results are seen from spectrophotometric absorption Å230 nm, Å260 nm, Å280 nm, Å320 nm, ratio Å260/Å230, ratio Å260/Å280 to see DNA purity, protein concentration before purification step, and DNA concentration produced. The results showed that there were no statistically significant differences in the results of DNA isolate spectrophotometry. However, the use of KIT with modified protocols is more recommended if researchers want to carry out DNA analysis more precisely and accurately. Abstract

Ageratum conyzoides L. leaf has the main compound of flavonoids and alkaloids that contribute to accelerate the healing process of diabetic ulcers with Staphylococcus aureus infection that can affect the quality of life of patients. This study aims to test the leaves of Ageratum conyzoides L. on the recovery of diabetic ulcers of mice infected with Staphylococcus aureus. The method used is the extraction of Ageratum conyzoides L leaves as the material of spray 15%, induction of diabetic compound (Alloxan monohydrate), length wound (1cm) and bacterial infections, and spray application with 1 times daily frequency, 2 times daily, 3 times a day for 9 days. The results showed that the lenght of wounds in group A, B, C, D, and E were 0,5; 0,8; 0,4; 0,2; 0,0. The administration of Ageratum conyzoides L. spray which has active flavonoide substances, alkaloid substance, and saponin substances has an effect on decreased length of diabetic wounds and restoration of dermis tissue structure (increases epithelial cells proliferation, initiates fibroblast cells, and stimulates type 1 collagen). Spray treatment 3 times a day (group E) was more influential than the other treatments, which experienced wound closure in 9 days. Thus, Ageratum conyzoides L. leaf spray is potential for therapy of diabetic ulcers infected with Staphylococcus aureus.

Abstract

Aspartame is safe for consumption according to the prescribed ADI, but this artificial sweetener is not nutritious or does not contain calories so it is not recommended to be consumed during pregnancy. This study was to analyze cariometric effect of aspartame doses in the development of fetal mice (Mus muculus L.). The type of research used was experimental laboratory research on 24 adult mice with 4 treatment groups, control, aspartame dose 13 mg / Kg BB, 39 mg / Kg BB and 78 mg / Kg BB. Data was analyzed using SPSS 16 with one way ANOVA. Based on the results of the study there were changes in the number and weight of the fetus due to aspartame doses, there was no change in fetal length due of aspartame dose variations. Consumption of aspartame during pregnancy should be avoided because it can cause teratogenic effects. For further research, it is necessary to observe the morphology and histology of the fetus which is expressed by aspartame to determine the types of congenital abnormalities.

Abstract

The objective of this work was to isolate and identify of 1-aminocyclopropane-1-carboxylic acid (ACC) deaminase producing endophytic bacteria from root of local agricultural plantation by using 16S rRNA gene as genetic marker. Twelve root samples were collected from local agricultural plantation in Java area. After surface sterilization, each root sample was inoculated in nutrient agar media and the grown colonies were further purified and tested for the ability to grow in N-free minimal medium containing AIB as a sole of nitrogen source. The selected colonies were further tested for their ACC deaminase activity by measuring the rate of conversion of ACC into α-ketobutyrate. From this work, 12 bacterial strains that exhibited the ACC deaminase activity by 123.75 to 1461.44 nmol α-ketobutyrate/mg/hour were successfully isolated. Based on the 16SrRNA gene sequences, those bacterial isolates were identified as Sphingobacterium multivorum BK1, Bacillus mycoides CB2, Pantoea dispersa CK4, Pantoea agglomerans KD6.2, Enterobacter ludwigii KW3, Bacillus aryabhattai TW7, Pseudomonas monteilii KS12, Pseudomonas plecoglossicida KS16.2, Pseudomonas putida PIR3C, Stenotrophomonas maltophilia PIR5, Lysinibacillus pakistanensis PIC5, and Raoultella terrigena PCM8. Pseudomonas putida PIR3C and Pseudomonas monteilii KS12 showed promising ACC deaminase activity and therefore it could be as a good candidate for further application in plant growth promoting in stress conditions.
