Abstract

Cyclosorus parasiticus (L.) Farw. from Mount Gumitir dominates the understory ferns vegetation along the roadside of a pine-mixed coffee plantation. Previous studies reported the metabolite profile with the flavonoid and alkaloid content. Current observations based on morphological structure of the specimens collection indicated a high variations, therefor molecular identification was required to support the phylogenetic data of the species. The aim of the study was to identify the Cyclosorus parasiticus sample originate from Gumitir using DNA barcode. We used three primers, i.e. ribulose 1,5-biphosphate carboxylase (rbcL), maturase K (matK), and Internal transcribed spacer 2 (ITS2) for molecular identification. The result showed that Cyclosorus parasiticus from Gumitir has a highest similarity with Christella latipinna (NC070300) from China with a value of Percent Identities value of 99.11% based on the ITS2 marker. The rbcL data in this study might contribute to supporting the molecular genus concept from recent Thelypteridaceae phylogeny.

Abstract

Several references indicate that active metabolites found in natural plant resources such as flavonoids and saponins have the potential as natural insecticides. One of natural plant resource that has the potential as a natural insecticide is durian, because in durian albedo there are flavonoids and saponins. In addition, durian seeds also contain flavonoids of 29.74 μg / mL. Based on this background, the author took the initiative to test the activity of flavonoid content in durian seed extract and albedo to determine its effectiveness as a natural insecticide for mealybug pest control. This research consists of 2 stages, including; extraction of flavonoids from durian seeds and albedo (peels), and testing of flavonoid activity of durian seed and peels extracts against mealybug mortality. Extraction of flavonoids from durian seeds and peels using socletation method and spectrophotometry tested. The results of flavonoid and phenolic analysis of durian seed extract are higher than durian albedo extract. The effectiveness test was carried out on the population of mealybug pests that had been acclimatized for 24 hours, placed on cocoa peels media that had been soaked in the treatment solution for 15 minutes, then the mortality rate was observed. The treatment given was albedo extract 12.5 g/100ml; albedo extract 25 g/100 ml; durian seed extract 12.5 g/100 ml; 25 g/100ml; albedo and seed extract 12.5 g/100 ml; albedo and seed extract 25 g/100 ml; positive control and negative control. The test results showed that the mortality of mealybugs in the treatment of seed extract 12.5 g /100 ml was not significantly different from the positive control treatment, this showed that durian seed extract 12.5 g / 100ml is expected to replace the use of chemical insecticides in the control of mealybug pests.

Abstract

Citronella essential oil (Cymbopogon nardus (L.) and basil leaf essential oil (Ocimum basilicum (L.) offer various benefits, including antibacterial properties. This study aims to identify the chemical compounds present in these oils obtained through local distillation at PT Syailendra Bumi Investama in Wonorejo Village, Gondangrejo, Karanganyar, Central Java. Gas Chromatography-Mass Spectrometry (GC-MS) analysis was conducted at the BRIN (National Innovation Research Agency) in Serpong, South Tangerang, Banten, to characterize citronella essensial oil and basil essential oil. The findings indicate that citronella essential oil meets the criteria of SNI 06-3953-1995, with three compounds showing significant concentrations: Geraniol (26.49%), citronellol (14.53%), and citronellal (10.95%). Similarly, research on basil leaf essential oil conforms to ISO 11043 - 1998 standards, revealing high concentrations of two compounds: Estragole (55.80%) and linalool (22.35%).

Abstract

Changing perceptions regarding the importance of conservation can be an effort to increase conservation awareness which is still low in society. This research aims to determine the effectiveness of implementing the Transformative Green Economy in Biology (T-GEB) Learning Model in changing students' perceptions of conservation. The sample for this research is class X students of State Senior High School 1 Turen for the 2022/2023 academic year. The type of quantitative research is pre-experimental using pretest-posttest and questionnaires, while qualitative data collection uses reflective essays. The application of the T-GEB Learning Model shows that learning is quite effective in improving students' cognitive learning outcomes, with an N-Gain Score of 58% on biodiversity material and 6.4% on environmental change material. the application of the T-GEB model can change students' perceptions of conservation from previously having a strong percentage of 77.7% to 91.3% which is classified as very strong based on the results of the conservation awareness questionnaire. Changes in students' perceptions of conservation are also supported by the results of reflective essays, with three themes formed, namely awareness of the important role of oil palm, perception of the impact of oil palm expansion, and awareness of conservation through oil palm expansion.

The results obtained from material validation are 100% with very valid criteria. Furthermore, media validation is 95.47% with very valid criteria, practitioner validation with a percentage value of 97.27% with very valid criteria. Practicality test averaged 96% including very practical criteria. The effectiveness test was obtained through the e-module implementation stage with the Nonrandomized Control-Group Pretest-Posttest Design research design using the N-gain score calculation which was 0.70 with high criteria on experimental class. 0.29 for control class which was on low criteria. Based on the test results that have been carried out, the development of e-modules based on Problem-based learning in critical thinking skills is said to be valid, practical, and effective in improving students' critical thinking skills.

Abstract

The safety of using liquid soap to wash fruit or vegetables is unclear. The purpose of this study was to determine the effect of liquid soap on the body weight of mice (Mus musculus). This research is pure experimental using a completely randomized design. The treatment given was soaking fruits and vegetables in liquid soap with a concentration of 20 mL/L for 30 and 60 seconds, each treatment had 10 replications. The mice used were male, aged 4 weeks, with a total of 30 mice selected by cluster random sampling. Fruit and vegetable extracts were administered once daily, alternating fruit and vegetable extracts for 8 weeks in addition to the main food. Mice’s body weight was measured using a digital scale. The data were analyzed using One Way Anova and Duncan's advanced test. The results showed that liquid soap had a significant effect on the body weight of mice, but the average body weight of fruits and vegetables soaked in liquid soap for 30 seconds and 60 seconds was not significantly different. The conclusion of this study supports other research that the use of liquid soap containing surfactants for a long time continuously has a negative effect on body weight.

The methodology of this research is an exploration of collecting data techniques using the sweeping net. This research was held in January to December. Sampling was done with 2 sessions are 09.00-11.00 WIB and 15.00-17.00 WIB with three times repetition each session. Sampling was done at two places especially in Sukatani and Cikedung. The procedures of this research are samples were taken by insect net and the abiotic components were calculated such climatic (humidity, air temperature, light intensity, wind velocity) edaphic (soil moisture, soil Ph). Based on the results, Sukatani obtained a diversity index was 1.98 and Cikedung was 2.46 with both categories are medium. Then, Sukatani obtained an evenness index was 0.67 and Cikedung was 0.87 with both categories are high. Environmental conditions are in the optimal range for the life of dragonflies. The final results of this research were made as a field guide book based on conservation education species diversity of dragonfly at Rawa Danau Nature Reserve with a score of expert test assessment obtained 80,39% with the category is good as an educative reading source.
