Abstract

Tramadol HCl is a synthetic opioid of cyclohexanol group which acts as an analgesic. To improve the comfort and compliance of patient, therefore tramadol HCl is made in sustained release tablet. In this study, sustained release tablet of tramadol HCl was combined with PVP and xanthan gum mucoadhesive matrix. This study was made in four formulas, they were: control formula (without the matrix), F I (25% PVP : 75% xanthan gum), F II (50% PVP : 50% xanthan gum), and F III (75% PVP : 25% xanthan gum). The tablets were made by dry granulation and compressed to 250 mg weight of tablet. The obtained granules and tablets were tested for the physical properties. The properties determination of the optimum formula used Simplex Lattice Design method (SLD) with Software Design Expert 8.0.6. The parameters used were: flowing time, hardness, friability, and disolution. The experimental results and the theoretical optimum formula were analyzed using t-test. The optimum formula sustained release tablet of tramadol HCl with PVP and xanthan gum mucoadhesive matrix combination by Simplex Lattice Design was obtained in proportion of 55,031% PVP and 44,969% xanthan gum. Pattern of tramadol HCl release on the optimum formula followed diffusion mechanism and zerro orde kinetic. The response of physical properties of optimum formula was obtained from prediction result and examination showed that there was not different significant.

Abstract

Obesity is excess weight due to the accumulation of fat cells. The adverse effects of obesity are high chances for the spread of degenerative diseases and in terms of aesthetic body look unattractive. This research was aimed to use bilimbi leaves as weight loss with varying doses in two dosage forms, namely infusion and tea bag. The test animals used were female white rats were grouped into: negative control group were given 0.5% CMC; positive control group were given xenical® a dose of 2.16 mg / 200 g BB rats; and a test group with a dose preparations bilimbi 5.0 mg/200 g BW, 10 mg/200 g BW, and 20 mg/200 g BW. The treatment were given for 30 days and observed weight. At the end of the treatment, animals dissected after anesthetized. Fats contained around abdominal weighed and the results recorded and compared. Results this study were the bilimbi leaves that has been made in the form of infusions and steeping tea bags in which both had effect as a weight loss in female rats wistar strain.

Abstract

Chalcones has been known to have many biological activities, for example as an antitumour. These research synthesized and observed the cytotoxicity effects against HeLa cells of the 3,4- dichlorochalcone analog compound. These compounds were synthesized by modifying in the aromatic ring which attached at β position to the keton carbonyl. The aromatic ring was furanylaromatic. Synthesis of 3,4-dichlorochalcone analog compound was carried out by Vogel method using starting materials of furanylbenzaldehide and 3,4-dichloroacetophenone. These purity test of synthesized compound was analyzed using thin layer chromatography, melting range, and GC methods. Structure elucidation was determined by using physical methods, such as UV spectrophotometer, IR spectrophotometer, H1-NMR spectrometer, and MS spectrometer analysis. The compound was then observed the cytotoxicity effects against HeLa cells by MTT method. The result indicated that the 3,4-dichlorochalcone analog compound had been able to be synthesized, purified, and it had structure as a predicted compound, furanyl-3’,4’-dichlorochalchone. The cytotoxic results shown that the 3,4-dichlorochalcone analog compound with the IC50 value of 23,5 μg/ml.

Abstract

Research has been conducted in year 1st with the manufacture capsule of noni fruit (Morinda citrifolia L.) and papaya leaf (Carica papaya L.) as the active material to cope with damage to the liver (hepatoprotective) for the treatment of tuberculosis (TB). Year 2nd pre-clinical tests have been conducted using rats as test animals given INH and rifampicin that are often used in the treatment of tuberculosis for 28 days. Test animals consisted of 30 rats were grouped into six groups. Group I (normal control) were without any treatment, rat were given only the usual food and drink. Group II (negative control) were given INH-rifampicin suspension (INH dose of 10 mg/200 g BW and rifampicin dose of 10 mg/200 g BW. Group III (positive control) were given INH-rifampicin suspension and drug antihepatotoxic methicol®. Group IV were given formula capsule 1, group V were given formula capsule 2, and group VI were given formula capsule 3. Parameters of liver damage was observed by measuring the levels of serum bilirubin, ALT and AST values. The parameters were measured on day 0 (before treatment), day 14, day 21, and day 28. There were observed liver histologically by histological methods-microscopic photograph using hematoxylin eosin staining. The data obtained were statistically evaluated using ANOVA one way for percentage of liver necrosis and two-way ANOVA for ALT, AST and serum bilirubin, with a 95% confidence level. The results showed that the formula capsule 1 of extract combination of papaya leaf and noni fruit with PVP binder 1 % was able to provide reduced liver damage on rats induced isoniazidrifampicin which was better than formula capsule 2 and 3.

Abstract

The use of natural materials as active materials in the treatment of benefits in addition to minimizing the adverse effects of natural materials also tend to be cheaper and easier to obtain. The aloe vera plant and snail slime proved to be used in the treatment of burn. Aloe vera contains potent compounds such as fats, carbohydrates, proteins, amino acids, minerals and enzymes Section meat has been widely used for the topical treatment of wounds, burns and skin irritation. Aloe vera can stimulate wound healing because it can increase the formation of collagen in skin tissue The use of snail slime and aloe vera directly considered less practical, so that made gel preparation. Selection of the appropriate base in a formulation will determine the ease or not the drug / active substance effect. Snail mucus gel formulation and a combination of aloe vera are made with a variety of combinations of active ingredients used in the total amount of active ingredient as much as 4%. The resulting gel in physical properties include organoleptic test, viscosity, dispersive power, shifting the viscosity and the antibacterial activity with the diffusion method. The test results were analyzed statistics one-way ANOVA test and Tukey's t-test with a level of 95%. The results showed that all three formulas give viscosity, dispersive power and adhesion are not significantly different. Results of tests performed for 4 weeks showed three formulas provide a stable gel preparation.

Abstract

Apple is suspected has an antioxidant. The use of apple fruit directly considered less practical, so that it made the gel preparations. This research was aimed to optimation by Simplex Lattice Design method to get the most optimum formula gel of apple fruit as antioxidant gel. The apple fruit was macerated. The apple fruit gel use carbopol 940 (C) and glycerin (G) with three formula, F1 (100%C), F2 (50%C: 50%G), dan F3 (100%G). Optimation gel was done based on viscosity, adhesion, and dispersive power by Simplex Lattice Design method with design- expert 8.0.6.1 program. The optimum gel was analyzed the antioxidant activity by DPPH method by probit analysis for calculate IC50 value and vitamin C was used as control. The study shown that optimum combination of carbopol 940 and gliserin as basis was carbopol 2.448 g and glycerin 2.552 g. The optimum gel of apple fruit had antioxidant activity (IC50 151,43 ppm). Storage time affected the viscosity of the gel which tends to decrease during the months of storage.

Abstract

Diabetes mellitus is a disease or metabolic disorder characterized with high blood glucose level accompanied with carbohydrate, lipid and protein metabolism disorder as a result of insufficient insulin function. In this study, ceplikan (Ruellia tuberossa L.) leaves contained flavonoid, saponin, and tannin with hypoglycemic activity. This research aimed to find out the hypoglycemic effect of ceplikan leaf extract and glibencamide combination, the effective dose of combination, and to find out the comparison of hypoglicemic effect between single extract and combination use. The extract of ceplikan leaves was obtained using maceration method with ethanol 70% solvent. The diabetic condition of animal was obtained using monohidrate alloxane induction 100 mg/kg BW. The treatment groups were divided into 6: negative control group with CMC, positive control with glibenclamide (0.45mg/kg BW), single dose of ceplikan leaves extract (937.5 mg/kg BW), combination of ceplikan leaves extract and glibenclamide (25%:75%; 50%:50%; 75%:25%). The blood glucose level was measured using glucometer on the day-7, and day-14. The result of research showed that the combination of ceplikan leaves extract and glibenclamide had hypoglycemic activity with the effective doses of 50%:50% and 25%:75%). The combination of ceplikan leaves extract and glibenclamide had hypoglycemic effect lower than the single extract did.

Abstract

Putri malu (Mimosa pudica L.) and pacar kuku (Laswonia inermis L.) leaves can be used as diuretic agent. This research aimed to find out the diuretic effect of combination of putri malu and pacar kuku leaves ethanol extract as the effective dose on Wistar male rat. The tested animal used consisted of 35 rats, divided into 7 groups: furosemida positive control (1,44 mg/200 g BW), negative control CMC 0.5%, extract of putri malu leaves (20 mg/200 g BW), extract of pacar kuku leaves (50 mg/200 g BW), combination of putri malu and pacar kuku leaves ethanol extract (10 : 25 mg/200 g BW; 5 : 37,5 mg/200 g BW; 15 : 12,5 mg/200 g BW). Observation was conducted every hour for 6 hours, and then at 12 and 24 hours. The diuretic effect was obtained by estimating cumulative urine volume and AUC (Area Under The Curve). The data was processed using oneway variance analysis followed by Tukey HSD test. The result of research the combination of putri malu and pacar kuku leaves ethanol extract has a diuretic effect. The extract combination had diuretic effect higher than the single extract. The combination of putri malu and pacar kuku leaves ethanol extract dose of 5 mg/200 g BW : 37,5 mg/200 g BW and 15 mg/200 g BW : 12,5 mg/200 g BB had diuretic activity compatible to the positive control.

Abstract

Typhoid fever is one of healthy problem in Indonesia and other developing countries. Complications and mortality of thyphoid fever decrease with rapid diagnose and appropriate antibiotic treatment. Highly frequency of antibiotic utilization without provisions may result in negative impacts. One quantitative study to analyze drugs utilization is by using the ATC/DDD method. The aim of this study was to determine the quantity of antibiotic utilization in hospitalized patients with typhoid fever in Sukoharjo General Hospital 2013 with ATC/DDD method and compatibility analyze with hospital formularies and hospital medical care standard. This study used a retrospective analysis. The samples used were medical records of hospitalized patients with typhoid fever (≥ 15 years of age) in Sukoharjo General Hospital 2013. Based on the results of this study concluded that the quantity of antibiotic utilization in hospitalized patients with typhoid fever in Sukoharjo General Hospital 2013 calculated based on ATC/DDD method is cefotaxime (36,03%), ampicillin (22,81%), amoxicillin (12,73%), ceftriaxone (10,01%), cefazolin (9,79%),levofloxacin (4,16%), ciprofloxacin (3,95%), and chloramphenicol (0,52%). Compatibility percentage with hospital formularies 100%; hospital medical care standard 10%.

Abstract

Diabetes mellitus is a metabolic disorder characterized by hyperglycemia and related with abnormality metabolism of carbohydrate, lipid, and protein cause a decrease of insulin secretion or insulin sensitivity decrease or both. Yakon leaves is traditional medicinal plants that one of benefits could treat diabetes mellitus. The experiment was purpose to find antihyperglycemic activity of combination of yakon leaves ethanolic extract and metformin in mice induced alloxan. Test animals were divided in 6 groups each group consist of 5 mice. Groups I was given CMC 0,5%, group II was given metformin (65 mg/kg BB), group III was given yakon leaves ethanolic extract (840 mg/kg BB), group IV was given combination of yakon leaves ethanolic extract (420 mg/kg BB) and metformin (32,5 mg/ kg BB), group V was given combination of yakon leaves ethanolic extract (630 mg/kg BB) and metformin (16,25 mg/ kg BB). The treatment were given during 14 days, with measurement of glucose levels at 7 days and 14 days, then it was statistically tested by one way analysis of variance at 95% confidence continued with Tukey test. The result showed that the combination of yakon leaves ethanolic extract (630 mg/kg BB) and metformin (16,25 mg/ kg BB) was better than metformin or yakon leaves ethanolic extract.
