Abstract

Herba sambiloto (Andrographis paniculata Burm. Nees) mengandung banyak komponen senyawa kimia yang telah banyak di teliti dengan aktivitas sebagai hepatoprotektor dan salah satu kandungan zat aktifnya yaitu andrographolid. Penelitian ini bertujuan untuk mengetahui aktivitas dari A. paniculata yang mempunyai aktivitas sebagai imunomodulator dan hepatoprotektor terhadap mencit Balb/c yang telah diinduksi vaksin hepatitis B. Ekstraksi herba sambiloto menggunakan alat Soxhlet dengan pelarut yang berseri. Vaksin hepatitis B digunakan sebagai penginduksi mencit Balb/c. Sebagai tolok ukur imunostimulator adalah peningkatan jumlah antibodi IgG dengan ELISA tidak langsung. Hasil penelitian menunjukkan bahwa ekstrak n-heksan (2,7 mg/20 g BB mencit) menunjukkan peningkatan IgG tertinggi. Tiga fraksi teraktif dengan peningkatan jumlah IgG tertinggi sampai terendah yaitu F2 (0,569 mg/20 g BB mencit), F1(0,126 mg/20 g BB mencit), F4(0,094 mg/20 g BB mencit). Kata Kunci : Herba sambiloto (Andrographis paniculata Burm. Nees), Imunomodulator, Hepatoprotektor, Vaksin Hepatitis B Abstract Andrographis herbs (Andrographis paniculata Burm. Nees) has many active compounds , which shown hepatoprotector activity and one of its component is andrograpolide. Therefore, the aim of this study were to search the active compounds from A. paniculata as imunomodulator from the BALB/c mice was induced by hepatitis B vaccine. Andrographis herbs was extracted using Soxhlet by series of solvents. The isolation process was guided with in vivo imunommodulatory assay induced by hepatitis B vaccine in Balb/c mice. The imunostimulant activity was assesed through the increasing of IgG antibody. Hexane extract (2.7 mg /20 g mice) was increasing the titer of IgG higher than others. The fractions activity on increasing IgG titer from highest to lowest were F2 (0.569 mg/20 g mice), F1(0.126 mg/20 g mice), F4(0.094 mg/20 g mice). Abstract

Tanaman mundu merupakan salah satu genus garcinia yang potensial sebagai antimalaria. Berdasarkan penelitian sebelumnya fraksi diclormetan-etilasetat kulit batang mundu mempunyai aktivitas antiplasmodium dengan kandungan metabolitnya flavonoid, xanton dan fenol (Rahman et al. 2012). Penelitian ini bertujuan untuk menetapkan kadar fenolik total pada ekstrak dan fraksi kulit batang mundu (Garcnia dulcis. Kurz) Kulit batang mundu di ekstraksi dengan metode maserasi bertingkat dengan pelarut n-heksan dan etil-asetat dan dilakukan fraksinasi menggunakan seperangkat alat kromatografi kolom vakum untuk ekstrak etil asetat kulit batang mundu. Fraksi diclormetan – etil asetat terdiri dari eluen penyusun n-heksana-diklorometana-etil asetat Kadar fenolik total ditetapkan menggunakan metode Spektrofotometri visibel dengan pereaksi Folin Ciocalteau. Prinsip dari metode ini adalah terbentuknya senyawa kompleks berwarna biru dari fosfomolibdat-fosfotungstat yang direduksi senyawa fenolik dalam suasana basa yang dapat diukursecara spektrofotometri. Sebagai pembanding digunakan asam galat. Dari hasil penelitian, diperoleh kadar fenol padaekstrak etil-asetatkulit batang mundu adalah 1,4998 mgGAE/100mg, fraksi diklorometana-etil asetat 0,5545mgGAE/100mg. Dapat disimpulkan bahwa ekstrak etil-asetat memiliki kadar fenol total yang lebih tinggi dibandingkandengan fraksi dichloromethan-etil asetat kulit batang mundu.

Abstract

Abstrak Pengobatan malaria di Indonesia saat ini di beberapa daerah masih menggunakan obat standar kloroquin dengan dosis 600 mg hari pertama dan kedua serta 300 mg hari ketiga. Efektivitas pemberian kloroquin saat ini sudah mulai diragukan karena telah banyak ditemukan resistensi terhadap obat ini. Salah satu usaha menemukan antimalaria baru adalah melalui penelitian terhadap tanaman obat yang digunakansecara tradisional oleh masyarakat untuk mengobati malaria. Tanaman mundu termasuk familia garcinia, Marga Garnicia diketahui kaya kandungan senyawa xanton dan menunjukkan aktivitas biologi termasuk antimalaria (Merza, et al.,).Penelitian ini bertujuan untuk mengetahui aktivitas antiplasmodium ekstrak etil asetat kulit batang mundu. Ekstraksi diperoleh secara maserasi bertingkat dengan pelarut n heksan dan etil asetat. Uji aktivitas antiplasmodium dilakukan secara in vivo pada mencit jantan galur Swiss yang diinduksi Plasmodium berghei dengan parameter pengujian persentase parasitemia, persentase penghambatan parasitemia dan penentuan dosis efektif 50% (ED50) menggunakan metode The 4-daySuppresive Test (Peters, 1970). Dosis yang diberikan ke hewan uji untuk ekstrak etil asetat kulit batang mundu yaitu 12,5; 25; 50; 100 mg/gr BB mencit Swiss. Dosis tablet klorokuin 5 mg/kg bb didapatkan 0.91 mg/ 20g bb dan sebagai kontrol normalCMC 0.5%. Pengujian aktivitas antiplasmodium ekstrak etil asetat kulit batang mundu diperoleh dosis ED50 adalah 5,01 mg/kg BB. Berdasarkan data tersebut, ekstrak etil asetat kulit batang mundu memiliki aktivitas antiplasmodium. Keywords: Antiplasmodium, ED50, Kulit batang mundu, etil asetat

Abstract

Pain is only a symptom and a mechanism in the body that serves as a danger signal about a disturbance in the tissue such as inflammation, infection, or muscle spasms. This study aims to determine the effect of the analgesic effect of the ethanol extract of the karamunting stem. Karamunting stems were extracted by the maceration method using 96% ethanol as solvent. Each analgesic test was divided into five groups, namely a negative control group (CMC Na), a positive control group (Tramadol), a karamunting stem extract test group (a dose of 100 mg/kg BW, 200 mg/Kg BW, and 400 mg/Kg BB). Analgesic test using Tail flick method. Data analysis used the Shapiro-Wilk test and then continued with the one-way ANOVA test. The results showed that the ethanol extract of the karamunting stem could provide an analgesic effect at a dose of 200 mg/Kg BW.

Abstract

Di Indonesia kanker serviks merupakan salah satu kanker penyebab kematian wanita setelah kanker payudara. Herba kemangi (Ocimum basilicum L) banyak digunakan sebagai alternatif pengobatan kanker. Tujuan: mengetahui efek sitotoksik dari herba kemangi terhadap sel HeLa dan sel Vero. Metode: ekstraksi herba kemangi dilakukan dengan maserasi kemudian difraksinasi menggunakan pelarut n-heksan, etil asetat dan air. Ekstrak dan fraksi diuji efek sitotoksiknya menggunakan metode MTT dengan seri konsentrasi 500; 250; 125; 62,5; 31,25; 15,625 µg/mL. Cisplastin digunakan sebagai kontrol positif dan kontrol sel sebagai kontrol negatif. Hasil: aktivitas sitotoksik ekstrak, fraksi n-heksan, etil asetat dan air herba kemangi terhadap sel HeLa menunjukkan nilai IC50 berturut-turut yaitu (156,27; 120,55; 170,17; 186,62) µg/ml dengan indeks selektivitas lebih dari 3 untuk ekstrak dan ketiga fraksi. Kesimpulan: ekstrak dan fraksi herba kemangi memiliki efek sebagai agen kemopreventif terhadap sel kanker serviks.

Abstract

In-vivo antimalarial activity of ethyl acetate extract of Garcinia dulcis Kurz stem  bark  have  been  evaluated  against  Plasmodium  berghei  induced  mice. Antimalarial  test  was  conducted  by  paracitemia  investigation  and  leucocyte counting  of  paracite  induced  mice  blood,  after  oral  administration  of  Garcinia dulcis  stem  bark  extract  dose  of  25  mg,  50  mg,  as  well  as  75  mg/kg  bw.  The highest  antiplasmodial  activity  and  decresing  of  leucocyte  amount  was  showedby  extract  of  dose  of  50  mg/kg  bw.  The  compounds  identified  in  ethyl  acetate extract  of  Garcinia  dulcis  stem  bark  were  flavonoid,  saponin  and  tannin.  The compounds that have antimalarial activity weren’t yet known.Keywords: antimalarial, Garcinia dulcis Kurz, stem bark, paracitemia

Abstract

Mundu (Garcinia dulcis Kurz.) is one of the potential with the chemical compound xanton and triterpenoid for antiplasmodial. The purpose of this study was to determine the activity of n-hexane extract of stem bark Mundu (Garcinia dulcis Kurz.) as the antiplasmodial in mice induced by Plasmodium berghei through a reduction in parasitemia and to find out the most effective doses that have antiplasmodial activities. The animal test mice before treatment with extract induced by Plasmodium berghei to the number of erythrocytic in the blood of 107 and antelmintika were given pirantel pamoat. The concentrate extract was tested in each group of animals test with a dose of 50 mg/kgBW mice, 100 mg / kgBW mice and 150 mg/kgBW mice, negative control group were given 0,5 % CMC and the positive control were given chloroquine be suspention in CMC solution. Antiplasmodial effect was observed by calculating the decrease of the amount of paracitemia. The results showed that the n-hexane extract of Mundu’s stem bark dose 50 mg / kgBW, 100 mg/kgBW and 150 mg/kgBW provide as antiplasmodial activity. Dose of 50 mg/kgBW provide antiplasmodial activity more effective.

Abstract

Diabetes mellitus is a disease or metabolic disorder characterized with high blood glucose level accompanied with carbohydrate, lipid and protein metabolism disorder as a result of insufficient insulin function. In this study, ceplikan (Ruellia tuberossa L.) leaves contained flavonoid, saponin, and tannin with hypoglycemic activity. This research aimed to find out the hypoglycemic effect of ceplikan leaf extract and glibencamide combination, the effective dose of combination, and to find out the comparison of hypoglicemic effect between single extract and combination use. The extract of ceplikan leaves was obtained using maceration method with ethanol 70% solvent. The diabetic condition of animal was obtained using monohidrate alloxane induction 100 mg/kg BW. The treatment groups were divided into 6: negative control group with CMC, positive control with glibenclamide (0.45mg/kg BW), single dose of ceplikan leaves extract (937.5 mg/kg BW), combination of ceplikan leaves extract and glibenclamide (25%:75%; 50%:50%; 75%:25%). The blood glucose level was measured using glucometer on the day-7, and day-14. The result of research showed that the combination of ceplikan leaves extract and glibenclamide had hypoglycemic activity with the effective doses of 50%:50% and 25%:75%). The combination of ceplikan leaves extract and glibenclamide had hypoglycemic effect lower than the single extract did.

Abstract

Hypertension is a condition in which an individual develops above normal blood pressure, when systolic blood pressure exceeds 140 mmHg and diastolic exceeds 90 mmHg. Hypertension becomes health problem in the world. Dewandaru leaves contained flavonoid, saponin and tannin. This research aimed to find out the antihypertensive activity of dewandaru leaves ethanol extract and to find out the most optimum dose of dewandaru ethanol extract (Eugenia uniflora L.). Dewandaru leaves ethanol extract was obtained through extraction using maceration method and using ethanol 96% solvent. This study employed 25 male white rats divided into 5 groups: groups I (control with kontrol negatif, II (control with captopril), III (ethanol extract at 0.3 mg/200 gBW dose), IV (ethanol extract at 0.6 mg/200 g BW dose), V (ethanol extracts at 1.2 mg/200 g BW dose). The tested animal was made hypertensive with adrenaline induction intraperitoneally. The blood pressure measurement was conducted on the week-3 using blood pressure analyzed. The analysis used in this research was ANOVA with Dunnet T3 advanced test. From the result of research, it could be found that all doses of dewandaru leaves ethanol extract had antihypertensive activity. Dose 1.2 mg/200 g BW had effect equivalent to captopril’s.

Abstract

Litsea cubeba Pers or Krangean is a traditional medicinal plant widely used as an antidiarrheal, skin diseases, as well as antiseptic agent. In addition to essential oils, this herb also contains various components such as tannins, saponins, flavonoids and alkaloids. The purpose of the study was to determine the activities of n-hexane fraction, ethyl acetate, and water from the ethanolic extract of the Krangean stem bark as antibacterial and antifungal properties against E. coli ATCC 25922, S. aureus ATCC 25923 and C. albicans ATCC 1023. Krangean stem bark extraction was conducted by maceration method using 70% ethanol as solvent, followed by fractionation with n-hexane, ethyl acetate, and water as solvent. Antibacterial and antifungal activities were determined by diffusion and dilution method. Diffusion method was used to measure the diameter of the inhibitory area against bacteria colonies with concentrations used were 50%, 25%, as well as 12.5%. Dilution method (dilution tubes), was conducted in serial dilutions of various concentrations, 50%, 25%; 12.5%, 6.25%; 3.12%; 1.56%; 0.78%; 0.39%; and 0.195%. The results showed that n-hexane fraction of the ethanolic extract of the Krangean’ stem bark had the highest activity against E. coli ATCC 25922 and C. albicans ATCC 10231 compared to other fractions, based on the average of diameter inhibition values. The diffusion method showed that nhexane fraction at concentration of 50% had inhibitory activity against E. coli ATCC 25922, and C. albicans ATCC 10231 with the average inhibitory power were 30 ± 0,00 mm and 20,4 ± 0,58 mm, respectively. n-Hexane fraction could kill E. coli ATCC 25922, and C. albicans ATCC 10231 with the Minimum Killing Inhibitory Concentration (MKC) value were 1.56%, and 0.39%, respectively.The presence of essential oils and triterpenoid in n-hexane fraction were detected in the preliminary phytochemical tests.
