Abstract

Aspergillus flavus LS03 is a type of fungus isolated from rhizosphere soil in West Lombok, which showed potential as an α-glucosidase inhibitor and free radical scavenger in the early stages of screening activity. This study aimed to identify and characterize secondary metabolite as antidiabetic and antioxidants from A. flavus LS03 fungus grown on liquid medium of potato broth (PDB). The testing of antidiabetic activity is based on the inhibition of the α-glucosidase enzyme while the antioxidant is based on the ability to reduce DPPH free radicals. The result of separation and purification was obtained by active isolate (1) which was identified as 5-hydroxy-2- (hydroxymethyl) -1.4-piran (kojic acid) based on spectroscopic data. Kojic acid (1) shows strong potential as an α-glucosidase enzyme inhibitor and free radical DPPH with IC50 values of 35.34 and 149.9 μg / mL, respectively.

Abstract

Red onion tunic waste contains quercetin which has anti-inflammatory activity. The formulation of anti-inflammatory preparations with the transdermal route is a solution towards the effect of gastrointestinal in the oral route. Transfersome is a nano vesicle particle that can increase the penetration of transdermal formulation. This study aims to characterize red onion tunic waste transfersome based on the ratio of phospholipids and surfactants used. The formulation of the best transfersome will be made into gel based formulation and the particle will be evaluated. Transfersome is made in 3 formulas namely Formula 1 (F1) with a ratio of phospolipids:surfactant (90:10), F2 (85:15), and F3 (70:30). This characterization included particle size distribution, potential zeta, entraptment efficiency, deformability index, and vesicle morphology. The best transfersome formulas are then formulated into gel preparations. The results showed that the best transfersome formula was F1 (90:10), with an average particle size of 357.9 ± 6.3 nm, zeta potential of -24.9 ± 0.9 and the entraptment efficiency results reached 77.964%. The morphology of the vesicles produced is spherical according to the expected vesicle criteria, but in gel-based formulation, the particle size not distributed homogenously.

Abstract

The high prevalence and risk of death from cardiovascular disease has led to the development of alternative treatments derived from nature. Green coffee has not undergone a roasting process. The product is claimed as a weight loser, even though the cardiovascular effects are still controversial. This study aim was to determine the impact of green coffee bean extract on rat lipid profiles and aortic histopathology. Green coffee bean extract made by a re-maceration method using 96% ethanol. Rats were induced with high-fat diet then treated with green coffee bean extract at a dose of 200; 400; and 800 mg/kg BB. Rats were sacrificed, blood was taken for lipid profile measurements and observations were made on histopathological preparations of rat aorta. The results showed that administration of green coffee bean extract (especially doses of 200 mg/kg BW) for 14 days in hyperlipidemic mice could significantly improve lipid profiles, except HDL levels. Microscopic observations showed that green coffee bean extract could enhance the rat aorta histopathology even though it did not match normal conditions. We can conclude that green coffee bean extract has potential effects to be developed as an alternative to traditional medicine that has antihyperlipidemic effects and cardiovascular protective effects.
