Abstract

Land insects function in the soil fertility process, assist the soil decomposition, and to accelerate soil material cycle so as there are equilibrum on food chains  and food webs cycle. The study aimed to examine of land insects in the South Sulawesi Regional Polda plantation and their surrounding environment benefits. The sampling technique was purposive sampling with an exploratory method, namely direct observation or observation in the field using the Pitffal Trap method at 5 stations. The study results at the South Sulawesi Regional Polda plantation obtained 172 individuals of soil insects consisting of 14 genera in 13 families. The land insects of Carabidae and Gryllidae families are insect genus with high abundance, while the families of Pentatomidae, Alydidae, Syrphidae, Saturniidae, Stratiomyidae, Blattidae, and Gryllotalpidae families has low abundances. The causing land insects genera high abundance at several research stations are the presence of supportive environmental conditions, where the habitats of some of these land insects are still preserved or natural, abundant food sources, and locations far from residential areas. The land insects found act as pollinators, decomposers, predators, parasitoids and bioindicators.

Abstract

Mangrove areas are widely reported as a place of abundance of microalgae producing omega-3 docosahexaenoic acid (DHA) and other bioactive materials of high economic value from the genus Aurantiochytrium. The search for microalgae was carried out in the mangrove area of Pari Island, Seribu Islands, Jakarta, in April 2022 and identified at the Biology Education Laboratory, FKIP UHAMKA. The purpose of this study is part of the long-term goal of collecting new strains or isolates from the genus Aurantiochytrium and other members of the family Thraustochytriidae from forests in Indonesian mangroves. This research was conducted by isolating microalgae with the pure streaking method and identifying them morphologically. This study succeeded in obtaining eight isolates (UHA1, UHA3, UHA5, UHA6, UHA10, UHA11, UHA12, and UHA13) which in the appearance of colonies and microscopic morphology lead to the species Aurantiochytrium sp. One of the isolates obtained (UHA12) was cultivated by batch culture for five days in an Erlenmeyer flask using glucose, yeasts extract, and seawater as a medium. The result is that the dry biomass obtained is almost equivalent to that of the Aurantiochytrium sp. other famous

Abstract

Indonesia is ranked 7th out of 10 countries with the highest number of people with diabetes mellitus, which is estimated to increase every year.α -amylase is an enzyme that catalyzes the hydrolysis of α-1,4 glycosidic bonds of polysaccharides to produce dextrins, oligosaccharides, maltose and D-glucose. Apis dorsata Binghami honeycomb contains secondary metabolites that have antidiabetic potential, including inhibiting the action of the α-amylase enzyme. The purpose of this study was to determine the in vitro antidiabetic potential of Apis dorsata binghami nest extract. This study uses a descriptive research method where the research data are obtained through laboratory experiments. Honeycomb extraction using maceration method. Analysis of total flavonoid content using UV-Vis spectrophotometric method. Antidiabetic potency test using -amylase enzyme inhibition method. The results of the extraction study showed that the % yield of the extract was 11.82% with a brownish yellow color. Analysis of the total flavonoid content showed the results of 3.33 mgQE/g. The inhibitory activity of the -amylase enzyme, the IC50 value of the extract obtained was 158.48±7.42 g/mL, the acarbose value was 165.96±7.08 g/mL. These results indicate that the extract of A.dorsata Binghami nest has hyperglycemic activity by inhibiting complex carbohydrate hydrolyzing enzymes such as inhibition of the -amylase enzyme which is better than acarbose because the IC50 value of the extract is smaller. If the IC50 value is smaller then the enzyme inhibition is stronger. For further research, it is recommended to use a more specific enzyme, namely the -glucosidase enzyme.
