Abstract

Pummelo (Citrus maxima (Burm.) Merr.) (Rutaceae) is a cultivated species with a high economic value and several cultivars are found in Indonesia. Bageng Taji is a seedless pummelo cultivar found in Bageng village, Pati regency, Central Java, Indonesia. This research aims to measure the effect of colony addition of two bee species, Apis cerana and Tetragonula laeviceps in pollination and fruit set of pummelo. We set-up four treatments of pollination, i.e., bagged flowers (control), open flowers , colony addition of A. cerana (honey bee), and colony addition of T. laeviceps (stingless bee). Visiting activity of bees were observed by using focal sampling method in the sunny days. Pollen load was measured using the acetolysis method. The effectiveness pummelo pollination was measured based on fruit formations. Results showed that the longest visitation and the number of flowers visited of A. cerana (15.2 seconds/flower and 37.3 flowers/5 minutes) and T. laeviceps (45.5 seconds/flower and 17.1 flowers/5 minutes) occurred at 09.30 am. The average number of pollens carried by A. cerana (19,908 pollen grains) was higher than T. laeviceps (4,308 pollen grains). Addition of A. cerana and T. laeviceps colonies increased 34% and 32% of fruit formation compared to control plants.

Abstract

The distribution of Culex quinquefasciatus as a vector for several tropical diseases in Indonesia could affect the transmission of various pathogens, including filariasis, West Nile, encephalitis, and Rift Valley fever. The use of insecticides to control mosquito populations is carried out massively. However, this effort is threatened by an increase in mosquito resistance to insecticides, in this case, pyrethroids that target Voltage-gated sodium channel (VGSC) as a complex protein in mosquito nerve cells that plays a role in the movement of sodium ions, where these compounds can have a knockdown effect and lead to death. The existence of a single nucleotide mutation in this specific codon results in knockdown resistance (kdr) in mosquitoes. This study aims to determine whether there is a kdr mutation in the Vgsc-L1014F target using the latest molecular method ETAS-PCR to type tri-allelic variation at Vgsc-1014 in Cx. quinquefasciatus samples collected in three areas in the cities of Surabaya and Sidoarjo, namely Wonocolo, Sukodono, and Wonoayu. The result showed ten male pool samples (C1-C10) of Cx. quinquefasciatus did not carry mutations or present as homozygous wild type (TTT/TTT), with a DNA band size of 181 bp. Hence, this preliminary study could be extended for further research to build vector control monitoring programs.

Abstract

Anti-thyroid peroxidase is an autoantibody generated by the immune system, its primary function is target and neutralize the enzyme thyroid peroxidase, is essential for production of thyroid hormones. Inhibin-B is a peptide belonging to TGF-β family produced by granulosa cells of the growing follicle cohort. It reflects the health of follicles, their overall numbers. The objective of this article is evaluation of Anti-TPO, Inhibin-B for female infertility. A case-control study includes 120females between 20-45 years divided into 60 apparently healthy females used as controls and 60 infertile females distributed 37 with Polycystic ovarian syndrome, 23 with Premature ovarian failure. Serum levels of Anti-TPO, Inhibin-B, LH, FSH, Testosterone, Prolactin and Estrogen were all measured using an ELISA assay. Inhibin-B, Anti-TPO higher in patients than in controls. Inhibin-B significantly increases(p≤0.0001) in PCOS above and under 40years (273.55±75.25 pg/mL), (234.91±52.43 pg/mL), but decreases in POF (18.96±1.21 pg/mL) than in controls (35.58±10.48 pg/mL), (42.04±10.04 pg/mL). AntiTPO increases in PCOS and POF above and under 40 years (16.08±2.14 ng/mL), (17.63±1.93 ng/mL), (14.72±1.19 ng/mL) than in controls(5.92±0.99 ng/mL), (5.93±0.84 ng/mL). The levels of FSH, Prolactin, Testosterone, Estrogen and LH were increased (p≤0.0001) in PCOS, POF as in controls. The results showed a negative correlation (p≤0.0001) between Inhibin-B, Anti-TPO (r = -0.520) in PCOS. The Receiver operating characteristic (ROC) analysis demonstrated Inhibin-B 95%CI:0.907-1.000; P-value:0.001; Cutoff Point:77.347; AUC:95.417% with Sensitivity to Specificity 91.667-98.333%; Accuracy: 95.000%. Anti-TPO 95% CI: 0.907-1.000; P-value: 0.001; Cutoff Point: 9.306; AUC: 98.861% with Sensitivity to Specificity 96.676-96.667%; Accuracy: 97.000%. Anti-TPO, Inhibin-B are useful biomarkers for evaluating fertility status and ovarian health, can used as sensitive indicators for early diagnosis of PCOS and POF in women of reproductive age.

Abstract

Exosomes have emerged as a cell-free alternative to mesenchymal stem cells (MSCs). Hypoxic preconditioning might result in the production of exosomes with ideal properties. As such, the effect of hypoxia on hsa-mir-21-5p expression, which is known to contribute to the immunomodulatory activity of exosomes, should be investigated. This research aimed to enrich and characterize exosomes from human umbilical cord MSC (hUC-MSC) secretome samples secreted under hypoxia and normoxia, as well as to analyze the expression of hsa-miR-21-5p in each sample. Secretomes were collected from cultured cells under both hypoxic and normoxic conditions, and exosomes were subsequently enriched from the secretomes through ultrafiltration. In this research, it was found that particles in N-Exo and H-Exo exhibited round morphology, expressed exosomal CD81 and CD63 markers with an average size of 153.7 ± 31.3 nm and 137.7 ± 25.0 nm, respectively. N-Exo and H-Exo can also be internalized by mesenchymal stem cells (MSCs). qRT-PCR results then indicated that hsa-miR-21-5p expression was significantly lower in H-Exo (p < 0.05). In conclusion, hypoxic preconditioning, as performed in this research, was also found to affect hsa-miR-21-5p and downregulate its expression in hypoxic samples relative to controls.

Abstract

Many studies have reported that intermittent hypobaric hypoxia (IHH) can cause oxidative stress and tissue damage in the kidneys, which can lead to impaired kidney function. However, a study using simulated intermittent hypobaric hypoxia was considered to have a protective role for the kidneys. This study aims to determine the effect of IHH on kidney adaptation to oxidative stress and kidney damage. Twenty-five rats were divided into five groups: control, one-time hypoxia (AHH), two-time hypoxia (IHH 1), three-time hypoxia (IHH 2), and four-time hypoxia (IHH 3). The intermittent interval was one week, and at the time of treatment, the rats were placed in a hypoxic chamber. At the end of treatment, the rats were sacrificed, and the kidneys, urine, and blood were collected. The kidney tissues were used for protein assay, ELISA (HIF-1α and VEGF), qPCR (cytoglobin (Cygb) and renin), SOD and GPx activity assay. Blood was used for creatinine and urea assay, and urine was used for KIM-1 ELISA. Formalin-submerged tissues were used for histopathological analysis. The level of HIF-1α and VEGF increased significantly from AHH to IHH 1. Cygb and renin expressions peaked at AHH and decreased at subsequent IHH groups. The Nrf2 level and GPx activity didn’t show any difference, but SOD activity peaked at IHH 1. The creatinine level only peaked at IHH 2; other groups remained the same as the control. Urea levels decreased with more IHH sessions, and KIM-1 didn’t show any difference. Our findings exhibit that hypoxia preconditioning by IHH treatment leads to the kidney’s adaptability to hypoxia and does not cause kidney damage.

Abstract

The effect of electric cigarettes (e-cigs) on health should be a concern since their legality has occurred in Indonesia. Young adults use e-cigarettes not as therapy but as a lifestyle. The vapor component contains propylene glycol, vegetable glycerin, flavor, and nicotine, which may harm human health. This study examined the health risks of e-cigarettes on body weight, erythrocytes, and sperm morphology in mice. We used twenty healthy male mice exposed to the cigarette's vapor for thirty days. Mice were divided into room air as the negative control, nicotine-free, and 18 mg nicotine-containing groups. Our results showed that the body weight of the nicotine-free and nicotine-containing groups increased, while micronuclei formation increased after e-cig exposure. Furthermore, abnormal sperm morphology in the nicotine-containing groups increased significantly, yet the nicotine-free group was not significantly different compared to the negative control. These results were supported by the molecular prediction that exposure to e-cigs might increase oxidative stress and decrease the expression of TGF-β. Moreover, studies on it are ongoing to gather scientific evidence.

Abstract

The nuclear factor erythroid 2-related factor 2 (Nrf2) is a master regulator of most cytoprotective genes that respond to oxidative stresses. Thus, Nrf2 depletion is usually associated with excessive oxidative stress (OS). We sought to assess Nrf2 concentration as an indicator of elevated OS in women with PCOS. Likewise, study the effects associated with this depletion on the balance of gonadotropin-releasing hormone (GnRH). The case-control study comprised ninety women of reproductive age. Our results revealed that women with PCOS had significantly higher blood Nrf2 depletion compared with the non-PCOS group (P = 0.000). So, there was a significant increase in both Luteinizing hormone (LH) concentration and LH\FSH ratio (P = 0.000 and P = 0.007, respectively. In the PCOS group, Nrf2 depletion was adversely correlated with elevated LH level (-0.551) at (P = 0.012), and elevated LH\FSH ratio (-0.628) at (P = 0.003), infertility (-0.460) at (p = 0.041) and menstrual irregularity (-0.575) at (P = 0.008). Meantime, according to the odds ratio analysis, Nrf2 depletion had a high relative risk in the etiology of syndrome 0.967 (0.932–0.945) at (P = 0.048). In conclusion, Nrf2 depletion had a role in the GnRH imbalance in PCOS through several mechanisms. Thus, we can suggest that enhancing the Nrf2 pathway can be a possible therapeutic approach for PCOS treatment.

Abstract

This research investigates the ocean's potential as a source of natural active ingredients with antioxidant, antibacterial, and sun protection properties. Marine gastropods are known to have symbiotic relationships with associated bacteria capable of producing pigments, which hold promise for the cosmetic and health sectors. The study aims to identify and analyze pigments produced by bacteria linked to marine gastropods and evaluate their effectiveness as antioxidants, antibacterials, and sun protectants. The method involved isolating and analyzing pigments from gastropod-associated bacteria, followed by tests to assess their properties. Results revealed two gastropod species: Telescopium telescopium (Linnaeus, 1758) and Cassidula nucleus (Gmelin, 1791), from which 21 bacterial isolates were obtained—10 from T. telescopium and 11 from C. nucleus. Among these, Micrococcus yunnanensis, a bacterium with high pigment production, was successfully isolated. At 1,000 µg/mL, its pigment's crude extract had low antioxidant activity and had a low SPF category as a photoprotective agent. Antibacterial tests showed efficacy against Pseudomonas aeruginosa and Staphylococcus aureus. HPLC and FTIR analysis suggested the pigment contains carotenoid compounds. These findings highlight the potential of M. yunnanensis pigment for sustainable health and cosmetic applications.

Abstract

Mangosteen peel simplicia (Garcinia mangostana L.) contains potential ingredients used in medicine. A toxicity test of mangosteen peel simplicia is needed before it is used. Wader pari (Rasbora lateristriata) is a potential animal model for toxicity testing. Thus, this study aims to determine the effect of mangosteen peel extract at various concentrations on the behavior and bone structure of R. lateristriata. In this research, fish behavior was observed at 48 hpf (hours post-fertilization) and 30 dpf (days post-fertilization). Bone structure observed at 96 hpf, 7 dpf, 14 dpf, 21 dpf, 28 dpf, 35 dpf, and 42 dpf using the Alizarin Red ‒ Alcian Blue method. Observation data on behavior and bone structure were analyzed descriptively, while the number of vertebrae was analyzed quantitatively using One-Way ANOVA (P < 0.05). The results showed that the ossification of cranium and caudal complex skeleton components in the 1 and 5 μg/mL treatments was significantly behind that of the control and 0.5 μg/mL treatments. Furthermore, 5 μg/mL treatment caused some behavioral and swimming pattern abnormalities in 48 hpf larvae. In conclusion, exposure to 1 and 5 μg/mL mangosteen peel simplicia resulted in skeletal growth inhibition in R. lateristriata. Exposure to 5 μg/mL mangosteen peel simplicia caused abnormalities in the behavior and swimming pattern of 48 hpf larvae.

Abstract

Plant-derived exosome-like nanoparticles (PDENs) have emerged as promising natural nanocarriers with potential applications in biomedicine and cosmetics due to their biocompatibility, stability, and intrinsic bioactivity. The objective of this research is to isolate, characterize, and analyze the effects of MZ-PDENs on 1BR3 cells. MZ-PDENs were isolated using filtration, differential centrifugation, and polymer-based precipitation. MZ-PDENs were characterized for their physiochemical properties, size, shape, and antioxidant activity. In vitro, bioactivities were conducted by measured cytotoxicity, proliferation assays, UVB protection, and migration assays. Results showed that MZ-PDENs possess strong antioxidant properties, are efficiently internalized by cells, and promote skin cell regeneration after UVB-induced damage. These findings suggest that MZ-PDENs have the potential as antioxidant agents for cosmeceutical applications and warrant further investigation in vivo models.
