Abstract

 Salah satu yang menyebabkan kegagalan budidaya rumput laut Kappaphycus alvarezii  yaitu terserangnya penyakitice – ice . Penyakit ice-ice merupakan penyakit yang umum terjadi pada rumput laut kappaphycus  dengan gejalaterjadinya pemutihan pada bagian thallus  dan penyakit ini dapat menyebabkan kematian massal pada produksirumput laut kappaphycus alvarezii . Salah satu cara yang dapat dilakukan untuk menganalisa bagian thallus  rumputlaut Kappaphycus alvarezii  yaitu uji histopatologi. Penelitian ini bertujuan untuk mengetahui perubahan bentuksel pada kondisi jaringan thallus  rumput laut K.alvarezii  yang terserang penyakit ice- ice  menggunakan metodehistopatologi. Pengambilan sampel rumput laut yang digunakan jenis Kappaphycus alvarezii  varietas coklat danhijau dengan 1 titik masing – masing varietas. Sampel rumput laut terdiri atas 2 sampel yaitu bagian thallus  yangterserang penyakit ice-ice  dan bagian thallus  tanpa gejala ice-ice . Sampel berumur 15 hari. Sampel yang dipilihuntuk pemeriksaan, dipotong setebal 0,5 - 1 cm. Sampel dimasukkan kedalam botol sampel untuk difiksasi ± 1minggu, yang berisi formalin 10%.Tahapan pembuatan preparat histopatologi terdiri atas fiksaksi, dehidrasi,clearing , impregnasi , embedding , cutting , staining , dan pemeriksaan dibawah mikroskop. Penelitian ini dianalisissecara dekskriptif kualitatif dengan melakukan pengamatan gambar histopatologi pada jaringan rumput laut K.alvarezii . Jaringan thallus  yang yang terserang penyakit ice-ice  mengalami perubahan bentuk sel, bagian lapisanluar terlihat tidak utuh, hancur, sehingga mengalami kematian sel dan bagian korteks dalam dinding sel terlihattidak teratur dan renggang, tidak terbentuk bulat atau lonjong, sebagian sel mengecil. Jaringan thallus  yang tidaknormal mengalami gejala Hipoplastik (terhambatnya atau terhentinya pertumbuhan sel) dan Nekrotik (kematiansel).Kata kunci : Kappaphycus alvarezii, penyakit ice – ice, Histopatologi, Hipoplastik, Nekrotik

Abstract

Sandeq is a kind of marine transportation , since long has been operated by Mandar seaman and fishers alongthe coastal water area, as well as for fishing in Sulawesi Barat . This research was carried out to study thefeasibility of utilizing the Sandeq boat in marine ecotourism in Pasir Putih Island of the Polewali MandarRegency. SWOT Analisys has been applied to reveal the level of support of stakeholders regarding this idea, andthe strategies to be carried out in developing this specific ecotourism. The analysis shows that the internal andexternal factors are strongly support the program with a strong and aggressive manner and having a positifopportunity. A scenario on commercially operated ecotourism was designed and financially analyzed for 5 yearsperiod of operation: (1) Net present value of Rp 705.121.922,-, (2) Internal rate of returnof 100,99 %, and B/Cratio range from 1.7 at 3nd year to 3.7 at the 5th year. Those values showed that the project is financiallyfeasible. Three alternatives routes of marine ecotourism werw iidentified: (1) Ballanipa Village to KaramasangIsland, (2)From Balanipa Village to Battoa Island, (3) From Tonyaman Village to Karamasang Island and PasirPutih.Key words : Sandeq boat, ecotourism, Pasir Putih Island, Polewali mandar Regency, Swot Analysis, Financialanalysis.

Abstract

Ocean acidification afected marine organisms especially calcifying organisms, such as Halimeda sp. This study was conducted on June to September 2012 at laboratory of Research Center and Development for Marine, Coastal and Small Islands, Hasanuddin University. The aim of study  to determine the efect of acidic level on Calcium concentration and growth rate of calcifying macroalgae, Halimeda sp. The experiment design was used completely  random  design  with  three  treatments  and  three  replicates.  Analysis  variance  was  used  for  data analysis with advanced respon test. Tukey test was used to compare the diference between treatments. Water quality parameters were analyzed descriptively. The results showed that there was a significant diference of pH treatments on calcium concentration an d growth rate of Halimeda sp. The highest calcium concentration was found  at pH 8. In  conclusion, increasing pH level of media disturbed the calcifying  process of Halimeda  sp especially at pH level of 6, while growth rate of Halimeda sp was not afected wit h the decreasing of pH.

Abstract

Waters of the southearn of Makassar Strait is a region which relatively rich of organic matter because thephenomenon of upwelling along the eastern season. The purpose of this research was to analyze the distributionpatterns and the development of upwelling areas in the southern of Makassar Strait. This study used chlorophylladata and sea surface temperature from level 1 of Modis image for two years (2009 and 2010). The resultshowed that phenomenon of upwelling that occurs in the southern Makassar Strait appears since early June, thestrongest upwelling in August and disapear in October. The upwelling was indicated by declining of sea surfacetemperature and increasing of chlorophyll-a concentration. Analysis of wind direction and speed indicate thatthe upwelling occurs in the southern Makassar strait spread to southwest with and estimated upwelling areaaround 46.000 km2.Keywords: upwelling, Makassar Strait, chlorophyll-a, sea surface temperature.

Abstract

This study aims to study the performance of phytoecdisteroids from mulberry leaves in molting stimulating of mangrove crabs. The research was carried out at the Research and Development Center for Aquaculture at the Faculty of Marine Sciences and Fisheries at Hasanuddin University, located in Bojo Village, Mallusetasi District, Barru Regency, South Sulawesi Province. Mangrove crabs (Scylla olivacea) weighing 45-55 g and carapace widths 60-65 mm were used as test animals. There were five doses of mulberry leaf extract applied through feed, namely: A) 1.1 mg / g feed, B) 1.9 mg / g feed, C) 2.7 mg / g feed, D) 3.5 mg / g feed. Crabs were kept individually in plastic boxes that are floated above the surface of the pond. During rearing, crabs are fed 3% dry fish per day which has been enriched with mulberry leaf extract. The parameters observed were ecdysteroid compounds contained in mulberry leaf extract, ecdysteroid content in hemolymph before and after application of mulberry leaf extract, and molting percentage. Identification of the ekdisteroid compounds contained in mulberry leaf extract was carried out using Thin Layer Chromatography (TLC) and Measurement of the content of ecdysteroids in crab hemolymh was carried out using Ultra Fast Liquid Chromatography (UFLC).  The results showed that the 3.5 mg dose of mulberry leaf extract / g feed gave the highest increase in ecdysteroid hemolymph, which was approximately 1760 ekdisteroid per mL hemolimph, while the dose of 1.1 mg / g feed only provided an increase of 100 ng ekdisteroid per mL hemolymph. The higher the dose of mulberry leaf extract in the feed, the higher the increase in the concentration of hemolymph ecdysteroids, but the high concentration of ecdystoid in the hemolymph does not guarantee molting.  The optimal dose of mulberry leaf extract in the feed stimulating molting was 2.4 mg / g of feed.Keywords: mulberry, phytoecdysteroid, molting, crab, aquaculture

Abstract

Tidak dapat dipungkiri bahwa usaha budidaya udang intensif merupakan kegiatan ekonomi di kawasan pesisiryang sangat produktif dalam peningkatan kesejahteraan masyarakat, PAD dan devisa negara, namun legitimasikeberlanjutan sangat ditentukan oleh dampak kerusakan lingkungan yang ditimbulkannya. Penelitian inibertujuan untuk : (1) melakukan analisa kelayakan dalam pengembangan budidaya tambak pada lahan marjinal;(2) menetapkan kesesuaian pengembangan teknologi tradisional, semi-intensif, dan intensif pada lahan tambakmarjinal. Pengamatan data primer berupa pengukuran dan pengamatan kesesuaian bioteknik aspek saranaprasarana,dan data sekunder berupa hasil penelitian dan data-data dari beberapa instansi yang terkait.Hasilpenelitian ini menunjukkan bahwa analisis kelayakan bioteknik dalam pengembangan budidaya tambak padalahan marjinal dilokasi penelitian pada musim hujan untuk metode tradisional adalah seluas 79,9 Ha, untukmetode semi-intensif seluas 356,31 Ha, dan untuk metode intensif adalah seluas 365,03 Ha. Sedangkanpengembangan pada musim kemarau untuk metode tradisional adalah seluas 43,12 Ha, untuk pengembanganmetode semi-intensif adalah seluas 282,7 Ha dan untuk pengembangan metode intensif adalah seluas 475,4 Ha. Abstract

The existence of mangrove in coast areas have both ecologic and physical important functions.  Mangrove is a high productive ecosystem. Litters from fallen leaves and twigs are organic sources for macrozoobenthic life.  The aim of this research was to determine the response of macrozoobenthics on total organic matter content and other chemical-physico characters of sediments due to differences in mangrove conditions. This research was conducted from May to September 2016 in Sub-district of Ampallas, Distric of Mamuju, Province of West Sulawesi, Indonesia. Sediments in 20 cm2 transects were collected using small shovel to obtain the macrozoobenthic samples. The sediments contained macrozoobenthics were sieved using a 1 mm mesh size sieve net. Four sampling sites were chosen, each to represent areas with no mangroves, low, medium, and high densities.  The total of organic matter samples were collected using 2 inch diameter of PVC pipe cores, which were analyzed further using Loss by Ignition method.  Besides total of organic matter, several parameters (i.e. redox potential, sediment pH, salinity, temperature, and dissolved oxygen) were measured.  The results showed that gastropods dominated the existed macrozoobenthics.  Gastropods and oligochaete lived in greater amounts in high mangrove density site with higher organic matter content and fine substrate.Keywords: mangroves, macrozoobenthics, organic matter, Distric of Mamuju

Abstract

Zooplankton has an important role in waters, especially in the food chain, these organisms are consumers I which play a major role in bridging energy transfer from major producers (phytoplankton) to living things at higher trophic levels (fish and shrimp). This study aims to analyze the relationship between zooplankton abundance with phytoplankton and physicochemical parameters in the waters of Kastela, Ternate. The research was conducted from March to April 2017 in the waters of Kastela Ternate City North Maluku Province at 5 stations and 4 periods of sampling. Zooplankton samples were taken by filtering methods. The results showed that there were 20 zooplankton genera from 5 (five) classes namely Ciliate (4 genera), Crustaceae (8 genera), Hydrozoa (3 genera), Rotifera (3 genera), and Urocohordata (2 genera). Zooplankton abundance ranges from 1032 to 10942 cells.l-1, the highest at station 3 periods II (10942 cells.l-1), and the lowest at station 4 period I (1032 cells l-1). The range of values of the zooplankton biological indices is diversity index (0.5005 - 1.8662), uniformity index (0.6521 - 0.9601), and dominance index (0.1661 - 0, 6800). There was a low correlation between zooplankton abundance and phytoplankton abundance and physical-chemical parameters of water in Kastela waters, with determination coefficient is 0.236 and regression equation Y = 35079.107 + 0.030 phytoplankton - 791,251 temperature + 75,417 salinity - 1658,557 pH Keywords: mangroves, macrozoobenthics, organic matter

Abstract

Toman fish seeds (Channa micropeltes) generally come from natural catches in South Kalimantan and Central Kalimantan. A closed system is an effort to efficiency transportation costs by increasing the density of fish in transport media. Efforts to reduce stress levels are very important so that they can transport a lot, have small deaths, and take a longer time, and do not disturb the physiology of the fish after transportation. The purpose of this study was to determine the effect of giving 3 ppt salt in transport media to the survival of fish during transportation. This research consists of two stages of activity, that is stage one is transportation for 12 hours and stage two was post-transportation maintenance for 11 days. In stage one, the plastic bag is filled with 10 L of water and is packed according to the treatment. This study used a Completely Randomized Design (CRD) with 4 treatments 3 refitition to produce 12 experimental units. K treatment density of 30 heads / bag with no salt water (0 ppt) and treatment A: B: C with density (30: 50: 70) tail / bag with salt water (3 ppt). The calculation of the highest survival rate in treatment A was 97.80%. The results of analysis of varians (ANOVA) that the treatment has a real effect on survival rate in fish seed transport toman. Abstract

The use of probiotics (Lactobacillus  sp.) in feed is one alternative that is done to improve immunostimulants, growth stimulants, and can be used as a balance of microorganisms in digestion. This study aimed to evaluate the growth performance of vanamei shrimp fed with the addition of Lactobacillus  sp. with sweet potatoes in feed. This study used a completely randomized design (CRD) with 4 treatments and 3 replications. The treatment was by feeding with different sweet potato concentrations: A) The dose of sweet potato in feed was 0% (as control); B) The dose of sweet potato in feed was 10%; C) 1The dose of sweet potato in feed was 15%; and D) The dose of sweet potato in feed was 20%. Data were analyzed using variance analysis (ANOVA) and continued with W-Tuckey's further test. The research parameters were digestive enzyme activity, nutrient digestibility, and growth. The results of the variance analysis showed the addition of various sweet potato concentrations with Lactobacillus sp. has no significant effect (p> 0.05) of the growth rate. The specific growth rate of vannamei shrimp ranges from 3.06 to 3.23% / day. Based on the results of the analysis on the utilization of sweet potatoes as a prebiotic on the performance of Lactobacillus sp. in the vannamei shrimps (Litopenaeus vannamei) digestive tract can be concluded that the growth performance does not have a significant effect after being fed with the addition of Lactobacillus sp. and sweet potatoes in feed.
