Abstract

The influence of various types of sera i.e. Fetal Calf Serum (FCS), Bovine Post Estrous Serum (SPES) as heterolog sera; and Ewes Fetal Serum (EFS) and Ewes Serum (ES) as homologue sera on in vitro maturation and fertilization of sheep oocytes was investigated using Tissue Culture Medium 199TM (TCM 199. Gibco). Sovine sera were collected from cow at seven days post-estrus, while ewes sera were collected during estrous (ES-HO) and six days post estrous (ES-H6). The result shows that the maturation rate of oocytes cultured in medium supplemented with EFS, ES-HO and ES-H6 at  concentrations of 32.9%, 68.7%, and 67.6% respectively is better than those supplemented with FCS and SPES at concentrations of 22.2% and 28.9%, respectively. In vitro fertilization rate were significantly higher in medium supplemented with ES-HO and ES-H6 at concentration of 65.4% and 65.8% (P < 0.05) than that supplemented with SPES at concentration of 31.7%. In conclusion, ewes sera could be used in culture medium for promoting both in vitro maturation and fertilization rate of sheep oocytes.

Abstract

Contamination of aflatoxins in animal feeds is one of a major problem in the development of poultry industry in Indonesia. Aflatoxins may lead to losses in animal productivity, aflatoxicosis and residue in animal products. A series of investigation on aflatoxin contamination in animal feed in poultry had been carried out at Research Institute for Veterinary Science (Balitvet) between 1984 to 1995. It showed that more than 80 % of commercial chicken feeds were contaminated by aflatoxin B1 (AFBI) within a wide range of concentration. Besides AFB1, other aflatoxins such as AFB2, AFG1 and AFG2 were also found. The level of AFB1 more than 200 ppb was found in 13.5 % out of 193 feed samples, whilst 23.3 % and 63.2 % of samples showed concentration 100-200 ppb and 400 ppb respectively. It had approved from the investigation that corn was the most frequent foodstuff contaminated by aflatoxins compared to other animal foodstuff. Most of corn used for chicken feeds appeared to be the major source of aflatoxin contamination, where it could be indicated visually in bad or good kernel appearance. The levels of AFB1 contamination seemed to be higher in the wet season (39.5 ppb) than in the dry season (19.5 ppb) as well as at the lower altitude (39.8 ppb) was higher than at the higher altitude (24.13 ppb). Based on this view, further studies are required to control of the aflatoxins problems in poultry feed in Indonesia.Key words: aflatoxins, animal feeds, poultry

Abstract

Twelve native young goats were used in this study to observe the effects of concentrate supplementation in ration such as sweet potatoes and rice brand on consumption, digestion, and growth development of young goats. The results showed that sweet potatoes and rice brand supplementation, respectively, 300 g and 30 g per head per day in goat's ration increased the consumption of dry matter, protein, fat, N-free extract, digestion of dry matter and N-balance. This ration also improved daily body weight gain and feed conversion.Key words: concentrate, fattening ration, growth, goats

Abstract

The research was planned to yield the biologic potential synergism of MoAb-hCG in increasing the embryo number. Twenty five dairy cows were divided into five groups. The Control Group was superovulated with 2,500 IU PMSG intramuscularly (i.m.), the remaining of the Group II, III, IV and V consecutively after being superovulated by 2,500 IU PMSG were treated with MoAb intravenously (i.v.), hCG i.v., MoAb-hCG i.v. and MoAb i.v-hCG i.m. Data evaluation were analyzed using ANOVA and LSD test. The result showed that there were increasing in number of transferable embryo per donor produced (P<0.01) after application with either MoAb i.v. (9.6), hCG i.v. (9.4), MoAb-hCG i.v. (11.2), or MoAb i.v.-hCG i.m. (9.6). The Control Group produced only 2.4 embryo per donor. The combined MoAb and hCG in one treatment were not able to increase the embryo yield (P>0.05).Key words: MoAb, PMSG, hCG, superovulation, donor, embryo

Abstract

Six hundred and sixty five eggs of hatched failure and 150 of ducklings, from the hatchery of Alabio duck at the District of Hulu Sungai Utara (HSU), South Kalimantan, were examined for Salmonella sp. The results showed that 45 samples of eggs (6.8 %) and 18 samples of ducklings were positive to Salmonella sp (12 %). The serotipe of Salmonella sp. from eggs samples were 17 isolates of S. typhimurium (2.6 %),26 isolates of S. sentenberg (3.9 %), 1 isolate of S. hadar (0.2 %) and 1 isolate of S. virchow (0.2 %); and from duckling samples there are 5 isolates of S. typhimurium (3.3 %), 4 isolates of S. senftenberg (2.7 %), 5 isolates of S. hadar (3,3 %) and 4 isolates of S. virchow (2,7 %).Key words: Salmonella sp, hatchery, Alabio duck

Abstract

Response of oestrus in Boer goats and development of ovarian follicle to superovulatory treatment were studied. All goats (n= 17) received subcutaneous progesterone ear implant for 11 days to synchronize oestrus cycle. Forty eight hours before implant removal and the following 3 days. 16 AU pFSH containing 40 % pLH was injected 6 times (4, 4; 2, 2; 2, 2) in 12 h intervals. Goats received PGF2a twice synchronized with FSH treatments to induce luteolysis (5 mg PGF2a of each) 24 h after implant removal. Animals were checked for oestrus with an aproned buck 3 times a day. During standing oestrus animals were naturally mated 2 or 3 times. Follicular development and ovulation were monitored 2 times a day using transrectal ultrasonography equipped with a 7.5 MHz linear-array transducer. This evaluation began from the time of implant removal until 48 h after the end of oestrus. Eighty eight per cent of animal showed oestrus with mean of the onset of oestrus 33 h after implant removal and oestrus duration of 30 h. Eighty two percent of does had corpora lutea with number of 11,0 and persistent follicles of 2.1 at laparoscopic examination on day 6 after the end of oestrus. The maximum number of follicles with diameter ≥ 2 mm recorded at the observation of 36 and 48 h after implant removal, whereas those with ≥ 6 mm in diameter were 48 - 60 h. The number of follicles with diameter 2 - 3 mm and 4 - 5 mm during observation period were constant. The ovulation of follicles did not occur spontaneously but they ovulated in several periods. The pFSH+ 40 % pLH preparation yielded a sufficient result for superovulation means in Boer goats. For monitoring the response of ovaries to the superovulatory treatment, ultrasonography was a simple and effective method.

Abstract

Studies on the production of spermatozoa and oestrus synchronization in sambar deer (Cervus unicolor) were conducted using nine stags and 12 hinds. Prior to the semen collection via electroejaculator stimulation, stags were sedated using a combination of fentanyl citrate, azaperone and xylazine hydrochloride (Fentazin®, Parnell Lab. NZ). Collected semen were evaluated for its volume, motility and concentration. Collected semen were evaluated for its volume, motility and concentration, placed into straw and then kept in frozen, chilled and fresh container. In hinds, the synchronization was conducted using intravaginal Controlled Internal Drug Release type G (CIDR-G®, 9 % w/w, 300 mg progesterone; Agricultural Division, CHH Products Group Ltd, Hamilton, NZ), followed by i.m injection of 250 IU PMSG. The results showed that stags and hind did not responp very well to all treatments. Mean of ejaculated semen volume was low (0.91 ml) and some ejaculated semen had 80 % motility. Any semen collection should be conducted based on the individual performance, with the age time in hard antler condition as the main factors. Half of the hinds responded to the synchronization 59.5 hrs after the injection of PMSG.

Abstract

The effect of maturing systems (single vs groups) of ovine oocytes was studied to examine the oocyte development in vitro. Oocytes were aspirated from follicle ovaries collected at local abattoirs Cibinong West Java, using a 18-G needle. Aspiration medium consisted of H 199 + 2 % FCS + 50 μg/ml Heparin and the maturating medium (IVM) consisted of B 199 - 10 % FCS + 10 μg/ml FSH + 10 μg/ml hCG + 1 μg/ml Estradiol. The oocytes collected were divided into three groups and treated separately as follows: T1) oocytes were matured singly in 50 μl IVM medium. T2) every five oocytes was matured in 50 μl IVM medium and T3) every 10 oocytes was matured in 50 μl IVM medium. All oocytes were maintained in incubator with 5 % CO, and high humidity at 38 °C for 20 h. The resulting ova were stained in 1 % lacmoid then examined for meiosis division under a microscope. There was a signiticant effect among treatments on the proportion oocytes reaching metaphase II (P
