Abstract

The study aimed to evaluate the  utilization of cassava plantation waste suplemented with zinc and cobalt on ewe reproduction performance. Six pregnacy ewes were allocated at experimentrations, i.e. K1: control (50% fresh grass + 50%  concentrate) and K2: 50% fresh grass + 50% cassava plantation waste  + 30 ppm zinc + 5 ppm cobalt. The results showed that the concentration of zinc and cobalt in blood of ewe in group K1 and K1 were 7.03 vs 8.22 ppm and 0.48 vs 0.44 ppm, respectively, while in faeces were 147,72 vs 158.59 ppm and 8.3 vs 9.8 ppm. In addition, the concentration of alkaline phosphatase and progresterone in blood were 116 vs 196 u/l and 0.56 vs 0.59 mg/ml, respectively. The average of offsprings weight on 24 day postpartum in K1 and K2 was 3.48 vs 3.16kg. In conclusion, cassava plantation waste could be used as feed for pregnancy and lactating ewe. Supplementation of 30 ppm zinc is efectively increase the concentration of zinc and alkaline phosphatase in blood.Key words: cobalt, reproduction, cassava, zinc, ewe

Abstract

The purpose of this study was to determine the genetic characteristics of rabies virus based on phylogenetic relationship among rabies virus in various regions in Indonesia. The amino acid sequence of the nucleoprotein gene of rabies virus isolate from Banten (RV/Banten-01/dog/2007), Makasar (RV/MKS-26/dog/2010), Bukit Tinggi (RV/BKT-52/dog/2009 and RV/BKT-58/dog/2009), Medan (RV/Medan-27/dog/2007) and Bali (RV/Bali-1/dog/2009; RV/Bali-2/dog/2009; RV/Bali-3/dog/2009), Indonesia was determined. These isolates showed a high degree of homology among Indonesian isolates which reached 100%. Meanwhile, the level of homology between rabies virus isolates from cats rabies virus isolates from dogs reached 97%. Results of  phylogenetic analysis using the amino acid sequences of the N genes showed that all of  Indonesian rabies virus isolates were closely related to rabies viruses from China than those  from Thailand, Laos, Burma, and Vietnam which geografically closer to Indonesia. Data obtained from the phylogenetic analysis is expected to trace the source of rabies spread and the possibility to create a vaccines which more suitable with rabies virus that spreads in Indonesia. Based on the phylogenetic relationship analysis using the amino acid sequence of the rabies virus N protein gene showed that all of rabies virus isolated  from Indonesian regions share a high homology with others ranging from 97-100%.Key words: sequencing, rabies, nucleoprotein gene (N), homology

Abstract

The purpose of this study was to determine the prevalence, serovar, and risk factors of leptospirosis on cattle in Progo watershed. A total of 330 cattle samples were clinically examined and blood was collected for Leptospira examination using microscopic agglutination test (MAT). Results of MAT were used as dependent variable (Y), while breeders, cattle, shed, and feed factors were used as the independent variable (X). Univariate analysis, bivariate analysis with chi square, odds ratio, and multivariate with logistic regression were implemented to analyze the data. The results showed that all cattle were cinically health, 13.03% (43/330) cattle were positively leptospirosis which 37 out of 193 (9.17%) cattle belongs to the farmer. Mostly, leptospirosis cases were caused by hardjo serovar (38.0%). Risk factors influencing the prevalence of leptospirosis were caused by some factors such as the presence of rats in cage (OR 2.7), the distance of waste disposal which should be less than approximately 5 meters (OR 2.1), feed concentrates, grass and rice straw (OR 0.4), grazing (OR 0.2), and concentrate feed and rice straw (OR 0.1). It can be concluded that leptospirosis infection on cattle in Progo watershed is 13.03% (at livestock level) and 19.17% (at farm levels). Factors of rat in the cage and the distance of waste disposal (5 m) increase the risk of leptospirosis cases. Factors of feed combine with concentrate and rice straw as well as grazing reduced the risk of leptospirosis in cattle, but the addition of grass in concentrate feed and rice straw provides a greater risk of leptospirosis infection compare to concentrate feed and rice straw alone.Key words: beef cattle, leptospirosis, prevalence, risk factors

Abstract

The objectives of this research were to examine the effect of Sechium edule ethanolic extract to repair beta pancreas cells damage and tomeasure nitrogen oxide (NO) concentration in streptozotocin-induced rats. Type I diabetic rats were obtained by intraperioneally injected therats with multiple low dose streptozotocin (MLD-STZ) for 5 days. In this research, rats were assigned into 5 treatment groups: untreated rats ingroup 1 were considered as negative control, while MLD-STZ induced type 1 diabetic rats in group 2 were considered as positive control. Inanother 3 groups, type 1 diabetic rats were orally treated with three doses of Sechium edule ethanolic extract (14, 28, 42 mg/kg bw) respectivelyfor 7 consecutive days. Repairing of beta pancreas cells were analyzed descriptively whereas concentration of NO were analyzed using one wayANOVA and continued with LSD test (α= 0.01). The result showed that the administration of Sechium edule ethanolic extract significantlydecreased NO concentration (P0.01) up to 67.9 µM and repaired beta pancreas cells damage. Dose of 42 mg/kg bw of Sechium edule ethanolicextract is the effective dose for diabetic rat therapy. Key words: Sechium edule extract, diabetic mellitus, rat, beta pancreas cells

Abstract

The objective of this study was to identify the type of specific glycoconjugates and its distribution in testicular spermatogenic cells in muntjak (Muntiacus muntjak muntjak) based on lectins histochemistry. An adult male muntjak aged 4-5 years old in hard antler period was used in this study. Testicular tissue was fixed in Bouin solution and processed histologically. Histochemistry method was performed using six types biotinylated lectins such as peanut agglutinin (PNA), soybean agglutinin (SBA), wheat germ agglutinin (WGA), ricinus communis agglutinin (RCA), concanavalin A (Con A), and ulex europaeus agglutinin I (UEA I) with 20 µg/ml of concentration for PNA lectins and 15µg/ml for other type of lectins. The results showed that glycoconjugates were detected by all type of lectins except UEA I in testicular spermatogenic cells with variation in distribution pattern and also the intensity of lectins binding. Glycoconjugates β-galactose, β-glucose, mannose, Nacetylgalactosamine, N-acetylglucosamine and sialic acid were stained intensely by lectins in golgy-cap phase and acrosomal phase of spermatids. Glycoconjugate N-acetylgalactosamine was the sugar residues which distributed abundantly that marked by positive reaction with PNA, SBA, and RCA lectins. In conclusion, glycoconjugates are detected in testicular spermatids cells of muntjak indicated that glycoconjugates have an important role in spermatogenesis particularly in spermiogenesis. Key words: glycoconjugates, lectins, spermatid, spermatozoa, muntjak

Abstract

The aim of this study was to examine the potency of fibroblast cells transdifferentiated to neuron cells in vitro. Newborn rat neuron conditioned medium (NBRN CM) was collected from neuron cells cultured with mDMEM without serum for 48 hours. Fibroblast cells were collected from fetal rat muscle treated with trypsin. Fibroblast cells were culture with 3 kind of culture medium: mDMEM + 0.01 mM          β-mercaptoethanol; mDMEM + 50% NBRN-CM and  mDMEM + 0.01 mM β-mercaptoethanol + 50% NBRN CM . As control, cells was cultured with mDMEM +10% newborn calf serum (NBCS). The addition of NBRN CM into culture medium resulted in 12.97% newborn cells in fibroblast culture medium passage I. Newborn rat neuron conditioned medium in fibroblast culture medium resulted 12.97% neuron cells at passage 1. The percentage was increased (14.60%) when β- mercaptoethanol added into medium. The same result was found at passage 3 (12.67%; 13.17%). It showed that fibroblast cells has potency to transdifferentiated into neuron cells when cultured with NBRN CM. Further research is needed to know the fibroblast transdifferentiation potency. Key words: fibroblast, transdifferentiation, conditioned medium, neuron cells, in vitro

Abstract

The study aims to determine the histological structure of the testes, ductus epididymidis and ductus deferens of Varanus salvator bivittatus. The tissue samples were obtained by the histological techniques preparation with thickness section is 3-4 µm then stained using hematoxylin-eosin (HE) and Masson's Trichrome (MT). The result showed that testes are covered by tunica vaginalis and tunica albuginea (tunica fibrous and tunica vasculosa). Tubulus seminiferous as a parenchyma are found in lobules of testes which are formed by septum as extension of tunica albuginea. Tubulus seminiferous is composed by layers of germinal epithelium cells consist of spermatogenia, Sertoli cell, spermatocyte and spermatid which is developed further to be spermatozoa in the lumen of tubulus. Among the tubulus seminiferous, there are groups of epithelioid cells called Leydig cells. The end of tubulus seminiferous formed ductus epididymidis which then ended up and known as ductus deferens. Epididymis is covered by dense connective tissue. Ductus epididymidis is divided into three segments: cranial, medial and caudal. It is lined by pseudostratified columnar epithelium which is varied in its thickness, amount and diameter of lumen. Lumen of the ductus deferens was lined by pseudostratified ciliated columnar epithelium and thin muscularis mucosal layer and thick circular smooth muscle were covered externally.Key words: Varanus salvator bivittatus, testes, ductus epididymidis, ductus deferens, micromorphology

This study aims to find out the eeffect of Eurycoma longifolia Jack. roots on the liver damage of pregnant mice. This research used Balb/cstrain mice, 2-3 month, with the weight ranging of 27-33 gram. Samples were divided into three treatment groups consisted of three mice. GroupI (KI) was negative control (placebo), only treated with 0.06 ml/20 g bw of distilled water, group II (KII) was administered with roots ofEurycoma longifolia Jack extract at dose of 14 mg/20 g bw, and group III (KIII, positive control) was administered with Schizandrae at the dose0.06 mg/20 g bw. Treatment were done for 7 consecutive days in pregnancy period. On day 8th, blood samples were collected to examine the levelof alanine transaminase (AST) and aspartate transaminase (ALT) enzymes in serum and liver organ were collected to observe thehistopathological changes. The average of ALT and AST enzymes level on K1, K2, and K3 were 18.34+0.28, 19.68+0.15, and 19.20+0.08 U/l; and21.79+0.26, 23.42+0.41, and 22.23+0.52 U/l. The histopathological examination results showed that the administration of E. longifolia Jackroots extract and Schizandrae reveal the karyomegali and fatty change of hepatocytes. It is concluded that the administration of E. longifoliaJack roots extract at the dose of 14 mg/20 g bw could decrease the liver function of pregnant mice. Abstract

This study aimed to investigate the effectivity of goat milk fermentation as tyrosinase inhibitory with Lactobacillus rhamnosus TW 2. The examination of fresh milk contained the density, protein, and fat content. The culture of Lactobacillus rhamnosus TW 2 acid lactid bacteria starter was added with the concentration of 3, 4, and 5%, inoculated in pasteurized goat milk, then incubated at 37 C for 24 hours. Fermented milk were extracted by centrifugation, then supernatant was collected and was used for inhibition of tyrosinase enzymes activity on L-tyrosin and L-dopa substrate. The result showed that the density, fat content, and protein content of Etawah crossbred goat milk are 1.028, 3.73, and 5.45%, respectively. Re-identification of lactic acid bacteria showed similar morphology, physiology, and bio-chemistry with the isolated lactic acid bacteria. The growth curve of TW 2 was observed in 12 hours. The 5 % of Lactobacillus rhamnosus TW 2 was the best concentration to inhibit tyrosinase activity in L-Tyrosin substrate. In conclusion, fermentation of goat milk using Lactobacillus rhamnosus TW 2 at concentration of 5% as starter is effective to inhibit tyrosinase activity significantly.Key words: goat milk, fermentation, tyrosinase inhibitory
