Abstract

Semen was collected using an artificial vagina, and then examined macroscopically and microscopically. Semen with motility of 70% was diluted into different extenders: tris egg yolk-glucose (TEYG), tris egg yolk-glucose extender with 0.5 mM melatonin (TEYG + M0.5), tris egg yolk-glucose extender with 1 mM melatonin (TEYG + M1), tris egg yolk-glucose extender with 2 mM melatonin (TEYG + M2). All semen samples were stored at 4 C for 5 days (H0-H5) and examined every 24 hours. Semen was examined for spermatozoa motility, viability, abnormality, and membrane plasma integrity. The results showed that there were no significant changes of semen quality on H0 (P0.05). In general, the melatonin supplemented groups had a higher semen quality value than the control. Melatonin supplementation can protect and maintain the quality of chilled semen. In addition, the result showed that TEYG + M1 extender was the best extender treatment, which was indicated by higher values than other treatments.

Abstract

This study aimed to analyze flavonoid and 6-gingerol content and to test the hypoglycemic effects of industrial-grade Cajanus cajan (pigeon pea) leaf and Zingiber officinale rhizome (ginger) extracts on hyperglycemic rats. Extraction utilized maceration with 70% alcohol at PT. Insular Multi Natural. Extracts were characterized and analyzed for flavonoids and 6-gingerol content. Hypoglycemic activity was assessed using the oral glucose tolerance test (OGTT) on rats with induced hyperglycemia via 90% sucrose solution. The results indicated higher flavonoid content in C. cajan leaf extract (4090.09338.23 mg/100 g) compared to Z. officinale extract (622.74203.39 mg/100 g), while gingerol content in Z. officinale extract was 701.524.42 mg/100 g. This study concluded that combination of industrial-grade pigeon pea leaves extract and ginger extract reduce the level of blood glucose in hyperglycemic model rats at all combination dose (150:150, 200:100, and 100:200 mg/kg BW). The combination dose of 200:100 mg/kg BW showed the best blood glucose level profile.

Abstract

This study aimed to determine the concentrations of Follicle Stimulating Hormone (FSH) and Luteinizing Hormone (LH) in New Zealand White (NZW) rabbits after superovulation induction using FSH. Six female NZW rabbits and one male NZW rabbit were used in this study. Criteria for female NZW rabbits used include having previously given birth, not being pregnant, and having a body weight of 2-3 kg. The rabbits were divided into two treatment groups; the first group was injected with physiological NaCl (NZW1, n= 3), while the second group was injected with FSH (NZW2, n= 3). Injections of physiological NaCl or FSH were administered five times at 12-hour intervals and first injection was given at 20:00 p.m. In NZW1, each injection was given physiological NaCl with volumes of 0.2:0.2+0.2;0.4+0.4 mL, respectively whereas in NZW2, each injection was given FSH with doses of 4:4+4:8+8 mg, respectively. Twelve hours after the last FSH injection, NZW2 rabbits were injected with 100 IU hCG and mated with male rabbit, while NZW1 rabbits were mated without hCG administration. On days 1, 3, and 5 after mating, blood samples were collected for FSH and LH hormone examination using the Enzyme-Linked Immunosorbent Assay (ELISA) method. The FSH concentrations on days 1, 3, and 5 between NZW1 vs NZW2 were 34.781.46 vs 39.753.18; 41.434.90 vs 41.425.14; and 44.471.51 vs 30.130.19 mIU/mL, respectively (P0.05). The LH concentrations on days 1, 3, and 5 between NZW1 vs NZW2 were 33.784.01 vs 40.1812.36; 29.342.24 vs 31.3110.13; and 42.0911.38 vs 30.578.87 mIU/mL, respectively (P0.05). It is concluded that FSH administration does not affect the concentrations of FSH and LH in NZW rabbits. The FSH concentration five days after mating tends to fluctuate, while the LH concentration tends to decrease as the days increased.

Abstract

This study was aimed to observe the survival rate (SR) and clinical signs, and the acute immune response of Pacific white shrimp (Litopenaeus vannamei), including total hemocyte count (THC), bacterial clearance, and prophenoloxidase (ProPO) activity due to infection with the Vibrio parahaemolyticus bacteria and its metabolites. Shrimp were infected by bacterial isolates with culture media (A), bacterial isolates without culture media (B), and metabolites suspensions from bacterial culture media (C) for 8 hours with observation every 2 hours. The results showed that experimental shrimps in groups A and B showed clinical signs and reduced SR, 24.44% and 62.22%, respectively. Experimental group C still showed the clinical signs but did not cause the mortality (SR 100.00%). The THC did not show significant changes, either based on experimental groups or duration of infection, but all groups tended to show decreasing THC at 8 hours post-infection. Bacterial clearance and ProPO activity also did not show significant differences between experimental groups or duration of infection at 8 hours post-infection. However, there was an increase in bacterial clearance activity at 6 hours post-infection in the experimental groups A and B. Based on the study results, the acute immune response in shrimp from all of the experimental groups did not show significant changes at 8 hours post-infection. However, group A significantly decreased SR compared to groups B and C.

Abstract

The aim of this study was to determine the effect of stems and leaves ciplukan extract on the degree of liver lobe inflammation in dyslipidemia-induced wistar rats. This research was an experimental study with a post-test only design. A total of 24 Wistar rats were allotted into 4 groups with 6 rats per group. Hepatic organs were then histologically stained with Hematoxylin-Eosin stain. Inflammation reading was done by looking for inflammatory foci in the preparation. The result was then analyzed using Kruskal-Wallis test. The results showed that groups I and II had one severe inflammation. Group III had two severe degrees of inflammation. Group IV had no severe inflammation. The analysis revealed that there was no significant difference in the degree of inflammation between groups (P= 0.964). Administration of ciplukan stem and leaf extract did not affect the degree of liver lobe inflammation in Wistar rats induced by dyslipidemia.

Abstract

This study aims to determine the effectiveness of combination of snakehead fish, ginger and meniran extract (GTM) on healing wounds after ovariohysterectomy (OH) in cats. Twenty healthy female cats aged 1-4 years, weighed 2-4 kg were divided into three groups with different doses of GTM and compared with the group without treatment (n= 5 cats/group). The GTM dose was extrapolated from the human dose using allometric scaling, with the basal dose for G:T:M were 40:3.2:4.8 mg/kg BW (group 1, GTM1x). Treatment groups 2 (GTM2x) and 3 (GTM4x) were given a dose of GTM that was multiplied by 2 and 4 of the basal doses, respectively. OH was performed via a flank approach with the same operator and anesthesia. Discharge, swelling and erythema scores were significantly lower on days 3, 4 and 5 post-operativeamong all GTM groups compared with control. Pain evaluation using Baseline Tactile Semmes Monofilaments showed significantly lower in GTM2x and GTM4x compared to GTM1x and control. The 4A-vet pain scale showed lower pain in GTM2x from 1 to 5 days postoperative. The number of leukocytes, erythrocytes and platelets was in the normal range indicates the absence of postoperative infection. Postoperative albumin levels showed a significantly higher difference in GTM2x compared to control. The treatment given GTM2x showed the fastest healing interval (3.20.44 days). The combination of GTM can reduce pain, reduce wound scores and accelerate post-OH wound healing with the most effective dose was GTM2x. ________________________________________________________________
