Abstract

Non-infectious infertility is particularly critical for sire rams/bucks, as seasonal breeders. This seasonal heavy use of males may lead to sexual exhaustion such that the regular semen production, libido, ejaculation, and litter yield can be seriously impaired. In severe cases, the reproductive processes may stop completely. For avoiding infertility risk, management and feeding should firstly be arranged with a flock-based approach. In reproductive management, the issues of using teaser ram/buck, male/female ratio, weekly mating number, exercise, and experience need attention. Also, inbreeding and using old breeders should be avoided. In oestrus synchronisation, necessary precautions against extremely hot/arid climatic conditions should be considered together with a sufficient number of rams/bucks used rotationally. For sustainable sheep/goat breeding, protective-preventive measures are preferred to problem-solving approaches. In enterprises where female breeder welfare is a priority, sustainable individual yield increases. Further, only the comfort level of males used heavily in season allows the expected libido and fertility to occur. In small ruminants, the necessary interest and responsibility in production provide convenience in the ideal solution with the opportunity of early interventions against undesirable fertility problems in breeders. Undoubtedly, it is vital to consider sustainable animal health and welfare for profitable breeding and husbandry in sheep/goats.

Abstract

The aim of this study was to determine the diversity of the sex-determining region Y (SRY) gene in Madura cattle bulls that had specifically selected for the production of frozen semen. DNA was isolated from the whole blood derived from 5 Madura cattle bulls and the SRY gene amplification carried out using polymerase chain reaction (PCR) with SRY-4 as a primer followed by DNA sequencing. Results revealed the proximity of the Madura cattle SRY genes to the Bos indicus SRY genes, furthermore this study also proved the existence of variation in Madura cattle SRY gene caused by mutation and deletion of nucleotides. It was concluded that the variations in Madura cattle SRY genes still persist even in the specifically selected populations with similar phenotype.

Abstract

Tujuan penelitian ini adalah mengetahui keragaman genetik gen COX1 pada Tarsius spectrum (T. spectrum), Tarsius dianae (T. dianae), dan Tarsius bancanus (T. bancanus). Sampel yang digunakan adalah 4 sampel jaringan T. spectrum asal Sulawesi Utara, 1 sampel jaringan T. dianae asal Sulawesi Tengah, 2 sampel jaringan T. bancanus asal Lampung, dan 3 sampel jaringan T. bancanus asal Kalimantan. Selanjutnya dilakukan isolasi deoxyribonucleic acid (DNA), amplifikasi dengan teknik polymerase chain reaction (PCR), pengurutan, dan data dianalisis menggunakan program MEGA v. 5.0. Hasil amplifikasi diperoleh produk PCR sebesar 1633 pasang basa (pb), hasil pengurutan DNA ditemukan 240 situs nukleotida dan 16 situs asam amino yang berbeda. Jarak genetika menggunakan Kimura-2 parameter paling tinggi 16,1%, paling kecil 0%, dan rata-rata 8,3%. Pohon filogenetika menggunakan metode Neighbor joining berdasarkan urutan nukleotida dan asam amino COX1 dapat membedakan antara T. bancanus, T. spectrum, dan T. dianae, namun tidak dapat membedakan antara T. bancanus asal Kalimantan dan T. bancanus asal Lampung.

Abstract

This research was conducted to find out the Wnt4 recombinant proteins which expressed by Escherichia coli (E. coli) BL21 carrying the recombinant DNA wnt4 (E. coli transformation). Research materials were E. coli BL21 transformation and E. coli BL21 non-transformation (negative control). The expression of recombinant protein was conducted by culturing E. coli for 24 hours in Luria-Bertani (LB) media with isopropyl β-D-1-thiogalactopyranoside (IPTG) induction. Recombinant protein was isolated by sonication of pellet bacteria. Protein analysis performed by 15% sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS-PAGE). The results showed that recombinant protein with a molecular weight of 33 kDa has been expressed by E. coli BL21 transformation successfully.

Abstract

Penelitian ini bertujuan mendeteksi keberadaan antigen Coxiella burnetii sebagai penyebab Q fever pada organ sapi yang dikumpulkan di rumah potong hewan (RPH) Kota Medan dan Kabupaten Deli Serdang. Pada penelitian ini telah dikumpulkan organ limpa, paru-paru, dan hati dari 162 ekor sapi. Sampel organ tersebut kemudian diperiksa secara imunohistokimia dengan metode streptavidin peroksidase untuk melihat keberadaan antigen Coxiella burnetii menggunakan antibodi poliklonal terhadap Coxiella burnetii. Hasil pemeriksaan imunohistokimia menunjukkan 62/162 (38,3%) sampel sapi imunoreaktif terhadap Coxiella burnetii. Berdasarkan asal pengambilan sampelnya, sebanyak 40/101 (39,6%) sampel sapi yang berasal dari RPH Kota Medan dan 22/61 (36,1%) sampel sapi yang berasal dari RPH di Kabupaten Deli Serdang menunjukkan hasil imunoreaktif. Dilihat dari jenis organnya, dari 162 sampel sapi, antigen Coxiella burnetii dapat dideteksi pada 61 (37,7%) organ limpa, 12 (7,4%) organ paru-paru dan 2 (1,2%) organ hati. Hasil ini menunjukkan telah adanya infeksi Coxiella burnetii pada sapi di Kota Medan dan Kabupaten Deli Serdang.

Abstract

ABSTRACT This study aimed to get specific genetic marker for Anisakis sp. identification on mackerel tuna using gene sequence cytochrome oxidase subunit II (COII) mitochondrial deoxyribonucleic acid (mtDNA) and to identify taxonomic affiliation between Anisakis sp. from Indonesia and others Anisakis sp. from GenBank database. This study started with sample collections at three fish auctions in Cilacap (Central Java), morphology classification, DNA isolation, and molecular based identification using polymerase chain reaction (PCR) and sequencing methods. Molecular based identification of Anisakis used gene amplification COII mtDNA as a cell target prior to sequence. Morphology characteristic results showed that Anisakis nematodes which infected mackerel tuna classified as type II L3 larvae. Molecular based identification showed significant result, which found 530 bp COII DNA gene fragment similar to target cell. Gene sequencing alignment results of COII Anisakis gene compared with GenBank showed 11 different nucleotide sites that can be used as genetic barcode for Indonesian Anisakis sp. This study showed that Anisakis sp. infected mackerel tuna in Java Sea is Anisakis physeteris and considered as zoonosis.

Abstract

Penelitian ini bertujuan mengetahui efektivitas amida minyak ikan dalam menurunkan konsentrasi kolesterol dan trigliserida plasma dan pengaruhnya terhadap nilai hematologi darah. Tiga puluh lima ekor tikus jantan, galur Sprague Dawley, umur 7 minggu dibagi secara acak ke dalam lima kelompok perlakuan. Kelompok kontrol (A) mendapat ransum semi purified yang mengandung 8% minyak jagung. Kelompok perlakuan masing-masing disuplementasi 4,5% minyak ikan (B), 3% minyak ikan+1,5% amida (C), 1,5% minyak ikan+3% amida (D), dan 4,5% amida (E). Ransum perlakuan diberikan selama 6 minggu. Pada akhir penelitian dilakukan pengambilan sampel darah untuk menganalisis nilai hematologi dan profil lemak plasma. Hasil penelitian menunjukkan bahwa suplementasi amida minyak ikan dapat mempertahankan jumlah eritrosit dan Hb, namun nilai hematokrit mulai menurun pada suplementasi amida 3% (D), dibandingkan suplementasi minyak ikan (B) dan suplementasi amida 1,5% (C). Jumlah leukosit pada kelompok yang mendapat suplementasi 4,5% amida (E) secara nyata (P0,05) lebih tinggi dibandingkan dengan kelompok yang diberi minyak ikan (B). Suplementasi amida minyak ikan tidak berpengaruh terhadap konsentrasi kolesterol total dan HDL plasma, namun pada suplementasi 3% amida mulai meningkatkan konsentrasi trigliserida dan LDL plasma secara nyata (P0,05). Disimpulkan bahwa suplementasi amida sebesar 3% mulai memperlihatkan pengaruh negatif terhadap nilai hematologi dan profil lemak plasma.

Abstract

The objective of this study was to identify the type of specific glycoconjugates and its distribution in testicular spermatogenic cells in muntjak (Muntiacus muntjak muntjak) based on lectins histochemistry. An adult male muntjak aged 4-5 years old in hard antler period was used in this study. Testicular tissue was fixed in Bouin solution and processed histologically. Histochemistry method was performed using six types biotinylated lectins such as peanut agglutinin (PNA), soybean agglutinin (SBA), wheat germ agglutinin (WGA), ricinus communis agglutinin (RCA), concanavalin A (Con A), and ulex europaeus agglutinin I (UEA I) with 20 µg/ml of concentration for PNA lectins and 15µg/ml for other type of lectins. The results showed that glycoconjugates were detected by all type of lectins except UEA I in testicular spermatogenic cells with variation in distribution pattern and also the intensity of lectins binding. Glycoconjugates β-galactose, β-glucose, mannose, Nacetylgalactosamine, N-acetylglucosamine and sialic acid were stained intensely by lectins in golgy-cap phase and acrosomal phase of spermatids. Glycoconjugate N-acetylgalactosamine was the sugar residues which distributed abundantly that marked by positive reaction with PNA, SBA, and RCA lectins. In conclusion, glycoconjugates are detected in testicular spermatids cells of muntjak indicated that glycoconjugates have an important role in spermatogenesis particularly in spermiogenesis. Key words: glycoconjugates, lectins, spermatid, spermatozoa, muntjak

Abstract

Tujuan penelitian ini adalah membuktikan potensi dan efektivitas rat bone marrow mesenchymal stem cell (rbmmsc) sebagai terapi pada kasus teratogenik selama kebuntingan dengan melihat ekspresi tumor necrosis factor (TNF-α), Bax, dan Bcl-2. Dua puluh empat ekor tikus bunting model teratogenik dibagi menjadi empat kelompok perlakuan yang berbeda yaitu P1-Kontrol (dipapar carbon black dosis 532 mg/m3 selama 4 jam pada umur kebuntingan ke-6 s/d ke-11 + Injeksi MEM 0,1 ml), P1-Terapi (dipapar carbon black dosis 532 mg/m3 selama 4 jam pada umur kebuntingan ke-6 s/d ke-11 + rbmmsc dengan dosis 1x106 sel/0,1 ml), P2-Kontrol (dipapar carbon black dosis 532 mg/m3 selama 4 jam pada umur kebuntingan ke-6 s/d ke-17 + Injeksi MEM 0,1 ml), P2-Terapi (dipapar carbon black dosis 532 mg/m3 selama 4 jam pada umurkebuntingan ke-6 s/d ke-11 + rbmmsc dengan dosis 1x106 sel/0,1 ml). Data yang didapatkan kemudian dianalisis menggunakan uji MannWhitney. Hasil penelitian ini menunjukkan bahwa pemberian terapi rat bone marrow mesenchymal stem cell pada tikus model teratogenikparticulate matter tidak berpengaruh dalam menurunkan parameter ekspresi TNF-α dan Bax, serta meningkatkan ekspresi Bcl-2 pada plasenta.

Abstract

Red tilapia (Oreochromis hybrid) is Indonesia's leading freshwater fishery commodity susceptible to streptococcal bacterial infection. Many studies have been conducted on various efforts to prevent and treat this disease, one of which uses the immunoglobulin Y (IgY) molecule from chicken egg yolk. This study aimed to observe the biological function of IgY against Enterococcus faecalis as a cause of streptococcal-like infection. The agglutinin function was conducted by observing the growth of Enterococcus faecalis in brain heart infusion (BHI) broth media which was added with IgY suspension. The function of inhibin was performed using a spectrophotometric method to measure the level of turbidity of the bacterial suspension inoculated with IgY suspension. The bactericidal potential through the complementary activation pathway for red tilapia serum was carried out using a scanning electron microscope (SEM) method to evaluate the topography of the bacterial cell wall. The results of the study can be concluded that IgY anti-Enterococcus faecalis has the potential as an agglutinin, inhibin, and bactericidal agent through its putative potential in complement activation in streptococcal bacterial infections in red tilapia commodities.
