Abstract

Tujuan penelitian ini adalah mengetahui waktu terbaik untuk mendapatkan akurasi tertinggi pada pemeriksaan kimia urin untuk diagnosis kebuntingan dini pada sapi lokal. Pemeriksaan urin dilakukan sesuai prosedur yang dikembangkan oleh Cuboni-Lunaas.Waktu koleksi urin adalah pada bulan ke-1, 2, dan 3 setelah inseminasi. Hasil positip dari pemeriksaan ditunjukkan oleh terbentuknya fluoresensi pada larutan. Hasil pemeriksaan ini dikonfirmasi dengan pemeriksaan kebuntingan secara manual. Akurasi metode diagnosis dengan kimia urin pada waktu pemeriksaan pada bulan ke-1, 2 dan 3 pasca inseminasi masing–masing adalah 75,0; 87,5; dan 100% untuk mendiagnosis sapi bunting dan 0,0; 100,0; dan 100,0 % pada untuk mendiagnosis sapi yang tidak bunting. Waktu pemeriksaan dengan akurasi terbaik untuk diagnosis bunting diperoleh pada bulan ke-3 sedang untuk diagnosis tidak bunting diperoleh pada bulan ke-2 pasca inseminasi.

Abstract

This study was aimed to find out the effect of ethanolic extract of ant plant (Myrmecodia sp.) to increase the activity of enzyme lipoprotein lipase (LPL) serum and to observe the histopathological changes of hypercholesterolemic rat liver. This study used 20 male rats grouped into 4 treatment groups, namely negative control group (K1), hypercholesterolemic group (K2), and hypercholesterolemic group that administered with ethanolic extract of ant plant 100 (K3) and 200 mg/kg bw (K4). The LPL enzyme activity were measured by the titration method and histopatological changes of liver were observed by calculated fatty degeneration and fatty infiltration. The data were analyzed using one way anova followed by Duncan test. The average of LPL enzyme activity on group K1, K2, K3, and K4 were 0.80±0.06, 0.45±0.10, 0.83±0.11, and 0.76±0.03 unit, respectively. The average number of fatty degeneration on hepatocyte and fatty infiltration were 1.80±0.83, 3.60±1.14, 23.00 ±1.22, and 40.20±1.30; and 9.20±0.84, 16.40±1.14, 2.60±0.54, and 4.80±0.83, respectively. The results showed that theraphy ethanolic extract of ant plant effects significantly (P0.01) on the increase of enzyme LPL and improve liver damage in hypercholesterolemic male rats. To conclude the administration of ethanolic extract of ant plant increases the LPL enzyme activity and improves liver damage on hyperch olesterolemic rats.Key words: Myrmecodia sp., LPL activity, histopathological liver, hypercholesterolemia

Abstract

Penelitian ini dilakukan untuk mengetahui efek suplementasi tepung daun jaloh (Salix tetrasperma Roxb) (TDJ) dalam pakan terhadap nilai diferensial leukosit dan ketahanan hidup setelah uji tantang dengan Aeromonas hydrophila pada ikan nila (Oreochromis niloticus) yang dipelihara pada akuarium diberi peningkatan suhu lingkungan. Penelitian ini menggunakan rancangan acak lengkap faktorial 2x4. Faktor pertama adalah penambahan jumlah tepung daun jaloh dalam pakan yaitu: 0% (P1), 5% (P2), 10% (P3), dan 15% (P4) dari berat pakan dan faktor kedua adalah suhu air dalam akuarium yaitu 29±1 C (S1) dan 35±1 C (S2), sehingga didapat 8 kombinasi perlakuan yaitu: P1S1, P2S1, P3S1, P4S1, P1S2, P2S2, P3S2, dan P4S2 dengan ulangan 10 ekor ikan per perlakuan. Sebanyak 80 ekor ikan nila dengan bobot badan 40-50 g secara acak dibagi ke dalam 8 perlakuan. Perlakuan dilakukan selama 30 hari. Pada hari ke 31 dilakukan pengambilan sampel darah dan uji tantang. Hasil penelitian ini menunjukkan bahwa suplementasi pakan dengan TDJ pada konsentrasi 5-15% tidak berpengaruh terhadap nilai diferensial leukosit ikan nila yang dipelihara pada suhu lingkungan yang berbeda. Suplementasi TDJ dalam pakan sebanyak 5-10% dapat mengurangi kematian ikan akibat infeksi Aeromonas hydrophila terutama pada suhu 35±1 C. Dari hasil penelitian ini dapat disimpulkan bahwa ikan nila yang dipelihara pada akuarium dengan suhu air 35±1 C dan diberi pakan yang disuplementasi TDJ 5-10% dapat meningkatkan daya tahan tubuh ikan nila.

Abstract

The purpose of this study was to evaluate the toxic effects of beach cabbage leaves [Scaevola taccada (Gaertn.) Roxb.]by observing the histopathological changes in the livers and kidneys of male white rats. This study used 20 white rats which were divided into 4 groups: 1 control group and 3 groups receiving treatments in the form of Scaevola taccada leaf ethanolic extract (STLEE) in graded doses (200 mg/kg BW, 400 mg/kg BW, and 600 mg/kg BW). Toxicity test was performed by administering STLEE for 14 days. The results of daily observations showed that the experimental animals experienced diarrhea. Meanwhile, histopathological observations showed the occurrences of hydropic degeneration, fat degeneration, necrotic liver cells, and dilatation of Bowman's capsule in the animals’ kidney cells. It was concluded that STLEE at a dose of 200 mg/kg BW is safe to use, but is toxic at a dose of 600 mg/kg BW, both to the livers and kidneys.

Abstract

This study was carried out to evaluate the seasonal variation on semen characteristics of Murrah buffalo bulls in Indonesia. A total of three Murrah buffalo bulls were included in this study. The semen was collected once a week during the rainy season (from January to April and from October to December) and the dry season (from May to September). Immediately after collection, the semen volume, sperm concentration, total sperm output, individual motility, post-thawing motility, and straw production were evaluated. The results showed that the season did not change (P0.05) the semen volume, individual motility, and post-thawing motility. However, sperm concentration, total sperm output, and straw production were decreased in the dry season (P0.05) as compared to the rainy season. In conclusion, several semen characteristics of Murrah buffalo bulls may vary depending on the season of collection. The semen collection during the rainy season is more profitable than the dry season.

Abstract

The objective of this study was to identify the five different viral infections including avian influenza, Newcastle disease, avian reovirus, avian encephalomyelitis, and Marek’s disease in the runting-stunting syndrome outbreak in several commercial layer farms in Sukabumi and Tangerang in November 2014 using polymerase chain reaction technique. As results, this study identified mix infection of three viruses in the field samples, including Newcastle disease, reovirus, and avian encephalomyelitis; however, it was negative for avian influenza and Marek’s disease viruses. Subsequently, the inoculation of several samples into embryonated chicken eggs confirmed the growth of these three viruses. As a consequence, disease control management should be conducted in the affected farms by implementing effective biosecurity and vaccination program.

Abstract

Penelitian mengenai pengaruh ekstrak batang Cissus quadrangula Salisb. (sipatah-patah) terhadap tingkat proliferasi dan diferensiasi sel-sel tulang tikus (Sprague Dawley) prepuber umur empat minggu menggunakan sistem kultur in vitro. Sel-sel tulang dikultur dalam medium dulbecco’s modified eagle’s medium (DMEM) yang diberi tambahan newborn calf serum (NBCS) 10%, non essential amino acid (NEAA) 10%, NaHCO3, ITS 1 µl/ml (mengandung insulin 5 μg/ml, transferin 10 μg/ml, selenium 5 μg/ml; Sigma St Louis USA), dan 50 µg/ml gentamisin (mDMEM), dengan dan tanpa ekstrak Cissus quadrangula (CQ). Penelitian terdiri atas lima perlakuan yakni kontrol positif (mDMEM + deksametason 10-8 M), kontrol negatif (mDMEM), dan tiga konsentrasi CQ: mDMEM + CQ 0,3 mg/ml; mDMEM + CQ 0,6 mg/ml; dan mDMEM + CQ 1,2 mg/ml. Kultur dilakukan dalam inkubator CO2 5%, pada suhu 37° C selama tujuh hari. Parameter yang diamati adalah konsentrasi sel, komposisi, diameter osteoblas, dan diameter osteosit. Konsentrasi sel dihitung menggunakan hemositometer Newbauer. Komposisi osteoblas dan osteosit ditentukan berdasarkan pengamatan morfologi di bawah mikroskop cahaya. Diameter sel diukur menggunakan eyepiece micrometer. Data dianalisis menggunakan analisis varians dan uji Duncan. Hasil penelitian menunjukkan bahwa pemberian ekstrak Cissus quadrangula Salisb. pada konsentrasi 0,3 mg/ml; 0,6 mg/ml; dan 1,2 mg/ml secara signifikan dapat meningkatkan proliferasi sel tulang; dan pada konsentrasi 0,6 mg/ml mampu menginduksi diferensiasi osteoblas menjadi osteosit (P0,05). Disimpulkan bahwa pemberian ekstrak Cissus quadrangula pada konsentrasi 0,6 mg/ml ke dalam medium kultur dapat meningkatkan proliferasi dan diferensiasi osteoblas.

Abstract

This study utilized conventional and immunohistochemical methods, to describe the morphology and distribution of glucagon and insulin cells in the pancreas of adult male babirusa (Babyrousa babyrussa). The results of research showed that the endocrine portion (Langerhans islet) spread evenly on the acinar portion of pancreas. Observation using immunohistochemical method showed that glucagon cells were distributed on the perifer, whereas insulin cells in the center of the Langerhans islet of pancreas. In general, the number of insulin cells was higher than glucagon cells. It is interesting to note, in the pancreas babirusa also found that neonatal elements similar to the Langerhans islet, but have larger sizes (ranging between 200 -700 mm). The picture is similar to the large Langerhans islet was reported in ruminant.  These cells give a negative reaction to insulin and glucagon antisera.

Abstract

Pada penelitian ini, purwoceng diberikan pada tikus betina bunting umur 1-13 hari dengan tujuan mengetahui bobot badan, ovarium, dan uterus serta mempertahankan titik implantasi yang terbentuk sebelum masa implantasi. Penelitian ini menggunakan 2 kelompok yaitu kelompok tikus bunting kontrol dan tikus bunting yang mendapatkan purwoceng dengan dosis 25 mg/300 g bobot badan. Hasil penelitian menunjukkan bahwa kelompok tikus yang diberi purwoceng cenderung memiliki bobot ovarium dan uterus lebih berat dibandingkan kelompok kontrol. Purwoceng juga menyebabkan jumlah titik implantasi tikus hampir mendekati jumlah korpus luteum yang sudah terbentuk, dan ini menunjukkan bahwa keberhasilan implantasi lebih baik pada kelompok tikus yang diberi purwoceng.

Abstract

This study aimed to estimate the gestational age of Etawa crossbred goats based on ultrasound images of embryo and fetal usingtranscutaneous ultrasonography. Etawa crossbred does (n= 5) were synchronized using double PGF2α injection with 10 days interval and matednaturally. Early pregnancy was diagnosed on 20th day after mating base on isoechogenic image of the embryo (2.65±0.75 mm) andhypoechogenic appearance of gestational sac. On 30th day of the first trimester, the embryo size was 8.31±0.42 mm. The embryo reached10.44±0.51 mm on 35th day. On 60th day, the existence of the fetal were clearly appeared along with isoechogenic of umbilical cord,hyperechogenic os cranium (25.5±0.1 mm), and body length (51.83±1.59 mm). On 75th day, the diameter of the os cranium was 33.03±0.20 mm,body length of 110±1.73 mm, and bones structures appeared hyperechogenic for os thoracalis and os vertebrae. The placenta was isoechogenoicwith diameter of 23.3±0.2 mm. On 90th day, the placenta dominated the uterus (22.67±1.70 mm) and reached its maximum diameter on 145th day(33.73±2.91 mm). In conclusion, early pregnancy detection of Etawa crossbred goats using transcutaneous ultrasonography could be performedon 20th day after mating
