Abstract

Penelitian ini bertujuan memperoleh profil fluktuasi kadar hormon ovari sepanjang siklus estrus. Sampel darah yang berasal dari sepuluh ekor tikus betina diambil selama tiga siklus estrus dan dianalisis menggunakan perangkat inframerah. Kadar progesteron selama tiga siklus memiliki kisaran 6-11 ng/ml pada masa estrus dan meningkat menjadi 32-49 ng/ml pada saat non-estrus.

Abstract

This research aimed to compare the morphology and carbohydrate content of parotid andmandibular glands of goat, cat and pig (herbivore, carnivore and omnivore). The microscopic observationwas done using histochemical method with hematoksilin eosin (HE), alcian blue (AB) pH 2.5 andperiodic acid Schiff (PAS) staining methods.The results showed that the parotid and mandibular glandsconsist of acinar cells and ducts. The parotid gland of goat and pig were serous while mixed in the cat.The mandibular gland of goat, cat and pig were mixed type. The acinar of mandibular gland is composedof serous and mucous cells in which the mucous cell predominates. The AB and PAS stain showed weakto high concentration of acid and neutral carbohydrates found in the mucous cells of mandibular gland.However, these carbohydrates were not found in the serous cells of both glands, while acid carbohydratewas found in the parotid gland of cat. The present result suggested that the carbohydrates in the salivaof the goat, cat and pig might be produced mainly by the mucous cells.Keywords: parotid gland, mandibular gland, PAS, AB pH 2.5

Abstract

Penelitian ini bertujuan mengetahui efek sentrifugasi semen domba terhadap persentase kapasitasi dan reaksi akrosom spermatozoa. Penelitian ini terdiri atas 3 kelompok semen domba yaitu kelompok kontrol (P0) semen tanpa sentrifugasi, P1 semen disentrifugasi selama 5 menit dengan kecepatan 1800 rpm, dan P2 semen disentrifugasi selama 10 menit dengan kecepatan 1800 rpm. Hasil penelitian menunjukkan bahwa pada kelompok P2 kapasitasi tidak berbeda nyata dengan kelompok P1, tetapi pada kelompok P0 efek kapasitasi dan reaksi akrosom paling rendah. Reaksi akrosom spermatozoa pada kelompok P2 menunjukkan hasil yang paling tinggi. Lama sentrifugasi semen domba ekor gemuk dengan kecepatan sentrifugasi 1800 rpm selama 10 menit dapat meningkatkan persentase kapasitasi dan reaksi akrosom spermatozoa.

Abstract

The aim of this research was to determine if the substitution of rosella filtrate (Hibiscus sabdarifa L.) in citrate-based diluent and egg yolks with the ratio 4:1 (16 mL citrate + 4 mL yolk) would help maintain the progressive motility of Kacang goat spermatozoa at room temperature. This study used a male Kacang goat with an average sperm quality of approximately ≥ 205 x 107 spermatozoa/mL and motility of 81.6%. An electro-ejaculator was used once a week to collect the semen. The treatments were labelled as follows: P0 was the negative control group, treatment P1 was given 5% rosella filtrate, treatment P2 was given 10% rosella filtrate, and treatment P3 was given 15% rosella filtrate. Examination of the motility of the spermatozoa was carried out at room temperature. The examination was carried out once every four hours. It was concluded that the rosella filtrate could maintain the progressive motility of spermatozoa of the Kacang goat at optimal concentrations. The optimal motility percentage was found at a concentration of 15% at a storage life of 0-4 hours.

Abstract

The purpose of this research was to study the changes in uterine support capability power due to increase of litter size in Kacang goat. In this study, 9 pregnant female Kacang goats were divided into 4 different groups based on litter size, namely 1, 2, 3, and 4. At 7 weeks of pregnancy, the experimental Kacang goats were sacrificed to observe the macro and micro parameters of the uterus. The results showed that the litter size had a quadratic relationship with micro parameters of uterus but it had a linear relationship with macro parameters of the uterus. The variables of uterine glands area, glands lumen area, and cytoplasmic area reached maximum condition at the litter size of 2.5. Litter size had a linear relationship with the volume, weight, and dimension of the uterus. It can be concluded that the optimal number of litter size in Kacang goat was two offsprings which was proven by the optimal function of the uterus to support fetal development.

Abstract

Penelitian ini bertujuan mengetahui pengaruh induksi superovulasi dengan pregnant mare’s serum gonadothropin (PMSG) dan follicle stimulating hormone (FSH) terhadap peningkatan level steroid sapi aceh. Penelitian ini menggunakan 6 ekor sapi aceh betina dengan status tidak bunting, minimal 2 bulan pasca partus, sudah pernah beranak, dan sehat secara klinis. Sapi dibagi atas dua kelompok, masing-masing 3 ekor untuk tiap kelompok. Pada kelompok I, sapi diinjeksi dengan 1.500 IU PMSG pada hari ke-9 yang diikuti dengan penyuntikan 5 ml prostaglandin pada hari ke-11. Pada kelompok II, hari ke-9 sampai hari ke-12, sapi diinjeksi dengan FSH dua kali sehari (pagi dan sore, 08.00 dan 16.00 WIB) menggunakan dosis bertingkat yakni 3-3, 2-2, 1-1, dan 0,5-0,5 ml. Pada hari ke-11 sapi diinjeksi dengan 2,5 ml prostaglandin (pagi dan sore, 08.00 dan 16.00 WIB). Koleksi darah untuk pemeriksaan estrogen dilakukan ketika sapi memperlihatkan gejala berahi (saat inseminasi) setelah pemberian PMSG dan FSH yang diikuti dengan pemberian prostaglandin (berahi sesudah superovulasi) sedangkan koleksi darah untuk pemeriksaan konsentrasi progesteron dilakukan pada hari ke-7 setelah inseminasi. Pengukuran konsentrasi estrogen dan progesteron dilakukan dengan metode enzymelinkedimmunosorbanassay (ELISA). Konsentrasi estrogen pada saat estrus setelah induksi superovulasi dengan PMSG dan FSH masing-masing adalah 89,46±2,46 dan 54,62+9,91 pg/ml sedangkan konsentrasi progesteron pada hari ke-7 setelah inseminasi masing-masing adalah 14,78±2,33 dan 17,40±5,8 ng/ml. Hormon PMSG mempunyai kemampuan yang lebih baik dalam meningkatkan konsentrasi estrogen pada saat berahi tetapi hormon FSH mempunyai kemampuan yang lebih baik dibandingkan dengan hormon PMSG dalam meningkatkan konsentrasi progesteron hari ke-7 setelah inseminasi.

Abstract

This study aimed to determine the prevalence and associated risk factors of coccidiosis. Samples were obtained from 400 dairy cattle (196 calves aged 6 months, 37 calves aged 6-12 months, and 167 calves aged 12 months). Feces samples were collected, examined and counted for prevalence and number of oocyst per gram faeces (OPG) by McMaster technique. A questionnaire was design to record information about animal health and husbandry, individually. Risk factors associated with the prevalence of Eimeria in cattle were analyzed by logistic regression model. The overall prevalence and the average of OPG of Eimeria in cattle was 179 (44.75%) and 286.75, while highest prevalence of Eimeria was observed in calves aged less than 6 months. Cattle aged more than 12 months showed significantly different relationship (P0.05) to the prevalence of Eimeria infection compare to calves aged less than 6 months and aged 6-12 months. The presence of an immature immune system in younger calves resulting in their higher susceptibility to coccidiosis. Among management and animal health practices, floor type and treatment of cattle influence the prevalence of Eimeria in cattle.

Abstract

Penelitian ini bertujuan mengkaji efisiensi pencernaan kerbau rawa dengan mengidentifikasi jenis dan distribusi glikokonjugat  pada daerah abomasum kerbau rawa. Enam ekor kerbau rawa jantan 2,5 tahun dan berat badan 300-400 kg digunakan dalam penelitian ini. Sampel diperoleh dari rumah potong hewan (RPH) Kabupaten Banjar, Kalimantan Selatan. Setiap bagian abomasum meliputi kardiak, fundus, dan pilorus diambil untuk pengamatan mikroskopis dengan pewarnaan hematoksilin-eosin (HE) dan alcian blue-periodic acid schiff (AB-PAS). Residu gula glikokonjugat pada abomasum dideteksi dengan pewarnaan histokimia lektin dengan menggunakan wheat germ agglutinin (WGA), ricinus communis agglutinin (RCA), concanavalin agglutinin (Con A), ulex europaeus agglutinin (UEA), dan soybean agglutinin (SBA). Hasil penelitian menunjukkan bahwa daerah kardiak mengandung glikokonjugat D manosa/D glukosa, D galaktosa, dan N asetilglukosamin.  Daerah fundus mengandung D manosa/D glukosa, D galaktosa, L fukosa, N asetilglukosamin, dan N asetilgalaktosamin. Daerah pilorus mengandung glikokonjugat L fukosa dan N asetilglukosamin. Pola reaktivitas daerah kardiak, fundus, dan pilorus kerbau rawa terhadap pewarnaan histokimia lektin memiliki pola yang berbeda dengan ruminansia lain. Jenis glikokonjugat yang dimiliki oleh kerbau rawa tersebut diduga berkaitan dengan fungsi peningkatan kemampuan efisiensi pencernaan kerbau rawa. Setiap bagian abomasum kerbau rawa memiliki jenis glikokonjugat yang spesifik dengan pola distribusi khas sesuai dengan fungsinya.

Abstract

This study was conducted to explore the potential of adjuvant for the production of immunoglobulin Y (IgY) as antistreptococcosis in layer chicken with mass production orientation. Enterococcus faecalis which causes streptococcosis in the red tilapia was selected as a candidateantigen. The production of immunoglobulin Y (IgY) was carried out on Isa Brown layer chickens and aged around 20 weeks. Furthermore, thechickens were grouped into four groups (A, B, C, and D groups), each consisting of three chickens based on the type of adjuvant, while twochickens were used as a control group. Each group was treated by giving MONTANIDE™ ISA 71R VG adjuvant (A), Freund's adjuvant (B), aluminum potassium sulphate adjuvant (KAl(SO4)2∙12H2O) concentration of 50 ppm in pH 7 (C), and only antigens without adjuvant (D). Chickens were kept for 35 days and each week was checked for presence the IgY antigen in the serum and egg yolk. Booster was conducted on 14th and 28th days of maintenance. The results showed that IgY in treatment group A was detected on day 28 in the serum and day 35 in the yolk. Whereas the treatment group B could be detected on day 35 in the serum. However, the IgY was not detected in the serum and yolk in C, D, and control groups until the end of the maintenance. Based on the results, it can be concluded that the appearance of IgY in serum and yolk in a relatively fast time is obtained in the combination of Enterococcus faecalis antigen with the emulsion of water-in-oil adjuvant (SEPPICMONTANIDE™ ISA 71R VG) compared to the other types of adjuvant that use in this study.

Abstract

The objective of this study was to evaluate the effect of storage time of epididymis at 5 oC on spermatozoa quality of Aceh local goat. This research was conducted at Reproduction Laboratory, FKH Unsyiah from May to June 2009. Nine epididymis from local goat at the age of 1.5-2.0 years were used in this study. The epididymis were collected from abattoir in Banda Aceh. The experimental design used in this study was completely randomized design with 3 treatments and 3 replications. Spermatozoa quality which consist of sperm concentration, motility, amount of life sperm, and sperm abnormality were examined after the collection of epididymis on day 0 (H-0), day 1 (H-1), and day 3 (H-3) post storage at 5 oC. Data were analyzed using one-way analyses of variance (ANOVA). The result showed that no significant difference (P0.05) seen in the spermatozoa quality after different storage time. The average of spermatozoa concentration on H-0, H-1, and H-3 were 318x107/ml, 282x107/ml, and 241x107/ml respectively. On the average, the percentage of spermatozoa motility on H-0, H-1, and H-3 were 82,27±2,75; 80,25±2,83; and 78,07±0,92%, respectively. Life spermatozoa observed on H-0, H-1, and H-3 were 82,29±2,71; 80,63±1,87; and 80,09±3,31%, respectively. Observation on spermatozoa abnormality showed that the average of spermatozoa abnormality on H-0, H-1, and H-3 were 7,23±0,27; 8,21±0,55; and 10,75±3,14%, respectively. It could be concluded that the spermatozoa quality were not affected by the storage time at temperature of 5 oC.
