Abstract

Aging is a natural process that occurs in every living organism, including humans. Skin wrinkle, caused by deterioration activity of the elastase enzyme, is one example of the aging process. The activity of the elastase enzyme could be inhibited by flavonoid and phenolic compounds. Zodia (Evodia suaveolens) from the family of Rutaceae is one of the indigenous Indonesian plants widely known as rich in flavonoid and phenolic compounds. The purpose of this study is to explore antioxidant and anti-aging activity from ethanolic extract of fruit and leaf Zodia. The extracts were prepared by refluxing fruit and leaf zodia in 96% ethanol. The obtained extracts were characterized for the phytochemical constituent and its antioxidant activity determined using the 2,2-diphenyl-1-picrylhydrazyl method as well as anti-elastase activity for anti-aging evaluation. Phytochemical analysis of both samples showed the presence of flavonoids, saponins, tannins, steroids and essential oils. The calculation of IC50 in antioxidant activity using the DPPH method showed that ethanolic extract of zodia leaf (264.15 ppm) was higher compared to fruit extract (121.59 ppm). The fruit extract was further tested for anti elastase activity with IC50 of 145.67 ppm. In conclusion, the presence of phytochemicals in zodia fruit may be responsible for the antioxidant and anti-aging activities.

Abstract

Allium cepa L. (onion bulb) is a cultivated onion extensively and types of onions that are often used for various cuisines in Indonesia. The onion bulb was efficient in reducing blood fat levels and diuretics. This study aims to complete the onion monograph with determine the physicochemical characteristics and antioxidant activity. Microscopic results obtained include: has fibrous roots and it’s white, and about 9.5 cm long. The stem is pseudo, and the water is whitish-green, cylindrical leaves are dark green, elongated like a pipe and hollow with a length of ± 20 cm, and the tip is tapered. Onion tubers are single layer bulbs, having a diameter of 6 mm, which is higher than onions. Microscopic results contained identification fragments, including hair covering and transporting beams with thickening stairs and spirals. From the results of research on ethanol extract, 70% Allium cepa L. shows a drying shrinkage of 9.69%, 5.16% total ash content, 0.07 % acid insoluble acid dust, 14.36% water-soluble extract, and soluble ethanol extracts 23.04%. Phytochemical screening contains flavonoids, saponins, phenols, dan triterpenoids. Total flavonoid content contained in ethanol extract of onion tubers was 1.48 ± 0.12 mg QE/g. Total phenolic concentrations obtained in onion tuber ethanol extract were 103.47 ± 3.09 mg GAE/g. The antioxidant activity of ethanol extract of onion tubers by the DPPH method received IC50 is 65.31 ppm.

Abstract

National data shows the prevalence of coronary heart disease is 3,6%. The prevalence of dyslipidemia is around 35,9%. Dyslipidemia has a causal relationship with cardiovascular disease. Based on previous research, 70% ethanolic extract of Ridge Gourd has been shown to have antihyperlipidemic activity. This study aimed to study the activity of 70% ethanolic extract fraction of Ridge Gourd in decreasing the levels of total cholesterol, low-density lipoprotein (LDL-C), and triglycerides in hyperlipidemic hamsters. A total of 32 hamster were divided into 8 groups. Negative control, normal control, positive control (using atorvastatin dose 1,24 mg/kg body weight), positive control (fenofibrate dose 12,35 mg/kg body weight), ethyl acetate fraction, n-hexane fraction, water fraction (fraction doses 36,75 mg/kg body weight), and ethanol extract group (extract dose of 240 mg/kg body weight). The results showed that fraction group (n-hexane, ethyl acetate, water) and ethanol extract group experienced a decrease in total cholesterol, LDL-C, and triglycerides levels. Statistical analysis with one-way ANOVA showed an effect on the treatment given (p = 0,005). Tukey's test showed that ethyl acetate fraction group had an effect comparable to positive control with a decrease in total cholesterol, LDL-C, and triglycerides level by 56,03%, 52,14%, and 59,51% consecutively. Ethyl acetate fraction (doses 36,75 mg/kg body weight) was the most effective fraction in reducing total cholesterol, LDL-C, and triglycerides.

Abstract

Erythrina variegata is one of the plants that have efficacy as thrombolytic agent. Isoflavonoid compound has the potential to accelerate the changes of plasminogen which plays important role in the forming of fibrin, that causes the formation of fibrin clots that can clog the arteries thus causing myocardial infarction. This study aimed to look at the activity of isoflavonoid compounds from Erythrina variegata bark which are expected to be candidates for thrombolytic drugs. This research aims at looking for the interaction of isoflavonoid compounds of bark  which is expected can be thrombolytic drug candidates. The method used in this research was the molecular docking with Autodock Vina and PyMOL software. The results showed that the value of ΔG binding affinity as ligands standard streptokinase -8.4 kcal/mol and isoflavonoid derivatives that have ΔG binding affinity value was lowest for the folitenol -11.5 kcal / mol and orientanol C -11.4 kcal/mol. Can be concluded that isoflavonoid derivatives in Erythrina variegata have better binding potential compared to streptokinase as standard ligands.

Abstract

Some studies state that curcumin analog compounds can improve the bioavailability and biological activity of curcumin. One of the methods to predict the bioactivity of curcumin was computational using molecular docking method. This study has done bioactivity tests of curcumin analog compounds as antiviral using the molecular docking method with the software used are PLANTS, YASARA, MarvinSketch, and Pymol for visualization. This study used analog curcumin compounds derived from previous research. This study used five different viral reseptor types. The maraviroc, docosanol, ribavirin, and zanamivir were used as compound control in this research. The validated target protein consists of 5 (five) receptors with PDB codes 1V2I, 4WEG, 2HWI, 2QAD, and 3ALP. Based on this research, compounds that are predicted active as antiviral on each receptors that are: 2,5-bis(3,5-ditertbutyl-4-hydroxy benzyl)cyclopentanone (1V2I), 1,7- diphenyl-1,6-heptadiene-3,5-dione (4WEG), 1,7-bis(3,4-dibenzyloxiphenyl)-1,6-heptadiene-3,5-dione (2HWI), and 2,5-bis(3,5-ditertbutyl-4-hydroxybenzyl)cyclopentanone (3ALP).

Abstract

Antioxidant prevents oxidative stress. One of the antioxidant sources is plant. Red and white galangal are herbal plants that are easy to grow and they may have antioxidant activity. The aim of this study was to compare antioxidant activity and phytochemistry screening of red galangal (Alpinia purpurata (Vieill.) K. Schum) and white galangal (Alpinia galanga (L.) Willd.). The method of this study used was the DPPH method and qualitative analysis. The results of this study showed that the value of antioxidant activity of red galangal was significantly higher than white galangal (p<0.05). The results of phytochemistry screening showed that red galangal contained alkaloids, flavonoids, phenolics, tannins, saponins, and triterpenoids. While white galangal contained alkaloids, flavonoids, phenolics, tannins, and triterpenoids. It is can be concluded that red galangal is more effective than white galangal.

Abstract

Seaweed (EucheumaCottonii J. Agardh) is one of the biological resources found in coastal and marine areas. Seaweed contains secondary metabolites such as flavonoids, alkaloids, polyphenols and tannins which can reduce cholesterol levels in the blood. This study aims to determine the effect of seaweed ethanol extract on reducing cholesterol and obesity levels by comparing plasma cholesterol levels, body weight, and weight of vital organs (liver, kidneys and jantumg) of obese rats, mice given seaweed ethanol extract with normal mice . Rats were grouped randomly into 6 groups. group one was given 0.5% CMC Na, group two was given high fat diet food, group three was given simvastatin, group four was given seaweed ethanol extract 100 mg / kgbb, group five was given ethanol extract of seaweed 200 mg / kgbb and group six was given ethanol extract of seaweed 300 mg / kg with a long suspension of extract for 14 days. Measurement of cholesterol levels in the blood using the CHOD-PAP method, body weight and organs carried out by weighing. The results showed that seaweed ethanol extract has the effect of inhibiting an increase in plasma cholesterol levels and has anti-obesity potential at a dose of 300 mg / kgbb.

Abstract

Piper retrofractum Vahl. (java chili or cabe jawa) and Piper nigrum L. (black pepper or lada hitam) are parts of the Piperaceae. One of the chemical constituents found in the fruit of the two plants is piperine, which has properties as antimicrobial, antidiabetic, anti-inflammatory, anticancer etc. This study aims to determine the piperine levels contained in 95% ethanol extract of java chili and black pepper fruits from regions with different altitude using UV-Visible spectrophotometer. Each sample were extracted by maceration method using 95% ethanol. Qualitative identification of piperine was carried out using thin layer chromatography (TLC). Determination of piperine levels was carried out with UV-Visible spectrophotometer at maximum wavelength of 253.8 nm. The results showed that the levels of piperine contained in the 95% ethanol extract of java chili fruit from low altitude (Lampung), medium altitude (Madura) and high altitude (Bogor) are 1.54&plusmn;0.02%; 1.44&plusmn;0.02%; and 1.41&plusmn;0.02% (w/v), respectively. Whereas the levels of piperine contained in the 95% ethanol extract of black pepper fruit from low altitude (East Luwu), medium altitude (Central Lampung) and high altitude (Bogor) are 5.63&plusmn;0.14%; 5.38&plusmn;0.05%; and 4.87&plusmn;0.09% (w/v), respectively.

Abstract

Daun keji beling (Sericocalyx crispus (L.) Bremek) [Sinonim Strobilanthes crispa (L.)] merupakan salah satu tanaman yang secara empiris digunakan oleh masyarakat untuk membantu pengobatan. Menurut penelitian daun keji beling diketahui mempunyai khasiat sebagai antidiabetes. Penelitian ini bertujuan untuk mengidentifikasi golongan senyawa yang terkandung dalam ekstrak, mutu ekstrak dan aktivitas penghambatan enzim α-Glukosidase secara in vitro. Serbuk simplisia diekstraksi dengan pelarut etanol 70% secara maserasi kinetik, kemudian dilakukan penapisan fitokimia, penetapan mutu ekstrak dan uji aktivitas penghambatan enzim α-Glukosidase. Hasil penapisan menunjukkan ekstrak mengandung senyawa flavonoid, saponin, steroid dan triterpenoid. Hasil penetapan mutu ekstrak menunjukkan bentuk ekstrak kental berwarna hijau kehitaman; rasa pahit; kadar sari larut air 60,46%; kadar sari larut etanol 73,45%; kadar abu total 15,06%; kadar abu tidak larut asam 3,10%; kadar abu larut air 11,29%; kadar air 7,68%; susut pengeringan 9,25%; sisa pelarut 0,38%; kadar Pb 0,4941 bpj; kadar Cd 0,0222 bpj; Angka Lempeng Total (ALT) 4,52x102 koloni/g; Angka Kapang Kamir (AKK) terlalu sedikit untuk dihitung (TSUD); dan kadar flavonoid total 2,39%. Hasil uji aktivitas penghambatan enzim α-Glukosidase pada akarbose dan ekstrak kental keji beling menunjukkan IC50 masing-masing sebesar 50 bpj dan 86,2 bpj. Dapat disimpulkan ekstrak kental daun keji beling memenuhi persyaratan mutu ekstrak dan memiliki aktivitas sebagai penghambat enzim α-Glukosidase. Keji beling memiliki potensi untuk dikembangkan lebih lanjut sebagai obat herbal antidiabetes.

Abstract

Daun pepaya mengandung senyawa alkaloid, flavonoid, saponin, dan tanin yang diduga memiliki efek antifertilitas. Tujuan penelitian ini adalah untuk mengetahui efek antifertilitas dari ekstrak daun pepaya dengan parameter kualitas sperma serta bobot testis dan vesikula seminalis pada tikus putih jantan galur Sprague Dawley. Penelitian ini dibagi menjadi 2 tahap yaitu ekstraksi dan uji antifertilitas. Ekstraksi dilakukan dengan metoda maserasi bertingkat menggunakan pelarut etanol, etil asetat dan n-heksana. Perlakuan uji antifertilitas terbagi menjadi delapan kelompok yaitu 2 kelompok kontrol dan 6 kelompok perlakuan (ekstrak etanol 100 dan 150 mg/kg BB, ekstrak etil asetat 100 dan 150 mg/kg BB, ekstrak n-heksana 100 dan 150 mg/kg BB). Ekstrak daun pepaya diberikan secara oral selama 20 hari. Pada hari ke-21 tikus dibedah dan dianalisa kualitas spermatozoa serta diukur bobot testis dan vesikula seminalis. Data yang didapat diuji secara statistik dengan ANOVA satu arah dan dilanjutkan dengan uji Tukey HSD. Hasil penelitian menunjukkan ekstrak n-heksana, ekstrak etil asetat dan ekstrak etanol 70% daun pepaya dapat menurunkan jumlah dan viabilitas spermatozoa serta menurunkan bobot testis dan vesikula seminalis. Ekstrak yang paling baik dalam mempengaruhi efek antifertilitas adalah ekstrak etanol 70% dosis 150 mg/kgBB.
