Abstract

The objective of this study was to determine the effectiveness of palmyra juice in maintaining motility,viability, and longevity of bali cattle sperm during in vitro storage. Semen collected from a four year oldbali bull had been trained to serve an artificial vagina. Good quality semen divided into four tubes to betreated with: without extender (negative control), citrate-egg yolk (citrate-EY) and coconut water-egg yolk(CW-EY) as positive control; and palmyra juice-egg yolk (PJ-EY), and then stored in a refrigerator attemperature 3 - 8oC. Each treatment was repeated five times to form 20 experimental units. The resultsshowed that up to the fourth day of storage, sperm preserved in PJ-EY had motility 44%, higher (P<0.05)than citrate-EY, CW-EY, and without extender, i.e 30%, 18%, and 0%, respectively. PJ-EY extender alsoyield sperm viability 60%, higher (P<0.05) than citrate-EY, CW-EY, and without extender i.e 50%, 32% and0%, respectively. Sperm livability achieve 11.20 days in PJ-EY, longer (P <0.05) than citrate-EY, CW-EYand without extender, i.e 9.60 days, 7.60 days , and 1.31 days, respectively. In conclusion that palmyrajuice-egg yolk extender effective to maintain motility, viability, and longevity of bali cattle sperm duringin vitro preservation.

Abstract

Calliandra calothyrsus leaf contains 17-28% protein which is hihly potential for use as source of protein supplement for animal feed. However, the plants also contain high level (>10%) of condensed tannins as antinutritional effects which may reduce consumtion efficiency of diet. The addition of protease (bromelain) into feed containing tannin is expected to decrease the negative effects of tannins. Bromelain can be obtained from the pineapple plant (Ananas comosus) including on the peels. This study was conducted to determine the effect of calliandra tannin combined with bromelain protease of pineapple peels in the feed on testicular histology and testosterone level of rats feeded during the growth period. The feeding experiment on post-weaning male rats was conducted using a completely randomized factorial design (4 main factors x 4 subfactors). The main factors were calliandra leaf meal substitution of 0; 10; 17.5 and 25% in the diets and subfactors were addition of pineapple peels, 0; 4.35; 8.70 and 13.05 g/rat/day. Rats were divided into 16 groups and were feeded the diets for two months (during the growth period). The results showed the susbtituion of feed with calliandra leaf had no sigbnificant effecton the thickness of the seminiferous tubules, but it appeared to have significant effect on the histology of seminiferous tubules. Increased pineapple peels level in the diets containing calliandra decreased wall thickness of the seminiferous tubules of the rat testes, indicating that bromalein fastened the maturation of spermatozoa. The addition of pineapple peels into the diet containing calliandra had a significant interaction effect on testoteron levels of male rats, but the testoteron levels among all treated rats were still within the normal range. ABSTRAK Daun kaliandra (Calliandra calothyrsus) mengandung protein 17-28% sehingga potensial sebagai sumber protein pakan ternak, namun tanaman ini mengandung condensed tannin cukup tinggi (>10%) yang bersifat antinutrisi. Penambahan enzim protease (bromelin) pada pakan yang mengandung tanin diharapkan dapat mengatasi dampak negatif tanin. Bromelin dapat diperoleh dari tanaman nanas (Ananas comosus) termasuk dari kulit buahnya. Penelitian ini bertujuan untuk mengetahui pengaruh tanin daun kaliandra dalam pakan yang dikombinasikan dengan protease bromelin dari limbah kulit nanas terhadap histologi testis dan kadar hormon testosteron tikus jika diberikan pada masa pertumbuhan. Penelitian ini merupakan percobaan pakan pada tikus jantan pascasapih selama masa pertumbuhan dengan Rancangan Acak Lengkap pola faktorial berjenjang 4x4 dengan level substitusi tepung daun kaliandra 0; 10; 17,5 dan 25% dalam ransum (main factor) dan dosis aditif kulit nanas 0; 4,35; 8,70 dan 13,05 g/ekor/hari (sub factor). Tikus dibagi menjadi 16 unit percobaan dan diberi perlakuan ransum selama dua bulan (masa pertumbuhan). Hasil penelitian menunjukkan bahwa tebal dinding tubulus seminiferus testis tikus tidak berbeda nyata, namun tampak adanya perbedaan pada gambaran histologi yang meliputi struktur membran basal, susunan antar sel-sel spermatogenik pada dinding tubulus, serta gambaran sel-sel spermatozoa di dalam lumen tubulus. Peningkatan dosis kulit nanas dalam ransum yang mengandung kaliandra menurunkan tebal dinding tubulus seminiferus. Aditif kulit nanas ke dalam ransum yang mengandung kaliandra menunjukkan interaksi yang memengaruhi kadar hormon reproduksi dengan menurunkan kadar hormon testosteron tikus jantan walaupun masih dalam kisaran normal.

Abstract

Trichinellosisis is a zoonotic disease caused by infection of Trichinella spp. and is found world-wide.The seroprevalence of trichinellosis in pig population has never been reported in East Nusa TenggaraProvince particularly Kupang City. The aim of this study was to determine the seroprevalence oftrichinellosis in pig at Kupang City, East Nusa Tenggara. A cross-sectional study was conducted byexamining 376 blood samples of pigs collected from slaughterhouses in Oeba, Kupang using simplerandom sampling method. Blood samples were tested by indirect enzyme linked immunosorbent assay(ELISA) using Screen®Trichinella indirect multi-species ELISA kit. Test results showed that 3 (0.8%)serum samples were positive of trichinella antibody. In conclusion ,some pigs in the city of Kupang hasbeen infected by Trichinella sp. and could be a threat for human health.

Abstract

The most common therapy on men who suffered fertility decline due to aging was called “T Therapy”, but that’s therapy has long-term risks of sexual dysfunction, metabolic syndrome, prostate, and cardiovascular system. Stem cells are an alternative therapy can be used for ameliorate testicular fuction because of their ability to differentiate into various cell types. The aim of this study was to evaluate the injection of hWJ-MSC in physiologic aging male rats on testicular function. This study was used 3 young male rats (8-12 weeks) and 6 physiological aging male rats (22-24 months) which divided into 3 groups, (i) the young rats, (ii) physiological aging male rats, and (iii) physiological aging male rats that injected with hWJ-MSCs. The young rat group did not give any treatment, physiological aging male rats received NaCl (0.9%) 0.4 mL, and the treatment group received 1x106 cells/kg BW of hWJ-MSCs. The observations were performed on the macroscopical and histological analysis. The result indicates that the younger group had the lowest body weight (154.6 g) and the percentage of the testis weight on the body weight was highest (2.2%) compared to the other groups (P>0.05). The physiological aging rats group had the smallest tubule (9726.9 ìm2) with a largest interstitial area (1117.1 ìm2) compared the other groups (P>0.05). After injection of hWJ-MSC, the tubule area became wider followed by narrowing of the interstitial area (P>0.05). The difference in the body weights is due to the different age of the rat. Improvement of tubule area and interstitial area due to the ability of hWJ-MSCs to improve spermatogenic cells within the tubule. Injection of hWJ-MSCs has been shown to increase fertility in aging rats.

Abstract

Vector infectious diseases such as Dengue Hemorrhagic Fever (DHF) are still a major health problem in Indonesia including Cirebon City. Cirebon seaport as one of the country’s entrances has the potential to transmit dengue disease. The aim of the study was to measure the population density of Aedes aegypti larvae and analyze the relationship of habitat characteristics with the presence of Ae. aegypti larvae in the perimeter region of Cirebon seaport. Analytic descriptive observation with a cross sectional study approach was conducted in the study. The sample were all the houses/ buildings in the perimeter region of Cirebon seaport are used as breeding habitats for Ae. aegypti. The results of the study, the types of positive containers of Ae. aegypti larvae are water closet (WC) tanks, bathtubs, drums, pails, ponds and used tires. Calculation results of the House Index (HI), Container Index (CI), Breteau Index (BI) and Density Figure (DF) (HI: 8.74%; CI: 9.05%; BI: 12.02; DF: 3). Based on chi square test, habitat characteristics that have a significant relationship with the presence of Ae. aegypti larvae were container type, container material, water volume, water source, water pH and air temperature (p<0.05). The results of the analysis with binary logistic regression had a significant effect on the presence of Ae. aegypti larvae, namely in pails (p=0.003; OR=18.972) and water pH (p=0.022; OR=2.489). Conclusion of the study is Cirebon seaport area has a risk of dengue transmission at a moderate level based on its vector density.

Abstract

Probiotics is a live microbial feed supplements that improve the microbial balance in the digestivetract of cattle host. Temulawak (Curcuma xanthorrhiza Roxb) is one of native Indonesian spices that areuseful for prevention and treatment of human disease but also can be used for broiler chickens supplement.Bioactive contents known acting as an anti-bacterial, anti-inflammatory, and anti-fungal. The study wasconducted to determine the effects of probiotic and temulawak supplementation on population of Salmonellasp and broiler chickens blood cholesterol. This study used 300 DOC final stock. Probiotic concentration 109cell/mL administered through drinking water every day while of temulawak extract dose 0.25 g/L 0.50 g/Lonce a week, for five weeks. The variables measured were Salmonella sp populations and blood cholesterollevels. The results showed that combination administered of probiotics with temulawak dose 0.50 g/Ldecreased of population of Salmonella amount to 87.04% and decreased of blood cholesterol amount21,48% at administered of probiotics with temulawak dose 0,25 g/L. The conclusions of this study showedthat the percentage decrease of Salmonella sp population and blood cholesterol levels in combinationprobiotic and temulawak supplementation so that broiler produced is safe for consumption.

Abstract

The information on pathogenicity and adaptation factors of avian influenza virus (AIV) in mammalsis very inportant in an effort to reduce the risk of avian influenza (AI) pandemic in the future. Polymerasegene complex appears to be the major factors for adaptation of AIV to certain animal species. A preliminarystudy on role of non-coding region (NCR) and amino-terminus of polymerase-basic 2 (PB2) is presented.Purified viral RNA of AIV isolated from chicken, duck, pig, and quail of Bali and Yogyakarta was reversetranscribed into cDNA and amplified using reverse transcriptase-polymerase chain reaction (RT-PCR)using PB-2 universal forward primer and specifically designed backward primer. The result showed thatall AIV’s H5N1 isolated from chicken, duck, quail, and pig, posed PB2 amino-terminus typical for IndonesianAIV H5N1. However, polymorphic amino acids of the protein fragment did not show any species specificmotive, with the exception of the pig isolate Sw/Tabanan/2006 which had specific substitution of D16E,H17Q, M40I, and H124Y.

Abstract

In a conservation effort, the identification of Tarsier species, on the bases of the morphological andmolecular characteristic is necessary. Up to now, the identification of the animals were based on themorphology and vocalizations, which is extremely difficult to identify each, tarsier species. The objective ofthis research was to study the genetic diversity on ATP6 gene of Tarsius sp. Based on sequencing of PCRproduct using primer ATP6F and ATP6R with 681 nts. PCR product. The sequence of ATP6 fragmentswere aligned with other primates from Gene bank with aid of software Clustal W, and were analyzed usingMEGA program version 4.0. Three different nucleotide sites were found (nucleotide no. 288, 321 and 367).The genetic distance based on nucleotide ATP6 sequence calculated using Kimura 2-parameter modelindicated that the smallest genetic distance 0%, biggest 0.8% and average 0, 2%. The phylogenetic treeusing neighbor joining method based on the sequence of nucleotide ATP6 gene could not be used todifferentiate among T. Dianae (from Central Sulawesi), T. Spectrum (from North Sulawesi), T. bancanus(from lampung, South Sumatera) and T.bancanus from West Kalimantan.

Abstract

The objectives of this study were to identify risk factors associated with historical of animals contact and animalproducts on the incident of cutaneous anthrax in humans in Bogor District. The research designed used in this study CaseControl of Observational Epidemiology. Patients of cutaneous anthrax disease record in Puskesmas (Center for HealthServices) were used as a case. Inhabitants in Bogor district living in the same area with patients of cutaneous anthrax andnot showing clinical signs of cutaneous anthrax. The data were collected by structured interviews and direct observations.Data analysis was carried out in three steps, consisting univariate for analysis of frequency distribution, bivariate withChi-square and also multivariate analysis for prediction model of logistic regression. All analysis processed by SPSS 13.0.It can be concluded that the first risk factor associated with the occurrence of cutaneus anthrax was holding susceptibleanimals with Odds Ratio (OR) of 6.648 (95% Confidence Interval (CI) = 2,914-15,167), the second risk factor was meathandling with OR of 5.318 (95% CI: 1,801-15,702). It showed that for people who live in endemic area of anthrax, holdingsusceptible animals sixth times more likely get cutaneous anthrax.
