Abstract

The aim of this study was to investigate effects of Spirulina phycocyanin on the total  blood cell count,phagocytic activity, and growth of humpback grouper fish, Cromileptes altivelis juvenil.  Fishes were fedwith a diet containing   0, 150, 250, 350 dan 450 mg  phycocyanin per kg diet for four weeks and eachtreatment was triplicates.  Initial body weight  of  grouper was  8.46 ± 0.22 g with a density of 10 fish per56 litre volume. The total count of  erythrocytes and leucocytes increased until the fourth week of rearingperiod. The highest of total erythrocyte and leucocytes were observed in fish treated with 150 mg phycocyaninper kg diet ( 13.17 x  105 cells/mm3 and 8.93 x 105 cells/mm3 respectively) which were not significantlydifferent (P>0.05) to those treated with 250 mg phycocyanin per kg diet. The total leucocytes and phagocyticactivity of fish fed diet containing  250 mg phycocyanin  per kg diet (8.49 x 105 cells/mm3 and 59.67%respectively) were significantly higher  (P <0.05) to those of control group. The highest of final weight(Wt=14.32 g) and weight growth (G=5.89g) and lowest of feed conversion ratio (FCR=1.13) were obtainedin fish treated with  250 mg phycocyanin per kg diet which were  significantly  higher  (P <0.05) than thosefed control diet. The data showed that  the addition of  phycocyanin 250 mg/kg diet enhances the totalleukocyte count, phagocytic activity and the growth of humpback grouper juvenil.

Abstract

The aims of this study were (1) to analyze the phenotypic performance of Ettawa Grade (EG) goat; (2)to estimate the heritability of birth weight (BW), weaning weight (WW), yearling weight (YW), and geneticcorrelation between two body weights on the third different period; and (3) to analyze the variance andcovariance component of body weight. The material used were the exiting records of 437 EG goats in BalaiPembibitan Ternak Unggul dan Hijauan Pakan Ternak Pelaihari, South Kalimantan. These goats originatedfrom the crossing between 19 males and 216 females from periods of 2009 - 2012. Nested Design methodwas used to etimate the phenotypic correlation, heritability and genetic correlation. Variance componentswere determined from heritability estimation, while covariance components were determined from geneticcerrelation estimation. Phenotypic correlation between BW and WW, between BW and YW, and betweenWW and YW were 0.19 (low); 0.31 (medium); 0.65 (high); respectively. Heritability of BW, WW, and YW were0.43±0.23 (high); WW 0.27±0.19 (medium); and YW 1.01±0.38 (excludeof the h2 value), respectively.Genetic correlation between BW and WW, between BW and YW, and between WW and YW were -0.04(negative low); 0.49 (positive medium); and -0.41 (negative medium), respectively. Variance components ofbuck, ewes, and kid for BW were 10.76%; 37.16%; and 52.09%, respectively, for WW were 6.67%; 38.52%;and 54.81%, respectively, and for YW were 25.15%; 58.37%; and 16.43%, respectively. Covariancecomponents of buck, ewes, and kid between BW and WW were -3.91%; 66.45%; and 37.46%, respectively,between BW and YW were 65.68%; 16.50%; and 17.82, and between WW and YW were -5.14%; 83.87%; and21.28%, respectively. In conclusions variance component of ewes and kid were high in body weight at birthand weaning time. Therefore, selection should be conducted for body weight at birth and weaning time.

Abstract

Bovine cysticercosis is one of the animal and public health problems throughout the world. Besides this disease has a negative impact on public health, also causing economic losses for farmers due to heavy infected carcasses should be rejected. Until now the availability of data related to this parasitic disease, especially in the Nusa Tenggara region is very limited.The purpose of this study was to obtain information on the prevalence and distribution of bovine cysticercosis on bali cattle in West Nusa Tenggara, Indonesia. The study was conducted by examining of bali cattle sera obtained from the farmer by ELISA (Bio-X Diagnostics’s Cysticercosis Antigen ELISA Kit). The results of ELISA examination of 92 serum samples with cut off 0.295, shows that there are 4 positive sera (4.35%). Three positive sera are originated from Lombok and one positive sera from Sumbawa. These results indicate that Lombok Island and Sumbawa Island in West Nusa Tenggara are not free from bovine cysticercosis. Since C. bovis infection is zoonotic, studies of risk factors are necessary, as well as the availability of information about the estimated burden of disease and the economic loss of the disease. It is recommended that veterinarians be more accurate when conducting post mortum examination, especially on the cattle which comes from a positive area.

Abstract

The aim of this study were to evaluate the effect of tannin concentration in protein protection ofsoybean meal and its influence on the change of body composition with urea space method (estimation)and cutting (direct). Sixteen male thin tail rams of eight month old with an average body weight of 11.81±1.65kg were used in this study. Treatment Groups of different tannin level were allotted according to a completelyrandomized design with four replicates for each treatment. The treatmen groups were T0 (complete feed +soybean meal not protected), T1 (complete feed + tannins protected soybean meal 0.5%), T2 (complete feed+ tannins protected soybean meal 1%), and T3 (complete feed + tannins protected soybean meal 1.5%).The treatment was performed in 10 weeks. However, In the week 0, fifth and tenth the rams body compositionwere measured with urea space technique. Result of the study showed that the feeding protected proteinwith different tannin levels did not significantly influence (P> 0.05) body composition of the samples. Theaverage content of body composition measured with the urea space method for body water, body proteinand body fat were 58.53%, 11.01%, and 20.74% respectively. Where as, the body composition measurementafter slaughtered for the body water, body protein and body fat were 3.08 kg, 0.50 kg and 0.89 kg respectively.The average increase of body protein and body fat that occurred during the 10-week study period were 0.37kg and 0.51 kg respectively. Based on these results it can be concluded that feeding protected proteincontained with tannin up to 1.5% may not be able to improve body composition of thin tail rams.

Abstract

Obesity and dyslipidemia are health problems that can increase the risk of cardiovascular disease. This study aims to analyze the potency of antiobesitas and antidislipidemia of jamu galian singset (JGS). The research method used complete randomized design with 8 treatment groups and 4 replications as follows: P1 (Standard Feed + Aquadest), P2 (Standard Feed + 200 mg Orlistat / kg BW), P3 (Standard Feed + 176,4mg JGS / 200g BW) P4 (Standard Feed + 352,8mg JGS / 200 g BW), P5 (High Fat Feed + Aquadest), P6 (High Fat Feed + 200 mg Orlistat / kg BW), P7 (High Fat Feed + 176,4 mg JGS / 200 g BW) and P8 (High Fat Feed + 352,8 mg JGS / 200 g BW). The parameters observed were feed intake, body weight gain, body weight gain ratio, fat intake, fat absorption, fat digestibility, feces fat, abdominal fat, muscle fat, body weight, body fat percentage and lipid profile, include Cholesterol total (TC), triglycerides (TG), HDL and LDL. The results showed that JGS significantly (P <0,05) decreased the body weight gain of the rat, the ratio of body weight gain per feed intake and the weight of abdominal fat. Giving JGS increases the absorption of fat, but not found high fat in the feces. Jamu galian singset has no effect (P> 0.05) on rat lipid profile. Baced on the results of this study it can be concluded that JGS can be used as antiobesity and more effectively decrease body weight gain on high fat diet than standard feeding diet. Jamu galian singset are suspected to have a different working mechanism with orlistat as antiobesitas.

Abstract

Dangke is a traditional food in Enrekang, a district in South Sulawesi. Its made from buffalo’s milk orcow’s milk. Dangke could be contaminated during the process with Escherichia coli which causes diarrheain children and adults. It was known that supernatant of Lactobacillus plantarum has antibacterial capacity,and it may be used as a biopreservative agent. The research aims were to determine the minimum inhibitoryconcentration (MIC) of L. plantarum supernatant in inhibiting the growth of E. coli ATCC 25922, determinethe nutrients level of cow’s milk dangke after the addition of 1% and 2% milk fat, as well as determine theeffect of L. plantarum supernatant and cow’s milk fat addition into dangke inhibited the growths of pathogenicbacteria of E. coli ATCC 25922. MIC value was determined based on the value of the lowest concentrationof supernatant that shown with no any bacteria growth in the media. Data of pathogenic bacteria growth analyzed with analysis of variance test with a 2x2 factorial design, which 1st factor was the addition of L.plantarum supernatant (with or without addition of supernatant) and the second factor was the additionof fat content (1% and 2%) and time observation was made on days 0, 2nd, 4th, 6th, and 8th. Resultsshowed that the filtrate of fermented L. plantarum was able to inhibit the growth of E. coli ATCC 25922in vitro and had 10% minimum inhibitory concentration (MIC). Level of fat and protein in dangke whichadded 1% cow’s milk fat, was higher than with 2% cow’s milk fat. The L. plantarum supernatant is provedto be able to inhibit the growth of E. coli ATCC 25922. Therefore, it is potentially used as a naturalbiopreservative agent in making dangke.

Abstract

Streptococcus agalactiae atau yang juga dikenal dengan Streptokokus grup B (SGB) telah diketahui sebagai agen penyebab pneumonia, septikemia dan meningitis neonatal pada manusia dan hewan. Hialuronidase merupakan produk ekstraseluler dari SGB yang menentukan virulensinya. Penelitian ini dilakukan untuk menguji akitvitas hialuronidase dari 10 isolat SGB yang diisolasi dari kasus komplikase obstetri, serta memurnikan dan mengkarakterisasi hialuronidase yang diisolasi dari SGB SV-14 pada substrat asam hialuronat.

Abstract

The purpose of this study was to verify the effectiveness of cumulus cells as a feeder layer in supportingthe growth of mouse embryonic stem cells. Embryos at blastocyst stage were exposed in pronase solution,and then incubated in rabbit anti-mouse antibody and guinea pig complement to lyse and separate thetrophoblast cells from the inner cell mass. Inner cell mass subsequently cultured in a petri dish containinga cumulus feeder layer, mouse embryonic fibroblasts, or without a feeder layer, in stem cells medium. Theresulting stem cells colony passaged in trypsin solution, pipetted repeatedly to produce subcolonies orsingle cells, and cultured as before in some new petri dishes. Characterization of stem cells was identifiedby using alkaline phosphatase staining. The results showed the effectiveness of cumulus cells as feederlayer for culture of mouse embryonic stem cells was comparable with mouse embryonic fibroblasts, andboth of them were better than without a feeder layer method. The number of primary colony, cell lines, andcolony growth rate increased 41.30, 8.70, and 54.20%, respectively, while doubling time was shorter 10:21hours compared to the growth without feeder layer method. These results prove that the cumulus feederlayer effectively supports the growth of mouse embryonic stem cells.

Abstract

This experiment was designed to study the impact of mammary carcinogenesis induced by 7,12-dimetilbenz (?) antracene (DMBA) on histopathological features of the gastric in Sprague Dawley rat.Eight female Sprague Dawley (SD) rats, of four week-old, were used in this study. The rats were randomlydivided into 2 groups of four. Group I were treated orally with 20 mg/kg BW of 7,12-dimetilbenz (?)antracene (DMBA) treatment, twice a week for 5 weeks. Group II were used as negative control withoutDMBA treatment. At 5 week after the end DMBA of, the rats of the both groups were sacrificed and thegastric were fixed in 10% buffer formalin. Subsequently, tissue sections were stained with hematoxylinand eosin (H&E) for microscopic examination. The results of the experiment show that oral administrationof DMBA (20 mg/kg BW) twice a week, for 5 weeks result in epithelial hyperplasia of cardia. Hemorrhagicand inflammation reactions are also found on the mucous membrane of fundic. However, macroscopicexamination shows no specific changes of the gastric. Histopathological changes were suspected as a precancer lesion of the gastric due to chronic exposure of DMBA.

Abstract

The use of antibiotics is one way to reduce the number of deaths in poultry industry. However, use maybe reduced because the impact for consumers. This study aims to find out the effect of the combinationbetween herbal turmeric (Curcuma domestica Val.), Garlic (Allium sativum Linn.) with Zn in the feed tothe number of erythrocyte, hematocrit and hemoglobin in chickens that is infected with Escherichia colibactery. Two hundred head of DOC were divided into 5 treatments and 4 repetitions, each repetitionconsisted of 10 chickens. Ration treatment K0 (basal ration as a healthy chickens), K (negative control)(basal ration chickens infected with Escherichia coli), P1 (basal ration + 1.5% turmeric powder + 180 ppmof ZnO / chickens infected with Escherichia coli), P2 (basal ration + 2.5% garlic powder + 180 ppm of ZnO /chickens infected with Escherichia coli), and K (positive control) (a basal ration + antibiotic / chickensinfected with Escherichia coli). At the age of 3 weeks, all treatment groups were challenged with E. coliwhich treated  orally at a dose of 108 CFU/mL. K(+) group was given antibiotics a day after the challenge. Antibiotics are given for 3 days with a dose of medication. Blood sampling performed at the age of 3 weeks(before the challenge), 1 and 2 weeks after the challenge. The blood test includes the number of erythrocytes,hematocrit values and level of hemoglobin  . The results showed the number of erythrocytes, hematocritvalues, and level of hemoglobin at treatment P1 (ration basal + 1.5% turmeric powder + 180 ppm of ZnO+ infected with Escherichia coli) tend to be higher than other treatments and have equal outcomes withtreatment K(+) (ration basal + antibiotics + infected with Escherichia coli). From the results of this studyconcluded that the use of combinations of herbs turmeric and Zn in the feed can be used as an alternativeof colibacillosis treatment.
