Abstract

Primary lung cancers, or tumors originating in the lung, are relatively uncommon in dogs. The objectiveof this study was to describe the canine lung carcinoma that serially transplanted into severe combinedimmunodeficiency (SCID) mice, in order to established cell line from this tumor cell.  Morphology andcharacteristic of this canine lung carcinoma in SCID mice by histopathological and ultrastructuralexaminations with metastatic lesion in lung were also examined.  Histopathologically, the tumor masswere consisted of cuboidal to columnar cells with papillary pattern, uniform in size, the nuclei were oftenvariable in size, and some cells have vacuole on their cytoplasms.  Glandular forms were predominant withlobulated pattern, ductal pattern with papillary injected into tube-like structure were also encountered.Mitotic figures commonly found with inflammatory reaction were sometimes present in the interstitiumand lumen gland.  Ultrastructural analysis of the tumor cells showed round to oval cells with one or moreprominent nucleoli.  The cells possessed numerous mitochondria, smooth endoplasmic reticulum, andindividual cells which were interconnected via desmosomes.  Tonofilament characterize by long cytoplasmicmaterial was encountered.  Positive reaction of the round to oval tumor cells to anti keratin antibodyconfirmed that their epithelial cell nature.  Lung metastatic lesions were found in SCID mice aftertransplantation and this phenomenon indicated that canine lung carcinoma is tumorigenic to SCID mice.

Abstract

There were three kinds of goat developed in Bali since a long time ago i.e. Gembrong goat, EtawahCrossbred goat, and Kacang goat. Gembrong goat is nearly disappearingrecently due to decrease of itspopulation from year to year. The Gembrong goat has very interesting performance, funny, and this speciesis Balinese native goats. The objective of thisexperiment was to know the blood illustration among thethree speciesof the Balinese native goatwhere in the future it can be used as a guide to develop each ofthem. The experiment was used nine adults goats namely three adult male Gembrong goats, three adultmale Etawah Crossbred goats, and three adult male Kacang Goats respectively. The animal fed dailygreen feed grows around them and additional rice bran. Blood samples of the animals were collected forabout10-15 mL each with venoject, and then was kept in a cooler box and subsequently sent to the ProdiaClinic Laboratory at Denpasar. The results of the experiment showed that blood illustration of the threekinds of goat was not significantly different (P > 0.05) except the triglyseride.

Abstract

Preeclampsia has remained a major cause of morbidity and maternal-perinatal mortality. Oxidative stress that occurs in preeclampsia increases HSP-70 expression. Therefore, VEGF therapy is expected to recover this oxidative stress. This research aimed to determine the role of VEGF-121 recombinant on HSP-70 expression in mice model of preeclampsia. This research was an explanatory study conducted at Animal Cage Experiment, Faculty of Veterinary Medicine, Airlangga University. The samples were 30 mice that were divided into 3 groups, namely a group of 10 normal pregnant mice, a group of 10 mice model of preeclampsia, and a group of 10 mice model of preeclampsia with VEGF-121 recombinant therapy. On the 16th day of gestation, HSP-70 expressions in the placenta of all mice were examined using immunohistochemical methods. The data were analyzed using Kruskall-Wallis test of SPSS program. The mean of HSP-70 expression in normal pregnancy group was 1.69 ± 0.68, in preeclampsia model group was 3.50 ± 0.95 with p = 0.00, and in preeclampsia model group with VEGF-121 recombinant therapy was 2.24 ± 0.84 with p = 0.00. In short, VEGF-121 recombinant has a role in lowering HSP-70 expression in mice placenta model of preeclampsia

Abstract

Jembrana disease is an infectious disease in Bali cattle cause by a member of lentivirus calledjembrana disease virus (JDV). It causes an acute and severe disease syndrome with short incubationperiod. As the disease has spread to several areas in Indonesia, a simple and rapid detection method isrequired. The objective of this study to apply rapid diagnostic method for JDVTabanan 1995 strain basedon Nucleic Acid Sequence Based Amplification (NASBA) methods targeting env-tm gene. The steps of thisresearch consisted of viral RNA isolation from organ and blood of cattle experimentaly infected withJDVTabanan 1995 strain . RNA amplification was conducted by NASBA using waterbath. The NASBAproducts were then separated on 2 % agarose gel. Using this technique JDV positive result was obtainedfrom organ samples such as spleen, liver, lung, prefemoralis lymph node, prescapularis lymph node andblood generating a RNA fragment of 207 bp. In this study, diagnosis method for env tm of JDV Tabanan1995 strain can be conducted by isothermal amplification NASBA.

Abstract

Putak is a local feed in west Timor, East Nusa Tenggara Province. It is obtained from soft core of palm tree which is called gebang tree (Corypha gebanga). Putak contains high carbohydrate and fiber but low in protein. As an effort to increase the ptotein level in putak a research was conducted by fermenting it with mixed-cultured of Trichoderma reesei and Aspergillus niger. A laboratorium experimental method was used and arranged factorially in Completely Randomized Design 3 x 4 x 3 namely three inoculant levels of T. reesei (T): 5,0; 7,5 and 10,0 % (w/w), and four levels of A. niger (A) 0,5; 1,0; 1,5 and 2,0% (w/w). Mixed-cultured was done by: first, T.reesei as treatment mix with putak and incubation for two days, after that mix with A.niger, incubation for two days. The best result of this study was on combination of T.reesei 7.5% level and 1,5% of A.niger were crude protein (23.62%) and soluble protein (14.92%) was highest and fiber 10,17%.

Abstract

The objective of this study was to determine genetic diversity of rabbit by blood plasma protein variance using electrophoresis at loci of pre-albumin (Pa), albumin (Alb), ceruloplasmin (Cp), transferrin (Tf), post-transferrin (P-tf) and amylase-I (Am-I). The blood analysis was performed at the Laboratory of Physiology and Biochemistry, Faculty of Veterinary Medicine, Gadjah Mada University, Yogyakarta. The materials used were 63 blood samples of rex, local and new zealand white (NZW) rabbit, in which the number of sample in each breed was 21, respectively. Materials were taken randomly. The protein bands resulted from electrophoresis were used to calculate the value of the gene frequency. Genetic diversity was determined by using individual heterozygosity (h) and the average heterozygosity (H) formulas. The genetic distance among breeds of rabbits was analyzed by using DISPAN and MEGA 6.06 programs. Chi-square (X2) was used to test of Hardy-Weinberg Law (HWL) equilibrium. The results showed that the loci of prealbumin (Pa), albumin (Alb), ceruloplasmin (Cp), transferrin (Tf), post-transferrin (P-tf), and amylase-I (Am-I) in rex, local and NZW rabbits were polymorphic. Rex and NZW rabbits had close genetic relationships based on phylogeny tree analysis. Chi-square test showed that rex, local, and NZW rabbits were in HWL equilibrium for locus of Pa and Cp, while at the locus of Alb, Tf, P-tf, and Am-I were in HWL unequilibrium. In conclusion, there are genetic variability of six locuses in rex, local, and NZW rabbits. ABSTRAK Tujuan penelitian ini adalah untuk mengetahui keragaman genetik kelinci melalui analisis keragaman protein plasma darah dengan metode elektroforesis pada lokus pre-albumin (Pa), albumin (Alb), ceruloplasmin (Cp), transferin (Tf), post-transferin (P-tf), dan amylase-I (Am-I). Analisis darah dilakukan di Laboratorium Fisiologi dan Biokimia, Fakultas Kedokteran Hewan, Universitas Gadjah Mada, Yogyakarta. Sebanyak 63 sampel darah kelinci rex, lokal dan New Zealand White (NZW) masing-masing sebanyak 21 sampel digunakan dalam penelitian ini. Penentuan kelinci dilakukan dengan metode observasional dengan cara kelinci dipilih secara acak. Pita-pita protein yang dihasilkan dari elektroforesis digunakan untuk menghitung nilai frekuensi gen. Perhitungan nilai ragam genetik ditentukan menggunakan rumus heterosigositas individual (h) dan rataan heterosigositas (H). Hubungan dan jarak genetik antar bangsa kelinci menggunakan program DISPAN dan MEGA 6.06. Analisis statistika menggunakan tabel X2 untuk menguji lokus-lokus dalam keseimbangan Hukum Hardy-Weinberg (HHW). Hasil penelitian menunjukkan bahwa lokus pre-albumin (Pa), albumin (Alb), ceruloplasmin (Cp), transferin (Tf), post-transferin (P-tf), dan amylase-I (Am-I) pada kelinci rex, lokal, dan NZW bersifat polimorfik. Kelinci rex dan NZW memiliki hubungan genetik yang dekat berdasarkan analisis pohon filogeni. Hasil uji X2 pada kelinci rex, lokal, dan NZW menunjukkan keseimbangan HHW pada lokus Pa dan Cp, sedangkan pada lokus Alb, Tf, P-tf, dan Amy-I memiliki keseimbangan HW yang berbeda-beda. Simpulan dari penelitian ini adalah terdapat keragaman genetik enam lokus pada kelinci rex, lokal, dan NZW.

Abstract

The rate and risk factors on protective immunity against rabies were conducted in Makassar city. Theaims of this study were to determine the level of immunity of dog and find out the association betweenmanagement factors and the level of maintenance of the dog immunity against rabies. Two hundred andforty-six serum samples were collected for measurement of dog immunity level with a method of enzymelinked immunosorbent assay (ELISA). Maintenance management data were gathered throughquestionnaires and direct observation. Analysis of data obtained by the level of immunity was donedescriptively. The significance association between the risk factors and the strength of immunity level wascalculated by using Chi square (X2) and odds ratios. The results showed only 12.2% of dogs had a protectiveantibody against rabies at > 0.5 IU/ml. Factors affecting the level of protective immunity against rabieswere the first vaccination at 1-6 months age (OR = 18.6), post-vaccination after 1-6 months (OR = 6.0), themaintenance location of dogs in urban areas (OR = 4.5), dog caring management in house (OR = 3.8), andowner income above Rp. 2.000.000/month (OR = 3.0). It is concluded that the low level of protective immunityagainst rabies and the dog bites reported each year indicates that Makassar, as endemic areal of rabies,is a threat to the outbreak of the disease in animals and humans to the surrounding areas.

Abstract

The objective at this experiment was to study the role of zinc supplementation (Zn) in minimizingstress at parturition. Thirty etawah cross breed age 3-6 years with average body weight 30-50 kg were usedin this study and divided into three groups ; i) animal were given grass + feed concentrate + ZnSO4 40 mg/kg dry matter ( control group) ; (ii) grass + feed concentrate + ZnSO4 60 mg/kg dry matter (treatment 1), and(iii) grass + feed concentrate + ZnSO4 80 mg/kg dry matter (treatment 2), respectively drinking water weregiven ad libitum. To monitor zinc and cortisone level, blood samples were withdrawn from each animalevery two weeks starting at age of pregnancy 3 months up to two months post parturition, in addition, tomonitor the cortisone level blood samples were also collected at 1,2,3 and 7 days post parturition. The Znand cortisone serum were analyzed using atomic absorbent spectro photometric (AAS) andradioimmunoassay (RIA), respectively the result showed that generally pregnancy at the beginning andget serum level at cortisone was relatively similar between the three animal groups and there was atendency to increase and reached peak at parts then gradually decrease until the and of the observation. Atparturition the lowest serum level at cortisone (19.00±18.72 mg/ml) was observed in animals receiving the60/ dry matter zinc supplement compared to animal receiving 80 mg/ dry matter zinc ( 52.65 ± 30.83 mg/ml) and control animal ( 75.92 ± 42.88 mg/ml). in addition serum level at Zn was significantly higher inanimal receiving 60 mg and 80 mg Zn in their diets compared to control animals. The best profiles wereseen in animal given 60 mg/kg dry matter Zn supplement. In conclusion, the addition of Zn in the diets hadsignificant effect in minimizing stress as pictured by the low serum level at cortisone at parturition.

Abstract

Twenty growing cross breed etawah goat were treated with clenbuterol every two days. Three levels ofclenbuterol , 5, 10 and 20 ?g/kg BW were injected intramuscularly from the thigh side. The experimentwould like to see the effect of clenbuterol on the metabolism and carcase quality. It was found thatclenbuterol did not affect the gross energy, digestibility, metaboliseable energy and urinary nitrogenexcretion.Clenbuterol will increase the feed intake up to 51.49%. Althoush not affecting protein intake, itsignificantly increased the retain protein from 70.51 up to 149.37%. Subcutaneous carcase weights werereduced between 29.6 up to 51%. Data from carcase quality showed that clenbuterol increase half emptybody weight indicating that clenbuterol is affecting protein metabolism. Dose -respond curve of clenbuterolagainst half empty body weight follows the Michaelis Menten equation and solved by Lineweaver-Burks.Result indicated that the half weight maximum was 5120 gram and Km of clemnbuterol was 9.50 x 10-8M. Clenbuterol is affecting the bone weight carcase but not affecting the intermusculer carcase.

Abstract

A research was conducted on the performance of broilers fed with diet supplemented with Achatina sp.meal as protein substitute of fish meal.  The aim was to increase the use of  Achatina spp. in diet byevaluating the feed consumption, the average daily gain (ADG), the feed conversion, and the dry matterand crude protein digestibility of 120 experiment broilers. The experimental design used was completelyrandomized design (CRD) consisting of five level of Achatina spp. concentration as  substitution of fishmeal.  The levels were: 0% as control, 10%, 20%, 30% 40%, and 50% of Achatina spp meal substitute for fishmeal. The result showed that substitution fish meal by Achatina spp. meal was able to increase significantly(P<0.01) of feed consumption and ADG, but it has no significant (P<0.05)  effect on feed conversion, and drymatter and crude protein digestibility. It was concluded that Achatina spp. meal can be used in diet as asubstitute for fish meal up to 30%, with ADG about 25.71 g (highest level).
