Abstract

Peningkatan keamanan pangan menggunakan bahan alami, seperti kunyit, kencur, dan temulawak sebagai suplemen pada pakan disebabkan karena bahan tersebut memiliki sifat yang ramah lingkungan, biodegradable, dan mudah ditemukan di alam. Penelitian ini bertujuan untuk mendapatkan dosis suplemen herbal dan padat tebar yang efektif dalam meningkatkan status kesehatan ikan ditinjau dari profil hematologis. Rancangan yang digunakan adalah Rancangan Acak Lengkap pola Faktorial. Perlakuan yang diberikan adalah dosis suplemen herbal (D1= 100 mL/kg pakan, D2= 200 mL/kg pakan, dan D3 = 300 mL/kg pakan) dan padat tebar ikan P1 = 50 ekor/ m3 dan P2 = 75 ekor/ m3. Ikan patin (Pangasius hypophthalmus) yang digunakan berukuran 8-12 cm, pemeliharaan dilakukan selama 60 hari di keramba jaring apung berukuran 1,5x1x1m. Pengambilan sampel darah dilakukan sebanyak tiga kali, yakni sebelum perlakuan, kedua pada hari ke-30 dan hari ke-60 pemeliharaan. Hasil penelitian menunjukkan bahwa penambahan dosis suplemen herbal 200 mL/kg pakan dan padat tebar 75 ekor/m3 yang dipelihara selama 60 hari memberikan hasil tertinggi terhadap profil hematologis, yaitu total eritrosit sebesar 2,81 x 106 sel/mm3, nilai hematokrit 39,00%, kadar hemoglobin 10,73 g/dL, total leukosit 11,41 x 104 sel/mm3 dan kelulusanhidup 100%. Hasil penelitian dapat disimpulkan bahwa kombinasi padat tebar 75 ekor/m3 dan dosis suplemen herbal 200 mL/kg pakan adalah yang terbaik untuk peningkatan kesehatan ikan patin.

Abstract

Kintamani dog is the only Balinese breed has been recognized by Federarion of International Cynology (Fédération Cynologique Internationale). Kintamani dogs live around Sukawana Village, KintamaniDistrict, Bangli Regency, of Bali. The purpose of this study was to determine the hystologycal structure and histomorphometry of kintamani dog skin. The kintamani dog used in this study were 1-2 years old.Histological structure observed by carlzeiss teaching microscope with objective lens magnification 5, 10, 20, 40, and 100x. In this study, the hystologycal structure of kintamani dog consists of the epidermis, dermis, and hypodermis, except on planum nasale. Planum nasale of kintamani dogs do not havehypodermis. The other components found in the hystologycal structure is sebaseous gland, sweat gland, hair follicles and blood vessels. While the histomorphometry of kintamani dog skin have a different thickness depending on location of the body and gender. In this study, Kintamani dog skin is the thickest in female planum nasale with thickness from epidermis to dermis at 6437.040 ?m and the thinnest inmale stomach at 2047.378 ?m.

Abstract

Newcastle Disease or tetelo is one of main problem in poultry Industry in Indonesia. Prevention such as biosecurity control and routin vaccination program have been conducted to overcome this problem, but they have not given any great impact. Phyllanthus Niruri L. or meniran is well known as immunostimulatory. This research was aimed to reveal effect of Phyllanthus Niruri L. extract on chicken vaccinated with live vaccine LaSota. Administration of Phyllanthus Niruri L. extract was conducted on three different time which were 7 days before vaccination, 1 days after vaccination, and 3 days before and after vaccination. The amount of Phyllanthus Niruri L. extract administered were 2 ml, 2.5 ml, and 3 ml orally. Data of antibody titre were collected for 4 weeks after the treatment. It was obtained by measuring the antibody through Haemagglutination Inhibition test each week. According to the result Phyllanthus Niruri L. extract could increase the amount of antibody titre against Newcastle Disease. The amount of Phyllanthus Niruri L. extract given that capable to induced maximum of antibody titre was administered 1 days after the vaccination with amount 2.5 ml. It is suggested that Phyllanthus Niruri L. extract should be administered post vaccination to boost antibody synthesis.

Abstract

This study aims to determine the level of growth hormone given feed supplement and anabolic growth promoter. This research was an experimental research using Complete Random Design (CRD) with three factors. There were feed supplement, anabolic growth promoter, and sampling time. There were 20 youngmale Bali cattle and they were taken care intensively, divided into 4 group, there were control, feed supplement, anabolic growth promoter, and the combination of feed supplement and anabolic growth promoter for five months, and measured the level of growth hormone each month. The level of growth hormone was detected by using competitive Enzyme Linked Immunosorbent Assay method. The result of the research showed that feed supplement and anabolic growth promoter increased the level of bali cattle growth hormone where feed supplement with anabolic growth promoter (P1G1) was not significantlydifferent than given feed supplement without anabolic growth promoter (P1G0). Providing feed supplement optimally by farmers is highly recommended.

Abstract

Avian colibacillosis is an infectious disease caused by avian pathogenic Escherichia coli (APEC). APEC ability to cause disease depends on many pathogenic factors, one of them is iroN pathogenic gene. This study purposed to find out iroN gene sequence in APEC in Bali. Two APEC isolates from free range chicken in Tabanan dan Badung has been used. The isolates have been been purified and were available at Laboratory of Veterinary Bacteriology, Faculty of Veterinary Medicine, Udayana University. Chelex 10%was used for DNA isolation. DNA amplification using published DNA primer has been conducted with polymerase chain reaction (PCR) method. The PCR product was sequenced at First Base Laboratories, Malaysia using Sanger’s Dideoxy Nucleotide Termination method. The iroN gene of both isolates can be analyzed and have 659 bp in length. Both were 100% homologous. Phylogenic test using 52 DNA sequence of iroN gene from Escherichia coli and other bacteria in the world was conducted in MEGA 5.2. All data have 24 polymorphic sites of nucleotide acid and eight polymorphic sites of amino acid. The iroN gene of Bali isolates was in the same group as iroN gene from Australia (MF174860) and Hong Kong (MF474175). This gene can be used as pathogenic marker of APEC in Indonesia.

Abstract

Tulang femur kodok sawah (Fejervarya cancrivora) mengalami proses reabsorbsi dan resorbsi tulang pada bagian rongga sum-sum (marrow cavity-MC), karena pada penelitian sebelumnya terlihat Metamorphosis Line (ML) pada sebagian besar individu terlihat sudah tidak utuh atau menghilang. Pengukuran pertumbuhan lebar femur (MP) yang diukur tegak mulai dari tepi MC ke bagian tepi tulang bagian luar dilakukan dengan menggunakan metode skeletokronologi. Hasil analisis korelasi antara panjang tubuh (SVL) dengan MP memperlihatkan korelasi kuat pada kelompok jantan dan betina untuk data normal dan data konversi log10; tetapi hubungan antara MP dengan jumlah Line of Arested Growth (LAG) adanya dinamika pertambahan lebar tulang femur pada kelompok jantan dan betina, yang mana kelompok jantan pertambahan lebar tulang femur lebih besar dibandingkan pertambahan lebar tulang femur kelompok betina.

Abstract

Flu burung atau Avian influenza (AI) subtipe H5N1, termasuk kelompok Highly Pathogenic Avian Influenza (HPAI) yang masih mengancam industri perunggasan, kesehatan manusia, dan sejumlah spesies burung liar. Itik merupakan reservoir alami virus AI H5N1 dan diduga kuat dapat menularkan virus ke unggas lain maupun manusia. Penelitian ini bertujuan untuk mengetahui seroprevalensi virus AI subtipe H5N1 pada itik bali di dua pasar yang menjual hewan hidup yaitu Pasar Hewan Beringkit di Kabupaten Badung dan Pasar Umum Galiran di Klungkung, Bali. Total sampel yang diambil sebanyak 120 serum itik bali, yang diambil dari itik umur lebih dari tiga bulan dan tidak divaksinasi AI subtipe H5N1. Pengambilan sampel dilakukan sebanyak empat kali dengan interval waktu dua minggu dari masing-masing pasar tersebut. Semua sampel serum tersebut selanjutnya diuji dengan uji Hambatan Hemaglutinasi (HI). Hasil uji menunjukan bahwa seroprevalensi AI subtipe H5N1 di Pasar Hewan Beringkit sebesar 20,0%, sedangkan di Pasar Umum Galiran sebesar 23,3%. Hasil ini mengindikasikan bahwa itik bali yang disampling dari kedua pasar tersebut pernah terpapar virus AI H5N1 secara alami. Surveilans dan monitoring serta tindakan pencegahan secara berkala perlu dilakukan untuk mencegah penyebaran penyakit AI H5N1 di pasar hewan.

Abstract

Daun singkong yang berwarna hijau mengandung klorofil dan flavonoid yang memiliki potensi sebagai antioksidan. Penelitian ini bertujuan mempelajari pengaruh ekstrak daun singkong terhadap jumlah leukosit, diferensial leukosit serta rasio heterofil limfosit (H/L). Penelitian ini menggunakan ayam broiler, yang dibagi ke dalam empat kelompok, masing-masing kelompok terdiri dari enam ekor. Kelompok (K0) sebagai kontrol. Kelompok K1, K2 dan K3 masing-masing diberi ekstrak daun singkong secara berurutan sebanyak 47,23 mg, 75,56 mg dan 113,34 mg/1500 g bobot badan per oral selama 21 hari setelah diadaptasikan satu minggu. Data diolah menggunakan Rancangan Acak Lengkap (RAL). Semua kelompok Ayam broiler diberikan ekstrak daun singkong setiap hari selama tiga minggu yang dimulai pada hari ke-7 sampai dengan hari ke-28, kecuali K0. Pemberian ekstrak daun singkong dilakukan pada pukul 16.00-17.30 WIB. Sampel darah diambil setiap satu minggu, mulai hari ke-7 sampai dengan hari ke-28. Hasil penelitian menunjukkan bahwa rataan jumlah leukosit ayam broiler pada hari ke-28 setelah pemberian ekstrak daun singkong dengan dosis bertingkat tidak memberikan pengaruh nyata (P>0,05) pada parameter limfosit, monosit, eosinofil dan basofil serta rasio heterofil limfosit (H/L) antar perlakuan. Kelompok ekstrak daun singkong 47,23 mg/1500 g bb memberikan pengaruh nyata (P<0,05) pada parameter heterofil antar perlakuan. Simpulan dalam penelitian ini adalah pemberian ekstrak daun singkong dengan dosis 47,23 mg/1500 g bb dapat memperlihatkan profil leukosit yang terbaik berupa heterofil dan limfosit serta rasio H/L pada ayam broiler.

Abstract

This study was aims to exploit the potential waste of cocoa pod (Theobroma cacao L.) as a natural acaricide in ticks (Boophilus microplus). The study was conducted with a two-variable completely randomized design (CRD) with two repetitions. The sample does not differentiate between sex, body weight, and length of about 0.8 - 1 cm. Each 5 ticks were treated as follows: treat 1 extract of 10% cocoa pod husk; treatment 2 20% cocoa pod skin extract; treatment 3 30% cocoa pod husk extract; treatment 4 negative control; treat 5 positive controls. In the first variable the tick was treated with methanol extract, while in the second variable the ethanol extract was treated with 10%, 20%, 30% respectively, negative control and positive control. In treatment I used 25 ticks plus treatment II 25 ticks. Each treatment was repeated to avoid data bias so that a total of 100 ticks were used. The results showed that the cocoa pod husk extract had the best killing power at a concentration of 30% with methanol solvent at 6.21 hours while ethanol solvent at 5.91 hours. The use of extraction materials has not been able to compensate for the use of synthetic materials that can kill in minutes.

Abstract

Currently, mesenchymal stem cells (MSCs) for implementing regenerative medicine in aging become interest in medical research science, especially in degerative disease and other aging problems. This research was aimed to determine the effectiveness of hUC-MSCs on inhibiting the aging process through the lifetime of the rat and the effect of intravenous administration of hUC-MSCs in phisiologycally aging female rat on the blood analysis. This study was used 40 aged female rats with 29-30 months of age divided into four groups with 10 rats each. The control rat group was given physiological NaCl (0.9%) 0.5 mL, and the treated rat group was given hUC-MSCs 1x107 cells/kg body weight in 0.5 mL NaCl 0.9%, wasinjected intravenously in caudo lateralis tail vein with stratified frequency; one time injection (SC1), three times injections (SC3) and five times injections (SC5). Perifer blood was collected from retro-ortbital sinus vein 30 days before and after injection of hUC-MSCs for hematology and blood chemistry analysis. Based on the results were obtained, it indicated that hUC-MSCs increased the survival of aging rat werein treatment group, life span of rats was extended up to 40 months compared to the average life of control rat aged up to 34±2 months. The injection of hUC-MSCs 1x107 cells/kg of body weight with one, three andfive times injection were affected to blood profiles and blood chemistry with correlation were low. The conclusions are hUC-MSCs extend the lifespan of aging rat and were affect the blood in general but in normal range of aging rat, affect in ALT and creatinin as tissue repair and tolerated by aging rat.
