Abstract

Sunda porcupine (Hystrix javanica) is one of the endemic porcupine species in Indonesia. The importance of wild species preservation brings with it the requirement for more basic aspects of anatomy. The knowledge of the normal radiological anatomy of the lung is of crucial importance for the making of a correct diagnosis of lung diseases. This study aimed to provide information about the radiographic anatomy in the lung lobation of sunda porcupine. This study was an explorative study using four sunda porcupines. Thoracic radiograph was retrieved using conventional and digital X-ray machine. Standard projections for thoracic radiography used lateral and ventrodorsal projection. The evaluation result of thorax radiogram showed that interlobar fissures separate the different lobes. The porcupine right lungs consist of four lobes, i.e. cranial lobe (cranial and caudal segment), middle lobe, caudal lobe, and accessory lobe, while left lung consisted of only two lobes, i.e. cranial lobe (cranial and caudal segment) and caudal lobe.

Abstract

The aim of this study was to determine the type and concentration of sugar that was most suitable for use in semen extenders in freezing kampung rooster spermatozoa. Semen was collected from five roosters using massage method. Immediately after collection, the semen was evaluated macroscopically and microscopically. Semen was divided into twelve tubes. Each of them diluted with ringer lactate egg yolk added with various sugar such as glucose, trehalose, sucrose and lactose combined with sugar concentration of 20 mM, 50 mM and 80 mM. Liquid semen was packaged in a mini straw and was equilibrated, semen was frozen above the surface of the liquid nitrogen then it was stored in a container for 24 hours. Spermatozoa motility was evaluated after dilution, equilibration and thawing. The results showed that there were no significant differences in quality (P> 0.05) with the treatment of various types of sugars, but there were significant differences in motility (P <0.05) with differences in sugar concentrations. The best motility of spermatozoa was obtained at 50 mM and 80 mM sugar concentrations. Spermatozoa motility after thawing was 36.56-37.19% with a sugar concentration of 50 mM and 80 mM, whereas a sugar concentration of 20 mM was only resulting 34.06% of motility. The conclusion of this research is glucose, trehalose, sucrose and lactose can be added to the kampung rooster semen extender with a concentration of 50 mM for freezing.

Abstract

Enterobacteriaceae are Gram negative bacteria contain endotoxin and exotoxins which arerequirements for pathogenic bacteria and act as opportunistic pathogens. The purpose of this research was to determine the ability of Enterobacteriaceae bacteria to infect zebrafish (Danio rerio) by observing anatomical pathology, mortality, time death and cumulative time of death. The method used in thisresearch is a completely randomized design method (CRD). This research was conducted in three stages, first preparation of zebrasfish as animal model by average size 3-5cm and reach three months old. Second, bacterial cultures from 10 species of Enterobacteriaceae were Escherichia coli 1, E. coli 2, Enterobactercloacae 1, Acinetobacter calcoaceticus, Enterobacter amnigenus 1, Enterobacter amnigenus 1, Escherichia coli 3, Pantoea spp., E. cloacae 1, E. cloacae 2, E. cloacae 3, cultured in Brain Heart Infused Broth (BHIB) and later media washed with Phospate Buffer Saline (PBS) 0.45%. The third stage was bacterial infection to zebrafish using immersion method with each treatment repeated 3 times and observed for 120 hours. The results showed 10 species that used caused death (mortality), the highest mortality in fish that infected with E. coli 2 and E. cloacae 1 with a percentage of 66.67% infected by E. coli 2 and E. cloacae 1. Anatomicalpathology in the external organs and internal organs showed post-infectious symptoms. Infection occurred with mean time to death of 12-86 hours, as long as the cumulative time of fish death caused by E. coli 2, A.cinetobacter calcoaceticus and E. cloacae 1 causing death from 24 hours after infection until the end ofobservation.

Abstract

This study aims to early detect Salmonella spp. and identify serotypes in commercial chicken eggs from layer chicken farms and traditional markets in Bali. Early detection study of Salmonella spp. was carried out by conventional bacteriological methods, while serotype identification by duplex Polymerase Chain Reaction (d-PCR) test against the invA gene from Salmonella spp. and the sefA gene from Salmonella enteritidis. Egg samples in this study were taken from 10 layer chicken farms in Bali which included districts of Bangli, Gianyar, Tabanan and Karangasem. Egg samples from traditional markets were taken from 18 traditional markets from the districts of Bangli, Gianyar, Tabanan, Karangasem, Badung, and Denpasar City. Samples were eggshells and egg whites. Analysis of positive results from Salmonella spp. described descriptively. The results showed that eggshells and white eggs from all of the layer chicken farms are negative contaminated with Salmonella spp. (0%). In eggshell samples taken from the traditional markets of Taman Bali and Tulikup from the districts of Bangli and Gianyar, positive with Salmonella spp. (11,1%) by conventional bacteriological tests. In the duplex Polymerase Chain Reaction test, S. enteritidis serotypes were identified. The finding contamination of Salmonella enteritidis in commercial chicken eggs from traditional markets require periodically detection to prevent the occurrence of salmonellosis due to consumption of contaminated chicken eggs in traditional markets in Bali.

Abstract

Abstract

Pengging ducks are one breed of Indonesia’s local duck has superior potential. This study was designed to evaluate the hematological parameters of Pengging ducks fed Moringa leaf meal as a feed additive. The data were obtained from a total of 80 female Pengging ducks of 35 weeks old were assigned into a completely randomized design and each experimental unit consisted of 4 replications. The treatment group consisted of basal fed without added Moringa leaf meal (control), basal fed added with 2.5, 5, 7.5, and 10% Moringa leaf meal. Data analyzed using one-way ANOVA. The results showed that the addition of Moringa leaf meal gave same effect (P>0.05) on the number of erythrocytes, hemoglobin, hematocrit, mean corpuscular volume (MCV), and mean corpuscular hemoglobin (MCH). However, the mean corpuscular hemoglobin concentration (MCHC) increased (P<0,05) to 15.91% at 5% moringa leaf meal. The number of leukocytes was lower (P<0.05) by 42.01% at 7.5% moringa leaf meal. Leukocyte differentials, the ratio of H:L, and trombocytes did not significant difference (P>0.05) between treatments. In conclusion, the Moringa leaf meal as a feed additive did not change the hematological parameters and could be increasing the health status of layer Pengging ducks.
