Abstract

Root disease controlling has been a challenge in increasing plant productivity. The soil-borne pathogens become the main concern which mostly infects the root surface. One of the most common soil-borne pathogens is Fusarium oxysporum (Fo). Cayenne pepper (Capsicum sp.) is one of the most abundant commodities and contributes significantly to the economy. This plant is also susceptible to Fusarium wilt infection. The use of endophytic fungi as biocontrol agents is an alternative against soil-borne pathogens, one of which is Trichoderma spp. fungi. This study aims to determine the effectiveness of various spore densities of Trichoderma spp. against the pathogen Fo. The research was conducted through an in vitro study by antagonistic testing between spore suspensions of Trichoderma spp. (103, 105, and 107 spores/mL) with Fo. The results showed the density of 107 spores/mL gave the highest percentage of inhibition (54.59%) compared to spore density of 103 (35.27%) and 105 spores/mL (44.44%). The spore density of 107 spores/mL gave a significant difference in response to the density of other spores according to the BNT test at a significance level of 5% and was able to inhibit the growth of Fo more than 50%. The results of this study are expected to be a reference for the formulation of Trichoderma spp. which is ideal for controlling Fo pathogens in the fields.

Abstract

Orchids of the Vanda helvola genus are extensively grown as ornamental plants. The method of propagation of plant seeds in tissue culture is an alternative that may be employed in the provision of a short period. Murashige and Skoog (MS) media and activated charcoal can optimize orchid cultivation utilizing tissue culture techniques. One of the keys to success in tissue culture is the application of natural types of growing regulatory substances (ZPT) at the proper concentration, which can lower the cost of orchid tissue culture.The objective of this study was to see how a combination of Murashige and Skoog (MS) media and activated charcoal influenced the Vanda helvola orchid subculture in vitro. The researchers employed a Complete Randomized Design (RAL) with four treatments and three repeats: negative control (K), ¼ MS (K1), ½ MS (K2), and 100% MS (K3) (K3). The observed parameters include the percentage of live explant, number of roots, and height of orchid plants.   The results showed that the explant had a 100% life percentage of over 21 HST and had no noticeable influence on the number of roots and the plant's height.

Abstract

Pencemaran lingkungan salah satunya disebabkan oleh limbah cair yang dihasilkan oleh industri virgin coconut oil yang dibuang langsung ke lingkungan tanpa diolah terlebih dahulu, sehingga menyebabkan beberapa tanaman mati dan bau tak sedap. Tujuan penelitian ini adalah untuk mengetahui pengolahan limbah cair virgin coconut oil menjadi pupuk organik cair dengan penambahan EM4 (Effective Microorganisme 4) dan kelayakan limbah cair virgin coconut oil untuk dijadikan pupuk organik cair. Penelitian ini menggunakan metode fermentasi. Limbah cair VCO yang digunakan diperoleh dari CV. Masyana Barokah, pemeriksaan yang dilakukan yaitu Nitrogen, Phosfor, Kalium dan C-organik. Hasil yang diperoleh yaitu kadar N (0,04%), Kadar P (0,03%), Kadar K (0,17%), Kadar C-organik (4,44%), namun belum memenuhi persyaratan minimal pupuk organik cair.

Abstract

Jamu gendong termasuk obat tradisional yang umum dikonsumsi masyarakat Indonesia, salah satunya jamu kunir asem. Tujuan penelitian ini mendeteksi ada tidaknya bakteri Escherichia coli dan Salmonella pada jamu kunir asem di Daerah Gamping. Metode yang digunakan adalah MPN (Most Probable Number), menghitung total Coliform pada jamu kunir asem. Uji positif Coliform selanjutnya di deteksi Escherichia coli dan Salmonella pada medium Endo agar dan Salmonella shigella agar, berturut-turut. Ke 6 sampel dari total sembilan sampel jamu kunir asem terkontaminasi bakteri Coliform dengan jumlah antara 7,40x10 hingga 1,10x104 Sel/mL. Deteksi Escherichia coli dan Salmonella pada sampel kunir asem memberikan hasil negatif.

Abstract

Tanaman ciplukan memiliki senyawa yang berpotensi sebagai antibakteri. Potensi senyawa antibakteri kemungkinan juga dimiliki oleh bakteri endofit tanaman ciplukan. Penelitian ini bertujuan untuk mengisolasi bakteri endofit dari daun ciplukan yang berpotensi sebagai antibakteri terhadap Escherichia coli dan Staphylococcus aureus. Metode penelitian meliputi sterilisasi, pembuatan media, pengambilan sampel, isolasi bakteri endofit, karakterisasi morfologi, dan uji aktivitas antibakteri. Sebanyak 8 isolat bakteri endofit berhasil diisolasi dari daun ciplukan dan memiliki morfologi koloni yang bervariasi. Berdasarkan uji aktivitas antibakteri, semua isolat mampu menghambat pertumbuhan S.aureus dengan diameter zona hambat berkisar antara 0,5-2mm dan 2 isolat terhadap pertumbuhan bakteri E.coli dengan diameter sebesar 2mm.

Abstract

Neurodegenerative is diseases which occur with age but no therapeutic approach is efficient enough to inhibit neurodegenerative progression which results from aging of the cells themselves. PSC is a stem cell which has the potential to differentiate into various types of cells in the body. In the expansion process, PSC encountered problems, one of the common obstacles. This research was conducted using a literature study method that aims to obtain information about the appropriate differentiation method so that it can be used as an alternative treatment for neurodegenerative disease in the future. The result showed that 3D technique is neuronal differentiation technique that has a good output and can resemble the conditions of its development in vivo. The use of growth factors such as FGF2, PDGF, IGF1, T3, FGF, EGF, and CNTF can also overcome the problem of differentiation of several target cells in neurodegenerative therapy such as neurons, astrocytes, and oligodendrocytes.

Abstract

Coronavirus disease 2019 (COVID-19) is an acute respiratory syndrome disease caused by the SARS-CoV-2 virus (severe acute respiratory syndrome coronavirus 2). This virus has mutations that cause its transmission to speed up so it requires a fast detection method to prevent such transmission. The detection method that becomes gold standard is RT-qPCR (reverse transcription-quantitative polymerase chain reaction). The disadvantage of the RT-qPCR method is that the long detection time and nasopharyngeal swab sampling cause discomfort in the patient. One alternative method of detection of the SARS-CoV-2 virus is colorimetric saliva based RT-LAMP (reverse transcription loop-mediated isothermal amplification). The advantage of the RT-LAMP method is that it is fast and accurate. The purpose of this study is to determine the ability of the RT-LAMP saliva-based colorimetric method to use thermal cyclers as an alternative to SARS-CoV-2 detection in Indonesia. This study used the primary design of SARS-CoV-2 Indonesia. The samples used were positive and negative saliva for COVID-19 based on RT-qPCR test results from PKU Muhammadiyah Hospital, Yogyakarta City. The research stages include optimization of RT-LAMP conditions, saliva sampling followed by colorimetric visualization of results and electrophoresis. Colorimetric testing was carried out with the addition of phenol red and observed a change in color from pink to yellow. The results showed that RT-LAMP can run optimally at 65 °C for 45 minutes. Colorimetric RT-LAMP testing in this study was ineffective because it could not detect positive samples compared to electrophoresis results. Factors that cause this include a small number of amplicons, incompatible RT-LAMP reagents used for colorimetric methods using phenol red, and phenol red which cannot work optimally so that it requires a new or more appropriate pH indicator. It is necessary to conduct further research by replacing other visualization methods such as replacing pH indicators or by fluorescence methods.

Abstract

Kelompok masyarakat kurang beruntung secara ekonomi sosial dan terpinggirkan seperti warga santri yang tinggal di panti asuhan adalah kelompok yang paling membutuhkan dukungan dan bantuan. Panti Asuhan Aisyiyah didirikan sejak tahun 1921 oleh Pimpinan Pusat Muhammadiyah dengan visi terwujudnya Panti Asuhan Putri yang Islami yang mempunyai Keunggulan Pengasuhan yang bermartabat dan menjadi Kebanggan Umat. Permasalahan yang dihadapi oleh kedua panti asuhan ini adalah (1) belum adanya pelatihan ketrampilan yang dibekalkan kepada warga panti asuhan sebagai bekal untuk hidup mandiri, (2) warga panti masih mengandalkan donasi dari yayasan dan donatur untuk kecukupan kebutuhan logistik (3) belum adanya infrastruktur ketahanan pangan mandiri di panti asuhan Kegiatan di luar panti lebih fokus pada kegiatan keagamaan antara lain menghafal qur’an dan pelajaran fiqih. Maka dari itu perlu diadakan pelatihan dan pendampingan kewirausahaan sebagai bekal mereka di masa depan agar lebih madiri sekaligus sebagai ketahanan pangan warga panti asuhan Aisyiyah di tengah kondisi ekonomi yang serba sulit. Beberapa solusi yang ditawarkan untuk membantu permasalahan mitra adalah (1) Pelatihan kewirausahaan pendampingan ternak lele dan budidaya sayuran organik dengan melakukan pembukuan dan pelaporan laba-rugi dan (2) Pelatihan branding dan marketing online. Kegiatan PKM ini bertujuan untuk meningkatkan pengetahuan ketahanan pangan warga panti asuhan di masa pandemi dan kemampuan kewirausahaan.

Abstract

The pandemic is having a negative effect on the economies of the most vulnerable households and increasing the risk of child marriage. Millions of children have been badly affected by the COVID-19 pandemic, the most affected are those who are under the socio-economic status of people who are already poor and neglected. The purpose of this study was to analyze in depth the role of Aisyiyah’s strategies to prevent child marriage during the COVID-19 pandemic in Indonesia. This research used a qualitative research type. This research was conducted in a Sub-District of Magelang Regency, Indonesia. The sampling technique used in this research were purposive sampling and snowball sampling. Sub-District Government is very concerned with the prevention of child marriage programs that have been carried out by involving cross-sectors such as Religious Affair Office (KUA), Public Health Center (Puskesmas), Village Advisory Advisor (Babinsa), Midwives, Teacher, and Community Health Workers (Kader Desa). Involving the community and using the rules and norms that have been regulated by the Ministry of Religion, they collaborate with the Religious Affairs Office in implementing these regulations. Suggestions for parents that the role of parents is very important in providing early education about reproductive health in children and adolescents.

Abstract

Pangan merupakan kebutuhan dasar manusia untuk hidup dan mempertahankan hidupnya. Isu ketahanan pangan juga menjadi salah satu permasalahan yang dihadapi Panti Asuhan Yatim (PAY) Putri ‘Aisyiyah Yogyakarta. Sumber pemasukan lain yang bersifat menopang kemandirian pangan diperlukan oleh mitra untuk memenuhi kebutuhan dan program lainnya di panti asuhan. Salah satu upaya pemenuhan kemandirian pangan yang dapat dilakukan yaitu melalui penerapan sistem aquaponik Budikdamber. Kegiatan pengabdian ini bertujuan untuk mengimplementasikan sistem Budikdamber di PAY Putri ‘Aisyiyah Yogyakarta dalam rangka meningkatkan ketahanan pangan, termasuk diversifikasi hasil panen Budikdamber. Metode yang digunakan pada kegiatan ini yaitu edukasi masyarakat melalui penyuluhan dan juga kegiatan pelatihan berupa demonstrasi Budikdamber dan diversifikasi olahan pangan. Berdasarkan kegiatan pengabdian masyarakat diperoleh hasil bahwa peserta mendapatkan pengetahuan dan ketrampilan mengenai Budikdamber dan cara pembuatannya. Hal ini ditunjukkan dengan adanya peningkatan pengetahuan dan pemahaman Budikdamber sebesar 23,6%. Selain itu, peserta pengabdian juga dibekali pengetahuan dan ketrampilan dalam mengolah ikan lele menjadi produk nugget. Dengan demikian, ketahanan dan kemandirian pangan di PAY Putri ‘Aisyiyah Yogyakarta dapat diwujudkan.
