Abstract

Hermetia illucens larvae, also known as the black soldier fly larvae, are best known as the decomposers of organic matter. There are many potential microbes found in the feces of BSF larvae. This research aimed to isolate the chitinolytic bacteria from chicken manure maggot waste, identify the antifugal activity, hydrolytic enzyme activity, phosphate and potassium solubilization, and bacterial species through 16S rRNA gene analysis. The initial screening focused on bacteria with chitinolytic ability. Antifungal activity tests were performed against phytopathogenic fungi, Colletotrichum sp. Isolate MKP02 showed the highest activity in inhibiting the growth of Colletotrichum sp. up to 100% and produced protease and cellulase enzymes, along with the ability to solubilize potassium. Furthermore, the potential isolate MKP04, the isolate shows the highest cellulolytic activity with a percentage of 300%. It can inhibit Colletotrichum sp. fungi, as well as having lipase enzyme content, and being able to dissolve potassium. The results of 16S rRNA gene amplification on the two isolates showed that both isolates were close to bacteria of the genus Lysinibacillus and Brevibacterium. This research is expected to provide valuable information about the bacterial content, levels of hydrolytic enzymes, and the ability to solubilize phosphate and potassium in BSF.

Abstract

This study aimed to determine whether there is a simultaneous difference in students' creative thinking and collaboration skills when using the Problem-Based Learning (PBL) model integrated with Team Assisted Individualization (TAI). The research employed a quasi-experimental design with a posttest-only control group design. The study population comprised tenth-grade students at SMA N 7 Surakarta during the 2023/2024 academic year. The sample included control and experimental classes selected through cluster random sampling. Data were collected using a creative thinking skills posttest, collaboration observation sheets, and documentation. The hypothesis was tested using MANOVA. The data analysis revealed that the creative thinking and collaboration skills of students in the experimental class were higher than those in the control class. The hypothesis test results indicated that the significance value was 0.021 (sig = 0.021 0.05), leading to the rejection of H0. This finding signifies that there is a simultaneous difference in creative thinking and collaboration skills when applying the PBL model integrated with TAI. It is concluded that the PBL model integrated with TAI creates a significant difference compared to the PBL model alone in fostering students' creative thinking and collaboration skills.

Abstract

Multidrug-Resistant Tuberculosis (MDR TB) is caused by an organism that is resistant to at least isoniazid and rifampisin, the two most potent TB drug. Immune response to against Mycobacterum tuberculosis infection is related to the function of immunity. The function of interferon-γ (pro-inflammatory) is to activate macrophages, to stimulate antimicrobial molecules (to reactive oxygen species and nitric oxide), and to inhibits interleukin-10. Interleukin-10 function is to triggers humoral immunity, to inhibit IFN- γ. This study aimed to analyze level changes and the correlation with clinical data, also months of MDR TB patients who received standard OAT therapy. This was an observational study using cross sectional design. There were 29 patients who received standard MDR TB OAT therapy from 1-24 months, who met the inclusion criteria. Then, the patients were divided based on duration of the therapy, which are the initial/intensive and advanced phase. The initial phase divided into 2: first one is for 1-4 months therapy's time (5 patients) and the second one is for more than 4-8 months (6 patients). Then, the advanced group divided into two groups again, which are third group with more than 8-16 months (13 patients) and fouth group with more than 16-24 months (5 patients). Then, measured serum concentration IFN-γ, IL-10 at the start of the study and 4 weeks later with the ELISA method. This research during the period July-December (6 months). IFN-γ post concentrations were decreased by 39.14 ± 139.12 pg/mL (p > 0.05). The concentration of IL-10 was decreased by 33.93 ± 109.20pg/mL (p>0.05). Based on the TB score bandim method during pre and posts results were 1 patient experienced severity change from severity class 1 to 2, 1 patient from severity class 2 to 1, 1 patient remained in severity 2 and 26 patient remained in severity 1. The results showed that serum IFN-γ and IL-10 levels in initial/intensive and advanced phase patients who received MDR TB regiment after four weeks did not changed,

Abstract

ABSTRACT. Due to their phytochemical constituents, Dewa leaves (Gynura japonica (Thunb.) Juel.) are often used in traditional medicinal herbs. However, in vitro and in vivo of antioxidative and anti-aging studies of Dewa leaves on yeast as a eukaryotic cell model have not been widely carried out. This study aims to determine the antioxidant, anti-aging, and antibacterial activities derived from G. japonica leaves extract. Extraction was conducted using 70% and 96% ethanol solvents, total phenolic content (TPC) was assayed using Folin-Ciocalteu method, flavonoid contents (TFC) was assayed using aluminum chloride method, antioxidant activity was tested using DPPH (1,1-diphenyl-2-picrylhydrazyl) and ABTS (2,2-azinobis-3-Ethylbenzothiazoline)-6-sulfonic acid) radicals. Subsequently, an anti-aging activity test was performed on the yeast Saccharomyces cerevisiae as a model organism following antibacterial activity. The antibacterial activity test was carried out using well diffusion agar, and the Minimal Inhibitory Concentration (MIC) was determined using the microplate method. We obtained that G. japonica leaves extracted from 70% and 96% methanol solvents, have TPC of 13.14 and 22.11 mg GAE/gr extract, and TFC of 8.04 and 14.09 mg QE/gr extract, respectively. As for DPPH and ABTS antioxidant activity, D70 showed the best activity with IC50 values of 1411.36±56.35 µg/mL and 2516.10±18.77 µg/mL, respectively. The anti-aging test showed that both 70% and 96% ethanol extracts were able to maintain the yeast cell viability under H2O2 oxidative stress. Further, 70% and 96% ethanol extract also showed antibacterial activity at the best value against Staphylococcus aureus with a MIC value of 390.62 µg/mL, it means that Gynura leaves extract has a potency as antibacterial agent. The results of this study indicate that the ethanol extract of G. japonica leaves can be developed for further investigation as an antioxidant and antibacterial therapeutic agent.
