Abstract

This research was conducted to determine the effect of skimmed milk concentration on characteristic of fermented tamarillo (Solanum betaceaum Cav.) probiotic juice with Lactobacillus rhamnosus SKG34 and the concentration of skimmed milk that produced the best characteristic. This research used a Completely Randomized Design (CRD) with factor of skimmed milk concentration that consists of 6 levels (0%, 2 %, 4%, 6%, 8%, 10%). Each treatment was repeated 3 times resulting 18 experimental units. The data were analyzed by Analysis of Variance (ANOVA) and continued with Duncan Multiple Range Test (DMRT). The results showed that skimmed milk concentration had a significant effect to total LAB, total sugar, hedonic of color, score of color, and overall acceptance. The concentration of 2% skimmed milk produced the best characteristic of fermented tamarillo (Solanum betaceaum Cav.) probiotic juice with total LAB 8,50 x 109 CFU/ml, total sugar 6,54%, total acid 0,56%, pH 3,72, preferred dark orange color, slightly likeable aroma, preferred sour and slightly sweet taste, and preferred overall acceptance.

Abstract

This research aimed to find effect of maceration time on antioxidant activity of lemon peel extract and to knew the best maceration time with highest antioxidant activity of lemon peel extract. This study used a Randomized Completly Design with treatment of maceration time of 6 levels: 18 hours, 24 hours, 30 hours, 36 hours, 42 hours and 48 hours. The experiment was repeated in three replication, so that 18 experimental units were obtained. The data were analyzed by ANOVA and then continued with DMRT. The results showed that the treatment of maceration time had significant effect on extraction yield, vitamin C, total phenol, total flavonoids and antioxidant activity of lemon peel extract. The best treatment was 36 hours with extraction yield was 26.96%, vitamin C was 5.22 mg AA/g, total flavonoids was 69.64 mg QE/g, total phenol was 16.73 mg GAE /g and antioxidant activity was 94.08%.

Abstract

This research aims to identify the effect of fermentation time on the characteristics of soursop juice (Annona muricata Linn) probiotic drink as well as to determine the specific fermentation time that produces good characteristics of soursop juice probiotic drink. This study used a randomized block design (RBD) of one treatment factor with different periods of fermentation time that consist of 18 hours, 20 hours, 22 hours, 24 hours, and 26 hours. The treatment was repeated 3 times to obtain 15 experimental units. The data was analyzed with analysis of variance (ANOVA) and if the treatment had a significant effect, then followed by the Duncan test. The results showed that the length of fermentation in soursop juice probiotic drink did not significantly affect the total LAB, total sugar, total acid, pH, and sensory tests. Fermentation time of 18 hours resulted the best characteristics of soursop juice probiotic drinks with an average total value of BAL 10,87 Log CFU / ml, total sugar 10,61%, total acid 1,37%, total sugar, pH 3,92, colour, aroma, and taste rather liked with rather acidic and rather sweet criteria and overall acceptance is rather liked. Abstract

This research was conducted in order to determine the type of solvent and ratio between sample and solvent to produce extract avocado seed with the highest antioxidant activity. The experimental design used in this research was a factorial completely randomized design which consisted of two factors. The first factor was the type of solvent consisting of ethanol, methanol, and acetone. The second factor was ratio between sample and solvent of 1:5, 1;10, and 1:15. The treatment was repeated three time to obtain 27 units of the experiment. Data were analyzed with analysis of variance, followed by Duncan test. The results show that the solvent of acetone and ratio between sample and solvent of 1:15 was highest antioxidant activity with total phenolic 803.60 mg/100g, IC50 was 540.95 ppm, and yield 41.36%.

Abstract

This research aims to identify the effect of adding purple sweet potato flour to the characteristics of health-promoting yogurt and to identify the right concentration of purple sweet potato flour to produce health-promoting yogurt with the best characteristics. This research used a Randomized Block Design (RBD) of the addition of purple sweet potato flour with the concentration of 0%, 2%, 4%, 6%, 8%. Each treatment was repeated 3 times, resulting in 15 experimental units. The data were then Analyzed of Variance (ANOVA) method and if the treatment had an effect on the variable, the Duncan Multiple Range Test (DMRT) were performed. The results showed purple sweet potato flour has significant effect to lactic acid total, pH, antioxidant activity, color, texture, and taste of health-promoting yogurt and did not significantly affect LAB total, hedonic taste, aroma, and overall acceptance. The addition of 8% purple sweet potato flour resulted in the best characteristic under the following criteria: 1.23% of lactic acid total, 3.73 of pH, 90.33% of antioxidant activity, 13.19 log CFU/ml of LAB total, pink color and indefferent color, thick texture rather like, sour taste rather like, flavor and overall acceptance rather like. Abstract

This study aims to determine the antimicrobial activity produced by lactic acid bacteria isolated from breast milk (ASI) against pathogenic bacteria Vibrio cholerae. The research design used in this study is the experimental method with 3 treatments, antimicrobial activity FCS (Free Cell Supernatant), FCSN (Free Cell Supernatant Neutral), and FCSHN (Free Cell Supernatants were Heated Neutral). The variables measured were the formation of inhibition zones produced by the presence of the antimicrobial activity of 9 isolates of LAB against pathogenic bacteria Vibrio cholerae. Data were presented in tables and figure and descriptive discussed. The results showed that the inhibitory activity against pathogenic bacteria Vibrio cholerae on FCS to isolate the A1, A3, A6, and B10b each produce inhibition diameter is 2.35 mm, 2.95 mm, 2.75 mm and 1.90 mm. the treatment FCSN inhibitory activity shown by A1 isolates diameter is 1.40 mm inhibition, and isolates A6 inhibition diameter is 1.00 mm. whereas the inhibitory activity on FCSHN treatment indicated by A1 isolates inhibition diameter is 0.40 mm.

Abstract

The research aims to investigate the effect of substitution wheat flour with brown rice flour to the characteristics of bakpao and determine the rate of its which produce the best characteristics of bakpao. The Completely Randomized Design used in this research with the comparisons of wheat flour with brown rice flour which consisted of 6 levels, such as 1) 100% : 0%, 2) 95% : 5%, 3) 90% : 10%, 4) 85% : 15%, 5) 80% : 20%, 6) 75% : 25% used as treatments. Each treatments was repeated 3 times to obtain 18 units. The datas were analyzed by analysis of variance. If there were any effect of the treatment, the analysis followed by Duncan's Multiple Range Test. The results showed that substitution of wheat flour with brown rice flour highly significant effect on the moisture content, antioxidant activity, crude fiber content, strength of expands, taste, color, flavor, texture and overall acceptance of bakpao. The best substitution of wheat flour with brown rice flour is 5% brown rice flour, with characteristics moisture content 31,55%, antioxidant activity 14,61%, crude fiber content 8,71%, strength of expands 55,81%, with criteria color 1,45 (white), texture 4,25 (soft), flavor 2,50 and taste 1,90 (not the typical brown rice) and overall acceptance of bakpao 5,70 ( mostly like).

Abstract

The aim of the study is to determine the quality of refill drinking water in district of KutaSelatan, Badung regency, Bali which includes smell, taste, color, pH, turbidity, hardness, amount ofdissolved solids, contamination of Coliform and Salmonella. The data obtained are presenteddescriptively in the form of table and compared to SNI standard quality of drinking water. Thedetermination depot sampling of drinking water refill is done by simple random sampling method.Population depot refill drinking water amount to 22 depots and 10 depots were taken as samples bylottery and analyzed in the laboratory. The results showed refill drinking water in the District of SouthKuta not meet drinking water quality standards and there are 8 depot refill drinking water that is notsafe for the health of depot A and B were contaminated by Salmonella, whereas, depot D, F, G, H, Iand J were contaminated by Coliform and Salmonella.

Abstract

This study aims to determine the effect of incubation time on milk clotting enzyme (MCE) activity produced by L. rhamnosus SKG 34 and determine the optimum incubation time to produce the highest its activity. This study used a randomized block design (RBD) with incubation time as a treatment consisting of 8 levels, that were 6 hours, 12 hours, 18 hours, 24 hours, 30 hours, 36 hours, 42 hours, and 48 hours. Data were analyzed with Variance Analysis (ANOVA) than followed by Duncan Multiple Range Test (DMRT). The analyzed were repeated 3 times resulting in 24 experimental units. The results showed that the incubation time significantly affected protease activity, MCE activity, specific protease activity and ratio of MCE to protease but did not affect the total LAB. The optimum incubation time of L. rhamnosus SKG 34 is 12 hours with total LAB 1.83 x 109 CFU/ml and protease activity 180.67 U/ml, MCE activity 595.06 SU, protease spesific activity 73.149 U/mg and ratio of MCE to protease 3.29 SU/U. Abstract

The aim of this study was to know the effect of adding soy protein isolate (ISP) to the characteristics of kamaboko barramundi fish (Lates calcalifer) and to know the right addition of ISP that can produce kamaboko with the best characteristics. The study used a completely randomized design with the addition treatment of ISP that consists of 5 levels: 0%, 4%, 8%, 12%, and 16%. The treatment was repeated 3 times to obtain 15 experimental units. The data was analyzed by variance and if it influential to the variable then continued with the Duncan test. The results showed that the addition of ISP on the producing of kamaboko had a highly significant effect to texture (gel strength), moisture content, protein content, ash content, hedonic test (color, texture, taste, overall acceptance) and scoring test (folding test and bite test). The addition of 12% ISP produced kamaboko with the best characteristics with gel strength 1404.76 gf/cm2, water content 74.42%, protein content 15.40%, fat content 3.64%, ash content 3.55%, color, texture, taste, and overall acceptances were liked, aroma rather liked, appearance of meat without fiber and without foreign things, folding test does not crack if folded 4 times, and bite test rather strong specific product elasticity.
