Abstract

Chlorophyll is tetrapyrole pigment, which is potential to be developed as photosensitizer agent on cancer photodynamictherapy. Its environments and properties during interaction are an important parameter to know the next fate of thesensitizer. Acidic environment is one of the key factors on cancerous patients. The research was carried out to determinethe acidification effect against photostability of chlorophyll a that was observed by its spectral pattern. Result showedthat titration of less than 250 μL acetic acid 3 M caused degradation of chlorophyll a more rapidly. Volume and type ofacid properties did not only influenced pheophytinization reaction, but also caused pheophytin a to deaggregate, thatwas characterized by hypsochromic shift of Qy band.

Abstract

World Health Organization (WHO) estimates both the number of patients and mortality rates due to cancer will continue to rise. Various researches were conducted in order to prevent and handle the cases of tumors and cancers, including the application of photosynthetic pigment molecules known as photodynamic therapy (PDT). Chlorophylls, the main pigment in photosynthesis, have an ability to capture light energy and control series of photobiology and photochemical processes1. In PDT, chlorophyll or its derivatives compounds act as the sensitizer which have energy excitation by light radiation (visible or near infra red), and generate some reactive oxygen species which triggers the death of cancer cells selectively (through apoptosis and / or necrosis pathway)2. Sensitizer compounds have been progressing from the first to third generation. The development of the third generation sensitizer was influenced by the advances of nanotechnology which lead to the improvement of PDT efficacy. The structure and size of nanoparticles can increase light absorption, and make the sensitizer accumulate in cancer tissues more specifically 3-4. Furthermore, nanoparticulatte targeting also interested to be studied because by conjugate functional groups, i.e. monoclonal antibody, on the sensitizer, it can improve the selectivity of therapy in targeting tumor and cancer tissues.

Abstract

Spirulina, ganggang hijau biru yang berbentuk spiral, mengandung fitonutrisi yang cukup lengkap dan berpotensi sebagai agen antikanker yang efektif. Fikosianin, klorofil a, dan karotenoid adalah pigmen-pigmen yang dikandung Spirulina. Artikel ini merangkum berbagai penelitian in vivo dan ex vivo yang melaporkan ketiga pigmen tersebut merupakan antioksidan kuat. Bahkan, beberapa laporan mencatat bahwa turunan klorofil a juga berpotensi sebagai antimutagen, induktor apoptosis, dan fotosensitizer untuk PDT (Fotodinamika Terapi Kanker). PDT diharapkan memberikan harapan baru dalam terapi kanker. Namun demikian, tidak hanya pigmen yang berperan sebagai antioksidan dalam terapi penyembuhan kanker dengan Spirulina, nutrisi lain seperti asam lemak GLA (gamma linoleic acid), enzim superoksida dismutase (SOD), dan oligosakarida juga turut berperan sebagai senyawa antikanker. Spirulina dijual dalam bentuk suplemen golongan CAM (Complementary and Aternative Medicine). Terapi dengan mengkonsumsi spirulina cair, tablet, kapsul, ataupun bubuk secara teratur menawarkan sebuah jawaban bagi mereka yang sedang bergumul dengan kanker stadium awal jika ingin menghindari pembedahan, radioterapi, ataupun kemoterapi.vKata kunci: spirulina, fikosianin, klorofil a, karotenoid, antioksidan, complementary and alternative medicine

Abstract

Astaxantin merupakan salah satu golongan karotenoid xantofil yang memiliki peran penting dalam menjaga kesehatan tubuh. Pada berbagai studi telah dibuktikan peran astaxantin dalam melindungi sel dan jaringan dari kerusakan terutama akibat stres oksidatif yang juga dapat memicu terjadinya kanker. Dalam tulisan ini akan dibahas mengenai potensi astaxantin sebagai senyawa antikanker.Kata Kunci: astaxantin, antioksidan, antikanker.

Abstract

Carotenoids are important biological compounds in cancer prevention because theycan inhibit the formation of free radicals, which react directly with oxygen. This researchwas aimed to determine the effect of antimutagenic â-carotene on 7.12-dimetilbenz(a)antracene (DMBA)-induced mice. From the observation of the mortality rate during theresearch period, the DMBA group showed the highest rate of mortality compared to thecontrol group and the groups of â-carotene isolates. The incidence of tumors showed thedeclining trend in the groups of test animals that were given with the increasing dosesof β-caroten isolate, i.e. 1.82; 3.64 and 7.28 mg kg-1 BW. The result may be caused bythe increasingly high levels of â-carotene in the test solutions, resulting in the lowertissue damage and the inhibited growth of tumors in the lungs. Based on Kruskal-Wallisstatistical analysis conducted on lung histology observational data for ten samples ofeach treatment group, it was confi rmed that there were signifi cant differences betweentreatments, which mean that â-carotene isolates can inhibit cancer growth in lung andskin of DMBA-induced animals.Keywords: β-carotene isolate, antimutagenic, DMBA (7,12-dimetilbenz(a)antracene)

Abstract

Current efforts to explore the potency of marine resources have been made to promote economic development in Indonesia, especially in the industrial sector. Marine cyanobacteria such as Arthospira sp. and Scytonema sp. are two potential candidates of the blue-green algae that can be used in the application for food and health industries. This study focused on the identification of the composition of the dominant chlorophylls and carotenoids. The results showed the presence of Zeaxanthin, Chlorophyll a, and β-carotene. The economic potency of those dominant pigments will be reviewed in this article.

Abstract

Sweet potatoes, especially the orange and yellow-fleshed, are functional local food because they contain carotenoids which serve as a pro-vitamin A. The processing of sweet potatoes into noodles, fermented cassava or “tape” and artificial rice is usually prepared through steaming. However, carotenoid is susceptible to degrade when it is subjected to high temperature, such as steaming. The objective of this research is to determine the effects of steaming on the color and carotenoid absorption spectra of local and excellent sweet potatoes which are correlated to the carotenoid content and to evaluate the difference of carotenoid spectral properties among sweet potatoes by principal component analysis (PCA). The steaming treatment decreased color values, such as lightness, redness, and yellowness. In addition, this process also influenced the spectral properties of carotenoid extracts of sweet potatoes. Steaming decreased absorbance and resulted in hypsochromic and bathochromic shifts. Madu Mojokerto, Manohara, local purple fleshed, Beta 1, and Papua Solossa sweet potatoes experienced some decreases in absorbance and a hypsochromic shift of ± 77 % and ± 2 nm, ± 40 % and ± 21 nm, ± 63 % and ± 28 nm, ± 44 % and ± 2 nm, and ± 20 % and ± 23 nm, respectively; while Antin 3 sweet potatoes experienced some decrease in absorbance of ± 36 % and a bathochromic shift of ± 28 nm. The PCA results showed that there were four groups of sweet potatoes based on spectrum shape and absorbance value.

Abstract

Suji (Pleomele angustifolia) is one kind of Indonesian typical plants which can be used as natural green food coloring agent. The susceptibility of natural pigment to external environment forces the protection in order to prolong its shelf life. Encapsulation has been known in the art of food preparation to provide protection for several ingredients including food coloring agent. The objective of this study was to observe the method for isolation and encapsulation of thylakoid, and to investigate the stability and characteristics of thylakoid of suji leaves encapsulated in maltodextrin during dark storage at 30 °C, 45 °C, and 60 °C. The degradation of the encapsulated pigments was identified through chromametric analysis which resulted in the increase of L* (lightness), a* (redness), and b* (yellowness) values. In addition, it was also indicated by the decrease of total chlorophyll (TC) which was determined using spectrophotometer. Chromatography analysis confirmed the presence of four major peaks in the fresh encapsulated thylakoid powder and five major peaks in the encapsulated thylakoid powder stored at the highest temperature (60 °C), with Chl a as the dominant pigments in both powder. The vivid green powder was able to preserve its color without any obvious change to an untrained eye up to 60 d of storage at 30 °C, becoming a promising ingredient to replace the synthetic colorants.

Abstract

Buah murbei kaya antioksidan karena kandungan antosianin yang banyak. Sayangnya,  pemanfaatannya sebagai sumber antioksidan di Indonesia masih  minim. Pemanfaatan tanaman murbei hanya sebatas daun yang dijadikan pakan ternak dan teh. Penelitian ini bertujuan untuk melakukan karakterisasi antosianin buah murbei spesies Morus alba dan Morus cathayana. Karakterisasi antosianin ekstrak buah murbei Morus cathayana dan Morus alba dilakukan menggunakan pelarut 0,1% asam klorida (HCl)  dalam metanol dan dianalisis menggunakan spektrofotometer UV-tampak dan Kromatografi Cair Kinerja Tinggi (KCKT). Hasil serapan spektra UV-tampak yang diperoleh dari ekstrak kasar menunjukkan spektra serapan ekstrak buah M. cathayana memberikan nilai yang lebih tinggi ± 1,3 a.u dibandingkan serapan yang diperoleh dari ekstrak kasar buah M. alba dengan serapan ± 0,4 a.u. Nilai total antosianin yang diperoleh dari M. cathayana sebanding dengan serapan ekstrak segarnya, yaitu 40,39 ± 7,64 mg/g berat kering dibandingkan  M. alba yang memiliki nilai total antosianin 11,57 ± 3,02 mg/g berat kering. Hasil kromatogram dari KCKT menggunakan kolom XR-ODS dengan pengelusi A adalah 0,1% asam formiat dalam asetonitril dan pengelusi B adalah 0,1% asam formiat dalam air menunjukkan baik M. cathayana maupun M. alba memiliki dua pigmen antosianin dominan. Walaupun bergitu, intensitas kromatogram M. cathayana lebih tinggi dibandingkan intesitas pada M. alba. Nilai intensitas M. cathayana adalah 50 mAU untuk waktu tambat 7,86 menit dan 15 mAU untuk waktu tambat 8,38 menit. Nilai intensitas M. alba adalah 10 mAU untuk waktu tambat 7,35 menit dan 3 mAU untuk waktu tambat 7,76 menit. Dua antosianin dominan pada M. alba dan M. cathayana  diprediksi  merupakan sianidin-3-O-glukosida dan sianidin-3-O-rutinosida.
