Abstract

This research was conducted on 12th May until 23rd July 2015 held at Parasite and Fish Disease Laboratory. The aims of this research was to determine the potency of mangosteen pericarp (G. mangostana L.) in feed to increase immunity of African catfish (C. gariepinus) on Motile Aeromonas Septicemia disease seen from the hematology parameters. The treatments were addition of mangosteen pericarp in feed with different concentration i.e : Kn (without mangosteen pericarp), Kp (without mangosteen pericarp and injected using A. hydrophila), P1 (addition mangosteen pericarp 2 g/kg of feed and injected using A. hydrophila), P2 (addition mangosteen pericarp 4 g/kg of feed and injected using A. hydrophila) and P3 (addition mangosteen pericarp 6 g/kg of feed and injected using A. hydrophila). Fish were maintained for 60 days. After 60 days treatment the fish were intramuscularly infected with A. hydrophila with density of 107cell/ml in a dose of 0,1 ml/fish. The best results showed that addition mangosteen pericarp in P3 treatment after challenged with A. hydrophila significantly different on haematocrit levels (22,94%), total leukocytes (104,83×103 cells/mm3), phagocytic activity (64,80%), and survival rate (100%). Total eritrocytes were not different significantly between treatment with 2,04×106 cells/mm3.

Abstract

 This research was conducted from Mei to September 2015 in the Laboratory of Parasitic Diseases of Fish Faculty of Fisheries and Marine Science, University of Riau. The purpose of this reseach was to knowing the addition of flour noni (Morinda citrifolia L) on feed to change the phagocytic activity, total of erythrocytes and hemoglobin of tilapia (Oreochromis niloticus) infected Streptococcus iniae. This research used five treatments, 1) Kn: negative control (without a solution of addition flour noni (Morinda citrifolia L)  and without the infection of Streptococcus iniae.), 2) Kp: positive control (without giving a solution of addition of flour noni (Morinda citrifolia L) and the infection Streptococcus iniae) 3) P1: addition flour noni (Morinda citrifolia L) 3 g/kg feed) P2: addition flour noni (morinda citrifolia l) 6 g/kg feed) P3: addition flour noni (Morinda citrifolia L) 9 g/kg feed). The research shows that the addition of flour noni (Morinda citrifolia L) on feed do impact the condition of fish blood because visible from the phagocytic activity is ranged between 35,33-56,66%, total erythrocytes 1,23-2,20 x106 cell/mm3, and hemoglobin is ranged between 4,66-6,33 gr/%.

Abstract

ABSTRAKDarah merah memiliki peran mengangkut oksigen, hormon, nutrien, dan hasil metabolisme tubuh, biji mangga harumanis merupakan limbah yang dapat dimanfaatkan sebagai antimikroba/antibakteri, antivirus, antijamur serta immune modulator karena mengandung senyawa sekunder seperti tannin, flavonoid, saponin serta asam ascorbat. Penelitian ini bertujuan untuk mencari dosis terbaik penambahan larutan biji mangga harumanis (Mangifera indica L) pada pakan dan pengaruhnya terhadap profil darah merah dan pertumbuhan ikan patin siam (Pangasius hypopthalmus). Metode penelitian yang digunakan adalah metode eksperimen dengan menggunakan Rancangan Acak Lengkap (RAL) terdiri dari Satu faktor dengan empat taraf perlakuan, P0 tanpa penambahan larutan biji mangga harumanis, P1 penambahan larutan biji mangga harumanis dengan dosis 1,6 mL/kg pakan, P2 penambahan larutan biji mangga harumanis dengan dosis 1,8 mL/kg pakan dan P3 penambahan larutan biji mangga harumanis dengan dosis 2 mL/kg pakan. Ikan dipelihara di kolam percobaan Fakultas Perikanan dan Kelautan Universitas Riau, dengan padat tebar 50 ekor/m3 dan ukuran 8-12 cm. Hasil penelitian menunjukkan bahwa pemberian pakan mengandung larutan biji mangga harumanis yang diberikan selama 60 hari pada ikan memberikan pengaruh terhadap profil darah merah. Dosis terbaik terdapat  pada perlakuan P3 (2 mL/kg pakan) dengan rata-rata total eritrosit 300,33 x 104 sel/mm3, kadar hemoglobin 11,13 g %, nilai hematokrit 29,33 %, pertumbuhan bobot mutlak sebesar 37.70 g, kelulusanhidup 95%.Kata kunci : Biji mangga harumanis, Hematologi, Pangasius hypopthalmus

Abstract

Penelitian telah dilaksanakan pada bulan Juli sampai dengan Oktober 2018. Penelitian ini bertujuan mengetahui bahan bioaktif yang terkandung di dalam ekstrak daun kersen (Muntingia calabura L), sensitivitas ekstrak daun kersen menghambat pertumbuhan bakteri Edwardsiella tarda, dosis Minimum Inhibitory Concentration (MIC) ekstrak daun kersen yang dapat menghambat pertumbuhan bakteri E. tarda dan  Lethal dosis50ekstrak daun kersen terhadap ikan lele dumbo (Clarias gariepinus) berukuran 7-9 cm.Daun kersen diekstraksi menggunakan metode maserasi dengan pelarut metanol.Dosis ekstrak daun kersen yang digunakan adalah 10000 ppm, 9000 ppm, 8000 ppm, 7000 ppm, 6000 ppm, 5000 ppm, 4000 ppm, 3000 ppm, 2000 ppm, 1000 ppm dan Novobiocin sebagai kontrol. Hasil uji fitokimia ekstrak daun kersen adalah terpenoid/steroid,flavonoid, fenolik, dan saponin. Hasil uji kromatografi lapis tipis dengan nilai Rf terpenoid/steroid 0,88 cm, flavonoid 0,6 cm, dan fenolik 0,6 cm. Ekstrak daun kersen mampu menghambat pertumbuhan E. tarda hingga dosis 700 ppm dengan rata-rata zona hambat 6,14 mm, Minimum Inhibitory Concentration (MIC) ekstrak daun kersen adalah 700 ppm dengan rerata jumlah koloni 261,66 CFU/mL. Hasil uji toksisitas LD50 ekstrak daun kersen terhadap ikan lele dumbo (Clarias gariepinus) dengan metode perendaman adalah pada dosis 720,06 ppm.Kata kunci : Ekstrak Daun Kersen, Edwardsiella tarda, antibakteri, LD50

Abstract

Aglutinasi merupakan reaksi antibodi dan antigen dapat digunakan sebagai dasar untuk mengembangkan test kit untuk mendeteksi keberadaan bakteri yang menyebabkan penyakit ikan dengan cepat. Untuk memahami apakah reaksi antigen-antibodi E. ictaluri dapat digunakan untuk mendeteksi penyakit Enteric Septicemia of Catfish (ESC), sebuah penelitian telah dilakukan dari September hingga Desember 2018. Antibodi diproduksi dari kelinci yang diimunisasi E. ictaluri yang tidak aktif (dosis bertingkat, periode 4 minggu, 0,5; 1; 2; dan 3 ml / minggu masing-masing). Serum Antibodi dipanen pada minggu ke-5. Antibodi itu kemudian digunakan untuk mendeteksi keberadaan E. ictaluri pada ikan yang terinfeksi. Sepuluh ikan patin diinfeksi dengan E. ictaluri aktif (1 ml, 106CFU / ikan, injeksi intraperitoneal). Tiga hari setelah infeksi dan ikan menunjukkan gejala penyakit ESC, isolat E. ictaluri diambil dengan mengekstraksi hati dan ginjal dan kemudian isolat tersebut ditanam di media TSA selama 24 jam. Isolat E. ictaluri kemudian disebarkan dalam kaca slide, dan dicampur dengan antibodi E. ictaluri. Reaksi antigen-antibodi itu dipelajari. Reaksi positif ditunjukkan oleh adanya butiran, sedangkan reaksi negatif tidak menghasilkan butiran-butiran seperti pasir. Hasil penelitian menunjukkan bahwa antigen E. ictaluri yang dikumpulkan dari ikan yang terinfeksi ESC bereaksi positif terhadap antigen E. ictaluri dan butiran terbentuk dalam waktu 1 menit setelah antigen - antibodi direaksikan. Antibodi E. ictaluri menunjukkan reaksi negatif terhadap antigen bakteri lain. The accuracy of Rapid Diagnostic Test based antigen – antibody agglutination is identic with the result of E. ictaluri detection using PCR. Berdasarkan data yang diperoleh, dapat disimpulkan bahwa Uji Aglutinasi Slide dapat digunakan sebagai Tes Diagnostik Cepat untuk mendeteksi penyakit ESC pada ikan patin.

Abstract

Pengendalian penyakit dapat dilakukan dengan cara meningkatkan daya tahan tubuh ikan melalui penambahan jamu fermentasi (kencur, temulawak, dan kunyit) ke pakan, karena pakan dengan jamu fermentasi memiliki beberapa keunggulan, seperti dapat meningkatkan nafsu makan, daya cerna, pertumbuhan, dan tahan terhadap penyakit. Tujuan penelitian ini adalah untuk mendapatkan dosis terbaik dari penambahan jamu fermentasi pada pakan guna meningkatkan status kesehatan ikan jambal siam. Metode yang digunakan adalah metode eksperimen dengan menggunakan Rancangan Acak Lengkap (RAL) satu faktor lima taraf perlakuan, dan tiga ulangan, sehingga didapatkan 15 unit percobaan. Adapun perlakuan yang digunakan adalah Kn: Kontrol negatif (pemberian pakan tanpa jamu fermentasi dan tanpa uji tantang A. hydrophila), Kp: Kontrol positif (pemberian pakan tanpa jamu fermentasi tetapi diuji tantang A. hydrophila), P1: Pemberian pakan mengandung jamu fermentasi dosis 200 ppm dan diuji tantang dengan A. hydrophila, P2: Pemberian pakan mengandung jamu fermentasi dosis 250 ppm dan diuji tantang dengan A. hydrophila. P3: Pemberian pakan mengandung jamu fermentasi dosis 300 ppm dan diuji tantang dengan A. hydrophila. Ikan dipelihara selama 46 hari dalam aquarium berukuran 40 x 30 x 30 cm dengan padat tebar 1 ekor/3 L air. Uji tantang dengan bakteri A. hydrophila kepadatan 108 CFU/mL sebanyak 0,1mL/ekor dilakukan pada hari ke-32. Setelah uji tantang ikan dipelihara kembali hingga hari ke-46. Hasil penelitian menunjukkan bahwa dosis jamu fermentasi yang terbaik untuk ditambahkan ke pakan adalah 250 ppm, yang ditandai dengan total eritrosit 233 x 104 sel/mm3, nilai hematokrit 38,66%, kadar hemoglobin 9,73 g/dL, dan pertumbuhan bobot mutlak 20,19 g. Hasil pengukuran kualitas air selama penelitian suhu berkisar antara 27-29,2oC, pH 6-7, DO 4 - 4,34 mg/L, dan NH3 0,028-0,048 mg/L.

Abstract

Most of the Kampung Harapan residents are farmers, namely farming corn, sweet potatoes, chilli, fruit, vegetables (such as spinach and kale) and fish farming. This village is prospective to be used as rearing sites for aquaponic systems. Aquaponic cultivation technology can be developed in limited area and water, and it also can increase business efficiency through the utilization of nutrients from the uneaten fish meal and the metabolism wastes of fish for vegetables, so it could be able to produce fish and vegetables optimally on narrow land and limited water sources, including in urban areas. Through the fish rearing with aquaponic systems, fish farmers will get two advantages in one production stage. This activity was carried out through discussion and practice, which the activity took place in the village office and in the home yard of the fish farmer group in Harapan Village. Extension activities involve fish farmers, plantation farmers and several students. The results of the activities showed that skills and knowledge of fish farmers increase regarding aquaponics fish farming and making pellets mix with gingger. Plastic fish ponds as a pilot project produce high yields of catfish and leafy vegetables, so it can be an alternative income for fish farmers.

Abstract

Koto Pangean village has an area of 15 ha, the topography of the area is flat and hilly land with a high of 300 m up to from marine water surface. Type of soil type is black on the top layer and yellow on the bottom layer. Width of the waters of Koto Pangean Village is 2,003.25 ha, has been used for fisheries activities ranging from 2.50 ha in lakes, 1.8 ha swamps, rivers of 180 ha and 10 ha for a fishpond. Several kinds of fish are reared in this village such as African catfish (Clarias batrachus), tilapia (Oreochromis niloticus) and carp (Cyprinus carpio). The main problems faced by fish farmers in this village are: the occurrence of fish deaths caused by decreasing of water quality, accumulation of organic matter, the number of fish attacked by disease and increasing number of pathogenic microorganisms that attack fish, and high commercial fishmeal prices which results in high operational costs for fish farming. To overcome these problems, knowledge innovations are carried out to farmer groups by conducting counselling about; 1. the use of probiotics to improve the soil quality and pond water, 2. Method to prevent fish from disease attacks, 3. how to make pellets by utilizing local ingredients of good quality. The results of the activities that have been carried out show that there is an increase in knowledge and skills of fish farmers, especially being able to increase catfish production with probiotic technology and decreasing operational costs for providing fish meal as a result of the increasing the income and welfare of fish farmer groups.

Abstract

The livelihoods of the people of Tangkerang Utara Village are; in agriculture, plantation, animal husbandry, self-employment and fisheries. In the fisheries sector, there are two groups of fish cultivators (POKDAKAN) in this kelurahan, namely; POKDAKAN Cahaya Utara Mandiri and Dolphin Farm, which conduct fish farming in soil and cement ponds and cages. The main problems faced by POKDAKAN are the limited availability of water intended for fish cultivation and the wate rquality that is not suitable for catfish growth. Throught his program of guided villages, knowledge and technology is given on the technique of applying probiotics to cultivation media to improve wate rquality and minimize water use. The method used in the community service activities of the Assisted Village is the method of lecturing, discussion and direct practice in the pools owned by POKDAKAN members. The results of the evaluation of 23 extension participants showed that 20 people (86.96%) had never known about the application of probiotics in cultivation media, while 2 people (8.70%) had known about the application of probiotics and 1 person (4.34%) had apply it to catfish farming ponds. After participating in this training activity, there was an increase in knowledge about the application of this probiotic by 69.57%. Meanwhile, 22 participants understood the dosage and frequency of feeding, and all participants had already known the criteria for healthy fish seeds. The partners' assessments of the implementation of this activity are: 52.17 strongly agree that this probiotic application can be done independently; 52.17% strongly agree that probiotic applications can improve water quality; 60.87% strongly agreed that it could be an alternative livelihood; 65.22% strongly agree to speed up the maintenance period.

Abstract

Chicken manure is an organic material that is abundant in Mentangor Village which has been used as fertilizer when fertilizing ponds, however the continuous use of fertilizers can reduce the quality of the bottom soil and pond water. One of the technological innovations to improve the quality of chicken manure is fermentation using EM4 to produce bokashi fertilizer. Bokashi fertilizer can increase the abundance of phytoplankton and zooplankton which are useful as natural fish food so that they can reduce the use of artificial feed, which in turn can reduce the operational costs of fish farming. Community service activities are carried out for the purpose of innovating knowledge and skills of fish farmers regarding techniques for making bokashi fertilizer from chicken manure and techniques for making fish feed in the form of pellets to increase cultivated fish production. This activity is carried out by lecturing, discussion and practice methods, which involve fish cultivator groups and Kukerta students. The results of community service activities show the high enthusiasm of farmers towards the activities carried out, resulting in increased knowledge, insight and skills of farmer groups. Thus, fish farming activities can be carried out by farmers by reducing operational costs for pond management and provision of feed so as to produce high fish production and increase the income and welfare of farmer groups.
