Abstract

Tyrosinase inhibitors are utilized as preservatives in the food industry and skin-lightening agents in the medical and cosmetic sectors. However, there has been little progress in clinical trials owing to challenges such as low bioavailability, significant skin irritation, and instability. Hence, the objective of this study was to evaluate the inhibitory activity of 3’,4’,5’-trimethoxychalcones through in vitro, molecular docking and molecular dynamics studies targeting tyrosinase. Five 3’,4’,5’-trimethoxychalcones (1-5) were evaluated their biological activity against tyrosinase for the first time. Compounds 4 and 5 were excellent inhibitory activity against tyrosinase with IC50 values of 1.9 and 1.7 μm compared with kojic acid and ascorbic acid. Isovanillin and catechol moieties are vital in this present study. This result was supported with molecular docking by shaping interaction in the catalytic site with histidine residues and the stability evaluation of the inhibitor-protein complexes using molecular dynamics simulation. The lipinski’s rules showed a fit with two potential inhibitors (4, 5). Therefore, 3’,4’,5’-trimethoxychalcones possessing isovanillin and catechol parts in the B ring are promising candidate for further study as tyrosinase inhibitors by evaluating their efficacy in vitro and in vivo.

Abstract

Abstrak: Penerapan pembelajaran kontekstual telah menjadi tren dominan dalam pendidikan di berbagai negara. Metode pembelajaran ini telah berkembang dengan beragam pendekatan yang menekankan kerjasama lintas disiplin, domain, dan wilayah. Penelitian ini bertujuan untuk memberikan kontribusi signifikan bagi peneliti dan pendidik dalam Pendidikan Sains dengan melakukan analisis bibliometrik global tentang pembelajaran kontekstual, menggunakan karakteristik publikasi sebagai dasar. Penelitian ini menggunakan metode analisis bibliometrik dengan mengambil database Scopus dan berhasil mengumpulkan 782 artikel yang diterbitkan dari tahun 2010 hingga 2022. Analisis bibliometrik yang dilakukan menggunakan alat seperti VOSviewer dan Microsoft Excel. Hasil penelitian menunjukkan bahwa Bialocerkowski, A. dan Olson R., adalah penulis yang paling banyak dikutip dalam penelitian tentang pembelajaran kontekstual dalam Pendidikan Sains. Artikel yang paling banyak dikutip adalah "Interprofessional Education in Allied Health: A Systematic Review". Berdasarkan peta kolaborasi antar negara, Amerika Serikat tampaknya menjadi pusat kolaborasi paling signifikan dalam penelitian tentang pembelajaran kontekstual dalam Pendidikan Sains. Selain itu, jurnal "Journal of Research in Science Teaching" terlihat menonjol dalam publikasi Pembelajaran Kontekstual dalam Pendidikan Sains, berdasarkan jumlah kutipan dan kuartil (Q1). Studi bibliometrik ini juga menemukan korelasi antara kata kunci yang digunakan oleh penulis, antara lain "Pendidikan Profesional", "Pendidikan Sains", "Pendidikan", "Konteks", dan "Pembelajaran Kontekstual". Temuan-temuan ini menyoroti pentingnya penelitian multidisiplin dan interdisiplin dalam memahami pembelajaran kontekstual dalam konteks pendidikan dan mengadvokasi untuk memasukkan konteks geografis yang lebih luas dalam penelitian tentang pembelajaran kontekstual.Abstract:  Implementing contextual learning has become a dominant trend in education across various countries. This learning method has evolved with diverse approaches emphasizing collaboration across disciplines, domains, and regions. This research aims to contribute significantly to researchers and educators in Science Education by conducting a global bibliometric analysis of contextual learning, using publication characteristics as the basis. This research uses bibliometric analysis method by taking the Scopus database and successfully collecting 782 articles published from 2010 to 2022. Bibliometric analysis is conducted using tools such as VOSviewer and Microsoft Excel. The analysis results show that Bialocerkowski, A. and Olson, R., is the most cited author in research on contextual learning in Science Education. The most cited article is "Interprofessional Education in Allied Health: A Systematic Review". Based on the map of collaboration between countries, the United States appears to be the most significant center for collaboration in research on contextual learning in Science Education. Furthermore, the journal "Journal of Research in Science Teaching" stands out in the publication of Contextual Learning in Science Education, based on the number of citations and quartile (Q1). This bibliometric study also found a correlation between the keywords used by authors, including "Professional Education," "Science Education," "Education," "Context," and "Contextual Learning". These findings highlight the importance of multidisciplinary and interdisciplinary research in understanding contextual learning in the educational context and advocating for including broader geographic contexts in research on contextual learning.

Abstract

Project-based learning is a global trend, widely adopted across countries. Project-based learning has developed diversely by emphasizing interdisciplinary, cross-domain, and regional collaboration. To provide in-depth resources for researchers and educators on project-based learning in chemistry experiments, this study aims to make a bibliometric analysis on the topic of project-based learning globally using publication characteristics analysis. The database used in this study used Scopus and obtained 5900 articles from 2018-2023. Bibliometric analysis was analyzed using VOSviewer and Microsoft Excel. The results of the bibliometric analysis show that Boone, C.D. is the author with the highest number of citations on the research topic, Project-based learning in a chemistry experiment. The United States is the most productive country in researching this topic, with 1351 articles. The most influential journal with the highest number of articles is the Journal of Physics Conference Series, with 243 articles. The major contextualized learning literatures in this study tend to fall into broad groups such as Project-based Learning, Education, and STEM Education. These findings suggest the need for multidisciplinary and cross-disciplinary studies on project-based learning research on chemistry learning and advocate for the inclusion of project-based learning research in education from a broader geographical context.

Abstract

Cancer poses a substantial global health challenge. Colorectal cancer (CRC) is the second leading cause of cancer-related mortality after lung cancer and is associated with high mortality rates worldwide. Chalcones have attracted significant interest because of their diverse biological properties, including potential anticancer effects. In this study, five 3′,4′,5′-trimethoxychalcones (1-5) were tested against HCT-116 colon cancer cells using an MTT assay for the first time. Molecular docking was conducted to predict molecular interactions targeting three proteins (tubulin, EGFR, and CDK2). Among the five, four compounds (1, 3, 4, and 5) exhibited strong inhibitory activity against HCT-116 colon cells, with IC50 values < 10 µM. Compounds 1-5 showed potency as drug candidates based on the Lipinski rules and pharmacokinetic profiles using SwissADME and pkCSM online tools. Moreover, molecular docking was performed on compound 5 against three protein targets (tubulin, EGFR, CDK2) with binding affinities of -7.4, -7.3, and -8.5 kcal/mol, respectively, and showed major H-bond interactions. Therefore, these results suggest that compound 5 could be a potential inhibitor to be developed in future studies, both in vitro and in vivo, to understand its inhibition mechanism and efficacy.

Abstract

Isovanillic acid and its derivatives serve as precursors in the synthesis of EGFR tyrosine kinase inhibitors, which are used to treat cancer cell lines. A crucial step in this process is the nitration of isovanillic acid through nucleophilic aromatic substitution, resulting in 6-nitroisovanilic acid and its derivatives, which act as intermediates for forming a quinazolinone ring. However, this study revealed that direct nitration of isovanillic acid derivatives led to unexpected products, such as 3-hydroxy-4-methoxy-2,6-dinitrobenzoic acid (1) and 4-(3-(2-methoxy-4-nitrophenoxy)propyl)morpholine (4). Additionally, the optimal conditions for etherification of 2 with N-(3-chloropropyl)morpholine to produce 3 involved using Cs2CO3 in DMF and refluxing for 7 hours, achieving an 89% yield. All synthesized compounds were characterized using NMR spectroscopy, and mass spectrometry was employed for two compounds (3, 4). Compound 1 represents the first report of direct nitration of isovanillic acid. Compound 4 was synthesized for the first time from 3 through a one-pot process involving hydrolysis and decarboxylation, followed by nitration at carbon C-1 without metal catalysis, as confirmed by a NOESY 1D experiment. Moreover, the application of 4 could hold promise for future advancements in medicinal chemistry.
