Abstract

Enzim termostabil dari bakteri termofilik merupakan enzim yang sangat potensial untuk mengatasi kendala teknis industri yang berhubungan dengan proses pada suhu tinggi. Salah satu sumber enzim adalah mikroorganisme termofilik yang banyak terdapat pada sumber air panas. Indonesia merupakan negara kepulauan yang banyak mempunyai sumber air panas, salah satunya adalah Kabupaten Solok. Penelitian ini mencakup skrining dan identifikasi bakteri penghasil enzim termostabil selulase dan amilase dari sumber air panas Bukit Kili Ketek di Kabupaten Solok. Suhu air panas adalah 52°C pada bulan Juni 2009 dan pH air adalah 8. Kultur murni yang diperoleh adalah sebanyak 22 kultur Dari hasil pewarnaan gram, diperoleh sebagian besar bakteri air panas adalah jenis bakteri streptobacilli (basilus) gram positif, hanya dua isolat yang merupakan gram negatif yaitu isolat S1B dan S5B. S2A adalah isolat yang mempunyai aktifitas selulase paling tinggi dibandingkan dari isolat yang lain karena mempunyai indeks zona bening selulase lebih tinggi yaitu 2,6. Isolat S2A juga menghasilkan amilase tinggi dengan indeks zona bening 2,5. Persen identitas bakteri air panas S2A yang diperoleh adalah 97% dengan Anoxybacillus flavithermus strain AE3.

Abstract

ABSTRAK. Studi ini bertujuan untuk mengidentifikasi keragaman gen FSH bagian ekson 2 menggunakan enzim restriksi TasI pada sapi Pesisir. Sampel darah yang digunakan yaitu 78 sampel darah dengan dua macam yaitu 13 jantan serta 65 betina yang berumur 1-5 tahun dari sapi pesisir yang didapat dari BPTUHPT Padang Mengatas. Protocol genomic DNA Purification system Kit dari Promega digunakan untuk mengisolasi DNA sampel darah. Amplifikasi gen FSH dilakukan dengan menggunakan teknik PCR atau Polymerase Chain Reaction. Enzim TasI digunakan untuk meretriksi produk amplifikasi. Hasil penelitian gen FSH bagian exon 2 pada sapi pesisir yaitu diperoleh terdapat dua macam genotip. Dua macam genotip tersebut yaitu heterozigot (+/-) serta homozigot tidak terpotong (-/-), untuk jantan berturut turut yaitu 0,38 dan 0,62 beserta frekuensi alel (+) dan alel (-) yaitu 0,19 dan 0,81, sedangkan untuk betina yaitu berturut-turut 0,43 dan 0,57 beserta frekuensi alel (+) dan alel (-) yaitu 0,21 dan 0,79. Sebaran genotip gen FSH|TasI exon 2 pada populasi Sapi Pesisir sifatnya polimorfik (beragam) dengan frekuensi alel yaitu kurang dari 0,99.Identification of Exon 2 FSH Gene Diversity Using TasI Restriction Enzymes in Coastal CattleABSTRACT. This study aims to identify the diversity of the exon 2 FSH gene using the restriction enzyme TasI in Coastal cattle. The blood samples used were 78 blood samples of two types, namely 13 males and 65 females aged 1-5 years, from coastal cattle obtained from BPTUHPT Padang Atas. The protocol genomic DNA Purification System Kit from Promega was used to isolate DNA from blood samples. The amplification of the FSH gene was carried out using the PCR or Polymerase Chain Reaction technique. The TasI enzyme was used to restrict the amplification product. The study results on the exon two portion of the FSH gene in coastal cattle showed two types of genotypes. The two types of genotypes are heterozygous (+/-) and untruncated homozygous (-/-); for males, 0.38 and 0.62, respectively, and the frequencies of allele (+) and allele (-) are 0.19 and 0. .81, while for females, it was 0.43 and 0.57, respectively, and the frequencies (+) and allele (-) were 0.21 and 0.79. The FSH|TasI exon two gene genotype distribution in the Coastal Cattle population is polymorphic (various) with an allele frequency of less than 0.99.Keywords: FSH Gene, Pesisir Cattle, TasI Enzyme

Abstract

The purpose of this study was to identify genetic diversity in the early exon 10 of the FSHR gene or follicle stimulating hormone receptor in Pesisir cattle. The blood samples used were 70 blood samples from female Pesisir cattle aged 2-5 years which were obtained from BPTU-HPT Padang Mengatas, West Sumatera. The isolated DNA was then amplified using a pair of primers L and R with PCR technique which produced a fragment with a length of 847 bp. The services of 1st Base Singapore are used for sequencing the amplified product. The results of the research on the identification of the early exon 10 FSHR gene in Pesisir cattle were found that there were 5 SNPs at positions --53T>C, +17A>G, +650C>T, +706A>C and +707 ins>A in some introns 9 to exon 10 first part. The results showed that the genotypic frequency of the Pesisir cattle population was in Hardy-Weinberg imbalance and polymorphisms were found in the early exon 10 FSHR gene diversity.

Abstract

The aims of this study were to detect Jembrana antigen with polyclonal antibody and to describe antigen distribution in the Bali cattle organ that positively infected with Jembrana disease. Spleens, lungs, and livers were harvested from 10 naturally infected Bali cattle whose infection was confirmed through positive Polymerase Chain Reaction (PCR) result for Jembrana virus. Immunohistochemistry test was performed using polyclonal antibodies produced in rabbits. The results showed that cells infected by Jembrana viruses displayed positive reaction with a reddish brown color. Immunohistochemistry methods using polyclonal antibodies can detect Jembrana antigens in the spleen, liver, and lung with the highest average detection score (P0.05) was found in spleen, followed by liver and lungs. There was significant difference in the distribution of Jembrana antigens between the spleen, liver, and lungs with spleen having the highest antigen density.

Abstract

Water offers many advantages that are essential for the preservation of human existence. Excessive quantities of water, such as flooding, can negatively impact human beings. The land utilization within a watershed (drainage area) plays a crucial role in the frequency and impact of flooding events. Instances of land use changes that resulted in flooding happened in the Bang Sub-watershed, located in Malang Regency. A comprehensive and up-to-date examination of flood management solutions is required. This study seeks to offer technical suggestions for flood management in the Bang Sub-watershed due to land-use alterations, utilizing the HEC-RAS software. Integrated treatment solutions are provided to mitigate the potential damage caused by future flooding. The flood simulation yielded recommendations for mitigating the impact of future floods. These recommendations include the construction of a retarding basin in the upper part of the river with a capacity of 1.4 million m3, as well as the construction of a 654 m long river embankment that is 50 cm. The alteration of the land leads to a decrease in the cross-sectional capacity of the river at the mouth of the Bang River, resulting in annual flooding occurrences. A non-technical approach to enhance the quality of the watershed is implementing reforestation measures to decrease the runoff coefficient.

Abstract

This paper attempts to provide an overview of the main findings and conclusions of the research conducted. The primary objective of this study is to provide a systematic, accurate, and impartial description of how the public participates in the oversight and growth of tourism attractions on Tabalinge Island, located in the Morotai Island geographical area. The research methodology employed in this study is qualitative descriptive research, which involves the collection of data through observations, interviews, and documentation. The findings of this study indicated that the general public did not engage directly in the initial planning stage of the management and development of Tabalinge Island as a destination for tourists. Rather, their involvement was observed to be limited to providing feedback and suggestions during public consultations held after the initial planning stage. This lack of direct engagement from the general public may have resulted in disconnect between the tourism attractions offered on Tabalinge Island and the preferences and needs of potential tourists. Additionally, it highlights the importance of incorporating community input and involvement in future management and development plans to ensure a more inclusive and sustainable tourism destination. During the subsequent implementation phases of management and service provision, which followed a top-down approach encompassing both tangible and intangible aspects, in the phase of evaluation and monitoring, there is restricted public participation in preventive measures aimed at mitigating security and order disruptions. This involvement primarily entails the dissemination of information, with the public acting as passive recipients of government updates regarding surveillance efforts. However, opportunities for public feedback are not provided.

Abstract

Pelaksanaan kegiatan pendampingan kelompok pengrajin batik di Kabupaten Jember bertujuan untuk meningkatkan ekonomi masyarakat khususnya kelompok usaha pengrajin batik. Proses pembelajaran yang diberikan kepada kelompok pengrajin batik di Jember dimulai dari strategi produksi, pengemasan, dan pemasaran. Pengrajin batik didorong dapat mengembangkan usaha dengan memanfaatkan teknologi informasi dan komunikasi berbasis internetisasi. Pengrajin batik disarankan dapat mengikuti berbagai kegiatan kegiatan sertifikasi kompetensi membatik yang diadakan oleh berbagai pihak yang kompeten. Kegiatan Pengabdian Kepada Masyarakat yang dilaksanakan Fakultas Ekonomi Universitas Jember ini telah membuka wawasan kelompok pengrajin sebagai mitra akan pentingnya menjaga kualitas produk melalui pengemasan produk batik sehingga produk batik mempunyai nilai jual tinggi dan mitra memperoleh pengetahuan dan keterampilan dalam pembukuan usahanya dan pemasaran melalui e-commece dalam menghadapi era digital 4.0 yang semakin kompetetif dalam dunia usaha.

Abstract

Perkembangan teknologi informasi telah mengubah paradigma pendidikan, termasuk dalam hal penilaian siswa. E-raport, sebagai pengganti rapor konvensional, menawarkan efisiensi dan kemudahan dalam pengelolaan data nilai. PKM ini bertujuan untuk mengkaji pemanfaatan Google Data Studio dalam memvisualisasikan data e-raport di SMAN 1 Gamping. PKM ini akan mengevaluasi efektivitas Google Data Studio dalam menyajikan informasi yang relevan dan mudah dipahami oleh guru, siswa, dan orang tua. Pelaksanaan PKM ini menggunakan metode demontrasi kepada guru SMAN 1 Gamping sebanyak 20 orang. Hasil PKM menunjukkan bahwa Google Data Studio efektif dalam memvisualisasikan data e-raport dan memberikan informasi yang bermakna bagi pengguna. Guru dapat dengan mudah mengidentifikasi tren kinerja siswa, membandingkan prestasi antar siswa, dan mengambil keputusan yang tepat berdasarkan data. Siswa dan orang tua juga dapat memantau perkembangan belajar siswa secara lebih transparan dan terstruktur.
