Abstract

The purpose of this study was to investigate the effect of α-tocopherol in tris aminomethane extender on the quality of Madura cattle semen during storage at room temperature. Fresh semen was collected from Madura cattle bull using artificial vagina. The semen with individual motility of sperm more than 70% was used in this study. Semen was diluted with tris-aminomethane extender supplemented respectively by 0.0 mM, 0.5 mM, 1.0 mM and 1.5 mM α-tocopherol. Data were analyzed using analysis of variance. The results showed that the addition of α-tocopherol using egg yolk diluent tris aminomethane could not maintain the quality of Madura cattle semen motility, viability, abnormality and membrane integrity of spermatozoa (P>0.05) during storage at room temperature. The conclusion of the study was the addition of the antioxidant α-tocopherol into tris aminomethane egg yolk on the quality of semen had no influence (P>0.05) on individual motility, viability, abnormality and membrane integrity of spermatozoa after dilution during storage at room temperature.   Keywords: Semen, α-tocopherol, Madura bull

Abstract

Artificial insemination (AI) in cattle using Ongole Cross Breed liquid semen with cauda epidydymal plasma-2 (CEP-2) diluent+0.6% Bovine Serum Albumin (BSA) aimed to improve the genetic quality of livestock with high sperm motility on beef cows. Addition of BSA as extracellular cryoprotectant to support egg yolk function to prevent membrane damage caused by cold shock during cold storage. This research was conducted in Tumpang, Pakis, and Singosari Sub-district, Malang Regency using 38 cows that inseminated using frozen semen of Limousine and Simmental breeds as control (19 cows) and liquid semen with CEP-2 + 0.6% BSA as a treatment (19 cows). The reproductive parameters were Non Return Rate (NRR), Service per Conception (S/C) and Conception Rate (CR). Data were analyzed descriptively and continued by chi-square test. The results showed that insemination using frozen and liquid semen did not affect the success rate of AI in beef cows. The NRR43-63 value of frozen was 89.40% and liquid semen was 78.94%; the CR value of frozen semen was 84.21% and liquid semen was 57.89%; and the S/C value of frozen semen was 1.21 and liquid semen was 1.42. The conclusions of the study were the values of NRR43-63, CR and S/C of frozen higher than liquidsemen.

Abstract

This study aimed to determine the accuracy of various thawing duration and temperatures to the quality of Simmental bull frozen semen. The experimental design used was a randomized group design with five treatments and ten replications, then analyzed using Chi-Square to compare the expected percentage of motility is 40%, and total motile spermatozoa are 10 million/straw value of SNI. A further test is carried out with Duncan's Multiple Range Test (DMRT) if there is a difference. The thawing treatments were 30 seconds at 37˚C water temperatures (T0) according to SNI and at 15 seconds (T1), 30 seconds (T2), 45 seconds (T3), and 60 seconds (T4) with 28˚C tap water temperatures. Variables observed involve motility, viability, abnormalities, concentration, and total motile spermatozoa. The data were analyzed using Analysis of Variance (ANOVA) with a significant difference in variance (P<0,05). The results showed that the different duration and temperatures of thawing influence motility (P<0,05) and the total motile spermatozoa, while the viability, abnormalities, and concentration had no influence (P>0,05). The motility and the total motile spermatozoa reach the highest results at the tap water temperatures T3 (P<0,05), while the lowest results were in T1 (P<0,05). It can be concluded that the different duration and temperatures of thawing influence motility and total motile spermatozoa. The best quality of Simmental bull frozen semen can be obtained with thawing using tap water at 28℃ temperatures for 45 seconds.

Abstract

The purpose of this research was to evaluate the use of onion extract (Allium cepa L) diluted in Ringer’s Dextrose base extender on the semen quality of Peranakan Etawah (PE) buck. Fresh onion was grinded and filtered with filter paper. The filtrate was then inactivated by heating it at 56oC for 10 min to avoid the negative or detrimental effect of its biological activity. The onion filtrate was added to the tube containing Ringer’s–dextrose solution with the concentration of 0%, 1%, 2% and 3% (v/v). Semen was collected with artificial vagina from a healthy selected PE buck reared at the Teaching Farm Laboratory of the Animal Husbandry Faculty Brawijaya University. The semen was diluted with prepared extender to rich the sperm concentration of 75x10 millions/ml. The semen parameters were observed after 1 and 2 hours during storage at room temperature. The results showed that concentration of onion extract (OE) affected the individual sperm motility (P<0.05), sperm viability (P>0.05) and sperm abnormality (P<0.05) during semen evaluation at 1 and 2 hours following the semen dilution time. The highest quality of semen following the dilution was shown by the dilution containing 2% of OE (2 ml/100 ml extender). It was concluded that it was possible to use the OE to improve the semen quality during storage at room temperature. A further study of the use of OE should be conducted to explore more positive effect of OE when it is used in combination with standard semen extender and other technique semen storage such as cold storage or freezing process.   Keywords: semen quality, onion extract, Ringer’s dextrose

Abstract

ABSTRACT: Antibody against progesgerone was produced from serum of local rabbit following low dose of progesterone injection: While a control group (Control; n=5) was injected with Freund's adjuvant solution in aquadest, the treatment groups were either firstly injected with progesterone conjugated to Freund's Adjuvant (P--CFA, 150 p.l : 150 pl) or progesterone conjugated to Freund's Adjuvant and bovine serum albumin (P-CFA-BSA; 135 p;l : 150 tt1 : 15 gl). Twice boostering injections were adminstered using incomplete Freund's Adjuvant on day 14 and 52 after first immunization. Weekly bleeding for serum collection were done from 1 week following first booster immunization to week 10, Using ELISA technique it was shown that the antibody titer to progesterone after first and second booster immunization in the P-CFA groug was higher than Control- and P-CFA-BSA groups. Abstract

The aims of this study are to 1) determine the causes of conflict between the Sidomulyo hamlet community and the Baluran National Park management; 2) find opportunities for conflict resolution through empowering peasant women. This research was carried out from April to June 2022. The method used is descriptive qualitative by direct field survey and interviews with Baluran National Park managements and Sidomulyo Hamlet farmers. Key informants were chosen using the purposive sampling method followed by snowball sampling. The data required includes the dynamics of conflict, the variety of conflicts, conflict actors, sources of conflict, and opportunities to empower women farmers. Based on the research data on the sources of conflict between the Sidomulyo community and the management of Baluran National Park, there are four different sources of conflict: differences in perceptions, knowledge, values, and values and differences in interests. Three resolutions are applied: Peacekeeping, Peacemaking, and Peacebuilding. In the Peacebuilding resolution empowerment programs is chosen because there is a big potential in for women to do entrepreneur activities. Women farmer empowerment can be done through entrepreneuship. The conservation village model through micro-credit assistance and the formation of women's farmer groups. The aim is to increase livestock yield products and the economy, income, and food security.

Abstract

Since the 1960s, or for four generations, farmers who live close to Baluran National Park have been grazing or raising beef cattle extensively illegally. Since there is not enough forage for beef cattle, their presence in the Baluran National Park area interferes with the preservation of wildlife. This is brought on by the shortage of forage. Farmers continue to use the traditional rearing method, which was self-taught and is not supported by systematic recording. As a result, there is a high risk of inbreeding, difficult calving, and disease contracting, particularly in cases of cattle collapses and outbreaks of FMD in even hoofed animals like cows. The absence of livestock extension staff in the region contributes to farmers' lack of understanding about reproduction, particularly estrus detection and illness prevention. The Animal Husbandry and Fisheries Service, Situbondo Regency, and the Community and Livestock Research Group (CLRG) Team of the Faculty of Animal Science, Universitas Brawijaya, Malang, have worked together to aid in the form of Indigofera sp. seeds as forage for animal feed, forage processing technology into silage, the concept of communal pens (groups) for farmers, and facilitate the formation of the Lembu Brawijaya. Direct observation of beef cattle and interviews with farmers and Baluran National Park personnel will be the research methods employed in the project. The outcomes of this study can serve as a guide for figuring out the best course of action for increasing livestock productivity and intensive rearing. It is also expected that cattle will not grazed in the Baluran National Park area and restore its function as a biosphere conservation area.  Farmers can monitor their cattle better when they raise them intensively, which is supported by systematic recording and disease prevention.

Abstract

Efforts to improve the quality of small and microenterprises (SMEs) depend not only on the availability of internal resources but also on innovation and worldwide regulation dynamics. The triple helix concept has become an alternative solution for improving the quality of SMEs, which consists of academicians, governments, and businesses (SMEs). Muza is an SME that is being fostered by lecturers in the Faculty of Animal Science Universitas Brawijaya in Blitar District; this SME produces smoked salted eggs and has become one of the featured products based on Blitar's Service of Fishery and Livestock Department. The communication between the academician and the Muza Management-Government on the development program was implemented separately. This research aimed to develop development strategies to increase the quality of Muza smoked salted eggs. The research was conducted in Blitar District in July 2022. The research used an explanatory method to identify problems in the field and categorize the problems into groups using the SWOT method. SWOT components are formed using in-depth interviews with the planner and implementer of the strategy, which consists of five people. The subjects were given a closed questionnaire that consisted of SWOT components. The SWOT analysis results show that the total IFE score is 4 and the EFE score is 4.45. The IE matrix shows that the empowerment strategy is on the “V” cell, which has a "hold and maintain" strategy. Thus, the programs given were not efficient, and strategic planners need to focus on product quality improvement. The design of GMP and HACCP systems is strongly recommended.
