Abstract

Diabetes mellitus (DM) is a metabolic disease which is becoming the first number of health problem in Indonesia, based on the results of the Basic Health Research of Ministry of Health in 2013. One of the diabetes complications affected by high levels of blood glucose is diabetic ulcers wich 85% the number of cases was overed by amputation as the result of improper handling. Herbal treatments could be an alternative treatment of diabetic ulcers, one of them is binahong plant. The results of previous studies have shown the ability of ethanolic extract of binahong leaf in accelerating wound healing in diabetic rats. This study would be a continued study to test five kinds of binahong leaf fractions (FDB) with gradual solvent polarity in accelerating wound healing in diabetic ulcer. Wound healing parameters observed were percentage of wound healing from the wound diameter contraction. The test begins with ethanolic extract of binahong leaf fractionation to produce fractions of hexane (FHDB), chloroform (FKDB), ethyl acetate (FEADB), and ethanol (FEDB). Water fraction (FADB) was obtained from ethanolic leaf extract residues of binahong leaf extraction. Each fraction activity was tested topically twice daily on dorsal of test animals that created wounds using a punch biopsy 5 mm diameters. Grouping of test animals were divided into 13 groups with normal control group (non diabetes), negative control (diabetes + topical application of biocream®), positive control (diabetes + topical application of madecassol), and 10 diabetes groups with topical application of 5 kinds of binahong leaf fractions with each fractions consist of a concentrations of 5% and 10% with biocream® as a vehicle. Diabetic parameters measured include blood glucose levels (KGD) and weight lost percentage (PB%) in day 1 and 10 during a 10-day treatment. The results showed the influence of KGD in the condition of diabetes on wound healing rats diabetic ulcers which topically treated with binahong leaf fractions. FADB 10% has significant differences potention in wound healing process in diabetic rats macroscopically that accelerates wound contraction compared with negative and positive control groups (p<0,05).

Abstract

Endophytic fungi are potential microorganisms in lead compounds investigation as research objects for antibacterial and antifungal drug candidates. These microorganisms are able to produce various secondary metabolites, moreover they can even be regulated according to the needs of researchers. This is an advantage of endophytic fungi with good adaptability response. This study aims to explore the antibacterial, antifungal and antioxidant activity of endophytic fungi from the branch of the Melinjo tree (Gnetum gnemon L.) by qualitative analysis. This study resulted in 13 potential endophytic fungi extracts to be explored further for their compounds. Based on the results of the TLC test of the fungal extracts with 10% vanillin in H2SO4 as a reagent, it showed that the dominant compounds were alkaloids and polyphenols. The antibacterial activity test of extracts of the endophytic fungi was carried out by the disc diffusion agar method against Staphylococcus aureus and Pseudomonas aeruginosa bacteria, while antifungal agents were used for Candida albicans and Aspergillus clavatus.. Meanwhile, the qualitative test of antioxidants from fungal extracts using the DPPH spray reagent showed positive results of 8 fungal extracts. Screening of endophytic fungi from melinjo represented a source of endophytes potential as antibacterial, antifungal and antioxidant to be developed in exploration of secondary metabolites from natural products of microbial.

Abstract

Diabetes Mellitus (DM) can lead to various complications, both macrovascular and microvascular. Diabetes education interventions play a crucial role in DM management, as they help improve patients' knowledge and adherence to diabetes management strategies. This study involved three sessions: a pre-test, an educational session, and a post-test. Among 53 respondents, the proportion of participants with a high level of knowledge increased from 30 (56.6%) to 43 (81.1%). Meanwhile, those with a low level of knowledge decreased from 10 (18.9%) to 1 (1.9%). This increase in knowledge levels demonstrates the effectiveness of the education provided. The average pre-test knowledge score was 69.06, compared to a post-test score of 86.23, demonstrating a significant improvement. Wilcoxon test analysis showed a significance value of <0.05. Thus, the "Patuhi, Obati dan Kendalikan Diabetes" education program for participants of the Chronic Disease Management Program (Prolanis) significantly enhanced participants' knowledge levels.
