Abstract

Aedes aegypti is known as the main vector of dengue fever in Indonesia which causes a high number of deaths and become an outbreak every year. This disease is generally controlled by breaking the life cycle of mosquitoes with restricted breeds and using synthetic insecticides. So far biological agents from natural enemies and natural pathogens have been developed to anticipate the negative effects of synthetic insecticides. Bacillus thuringiensis is well known as entomopathogenic bacteria that can control various insect pests of cultivated plants and vector insects, such as Ae.aegypti.  B. thuringiensis can form endospores that produced toxin to Ae.aegypti, while the advatages are safe against non-target organisms such as natural predators and parasites, safe for mammals and humans and do not pollute the environment. Effective duration of B. thuringiensis in infecting Ae. aegypti is an important thing to be studied, moreover it is important to know the effectiveness of the expired formulation on the market and the residual effect. This ensures that the use of B. thuringiensis remains safe and does not create new problems such as Ae. aegypti  resistance. The experimental research was used bioassay method, toxicity test was performed B. thuringiensis expired and non expire as comparison at concentration 0 ppm (control), 0,3 ppm; 0.1 ppm; 0.07 ppm; 0.05 ppm; 0.03 ppm; 0.01 ppm. The parameters observed were mortality for 48 hours of toxicity test that results were indicated by LC 50, LC90 and recidual values. The results obtained were LC50 and LC formulations B.thuringiensis expired 0.305 ppm and 1.513 ppm much higher than the non expire LC50 and LC formulations of 0.0363 ppm and 0.8971 ppm. The recidual effect of expired B. thuringiensis on larvae Ae. Aegypti was only effective until day 12, whereas B. thuringiensis  that  non expire formulation was effectively until day 14.Keywords : Ae. aegypti, B. thuringiensis, mortality, recidual values, expired formulation

Abstract

Penelitian mengenai pengaruh suhu pengeringan polyhedra terhadap efektivitas Helicoverpa armigera Nuclear Polyhedrosis Virus (HaNPV) subkultur dalam mengendalikan populasi Spodoptera litura Fabricus telah dilakukan. Penelitian dilakukan dengan metode eksperimental dengan menggunakan Rancangan Acak Lengkap (RAL) faktor tunggal dengan 5 taraf perlakuan (HaNPV cair dan HaNPV yang dikeringkan pada suhu 4oC, 25C) dengan empat kali pengulangan. Hasil penelitian menunjukkan bahwa berat badan maksimum larva S. litura instar tiga mencapai 0,967 g terjadi pada larva yang terinfeksi HaNPV yang dikeringkan pada suhu 4C dan pertambahan berat badan mencapai 0,732 g/ekor/hari pada larva yang terinfeksi HaNPV yang dikeringkan pada suhu 4C

Abstract

Ikan komet (Carrasius auratus auratus Linnaeus, 1758) merupakan ikan hias yang banyak dibudidayakan. Salah satukendala dalam budidaya ikan komet adalah serangan penyakit yang disebabkan oleh bakteri Aeromonas hydrophila.Penelitian ini bertujuan untuk mengetahui virulensi bakteri A. hydrophila pada ikan komet. Penelitian menggunakanmetode deskriptif eksploratif. Data hasil dianalisis secara deskriptif. Kepadatan bakteri yang digunakan pada uji perendaman sebesar 1,58×10⁷ cfu/ml dan 5,08×10¹¹ cfu/ml. Pengamatan tanda klinis dan mortalitas ikan dilakukan setiap12 jam selama 96 jam. Hasil penelitian menunjukkan bahwa ikan komet yang terinfeksi A. hydrophila dengan kepadatanberbeda akan menunjukkan tanda klinis dengan tingkat keparahan yang berbeda. Bakteri A. hydrophila dengan kepadatan5,08×10¹¹ cfu/ml mengakibatkan kemunculan tanda klinis yang parah dan bakteri dengan kepadatan 1,58×10⁷ cfu/mlmengakibatkan tingkat kelangsungan hidup (survival rate) yang rendah. Pada penelitian ini bakteri A. hydrophila yangmenginfeksi ikan komet memiliki tingkat virulensi yang tinggi.

Abstract

Bacillus and Lactobacillus are bacteria that can be used in dry-stock probiotics in vannamei shrimp feed. In this study, it was performed to encapsulate Bacillus and Lactobacillus probiotic bacteria in several carrier materials, including rice flour, talc, and maltodextrin. The purpose of this study was to determine the type of carrier material that can provide the best probiotic character in the encapsulation process which will then be used in vannamei shrimp feed. The method used in this study is experimental method. The study was conducted with the preparation stage and the initial probiotic characterization test. Furthermore, Bacillus and Lactobacillus probiotic encapsulation process used spray drying method to produce powder inoculum, as well as advanced characterization test of bacteria that have been encapsulated. The parameters observed were the viability, resistance to temperature, pH and bile salts. Data were analyzed using ANAVA and Duncan Multiple Range Test. The results showed that all types of carrier materials used maintained the probiotic character of Bacillus and Lactobacillus after the encapsulation process. This can be seen from the character of probiotics that are in good category and deserve to be used as a probiotic agent after encapsulation process in the material of rice flour, maltodextrin, and talc.
