Abstract

A new antiplasmodial compound, 2-(3H-diazirine-3-yl)benzaldehyde (1), has been isolated from the Papuan marine sponge Xestospongia sp. The structure elucidation of compound 1 was determined by spectroscopic evidences including UV, IR, MS, 1D and 2D-NMR analysis. Compound 1 showed moderate antiplasmodial activity against Plasmodium falciparum with IC50 value of 1.08 x 10-6 mM. DOI:http://dx.doi.org/10.15408/jkv.v4i1.6735

Abstract

Discovery of new antibacterial agent to treat oral disease caused by pathogenic oral bacteria is an attracted focus to more pay attention of researcher. The tropical natural product is the promising sources of new bioactive as antibacterial compounds. The epiphyte plant of Sarang Semut (M. pendans) empirically has been used as a drug to treat various diseases. Our previous research found that ethyl acetate extract of Sarang Semut inhibited the bacteria growth of Enterococcus faecalis. This study aims to isolate, structure determination and antibacterial activity evaluation of active compound against Streptococcus mutans ATCC 25175. The isolation by combination chromatography on normal and reverse phase resulted an antibacterial compound 1. Based on the analysis of the spectroscopic data including IR, FTIR, 1H-NMR, 13C-NMR, 2D-NMR as well as by mass spectroscopic (MS) and compare to reported data, structure of compound 1 was suggested as terpenoid type diterpene derivative. Antibacterial activity evaluation by Kirby-Bauer method of compound 1 against S. mutans ATCC 25175 showed inhibition zone values were 17.8, 14.5 and 11.1 mm at 10000, 5000 and 1000 ppm, respectively, while the MIC and MBC values were 18.125 ppm with MBC of 2500 ppm, respectively.

Abstract

Antioksidan merupakan senyawa yang menghambat atau menunda reaksi oksidasi molekul dengan cara menghambat proses inisiasi atau propagasi reaksi oksidasi berantai. Struktur kimiawi antioksidan, sumber radikal bebas, dan sifat fisiko-kimia sediaan sampel yang berbeda dapat memberikan hasil uji aktivitas antioksidan yang beragam. Oleh karena itu, diperlukan suatu metode analisa aktivitas antioksidan yang selektif untuk suatu jenis sampel tertentu. Studi perbandingan metode uji aktivitas antioksidan DPPH, FRAP, dan FIC telah dilakukan terhadap asam askorbat (AA), asam galat (AG), dan kuersetin. Ketiga metode uji antioksidan dibedakan berdasarkan pada jenis mekanisme reaksinya, sedangkan sampel standar antioksidan dipilih berdasarkan struktur polihidroksi atau polifenol yang umumnya mewakili dasar struktur antioksidan bahan alam. Metode uji aktivitas antioksidan terhadap radikal DPPH ditemukan paling efektif dan efisien diantara ketiga metode uji yang digunakan dengan nilai IC50 berturut-turut 1,27; 2,44; dan 2,77 mg/L untuk AG, kuersetin dan AA. Adapun metode FIC terbukti paling tidak efektif dan efisien karena sensitivitasnya yang sangat rendah dan daya kelatnya lebih kecil dari 20%. Korelasi antara metode uji untuk semua standar antioksidan terbukti sangat tinggi (R>0,98), khususnya antara FRAP dan DPPH. Hal ini mengindikasikan adanya keterkaitan sangat kuat antara daya hambat radikal bebas dengan potensial reduksi senyawa polihidroksi (polifenol) terhadap ion besi. Secara umum kedua metode ini sangat dimungkinkan bisa saling menggantikan satu sama lain.

Abstract

Pada penelitian ini dilakukan pengujian aktivitas penangkal radikal bebas superoksida secara in vitro terhadap senyawa kompleks polimerik [Mn(salen)N(CN)2]n, baik dengan metode non-enzimatik maupun enzimatik. Aktivitas penangkal radikal bebas superoksida yang didapatkan dari kedua metode ini dinyatakan dengan IC50.  Melalui metode non-enzimatik riboflavin-nitrobluetetrazolium (Rb-NBT), radikal superoksida yang dihasilkan dari penyinaran riboflavin, bereaksi dengan nitrobluetetrazolium sebagai indikator dan senyawa kompleks, lalu serapan NBT tereduksi diukur pada panjang gelombang 560 nm. Dengan metode non-enzimatik ini, dihasilkan nilai IC50 yang tinggi, konsisten, dan standar deviasinya sangat kecil, yaitu 3,4 ± 0,4 µM. Sedangkan secara enzimatik, radikal superoksida yang dihasilkan dari reaksi antara xantin dan xantin oksidase ditangkap oleh sitokrom C (sebagai indikator) dan senyawa kompleks, kemudian sitokrom C tereduksi diukur serapannya pada panjang gelombang 450 nm. Melalui metode enzimatik McCord-Fridovich (McCF) ini, dihasilkan beberapa nilai IC50 kompleks [Mn(salen)N(CN)2]n yang tidak konsisten, yang disebabkan oleh tidak berfungsinya enzim. Dengan demikian, metode non-enzimatik Rb-NBT lebih mudah dan lebih akurat untuk digunakan sebagai teknik penentuan aktivitas penangkal radikal anion superoksida dibandingkan dengan metode enzimatik.

Abstract

Biji kedelai merupakan sumber protein berkualitas tinggi, oligosakarida, serat makanan, mineral dan fitokimia terutama isoflavon. Isoflavon dan protein yang merupakan komponen utama dalam kedelai terbukti mempunyai beberapa efek yang menguntungkan. Isoflavon mempunyai efek menurunkan berat badan melalui peningkatan kadar hormon pencernaan Kolesistokinin (KSK). Kedelai varietas Detam 1 adalah kedelai unggulan berkualitas tinggi berdasarkan Surat Keputusan Menteri Pertanian November 2008, dan sebagai pembanding adalah kedelai lokal varietas Wilis yang banyak ditanam oleh petani di Indramayu. Tujuan penelitian ini adalah untuk menguji dan membandingkan kandungan makronutrisi (air, protein, lemak dan karbohidrat) kedelai dan tempe, mengetahui jenis ekstrak yang mengandung kadar isoflavon yang tertinggi. Pengujian kadar air dengan metoda Thermogravimetri, protein dengan metoda Kjeldahl, lemak dengan metoda Soxhlet, dan karbohidrat dengan metoda Antron. Kadar Isoflavon yang terdapat dalam ekstrak metanol, fraksi n-heksana, fraksi etil asetat, fraksi n-butanol, fraksi air dan ekstrak protein dari kedelai dan tempe dari Detam 1 dan Wilis diperiksa menggunakan High Performance Liquid Chromatography (HPLC). Hasil penelitian menunjukkan kadar protein yang paling tinggi terdapat dalam biji kedelai Detam 1 (41,82%). Proses fermentasi meningkatkan kadar air dari biji kedelai namun menurunkan kadar protein, lemak dan karbohidrat. Kadar isoflavon paling tinggi terdapat dalam fraksi etil asetat, kadar Daidzein tertinggi dalam fraksi etil asetat biji kedelai Detam 1 (Daidzein 0,669%). Ekstrak metanol tempe kedelai Wilis juga memiliki kadar isoflavon yang tinggi (Daidzein 0,580%, Genistein 0,576%). Ekstrak metanol tempe kedelai Wilis diharapkan mempunyai manfaat sebagai terapi untuk penurunan berat badan di masa mendatang.Kata Kunci: Kedelai Detam 1, kedelai Wilis, makronutrisi, isoflavon, HPLC

Abstract

Substansi organik dalam tanah dapat menyebabkan terbentuknya kompleksdengan ion logam. Asam humat yang merupakan salah satu substansi tanahberperan dalam transportasi ion-ion tersebut, sehingga dapat membentukkompleks ion logam melalui reaksi pembentukan kompleks. Penelitian inibertujuan menentukan konstanta stabilitas kompleks humat-besi danpengaruhnya dengan penambahan urea pada variasi konsentrasi terhadapkestabilan kompleks tersebut. Asam humat yang diperoleh dengan cara ekstraksidari tanah, mempunyai bobot molekul (BM) 17,987, spekturm inframerahmenunjukkan gugus karboksil, fenolik, dan karbonil. Dan spektrum ultravioletvisibelmemberi rasio warna (E4/E6) sebesar 5,47. Konstanta stabilitas humat-besisebesar 0,046. Penambahan urea akan mempengaruhi harga konstanta stabilitaskompleksnya. Penambahan urea yang akan memperbesar nilai konstantastabilitas tersebut, mempunyai nilai minimum sebesar 0,04 N.Kata Kunci: Urea, Asam humat, Kompleks humat-besi

Abstract

Two flavonoid compounds, quercetin (1) and quercetin-3-O-glucoside (2) have been isolated from the bark of Sonneratia alba (Lythraceae). Chemical structure of both compounds were determined on the basis of spectroscopic data and comparison with those spectra data previously reported. Compound 1 and 2 exhibited antibacterial activity against Gram-positive bacteria, Staphylococcus aureus and Streptococcus mutans with MIC values of 51.2; 48.8; 72.5; dan 100.7 µg/mL respectively.DOI :http://dx.doi.org/10.15408/jkv.v0i0.3151

Abstract

Background: Enterococcus faecalis (E. faecalis) is a gram positive oral pathogen that reported at the main agent infection of endodontic treatment. Its activities are influenced by the virulence factors facilitating the interaction process between agents with host cells. Like aggregation substance, cytolysin, extracellular superoxide, gelatinase, hyaluronidase, sex pheromones, and surface adhesions molecules. Plant extracts are reported as the material antibacterial as well as E. faecalis in pathogenesis of endodontic infections. Purpose: Purpose of this study was to analyse of sarang semut extracts (Myrmecodia Pendens Merr. & Perry) towards sensitivity of E. faecalis. Method: This research used the methanol extract of sarang semut, E. faecalis ATCC 29212, and fosfomycin also chlorhexidine as the positive controls. Whereas, Bradford protein method was measured the concentration of the surface protein of E. faecalis and active component of the sarang semut extract. Result: Generally, the sarang semut extract possessed low sensitivity toward E. faecalis (≤ 13 mm), but on the concentrations of 100 µg/ml and 75 µg/ml better than inhibition of other concentrations, round 10.6-11.6 (mm). Specifically, on 100 µg/ml has indicator the minimal bactericidal concentration (MBC) on E. faecalis. Whereas minimal inhibition concentration (MIC) on the concentration of 3,125 µg/ml. Conclusion: Based on MBC and MIC assay, the extract of sarang semut has potential effects to adherence growth of E. faecalis, mainly on the highest concentration 100 µg/ml also MIC on 3,125 µg/ ml.

Abstract

Background: Porphyromonas gingivalis (P. gingivalis) is a pathogenic bacteria present in the oral cavity involved in the pathogenesis of chronic periodontitis and biofilm. This mass of microorganisms represents one of the virulent factors of P. gingivalis which plays an important role as an attachment initiator in host cells. Sarang semut is a natural material possessing the ability to inhibit the growth of P. gingivalis. Purpose: This study aims to analyze the effect of sarang semut extract on the formation of P. gingivalis biofilm. Methods: The study used methanol sarang semut extract and P. gingivalis ATCC 33277 and phosphomycin as a positive control. Treatment was initiated by means of culturing. Biofilm test and P. gingivalis biofilm formation observation were subsequently performed by means of a light microscope at a magnification of 400x. Results: The formation of P. gingivalis biofilms tended to increase at 3, 6, and 9 hours. Results of the violet crystal test showed that concentrations of 100% and 75% of the sarang semut extract successfully inhibited the formation of P. gingivalis biofilm according to the incubation time. Meanwhile, the sarang semut extracts at concentrations of 50%, 25%, 12.5%, and 6.125% resulted in weak inhibition of the formation of P. gingivalis biofilm. The biofilm mass profile observed by a microscope tended to decrease as an indicator of the effects of the sarang semut extract. Conclusion: Sarang semut extract can inhibit the formation of P. gingivalis biofilm, especially at concentrations of 100% and 75%. Nevertheless, phosphomycin has stronger antibiofilm of P. gingivalis effects than those of the sarang semut extract at all of the concentrations listed above.

Abstract

The routine and long term use of chemicals to maintain oral and dental health have the potency to result in the emergence of side effects; therefore another strategy is needed as an alternative such as using antimicrobial agents extracted from plants. The purpose of this study is to review the effectiveness of lime (Citrus aurantifolia) peel extract as an antibacterial in preventing biofilm formation. Biofilm is a component consisting of bacteria in a self-produced polymeric matrix, attached to an inert surface, alive, and can survive because of its ability to capture nutrients and withstand adverse environmental conditions. Lime peel contains flavonoids which are the largest group of polyphenol compounds that can work as antioxidants and antibacterial by denaturing bacterial cell proteins and damaging bacterial cells. Flavonoids can also inhibit glucosyltransferase (GTF) activity of Streptococcus mutans to prevent biofilm formation. Lime peel extract inhibits the formation of the activity of the enzyme Streptococcus mutans. As a conclusion lime peel extract contains compounds with therapeutic potential and has the effect of inhibiting the formation of the activity of the enzyme Streptococcus mutans so that it can be used to inhibit the formation of biofilms.
