Abstract

Mefenamic acid is classified as a Biopharmaceutical Classification System (BCS) Class II drug. Crystallisation techniques can significantly influence the critical properties of mefenamic acid crystals, an essential consideration in the pharmaceutical industry. This study investigates the effect of recrystallisation on the morphology and particle size of mefenamic acid crystals using ethyl acetate as the solvent.  Recrystallisation was conducted using both conventional and ultrasonic-assisted methods. The yield was calculated, and characterization was performed to determine the crystal properties, shape, and particle size using polarized microscopy, X-ray Diffraction (XRD), and Fourier Transform Infrared Spectroscopy (FTIR). The results showed that conventional recrystallisation yielded 25.078%, while ultrasonic-assisted recrystallisation at 50 W, 60 W, 70 W, and 80 W yielded 43.47%, 47.71%, 51.90%, and 50.20%, respectively. Skewness and kurtosis values for the average crystal length and width indicated a normal distribution. XRD analysis revealed that the diffraction peak intensity of conventionally recrystallised crystals became narrow. In contrast, the diffraction peak intensity of ultrasonically assisted crystals broadened. FTIR results demonstrated that the recrystallisation process did not alter the fundamental chemical structure of mefenamic acid. Recrystallisation methods influence crystal shape and size, with ultrasonic-assisted techniques producing crystals of smaller average size than those obtained through conventional methods.

Abstract

UV-Visible spectrophotometry (UV-Vis) is one of the most widely used instrumental analytical methods in the fields of pharmacy, food science, and analytical chemistry, particularly for determining the concentration of active compounds in various types of samples. The working principle of UV-Vis spectrophotometry is based on the ability of a compound to absorb light at a specific wavelength, resulting in an absorbance value proportional to the concentration of the substance in solution. This method is considered efficient due to its relatively simple operation, short analysis time, and lower cost compared to other instrumental methods such as HPLC, AAS, or GC-MS. In addition, UV-Vis spectrophotometry can be applied to a wide range of compounds containing chromophoric groups, making it suitable for analyzing herbal products that consist of complex natural matrices. This study aims to review the application of UV-Vis spectrophotometry in the analysis of active compounds in herbal products through a literature review of ten relevant and recent scientific journals. Based on the results of the review, it was found that UV-Vis spectrophotometry is widely used to analyze compounds such as flavonoids, vitamin C, caffeine, paracetamol, and oxalates derived from natural sources such as tea, fruits, vegetables, as well as traditional herbal preparations such as jamu and supplements. In addition, this method is also applied in solubility studies of active compounds within drug delivery systems such as Self-Nanoemulsifying Drug Delivery Systems (SNEDDS), analytical method validation, and evaluation of compound stability in final formulations. Several studies also demonstrated the ability of UV-Vis spectrophotometry to detect the presence of synthetic contaminants in herbal products quickly and simply. From the overall findings, it can be concluded that UV-Vis spectrophotometry plays a significant role in the development, formulation, and quality testing of herbal-based products. This method is not only practical and efficient but also capable of producing reliable quantitative data that can be applied in various research and industrial contexts.

Abstract

Noni fruit containing polysaccharides, has the potential to be developed into pharmaceutical products such as SPF syrup. The content of noni fruit polysaccharides is known to increase the proliferation of T lymphocyte cells, the regulation of the immune system molecularly, especially with regard to antibodies (IgG), CD4+, and CD8+, is still not widely studied. The aim of this study was to assess the impact of SPF syrup on the modulation of immunoglobulin G (IgG) and T lymphocytes. The study was conducted on female Wistar rats aged 8-9 months, which were given SPF syrup orally at doses of 25, 50, and 100 mg/kgBB for 31 days. Mice were vaccinated against hepatitis B every week. Blood samples were taken on day 31 to measure IgG levels by the ELISA method and T cells (CD4+ and CD8+) by flow cytometry. The results showed that SPF administration had a significant effect on IgG levels (p<0.05), and there were no significant changes in T lymphocyte cells (CD4+, CD8+) (p>0.05). Histopathological examination showed that long-term induction of hepatitis B caused damage to the liver and kidney, but SPF did not affect the condition of these organs. SPF that contains polysaccharides can be an innovative component in immunostimulant formulations.

Abstract

Indonesia is a tropical country with high sun exposure every year. Sunlight has radiation that is bad for skin health, especially ultraviolet (UV) rays. Without protection, these rays have the potential to cause skin damage such as erythema, hyperpigmentation, premature aging, and even skin cancer. The use of sunscreen is an effort to protect the skin because it has a protective power expressed as Sun Protection Factor (SPF), the SPF value can be determined by using the UV-Vis spectrophotometric method. The purpose of this study is to explain the application of the UV-Vis spectrophotometric method in determining the SPF value in natural extract-based sunscreen preparations. The methodology used was a literature study of the last 10 years. The results of this study indicate that the application of UV-Vis spectrophotometric method is very effective and efficient to measure the sun protection value (SPF) of natural extracts. The analysis showed significant variation in SPF values of various plant extracts, with kecombrang (Etlingera elatior) flower extract recording the highest value (106.8) in the ultra of sunscreen protection category, while jejuru leaf extract showed effectiveness in the minimal protection category (3.8478). This study makes an important contribution in exploring the potential of local materials for sustainable sunscreen formulations.

Abstract

Mefenamic acid, classified as a Biopharmaceutics Classification System (BCS) Class II drug, exhibits high permeability but low dissolution rates, limiting its bioavailability. This study aims to enhance the dissolution rate of mefenamic acid through crystal structure modification using fast cooling recrystallization with three distinct solvents: acetone (AMA), ether (AMEt), and ethyl acetate (AME). The recrystallized crystals were characterized using advanced techniques, including polarized microscopy, X-ray Diffraction (XRD), Fourier Transform Infrared (FTIR) spectroscopy, and Differential Scanning Calorimetry (DSC), alongside solubility and dissolution testing. Recrystallization yielded crystals with varied morphologies and sizes, achieving high crystal yields of 92.66% (AMA), 85.54% (AMEt), and 94.59% (AME). FTIR analysis revealed that AME crystals exhibited stronger OH bonding, indicated by the highest peak intensity. XRD diffractograms demonstrated distinct peak patterns at specific 2θ positions for AME crystals, suggesting internal structural changes (polymorphism) while maintaining identical chemical composition. DSC thermograms further confirmed variations in melting points between recrystallized crystals and pure mefenamic acid. Dissolution tests revealed that recrystallized mefenamic acid exhibited significantly improved solubility and dissolution rates compared to its pure form. These findings highlight that rapid cooling recrystallization can produce mefenamic acid polymorphs with optimized dissolution properties, potentially enhancing its pharmacological performance and also provides a promising approach to improving the bioavailability of poorly soluble drugs, offering significant implications for pharmaceutical formulation development.
