Abstract

Hipertensi merupakan suatu keadaan seseorang ketika terjadi peningkatan tekanan darah sistolik ≥ 140 mmHg dan/atau tekanan darah diastolik ≥ 90 mmHg. Pengobatan jangka panjang hipertensi menimbulkan beberapa efek samping sehingga diperlukan suatu alternatif pengobatan untuk hipertensi, misalnya dengan protein isolat biji melinjo. Penelitian ini bertujuan untuk mengetahui efek antihipertensi dari protein isolat biji melinjo pada tikus hipertensi. Tikus wistar diinduksi dengan prednison 1,5 mg/kgBB dan NaCl 2% selama 7 hari kemudian diberi perlakuan. Terdapat 7 kelompok perlakuan, kelompok K merupakan tikus normal, kelompok K(-) adalah tikus hipertensi tapi tidak diberi perlakuan, kelompok K(+) adalah tikus hipertensi dan diberi kaptopril 2,5 mg/kgBB pada hari ke-8, kelompok P1, P2, P3, dan P4 merupakan tikus hipertensi dan masing-masing diberikan protein isolat biji melinjo dengan dosis 5 mg/kgBB, 10 mg/kgBB, 20 mg/kgBB, dan 30 mg/kgBB pada hari ke-8. Tekanan darah diukur sebelum dan setelah diberi perlakuan tiap 1 jam selama 5 jam. Data dianalisis secara statistik menggunakan uji Kruskal Wallis dan Mann. Dari hasil penelitian menunjukkan bahwa protein isolat biji melinjo tidak dapat menurunkan tekanan darah tikus model hipertensi.   Kata kunci: Antihipertensi, Gnetum gnemon, Protein Isolat, Tekanan Darah

Abstract

[ENGLISH] Protein was one of macromolecule in seed which played an important role in the process of germination. Storage protein in seed was hydrolyzed to amino acid which are then used in the translation process for a genetic message interpretating and protein building in pre-germination process. Pre-Germination was the first step of germination that have many biochemical changes. The objectives of this research was to know biochemical characteristic of soluble protein and protein pattern of melinjo seed during pre-germination. Two varieties of melinjo seed Gentong and Kerikil was used in this research with three stages of treatment (initial seed, 3 weeks warm stratification seed and 1 month seed seedling). Seed samples are extracted to determine total soluble protein and protein pattern with 15 % SDS-PAGE. The results showed that total soluble protein have a change different at each stage of the treatment. Protein patterns analysis indicates band protein with molecular weight 66 kDa and 14 kDa change during pre-germination. Protein yang tersimpan pada biji dihidrolisis menjadi asam amino yang kemudian digunakan dalam proses translasi untuk menginterpretasi suatu pesan genetik dan membentuk protein dalam proses awal perkecambahan. Awal perkecambahan adalah tahapan pertama dari proses perkecambahan dimana perubahan biokimia telah banyak terjadi. Tujuan dari penelitian ini adalah mengetahui karakteristik perubahan kandungan protein terlarut dan pola protein biji tanaman melinjo pada awal perkecambahan. Penelitian ini menggunakan 2 varietas biji melinjo yaitu varietas Gentong dan Kerikil dengan tiga tahap perlakuan yaitu biji awal, biji setelah perlakuan stratifikasi hangat selama 3 minggu dan biji setelah 1 bulan semai. Sampel biji melinjo diekstrak untuk menentukan kandungan protein terlarut dan pola protein menggunakan 15% SDS-PAGE. Hasil penelitian menunjukkan bahwa kandungan protein terlarut mengalami perubahan yang berbeda pada setiap tahap perlakuan. Analisa pola protein menunjukkan adanya 2 jenis pita protein yang mengalami perubahan pada awal perkecambahan yaitu protein dengan berat molekul 66 kDa dan 14 kDa. Abstract

ABSTRACT Green Deli Rose Apples (Syzygium samarangense) has several beneficial properties in terms of usability, cultivation, health, and economy. The intensive development of green deli rose apples plants is needed given increasing demand, needs, and has bright prospects. One of the efforts to develop green deli rose apples plants is through handling plant propagation. Propagation of green deli rose apples can be done in two ways, namely generative and vegetative. Generative propagation requires a relatively long time, while vegetative propagation have difficult to growth in rooted. Shoot-cuttings is one of the vegetative propagation that has been chosen because it is easier to root than the old parts of the plant. The use of plant growth regulators (PGR) is needed to stimulate green deli rose apples shoot-cuttings rooting. Alternative PGR is giving shallot extract and coconut water as a substitute for synthetic auxin which has a relatively expensive price. The purpose of this study was to determine the combination of the effect of the concentration shallot extract and coconut water, as well as to find out the concentration of shallot extracts and coconut water which was most influential on the growth of shoot-cutting of green deli rose apples. The extract shallot concentration used is control, 0,5%, 1,0%, and 1,5%. The coconut water concentration used is control, 20%, 30% and 40%. Data obtained next analyzed and tested using DMRT α 5%. The result showed te best treatment combination for concentration shallot extract of 0,5% and coconut water of 20%. Keywords: greend deli rose apples, shoot-cutting, shallot extract, coconut water ABSTRAK Jambu air madu deli hijau (MDH) (Syzygium samarangense) memiliki beberapa sifat yang menguntungkan baik dari segi kegunaan, budidaya, kesehatan, maupun ekonomi. Pengembangan tanaman secara intensif diperlukan mengingat permintaan dan kebutuhan yang semakin meningkat, serta memiliki prospek yang cerah. Salah satu usaha pengembangan tanaman yaitu melalui penanganan perbanyakan tanaman. Perbanyakan dapat dilakukan dengan dua cara yaitu generatif dan vegetatif. Perbanyakan secara generatif membutuhkan waktu yang relatif lama, sedangkan perbanyakan vegetatif memiliki kendala sulit membentuk perakaran. Stek pucuk merupakan salah satu perbanyakan vegetatif yang banyak dipilih karena lebih mudah berakar dibandingkan bagian tanaman tua. Penggunaan zat pengatur tumbuh (ZPT) diperlukan untuk merangsang perakaran stek pucuk jambu air MDH. Alternatif ZPT yaitu pemberian ekstrak bawang merah dan air kelapa sebagai pengganti auksin sintetis yang memiliki harga relatif mahal. Penelitian ini bertujuan untuk mengetahui kombinasi pengaruh konsentrasi ekstrak bawang merah dan air kelapa terhadap pertumbuhan stek pucuk jambu air MDH, serta mengetahui konsentrasi ekstrak bawang merah dan air kelapa yang paling berpengaruh terhadap pertumbuhan stek pucuk jambu air MDH. Konsentrasi ekstrak bawang merah yang digunakan antara lain kontrol, 0,5%, 1% dan 1,5%. Konsentrasi air kelapa yang digunakan antara lain kontrol, 20%, 30%, dan 40%. Data yang diperoleh selanjutnya dianalisis dan diuji menggunakan DMRT α 5%. Hasil penelitian menunjukkan kombinasi perlakuan terbaik untuk konsentrasi ekstrak bawang merah dan air kelapa adalah konsentrasi 0,5% dan air kelapa 20%. Abstract

Usaha untuk meningkatkan produksi tomat pada lahan tercekam genangan dapat dilakukan dengan menguji varietas-varietas tomat yang sudah beredar terhadap cekaman genangan. Uji dilakukan dengan mengamati parameter pertumbuhan dan hasil beberapa varietas tomat. Penelitian ini bertujuan untuk mengetahui toleransi beberapa varietas tomat terhadap cekaman genangan. Metode yang digunakan pada penelitian ini adalah Rancangan Acak Kelompok (RAK) faktorial dengan 3 kali ulangan. Varietas tanaman tomat merupakan faktor pertama dengan 5 taraf yaitu a) V1 Sakina F1, b) V2 Tora, c) V3 Rewako F1, dan d) V4 Mawar. Sedangkan faktor yang kedua adalah cekaman genangan dengan 3 taraf, yaitu a) T1, perlakuan kondisi cekaman genangan 5 cm, b) T2, perlakuan kondisi cekaman genangan 10 cm, c) T3, perlakuan kondisi cekaman genangan 15 cm. Hasil penelitian menunjukkan bahwa varietas Rewako mempunyai karakter lebih toleran terhadap genangan yang ditunjukkan pada parameter rerata tinggi tanaman, ukuran diameter buah, berat buah dan berat tajuk atas tanaman.

Abstract

Invertases (beta-D-fructofuranosidase, E.C. 3.2.1.26) are the key enzymes involved in sucrose metabolism in sugarcane plants. They are highly correlated with sucrose and reducing sugar contents during plant growth. The sugarcane plants have two kinds of invertases, namely neutral invertase (NI) and acid invertase (AI). They have different function in sucrose accumulation. The research aimed to study the role of AI and NI in accumulation of reducing sugar during storage of sugarcane stems. Plant materials of 18-month-old field grown sugarcane of the commercial variety R-579 (wet-land) and M 442-51 (dry-land) were used. Three internodes were sampled to represent immature (internode 1-8/F1), maturing (internode 9-16/F2), and mature (internode 17-24/F3) stem tissues. All tissues were stored for 0-9 days at room temperature (28-32oC) and each day, the samplewas extracted to determine invertase activity, total soluble protein, and sugar contents. This observation was valid for invertase activity expressed on a protein basis. At the initiateharvested (0-3 days), NI had a higher specific activity than AI in the sucrose-accumulating region of the sugarcane stems. Negative significant correlation was found between NI specific activity and sucrose accumulation (r2 = 0.41, P < 0.05). AI showed a higher specific activity after 4 days harvested and had negative correlation with sucrose accumulation (r2 = 0.40, P < 0.05). These results showed that NI could be more  responsible in sucrose hydrolisis than AI at early storage of sugarcane stems.

Abstract

Weather conditions often unstable changed and increase the floods of agricultural land. Land which was originally drought will experience wetness and need variety of a plant that wetness tolerant. This study were aimed to get cassava that wet tolerant and to study the morphological, physiological characters of cassava grown on wetness land. The experiment was conducted based on factorial random plot design that consisting two factors with five replications. The first factor were four varieties namely: V1 = Daun Ganja; V2 = Sawi Ketan; V3 = Kasesat Beracun; V4 = Gajah. The second factor was wet treatment consisting of 2 the condition that were K0 = as control, media with 100% field capacity; K1 = as wet treatment by watering of media maintain on -10 cm from media surface. The difference between treatments tested with Duncan Multiple Range Test (DMRT) with the probability 5%. Wet treatment started when plant aged was 2 months after planting, during eight weeks. The observations on variables based on morphological and physiological characters. Result showed that every varieties give the different response on wet treatment. Best response on wet tolerant variety shown by variety code of V2 (Sawi Ketan) were indicated by plant height, total leaf number produced, stem diameter, fresh weight of root, dry weight of root, fresh weight of shoot, dry weight of shoot, photosynthesis index, stomata conductivity and root volume.

Abstract

Cocrystal is a relatively new solid form of activepharmaceutical ingredient that offers an alternativeplatform in improving physicochemical properties ofactive pharmaceutical ingredients Padrela et al.,2009; Mashhadi et al., 2004. Cocrystal is defined asa stoichiometric multi-component system connectedby non-covalent interactions where all thecomponents neutral and solid under ambientconditions Thakuria et al., 2013. Cocrystal can beconstructed through interaction hydrogen bonding,pi-stacking, and van der Waals forces  Mashhadi etal., 2004. A pharmaceutical cocrystal is composed ofan API and an appropriate coformer as carboxylicacids and amides Qiao et al., 2011.Cocrystallization of active pharmaceutical ingredientis an opportunity for enhancement of importantphysiochemical properties of an activepharmaceutical ingredient without changing itsmolecular structure Maeno et al., 2014.Atorvastatin Calcium (AC), ([(R-(R*,R*)]-2-(4-fluorophenyl)-beta, delta-dihydroxy-5-(1-methylethyl)-3-phenyl-4-[(phenylamino)carbonyl]-1H-pyrrole-1-heptanoic acid), calcium salt (2:1)trihydrate ([C33H34FN2O5]2Ca.3H2O), is considered asone of the most effective of synthetic lipid loweringagent Shete et al., 2010. The drug is orally used toreduce of total cholesterol, low density lipoproteinand triglycerides Anwar et al., 2011. There are 42crystalline structures of AC Shayanfar et al., 2013.However, chance to create AC into another crystalstructure to improving physicochemical properties ofAC is still fully open Chadha et al., 2012.In the present study, we explored cocrystallization ofAC by solvent evaporation method. This study aimedto confirm whether AC was able to form cocrystalwith malonic acid (MA) as coformer. AC-MAcocrystal was prepared by solvent evaporationmethod by using methanol as solvent.Characterization of cocrystal was done by powder Xraydiffractometry (PXRD), differential scanningcalorimetry (DSC), fourier transform infrared (FTIR)spectroscopy and scanning electron microscopy(SEM).

Abstract

Experiments were conducted to determine the relationship between lipase activity and the metabolism of lipids during germination of sesame (Sesamum indicum. L) seed. During the course of germination, concentration of the non-polar lipid fractions and lipase activity increased. The glycolipids and phospholipid fractions were relative stable during early germination, but there were marked changes phospholipid fractions after 5 days germination. In a comparison among lipid fractions Non-polar lipids (NL), Glycolipids (GL), and Phospholipids (PL), there were no significant regressions between the quantitative changes in lipid fractions and lipase activities. It was concluded that the degradation in NL and increase in PL and GL observed during sesame germination were not controlled by the level of lipase activity. Rather, it was suggested that sesame may have another lipase present in the cells where the oil bodies are located that hydrolyzes lipids from the oil bodies during germination. Abstract

Ciplukan (Physalis angulata L.) is one of the plants from the class of herbal plants that is good to be developed because people are starting to choose herbal treatment. The content of ciplukan fruit is phenolic and flavonoid. Thus, technology for enlargement and increasing fruit content with the giberellin (GA3) hormone is needed. GA3 functions to stimulate the formation, development of fruit and increase the biosynthesis of secondary metabolites. This study aimed to determine the influence of the GA3 hormone on the phenolic content and antioxidant activity in ciplukan fruit. The experimental design used RAL 5 treatments: GA3 concentrations of 0 ppm (P0), 15 ppm (P1), 20 ppm (P2), 25 ppm (P3), and 30 ppm (P4). The parameters studied were the number of fruit, fruit size, fresh weight of fruit, total phenolic, total flavonoid, and antioxidant activity (DPPH method) in ciplukan fruit. The results showed that the concentration of 30 ppm (P4) of GA3 was showed the highest values on the number of fruits (33±3.53 g), fruit length (14.23±0.35 mm), width (12.87±0.39 mm), and fresh fruit weight (38.6±3.97 g) without calyx. The concentration of 30 ppm (P4) also affected the phenolic content (2.52±0.23 mgGAE/g), flavonoids (2.43±0.18 mgQE/g), and IC50 value of antioxidant activity (60.59 g/mL). Abstract

L-ascorbyl palmitate (AsA-Pal-Enz) was synthesized by using the immobilized lipase from Aspergillus niger. The antioxidative activity of AsA-Pal-Enz was investigated in coconut oil at different temperatures within a range of 30(RT)-120oC. The effectiveness of AsA-Pal-Enz in coconut oil was monitored by the rate of formation of hydroperoxides, conjugated dienes (K232) and decomposition of hydroperoxides (K270). Samples in storage experiments were periodically removed and analyzed for peroxide value, K232, K270 and radical scavenging activity (DPPHr). Application of AsA-Pal-Enz have markedly reduced the rate of peroxidation in coconut oil during incubation time at RT, 60 and 80oC but at 120oC the rate of peroxidation slightly increased. It has shown that the effectiveness of AsA-Pal-Enz antioxidant in coconut oil was strongly depend on temperature and the time of incubation.
