Abstract

Cardiovascular diseases (CVDs) still contribute as the main cause of mortality and premature mortality worldwide. In Indonesia, CVDs contribute to 35% of the main cause of death in non-communicable diseases followed by diabetes at 6%. The ischemic heart disease and acute ischemic stroke is the main cause of death in Indonesia due to atherosclerosis. Atherosclerosis is a multifactorial cause, with chronic inflammation which causes myocardial infarction and acute ischemic stroke. Research demonstrated that one of the underlying mechanisms of atherosclerosis is inflammation. The current research suggested that inflammation could activate a complex of cytosol proteins, namely nucleotide-binding oligomerization domain-like receptor family pyrin domain containing 3 (NLRP3) inflammasome. The NLRP3 inflammasome is known as a protein sensor as a response to danger signals known as pathogen-associated molecular patterns (PAMPs) or damage/danger-associated molecular patterns (DAMPs). The NLRP3 inflammasome will convert the pro-IL1β and pro-IL-18 into their mature forms which initiates the cell death (pyroptosis) through the cleavage of Caspase-1. This NLRP3 inflammasome has a central role in the initiation and progression of atherosclerosis by affecting the sequences of its cellular and molecular targets. To date, several small molecules and drugs have been identified as NLRP3 inflammasome inhibitory pathways and potential therapeutic agents for atherosclerosis. In this review, we will discuss further the structure of NLRP3 inflammasome and its roles in atherosclerosis, and the potential candidates of the small molecules and drugs targeted in the inflammasome pathway.

Abstract

Infant nutrition and immune protection require the consumption of human milk, with macrophages mainly contributing to immune defense and infant development. This study was performed to investigate the correlation between macrophage cell numbers in human milk and various maternal factors. Thirteen human milk samples were collected from lactating mothers and were evaluated for macrophage cell numbers using a hemacytometer. The recorded questionnaire containing several questions, including age, multivitamin consumption, coronavirus disease 2019 vaccination status, allergy history, and lactation duration, was used to determine the maternal factors. Pearson’s correlation and Mann-Whitney test was employed in statistical analysis, with p-values < 0.05 considered significant. The macrophage cell number reached 54.236 ± 7.456 cells per mL. Notably, a significant correlation was observed between maternal allergy history and the number of macrophage cells in human milk (p = 0.049). Meanwhile, no statistically significant associations were detected among multivitamin consumption (p = 0.833), vaccination status (p = 0.923), and lactation duration (p = 0.236). This study emphasizes the effect of maternal characteristics on the composition and immune properties of human milk. The specific mechanisms underlying these correlations and their potential influence on infant health and development must be explored in future research.

Abstract

Restaurant X is a small and medium enterprises (SME) engaged in the food and beverage sector by offering a variety of Indonesian food menu. SMEs have three main roles, namely as a means to improve the welfare of employees and the community, leveling the economic level of smallholders, and providing foreign exchange to the state. SME efforts as a means to improve the welfare of employees is through the evaluation of compensation. By providing better compensation, employees will be motivated to create a positive and productive work environment. Compensation evaluation is done by identifying the initial salary structure in SME, then analyzed to improve compensation using overlapping, adhered and factor points methods. The analysis showed that the most optimal method is overlapping, with a total salary of Rp137, 224, 291 from the previous Rp129, 850, 000. By using the overlapping method, SME experienced a salary increase of 5.68% or Rp7,374,297.

Abstract

Abstract

Introduction Cell-size area (CSA) becomes the standard parameter routinely tested in vitro for cardiac hypertrophy studies. Thus, staining is an essential tool for this purpose. As reported in a previous study, immunofluorescence staining is an established method for CSA. However, because it is expensive and requires a specialized microscope, e.g., immunofluorescence or confocal microscope, it is not applicable in a laboratory with limited equipment. Wright-Giemsa staining is a standard procedure in hematology laboratories and is inexpensive and convenient. Here, we shared our experience developing a CSA determination protocol using Wright-Giemsa in H9c2 cardiomyoblast. Methods The viability tests were performed on H9c2 to determine the effective dosage of angiotensin II and Irbesartan (standard drug). The H9c2 were divided into three groups: the control group (without either angiotensin II or irbesartan), the negative control (with angiotensin II), and the positive control (with angiotensin II and Irbesartan), triplicate for each group. The cells then are acclimatized overnight, serum-starved for one day, and incubated with angiotensin and irbesartan for 48 hours. Lastly, Wright-Giemsa was observed using a light microscope in three fields. The CSA was determined by three independent observers blindly, statistically different if the p<0.05 using ANOVA ways or Kruskal-Wallis. Results After the H9c2 induced by angiotensin-II 1 μM and Irbesartan 1μM, we found the CSA significantly differed among each group (p<0,0001). The negative control has a higher median and interquartile range (IQR) CSA (10.78 (6.79) um2) compared to the control group (median (IQR) 7.27 (4.91) um2) and positive control (median (IQR) 7.849(5.31) um2). Conclusion It can be concluded that the Wright-Giemsa might help determine the CSA for in-vitro hypertrophic studies.
