Abstract

ABSTRACT. Butanol-HCl4 (Bu-HCl4) and Protein-Precipitation (P-P) are methods that proposed to determine the content of free tannins, protein-bound tannins, and fiber-bound tannins. This study aims to determine the feasibility of tannin measurements by both methods and validation of Analytical Methods (VMA) evaluates specific parameters like accuracy, precision, specificity, and linearity through lab testing. The material used was twenty C. calothyrsus leaves which collected randomly from different trees. The samples were measured by both methods in triplicates for qantification and validation. The obtained data were analyzed by descriptive and t-test to compare both methods. The analysis of VMA shows that the P-P method yields higher accuracy compared to the Bu-HCl method in percentage recovery rates. The quantification of free tannins, protein-bound tannin, tannin fiber, and total tannin by the Bu-HCl4 method versus the P-P method showed significantly higher values (p<0.01) for the Bu-HCl4 method: free tannins (13.44±0.54 vs. 7.11±0.16), protein-bound tannin (2.80±0.44 vs. 1.52±0.20), tannin fiber (5.20±0.45 vs. 1.00±0.21), and total tannin (21.50±0.88 vs. 9.59±0.36). The methode validation result of Bu-HCl4 and P-P methods in the accuracy was different, but the linearity test showed a good response. In conclusion, the tannin quantivication by the P-P and Bu-HCl4 can be applied for C. calothyrsus analysis. The total tannin content was higher if quantified by the Bu-HCl4 method compared to P-P. The accuracy of the P-P found higher than the Bu-HCl4 method. Keywords: Butanol-HCl4, leaves C. calothyrsus, protein-precipitation methods, tannin.

Abstract

Background. The study of the relationship between Length, Circumference, and Volume of the scrotum with Viability and Abnormality of Spermatozoa in Etawa Crossbreed (PE). Goats was carried out from 15 November 2020 until 21 November 2020 at Tani Ternak Sato Guno, Gumelar District, Banyumas Regency, Central Java. Materials and methods. The purpose of this study was to determine the length, circumference, volume of the scrotum with the viability and abnormalities of spermatozoa in Etawa male goats, provide scientific information and expand knowledge in the field of animal husbandry, especially the quality of spermatozoa. This research uses experimental with linear correlation analysis method and linear regression analysis. Results. The results of this study indicate that together the length, circumference, and volume of the scrotum have a very strong relationship (P < 0.01) with spermatozoa viability with a coefficient of determination of 0.825 and a correlation coefficient of r = 0.908. This shows that the viability of spermatozoa is influenced by the length, circumference, and volume of the scrotum by 82.5%, while the remaining 17.5% is influenced by other factors. The results of the next study showed that together the length, circumference, and volume of the scrotum had a very strong relationship (P < 0.01) with spermatozoa abnormalities with a coefficient of determination of 0.918 and a correlation coefficient of r = 0.958. This shows that spermatozoa abnormalities are influenced by the length, circumference, and volume of the scrotum by 91.8%, while the remaining 8.2% are influenced by other factors. It was concluded that the mean length, circumference, and scrotum volume of Etawa Crossbreed Goats were 25.50 ± 1.73 cm, respectively; 15.00 ± 1.28 cm; 85.96 ± 20.39 cm3 with the mean viability and abnormality of spermatozoa were 82.12 ± 5.71% and 4.54 ± 2.35%, respectively. Conclusion. Length, circumference, and volume of the scrotum simultaneously have a very strong relationship with the viability and abnormality of spermatozoa, so that it can be used as a good indicator to estimate the quality of spermatozoa, especially the viability and abnormality of spermatozoa with the respective formulas being Y₁ + = 232,500 - 6,500 X₁ - 3,583 X₂ + 0.212 X₃ and Y₂ += 69,000 + 0.917 X₁ - 3.801 X₂ + 0.103 X₃

Abstract

Background. This research aimed to find out the effect of prolification levels on hematological profile in Batur sheep which includes erythrocyte, leukocytes, and hemoglobin levels. Materials and methods. The research material used 20 ewes of batur sheep with a minimum parity in 2 times and an average age of 3 years, then they were grouped into two groups of 10 each. The first change is low prolification (P1) with JAS = 1 and the second change is high prolification (P2) with JAS records ≥ 2. Grouping is done based on the results of interviews with breeders related to their reproductive history. Blood sampling of 1-2 ml through the jugular vein and poured into the EDTA vacuum tube then stored in the ice box to prevent plumping and damage of blood cells. Examination of hematological profiles is done using a hematology analyzer. Data is processed by using t-test unequal. Results. The results of statistical analysis showed that there was no significant difference (P>0.05) from the level of prolification to erythrocyte and hemoglobin counts, but has a significant difference (P<0.05) to the number of leukocytes in Batur sheep. Conclusion. It’s concluded that the levels of erythrocyte and hemoglobin were relatively similar between prolification levels, but leukocytes levels in low prolific were significantly higher than those of high prolific in Batur sheep.

Abstract

Penelitian ini berjudul Pengaruh Sentrifugasi Gradien Densitas Percoll dan Lama Inkubasi Terhadap Morfometri dan Membran Plasma Utuh Hasil Sexing Spermatozoa Kambing Peranakan Etawah. Tujuan penelitian dilakukan untuk mengkaji interaksi gradien percoll dan lama inkubasi terhadap morfometri dan Membran Plasma Utuh (MPU) hasil separasi seks kambing Peranakan Etawah (PE). Materi yang digunakan untuk penelitian yaitu semen kambing Peranakan Etawah (PE) jantan sebanyak 2 ekor dengan umur 3-4 tahun. Rancangan penelitian dilakukan menggunakan Rancangan Acak Kelompok (RAK) pola faktorial 2 x 3 dengan ulangan frekuensi penyadapan sebanyak 3 kali. Hasil analisis variansi perlakuan yang berpengaruh nyata kemudian dilakukan uji lanjut dengan uji Duncan’s Multiple Range Test (DMRT). Lokasi penelitian berada di Laboratorium Kesehatan Ternak dan Experimental Farm Fakultas Peternakan, Universitas Jenderal Soedirman, Purwokerto, Kabupaten Banyumas, Jawa Tengah. Hasil pengukuran morfometri rata-rata spermatozoa unsexing 30.88 µm2, spermatozoa sexing fraksi atas dan fraksi bawah masing-masing 28.44 µm2 dan 32.63 µm2. Hasil dari penelitian keutuhan membran plasma spermatozoa X diperoleh rataan sebesar X1 = 66.61% dan X2 = 68.57% sedangkan spermatozoa Y diperoleh rataan sebesar Y1 = 64,87% dan Y2 = 66.96%. Hasil perhitungan statistic dengan uji t-student menunjukkan bahwa spermatozoa fraksi bawah (X) memberikan perbedaan nyata (P<0,05) lebih besar dibanding spermatozoa fraksi atas (Y). Berdasaran analisis variansi diperoleh hasil tidak ada interaksi antara faktor gradien dan lama inkubasi (P>0,05) terhadap morfometri dan Membran Plasma Utuh (MPU). Faktor gradien berpengaruh nyata (P<0,05) terhadap morfometri fraksi atas (Y) dan bawah (X), sedangkan faktor gradien dan lama inkubasi masing-masing berpengaruh nyata (P<0,05) terhadap Membran Plasma Utuh (MPU) fraksi atas (Y) dan bawah (X). Kesimpulan yang didapat dari penelitian ini yaitu separasi seks menggunakan medium gradien percoll 45:60% dengan lama inkubasi 30 menit mampu memisah dengan baik morfometrik spermatozoa fraksi atas (Y) maupun fraksi bawah (X), bahkan diperoleh persentase membran plasma utuh (MPU) tertinggi baik untuk fraksi atas dan bawah.

Abstract

Background. Aims to determine and analyze the pregnancy rate of Etawah Crossbreed goats based on Non-return Rate method and Cubboni Reaction. Materials and methods. The materials used in this study were 20 Etawah Crossbreed goats, prostaglandins, urine, and concentrated H2SO4. The variables studied were the pregnancy rate of Etawah Crossbreed Goat based on Non-return Rate method and Cubboni Reaction. This study used an experimental method and the data obtained were processed using Chi Square analysis. Results. The analysis shows the number of x2<x2 tables (P>0.05), this means that the pregnancy detection method does not significantly affect the pregnancy rate of Etawah Crossbreed goats. Conclusion. Pregnancy rate of Etawah Crossbreed Goat based on Non-return Rate method amount 80% while based on Cubboni Reaction method amount 90%.

Abstract

Latar belakang. Penelitian ini bertujuan untuk mengetahui pengaruh dari tingkat prolifikasi terhadap kadar hematologis pada domba Batur yang meliputi kadar eritrosit, leukosit, dan hemoglobin. Materi dan metode. Materi penelitian menggunakan 20 ekor induk domba Batur dengan minimal paritas sebanyak 2 kali dan rata-rata berumur 3 tahun kemudian dikelompokkan kedalam dua kelompok masing-masing terdiri atas 10 ekor. Peubah pertama yaitu prolifikasi rendah (P1) dengan catatan Jumlah Anak Sekelahiran (JAS) 1 dan peubah kedua yaitu prolifikasi tinggi (P2) dengan catatan JAS ? 2. Pengelompokkan dilakukan berdasarkan hasil wawancara dengan peternak terkait riwayat reproduksinya. Pengambilan sampel darah sebanyak 1-2 ml melalui vena jugularis dan dituangkan ke dalam vacuum tube EDTA kemudian disimpan di dalam ice box untuk mencegah adanya kerusakan dan penjendalan pada sel-sel darah. Pemeriksaan kadar hematologis dilakukan menggunakan hematology analyzer. Data diolah dengan menggunakan t-test unequal. Hasil. Hasil analisis statistik menunjukkan bahwa tidak ada perbedaan signifikan (P>0,05) dari tingkat prolifikasi terhadap kadar eritrosit dan kadar hemoglobin, namun berbeda signifikan (P<0,05) terhadap kadar leukosit pada domba Batur. Simpulan. Disimpulkan bahwa kadar eritrosit dan hemoglobin relatif sama antar tingkat prolifikasi, namun kadar leukosit pada prolifik rendah nyata lebih tinggi dibandingkan dengan prolifik tinggi pada domba Batur.

Abstract

Background. This study aims to determine the effect of Induction gonadotropin releasing hormone hematological levels in Batur sheep which include the levels of erythrocytes (SDM), leukocytes (SDP), and hemoglobin. Materials and Methods. The research material used was 20 female Batur ewes with a minimum parity of 2 times and an average age of 3 years, then grouped into two groups of 10 each. The research method used is experimental methods in the field which are grouped into 2 (two) treatment groups, namely: P1 =Double injection of PGF2? (Dinoprost trometamol 5mg/ml) as much as 2 ml intramuscularly as a controland P2= Double injection of PGF2? (Dinoprost trometamol 5mg/ml) as much as 2 ml intramuscularly as a control plus 1 ml of gonadotropin releasing hormone (Gonadorelin 0.1 mg/ml) intramuscularly. Each treatment was repeated 10 times. Blood sample were taken as much as 2-4 ml through the jugular vein with a 5 cc syringe. Data was processed using unequal t-test. Results. The results of statistical analysis showed that there was a very significant difference (P<0.01) from injection of gonadotropin releasing hormone to leukocyte levels in Batur sheep, whereas there was no significant difference in erythrocyte and hemoglobin levels (P>0.05) and was still relatively normal. Conclusion. The induction of gonadotropin releasing hormone did not increase the levels of erythrocytes and hemoglobin but there was an increase in leukocytes that exceeded the normal limit, so the dose of GnRH should be adjusted according to the body weight of the livestock.
