Abstract

Full-thickness wound are areas damage of skin associated with loss of epidermis and dermis. The wound healing mechanism consists proliferation, migration and remodeling. Hypoxic conditional medium of mesenchymal stem cells (HMSCs-CM) contains lots of soluble molecules, such as protein growth factor and cytokine anti-inflammation. The soluble molecule of HMSCs-CM plays a critical role in wound healing by upregulation of VEGF and collagen synthesis. The objective of this study was to evaluate the effect of HMSCs-CM on VEGF and collagen concentrations in rats with incised wounds. The methods of this study were an experimental animal study with post-test only control group design was performed involving 24 Wistar rats. The rats were randomized into four groups consisting of sham, control and two treatment groups (gel of HMSCs-CM at doses of 200 μL and 400 μL). The VEGF levels and collagen density were analyses using ELISA assay and Masson-trichome specific staining, respectively. One-way ANOVA and Post Hoc LSD were used to analyses the data. The results of this study showed that a VEGF levels was significant increased on day 6 with doses-dependent manner. Interestingly, the VEGF levels gradual decrease on day 9. In addition, the decreased of VEGF levels on day 9 in this study in line with our findings in which we found there was a trend in the decreased of collagen density, it indicated the completion of remodeling phase and there has been an acceleration in wound healing. This study demonstrated that HMSCs-CM were able to regulate VEGF levels and collagen synthesis in accelerate wound healing. The role of HMSCs-CM stimulate cutaneous wound healing should be clarified further.Keywords: hypoxic conditional medium of mesenchymal stem cells (HMSCs-CM), vascular endothelial growth factor, collagen synthesis, paracrine factors

Abstract

Penelitian ini bertujuan untuk menganalisis implementasi penerimaan resep elektronik pada pengguna yang menerapkan model extended TAM. Metode yang digunakan dalam penelitian ini adalah penelitian kausal dengan pendekatan kuantitatif. Jumlah sampel yang digunakan sebanyak 74 responden. Penentuan sampel menggunakan teknik simple random sampling yang merupakan teknik yang dapat mengurangi bias dalam penelitian. Variabel yang diteliti adalah Penerimaan Penggunaan, Persepsi Kegunaan, Persepsi Kemudahan Penggunaan dan Sikap Penggunaan. Analisis data menggunakan software Smart PLS untuk mengukur data dengan auto coding. Temuan penelitian ini menunjukkan bahwa seluruh hipotesis yang digunakan dalam penelitian ini diterima dengan hasil uji reliabilitas dengan Cronbach’s Alpha. Penerimaan Penggunaan (0,730), Persepsi Kegunaan (0,877), Persepsi Kemudahan Penggunaan (0,887) dan Sikap Penggunaan (0,894). Persepsi kemudahan penggunaan berpengaruh positif dan signifikan terhadap persepsi kegunaan difusi elektronik. Persepsi kemudahan penggunaan berpengaruh positif dan signifikan terhadap sikap penggunaan infiltrasi elektronik. Persepsi kegunaan berpengaruh positif dan signifikan terhadap sikap penggunaan infiltrasi elektronik. Persepsi kegunaan berpengaruh positif dan signifikan terhadap penerimaan infiltrasi elektronik. Sikap terhadap teknologi ini secara positif dan signifikan memengaruhi penerimaan resep elektronik. Kegunaan yang dirasakan memediasi pengaruh persepsi kemudahan penggunaan terhadap sikap difusi elektronik. Kemudahan penggunaan mempengaruhi kegunaan infiltrasi elektronik. Persepsi kemudahan penggunaan berpengaruh positif dan signifikan terhadap sikap penggunaan infiltrasi elektronik. Implikasi dari penelitian ini hanya menganalisis penerimaan resep elektronik dengan menggunakan model extended TAM. Diharapkan ada penelitian lebih lanjut dengan menggunakan alat dan pendekatan lain.

Abstract

BPJS participants often experience discrimination by hospitals, hospital services for general patients and BPJS participants are very different. The purpose of this research is to analyze Position of Health Insurance Agreement and to analyze Liability of the Social Security Agency in the Case of Rejection of BPJS Participants by Hospitals in the Perspective of Civil Agreement Law. The method used in this study was normative method. Normative legal research in its development is research that uses the study of legal behavior concluded in legislation. BPJS neglect of its participants when there is a rejection by its partner hospitals is an act of default for BPJS participants. So it is clear that the neglect of BPJS towards its participants in the issue of rejection of BPJS participants by BPJS partner hospitals is clearly an action that violates the obligations, duties, and responsibilities of BPJS towards BPJS participants, this is not in accordance with what is promised in the health social security agreement made by BPJS with its participants.

Abstract

Burn injuries caused by thermal exposure trigger oxidative stress and inflammation, largely driven by reactive oxygen species (ROS) and systemic inflammatory response syndrome (SIRS). These mechanisms may disrupt the production of cytokines involved in tissue repair, such as interleukin-10 (IL-10) and signal transducer and activator of transcription 3 (STAT3). Clitoria ternatea L. (butterfly pea) contains potent antioxidant and anti-inflammatory phytochemicals, making it a promising candidate for modulating inflammatory responses and promoting wound healing. This study evaluated the effects of Clitoria ternatea extract gel on IL-10 and STAT3 expression in second-degree burn injuries using a Wistar rat (Rattus norvegicus) model. A post-test only control group design was employed, involving 30 male rats randomly assigned to five groups (G1–G5). Second-degree burns were induced using a 1.8 cm diameter heated metal plate applied to the dorsal skin for 3 seconds after heating for 15 minutes. Treatments with the extract gel were administered topically for seven days. Skin samples were collected 24 hours after the last application and analyzed for IL-10 and STAT3 mRNA expression using qRT-PCR. Data were statistically tested with One-Way ANOVA. The results showed no significant differences in IL-10 expression among all groups. However, STAT3 expression was significantly higher in the groups treated with 5% and 10% Clitoria ternatea extract gel, with the highest observed in the 10% group (6.73 ± 2.94). These findings indicate that while IL-10 remained unaffected, butterfly pea extract gel effectively increased STAT3 expression, suggesting its potential role in modulating regenerative pathways in burn-injured skin

Abstract

Ischemic stroke is a major global cause of disability and mortality. Inflammation plays a central role in its pathogenesis, characterized by elevated pro-inflammatory cytokines such as IFN-γ and reduced anti-inflammatory cytokines like IL-10. Mesenchymal stem cells (MSCs), especially those derived from the umbilical cord (UC-MSCs), exhibit enhanced immunomodulatory potential when preconditioned under hypoxia. This study aims to evaluate the effect of hypoxic-preconditioned UC-MSCs administration on IFN-γ and IL-10 levels in an ischemic stroke rat model. This in vivo experimental study employed a randomized posttest-only control group design with four groups of male Wistar rats (n=6 each), ranging from healthy controls, untreated group to stroke-induced groups treated with hypoxic UC-MSCs at different doses (1.5x106 and 3x106). IFN-γ and IL-10 levels in brain tissue of each group were measured via ELISA. Significant reduction in IFN-γ and elevation in IL-10 were observed in UC-MSC-treated groups, particularly at the 3×10⁶ cell dose compared to the untreated ischemic group (p<0.05). Hypoxic UC-MSCs reduce post-stroke inflammation by lowering IFN-γ and enhancing IL-10, indicating a promising immunomodulatory potential.
