Abstract

Cigarettes are a source of exogenous free radicals which disrupt the balance between free radicals and antioxidants in the body, causing oxidative stress. The aim of this study was to determine the effect of giving honey (Mel depuratum) on SOD and caspase-9 levels in Wistar rats exposed to cigarette smoke. This research used a laboratory experimental approach with a post-test only control group design. 24 Wistar rats were divided randomly into 4 groups then analysis tests were carried out using the One-Way ANOVA test and followed by the Post-Hoc LSD test. The results showed that the mean SOD levels in groups K1, K2, K3, K4 were 73.51 ± 1.68, 21.72 ± 2.23, 52.08 ± 2.03, and 73.51 ± 1.68. The results of the One Way Anova test showed a significant difference in SOD levels with a value of p = 0.000. The results of the LSD Post-Hoc Test showed that SOD levels between groups had significant differences in all groups (p=0.000). The IHK staining results showed that samples K1 and K4 were negative, while K2 showed strong positive and K3 was moderately positive.

Abstract

Polycystic ovary syndrome (PCOS) is a condition characterized by chronic anovulation and hyperandrogenism, which often leads to infertility. It is closely associated with chronic inflammation triggered by glucose and saturated fat, contributing to hyperandrogenism and negatively impacting a patient’s quality of life. Effective therapeutic approaches are essential to address these issues. The secretome of mesenchymal stem cells (MSCs) have demonstrated the ability to suppress pro-inflammatory cytokine secretion and regulate growth factors. The aim of this study was to investigate the effect of hypoxic mesenchymal stem cell secretome (MHSSCs) on the expression of interleukin-10 (IL-10) and signal transducer and activator of transcription 3 (STAT3) genes in a PCOS-induced mouse model. An in vivo experimental study was conducted using a post-test-only control group design. A total of 24 female C57BL/6 mice were divided into four groups: healthy control, negative control (PCOS mice injected with 0.9% NaCl), T1 (PCOS mice administered 200 μL of MHSSCs), and T2 (PCOS mice administered 400 μL of MHSSCs) for 33 days. Gene expression of IL-10 and STAT3 were quantified using quantitative reverse transcription polymerase chain reaction (qRT-PCR), normalized to the expression of the housekeeping β-actin gene. Statistical analysis using one-way ANOVA followed by the least significant difference (LSD) post-hoc test was then performed. The results showed a significant increase in IL-10 expression in the T2 group compared to the negative control group (p<0.001). STAT3 expression was also significantly higher in the T2 group compared to the negative control group (p=0.035). A dose-dependent effect was observed, with the T2 group demonstrating the highest upregulation of both IL-10 and STAT3 expression levels. The study highlights that the administration of MHSSCs effectively increased IL-10 and STAT3 gene expression, suggesting their potential as a therapeutic strategy to alleviate inflammation in PCOS.

Abstract

Ultraviolet B (UVB) radiation is known to induce skin damage, including erythema, roughness, sagging, wrinkles, and reduced skin moisture, contributing to premature aging. Turmeric (Curcuma longa Linn.) contains curcumin, an active compound with potent anti-inflammatory properties. This study investigated the effect of turmeric ethanol extract on IL-6 and IL-10 levels, as well as skin moisture, in UVB-exposed rats. A total of 24 Wistar rats were randomly divided into four groups: healthy controls (K1), UVB-exposed without treatment (K2), UVB-exposed treated with turmeric ethanol extract at 100 mg/kg BW (K3), and 200 mg/kg BW (K4). The extract was administered orally for 14 days. IL-6 and IL-10 levels were measured using ELISA, and skin moisture was assessed macroscopically. Statistical analysis was performed using one-way ANOVA. Rats in groups K2, K3, and K4 exhibited very dry skin, redness, visible wrinkles, and peeling following UVB exposure. IL-6 levels were significantly lower in treated groups, with the lowest levels observed in K4 (1.85 ± 0.24 ng/L), compared to K1 (4.36 ± 0.75 ng/L) and K2 (4.46 ± 0.74 ng/L) (p = 0.000). IL-10 levels increased in the treated groups, notably in K3 (376.48 ± 78.24 pg/mL) and K4 (351.06 ± 78.24 pg/mL), compared to K1 (285.34 ± 45.54 pg/mL) and K2 (306.89 ± 45.54 pg/mL) (p = 0.001). These findings suggest that oral administration of turmeric ethanol extract modulates inflammatory responses by reducing IL-6 and enhancing IL-10 in UVB-exposed skin, although significant dryness persists.

Abstract

Continuous exposure to ultraviolet B (UVB) radiation can induce photoaging and accelerate premature skin aging. Turmeric (Curcuma longa) is a medicinal plant that contains curcumin, an active compound known for its anti-inflammatory properties and its role in accelerating wound healing. This study aimed to evaluate the effect of turmeric ethanol extract on TNF-α levels and wound lesion areas in Wistar rats exposed to UVB radiation. A post-test-only control group design was employed, involving 24 Wistar rats divided into four groups: a healthy control group (K1), a negative control group (K2), and two treatment groups receiving oral turmeric ethanol extract at doses of 100 mg/kg BW (K3) and 200 mg/kg BW (K4) for 14 days, with concurrent UVB exposure. TNF-α levels were assessed using one-way ANOVA followed by Tukey's post hoc test, while wound lesion areas were analyzed using the Kruskal–Wallis test followed by the Mann–Whitney U test. The results showed that the highest TNF-α levels were observed in the healthy control group (129.14 ng/L), although no statistically significant differences were found between groups (p = 0.538). In contrast, a significant difference was observed in wound lesion areas (p < 0.001), with the largest lesions occurring in the negative control group. In conclusion, oral administration of turmeric ethanol extract did not significantly affect TNF-α levels but was effective in reducing the area of wound lesions caused by UVB exposure.

Abstract

Background: The digital era has markedly increased adolescents’ exposure to explicit content, including pornography, raising significant concerns regarding its impact on sexual and reproductive health. This is particularly critical among female adolescents in Indonesia, where access to comprehensive sexual education remains limited. Objective: This study aims to examine the exposure of female adolescents to pornographic content, with a focus on patterns of media consumption and their implications for reproductive health. Methods: A descriptive cross-sectional study was conducted among 58 female adolescents aged 13 to 18 years in Karanganyar, Indonesia. Data were collected using a validated questionnaire and analyzed through descriptive statistical methods to identify patterns of media usage and exposure to pornography. Results: The findings indicated that the internet and handheld devices were the predominant sources of media consumption (96.6%), significantly contributing to exposure to pornographic material (86.2% and 96.6%, respectively). Television and print media also played contributory roles, particularly through soap operas and Western films. Exposure to pornography was associated with limited awareness of reproductive health, especially among younger participants. The exposure was further linked to distorted perceptions of sexual behavior, negative body image, and potential psychological consequences such as anxiety and low self-esteem. Conclusion: This study highlights the urgent need for targeted interventions to enhance critical media literacy and provide comprehensive sexual education for adolescents. Addressing these issues is essential to mitigate the adverse effects of pornography exposure on reproductive health, while taking into account cultural and contextual factors. Future research should investigate the longitudinal effects and gender-specific outcomes to inform the development of tailored preventive strategies.
