Abstract

BackgroundMesenchymal stem cells (MSCs) are multipotent stromal cells that express CD73, CD90, and CD105 surface markers, but not CD14, CD45, CD34, CD11b, and HLA-DR. MSCs under hypoxic conditions have the essential role of maintaining the stemness capacity by releasing several growth factors into their medium, known as hypoxia conditioned medium (HCM). This study was performed to compare the effect of percentage of HCM to normoxic medium (NM) in increasing MSC proliferation marked by proliferation rate and surface marker expression.MethodsThis study was of post-test only control group design using human umbilical cord-MSCs (hUC-MSCs) as subjects. The HCM treatment group was obtained by culturing MSCs under 5% O2, whereas the NM control group was grown under 20% O2. The hUC-MSCs were divided into 4 groups with different dose ratios of HCM to NM (25%:75%; 50%:50%; 75%:25% for P1, P2 and P3, respectively and 100% of NM for the controls). All of these groups were maintained at 37oC and the data was collected after 72 hours incubation. MSC marker expression of CD73, CD90 and CD105 was analyzed using flow cytometry and MSC proliferation by trypan blue assay. ResultThere were significant differences in MSC marker expression of CD73, CD90 and CD105 and proliferation at all dose ratios of HCM to NM (p<0.05).ConclusionLow oxygen concentration promotes MSC proliferation and stemness thus it might be beneficial for maintaining the MSC physiologic niche in-vitro.

Abstract

Merkuri adalah logam berat yang toksik didam jumlah kecil, akan tetapi toksisitasnya bervariasi tergantung dari bentuk senyawa kimianya dan tempat pemaparannya. Dalam perairan, kandungan logam merkuri sekalipun kadarnya relatif rendah, dapat diabsorbsi dan terakumulasi secara biologis oleh ikan dan akan terlibat dalam sistim jaringan makanan. Pengamatan penelitian ini melihat kondisi air tambak oso yang meliputi suhu, pH, salinitas, kandungan merkuri dalam air dan kandungan merkuri dalam ikan mujair. Jenis penelitian ini adalah penelitian analitik yang bersifat observasional, yang dilakukan secara cross-sectional. Hasil dari penelitian ini menunjukkan bahwa kandungan logam merkuri dalam air rata rata sebesar 0,0024 ppm, di mana suhu berfluktasi, pH dan salinitas adalah relatif rendah. Rata rata suhu dari air 28,23°C, pH 7,28 dan salinitas 0,3084 o/ooo. Merkuri dalam sampel ikan mujair rata-rata sebesar 0,00084 ppm. Sampel ikan mujair yang berumur 3 bulan. 4 bulan dan 5 bulan mempunyai kandungan merkuri yang berbeda

Abstract

Corona Virus Disease 2019 atau yang biasa disingkat COVID-19 adalah penyakit menular yang disebabkan oleh SARS-CoV-2, yang merupakan salah satu jenis coronavirus. Covid-19 memiliki gejala ringan sampai berat, dari yang menyerupai flu pada umumnya sampai sesak nafas bahkan kematian. Penyebaran covid-19 terhitung sangat cepat. Daya tahan tubuh memiliki peran yang sangat penting dalam mencegah terjangkitnya virus tersebut. Salah satu yang dapat menunjang peningkatan daya tahan tubuh yaitu suplemen, baik vitamin, mineral maupun pemanfaatan tanaman herbal, seperti jahe. Tujuan kegiatan pengabdian ini adalah meningkatkan pemahamam para kader PKK di kelurahan Banjardowo tentang manfaat suplemen sebagai penangkal infeksi virus corona. Metode pengabdian ini dengan membagikan kuesiner kepada 24 kader PKK yang mengikuti penyuluhan dan pelatihan cara membuat suplemendengan jumlah pertanyaan sebanyak 12. Kuesioner diisi sebelum dan sesudah mengikuti acara. Hasil kegiatan tersebut adalah peningkatan pemahaman para kader akan covid-19 dan manfaat suplemen sebanyak 90%. Hasil tersebut dapat disimpulkan bahwa kader PKK telah memahami tentang covid-19 dan manfaat suplemen khususnya tanaman herbal sebagai penangkal covid-19. Keywords : Covid-19, Suplement, Pemahanam

Abstract

Bekam sangat berkhasiat untuk berbagai macam penyakit, bekam juga merupakan preventive medicine dan pengobatan simptomatis. Penelitian ini bertujuan untuk mengetahui pengaruh bekam basah terhadap Kadar Hemoglobin, Fe, dan Kadar TIBC pada pasien Anemia. Penelitian quasi experimental dengan pre dan post test tanpa kelompok kontrol, Sampel ditentukan sebanyak 21 orang dan mendapat terapi bekam basah. Analisis data pada penelitian ini dilakukan dengan uji Wilcoxon Signed Ranks Test dan uji T berpasangan. Terdapat perbedaan yang bermakna secara statistik dimana hasil uji didapatkan p value 0,000 pada masing – masing variabel dengan kadar hemoglobin medapatkan pengaruh tertinggi dengan selisih hasil rata – rata post test sebesar -0,266 (SD = 72), kemudian Fe sebesar -0,105 (SD = -0,048), dan selanjutnya pada kadar TIBC -0,67 (SD = -0,167). Terapi bekam basah berpengaruh terhadap peningkatan kadar Hb, Fe, dan penurunan kadar TIBC pada pasien anemia.

Abstract

Hiperkolesterolemia mengakibatkan terjadinya peningkatan produksi ROS yang menimbulkan stress oksidatif dengan peningkatan Lipid Peroksidase (LPO) yang menyebabkan infertilitas dengan penurunan kadar testoteron. Kondisi ini dapat dinetralkan dengan mengkonsumsi antioksidan dari luar tubuh seperti N-Acetylcysteine (NAC). Untuk mengetahui pengaruh pemberian NAC terhadap hormon testoteron yang diberi diet tinggi kolesterol. Penelitian ekperimental dengan pendekatan post test only control group design. Subyek penelitian berjumlah 24 ekor tikus jantan galur sparague dawley yang dibagi secara acak menjadi 4 kelompok. Kelompok K1 tanpa pemberian NAC dan diet tinggi kolesterol. Kelompok K2 tanpa pemberian NAC namun diberi diet tinggi kolesterol. Kelompok K3 dan K4 diberi diet tinggi kolesterol dan NAC masing-masing dengan dosis 3,14 mg/ml dan 6,3 mg/ml. Hari ke 25 dilakukan pengambilan darah untuk pemeriksaan kadar testoteron di IBL FK UNISSULA pada November - Desember 2020. Uji One Way Anova menunjukkan perbedaan bermakna pada kadar testoteron menunjukkan perbedaan bermakna (p=0,021). Hasil uji Post Hoc Tukey kadar testoteron pada K1 dan K4 menunjukkan perbedaan signifikan terhadap K2 (p<0,05). Pemberian NAC menunjukan secara bermakna dapat meningkatkan kadar hormon testoteron yang diberi diet tinggi kolesterol.

Abstract

Gangguan ginjal akut (GnGA) merupakan suatu keadaan dimana proses laju filtrasi glomerulus ginjal menurun secara cepat yang menyebabkan retensi nitrogen terutama kreatinin dan blood urea nitrogen (BUN). Kondisi ini dapat dinetralkan dengan mengkonsumsi antioksidan dari luar tubuh seperti Propolis (metode CMCE). Untuk mengetahui pengaruh ekstrak propolis (metode CMCE) terhadap kadar MDA dan degenerasi tubulus renalis. Penelitian ekperimental dengan pendekatan post test only control group design. Subyek penelitian berjumlah 25 ekor tikus jantan galur wistar yang dibagi secara acak menjadi 5 kelompok. Kelompok K1 tanpa diinduksi gentamisin. Kelompok K2 diinduksi gentamisin dan tanpa diberi ekstrak propolis metode CMCE. Kelompok P1, P2 dan P3 diinduksi gentamisin dan ekstrak propolis metode CMCE masing-masing dengan dosis 200, 400, 800 mg/k, BB per hari per oral selama 7 hari. pemeriksaan kadar Malondialdehid (MDA) di IBL FK UNISSULA dan pemeriksaan degenerasi tubulus renalis di RSI Sultan Agung Semarang pada Maret - Juli 2020. Uji One Way Anova menunjukkan perbedaan bermakna pada kadar MDA dan degenerasi tubulus renalis menunjukkan perbedaan bermakna (p<0,05). Pemberian ekstrak propolis metode CMCE menunjukan berpengaruh secara signifikan terhadap kadar MDA dan skor total degenerasi tubulus renalis yang diinduksi gentamisin.

Abstract

Introduction:Liver exposure to toxic substances will disrupt the metabolic system. Carbon tetrachloride (CCl4) is a hepatotoxic substance causing a liver damage. Continuous multi-stage countercurrent extraction(CMCE) propolis extract contains active balsamic substances, caffeic acid, phenethyl esters and flavonoids.Objective: to prove the effect of CMCE propolis extract on IL-1 levels and the hepatocyte histopathology findingsof male wistar rats’induced by CCl4.Methods:In the posttest only control group design, the total sample of 28 rats was divided into 4 groups: positive control group (K-0; standard feed); group P-1 (propolis3.6 mg/200 g); P-2 (propolis7.2mg/200gr and P-3 (propolis14.4mg/200g) were respectively given for 14 days,yet those on the 14thday were given CCl4. The IL-1 level was measured with the ELISA method, while the male wistar rats’hepatocyte histopathology findings was determined by the HE staining methods. IL-1 levelswere analyzed usingOne Way Anova test,followed by the Post Hoc LSD test. Hepatic cell histopathology was analyzed usingKruskal Wallis, followed by Mann-Whitney U.Results:Post Hoc LSD analysis shows that the levels of IL-1 in P-1 (983.54 ±19.67), P-2 (841.58±19.47), and P-3 (755.50±31.63) are significantlylower thanthosein K-0 (1131.69±14.91) withp<0.05. Mann-Whitney U analysis shows that the hepatocyte histopathology in the P-1 group (2.55±0.39), P-2 (1.24±0.94) and P-3 (1.20±0.93) is significantlylower thanthatinK-0 (2.94±0.16) with p<0.05.Conclusion: The administration of CMCE propolis extract may reduce IL-1 levels and improve the male wistar rats’hepatic cell histopathologyinduced with carbon tetrachloride (CCl4).

Abstract

Introduction: Leprosy reaction is an immunity declining reaction marked with inflammation which may cause permanent disability. Physical exercise may improve body’s immune system. Objective: To prove the effect of moderate-intensity physical exercise on the interleukin-6 level and lymphocyte count in leprosy reaction patient.Methods: The quasi-experimental research with pretest and posttest and a control group design used 30 respondents consisting of 14 interventions and 16 controls. The intervention group was given moderate-intensity physical exercise 60%-79% in the form of static cycle at a frequency of 4 times weekly, 150 minutes per week. Corticosteroid was always administered to the two groups as a standard therapy. The interleukin-6 level was measured using ELISA method and lymphocyte count was measured using hematology analyzer.Result: The Mann-Whitney analysis shows that the mean difference in IL-6 level between pretest and posttest with the intervention group (-7.1429±25.2369) is significantly lower than that of the control group (8.8125±11.2025), p<0.05. On the contrary, the Independent t-test analysis shows that the mean difference in lymphocyte count between pretest and posttest with the intervention group (0.7407±0.41236) is significantly higher than that of the control group (-0.4500±0.42415), p<0.05. Conclusion: Physical exercise at moderate intensity in the form of aerobics using static cycle at a frequency of 4 times weekly, with duration of 150 minutes per week decreases interleukin-6 level and increases lymphocyte count in leprosy reaction patient.

Abstract

 Introduction: Consumption of excessive calories can increase the incidence of degenerative diseases mediated by ROS. Caloric restriction, have been shown to increase levels of antioxidants superoxide dismutase (SOD) and Gluthatione Peroxidase (GPx). Fasting like Ramadan fasting (FLRF) is a form of calorie restriction, but its effect on levels of SOD and GPx remains unclear. Objectives: to investigate the effect of fasting on levels of SOD and GPx. Methods: in a post-test only control group design, sample of 24 rats Sprague Dawley Rats aged 3-month-old, weighing 250-300 grams, were randomly divided into 4 groups. Group 1 (P-70), 2 (P-100), and 3 (P-140) were fasted for 6 hours/day, each group received of 70%, 100% and 140% calories respectively. Group 4 (C-AL) received 100% calories, ad libitum. Day 16 blood was taken and levels of SOD and GPx were determined by ELISA. Data were analyzed using one-way ANOVAs, followed by post hoc LSD tests, p<0.05 was considered statistically significant. Results: the results showed that the levels of SOD and GPx occur significant differences between the groups, p = 0.000. The test results post hoc SOD (318.64) and GPx (89.16) group P-70, compared with group C-AL (278.60 and 57.20) was significantly higher (p = 0.00). SOD and GPx P-70 group compared with the group P-140 (92.03 and 48.79), significantly higher (p = 0.00). Compared with group P-100 (296.70 and 75.71) SOD and GPx in group P-70 was significantly higher, p = 0.000. Conclusion: Fasting with calorie intake of 70% and 100% for 15 days can increases levels of SOD and GPx in male rats.

Abstract

Background: Dental plague has been known to be the major agent causing dental carries and periodontal disease which are the most common dental and soft tissue diseases with the prevalence of 70-80%. The preventive measure including the use of chemical substance of catechin having anti-plague activity in the form of mouth wash has been shown to have effects on the dental plague formation. The objective of this study was to find out the duration of mouth washing with boiled gambir (Uncaria ambir) on the plague formation in Pondok Pesantren Al-Badriyah Mranggen, Demak.Design and Method: In this study using Pretest-Posttest Control Group Design, 44 samples of subjects was divided into 4 groups of 11 each. Group I, II, II were treated with the mouth wash of Uncaria gambir boiled 2, 3, 4 minutes respectively. Group IV serving as control was treated with mouth wash of water for 3 minutes. The data was analyzed with non parametric test using Kruskal-Wallis followed by Mann-Whitney test with p < 0.05.Result: Kruskal Wallis analysis showed a significant difference in mean of plague before and after the treatment among the four groups (p < 0.05). Mann-Whitney test showed no significant difference between group I and III ( p> 0.05) and significant difference for the rest of the groups (p < 0.05). Conclusion: The mouth rinse duration using boiled Gambir has been shown to have an effect on the dental plague formation. 3 minute mouth washing shows the optimum effect (Sains Medika, 4(1):57-62).
