Abstract

SARS-CoV-2 is a Coronaviridae virus with a single-stranded RNA genome. This virus is highly contagious and spreads rapidly, with mutations occurring at a rapid pace. Various preventive actions have been implemented to prevent the spread of Covid-19 instances. The World Health Organization recommended that everyone clean their hands, masks, and other items in their environment on a frequent basis. As a result of these measures, the virus's transmission will be slowed. Furthermore, keeping one's immune system is critical for limiting the spread of Covid-19. As a result of a weakening immune system, our body's defenses will deteriorate, making us more susceptible to sickness and unable to fight diseases caused by viruses or bacteria. If the immune system is strong, it will recognize and kill viral and bacterial compounds when they are exposed. Immunomodulators, such as natural vitamins and herbs, can help maintain and boost immunity. Indonesia is primarily a farming country. In Indonesia, many different varieties of plants can be grown. Turmeric is an Indonesian medicinal plant that may be found all across the country. Turmeric's ability to act as an immunomodulator has also been proven. As a result, the goal of this study is to determine the usefulness of the active chemicals found in the Curcuma longa plant as a natural immunomodulator in order to avoid mutations of the COVID 19 variation by looking at the background of the current situation. The approach employed in this study is an in silico Molecular Docking test, with the goal of determining the active side of the drug and the value of Binding Affinity from the Docking data. Tissue Pharmacology is used to depict the protein network found in turmeric plants, as well as the substances and diseases that are linked to these proteins. According to the findings, the compound quercetin acts as a ligand for the PIK3CG protein and has a high binding affinity of -7.4. As a result, it can be identified as a substance that acts as a natural immunomodulator in turmeric plants.

Abstract

ABSTRAK Biji Nigella sativa telah banyak diketahui khasiat farmakologisnya. Salah satu aktivitas farmakologis senyawa N. sativa adalah sebagai antimikroba. Untuk itu penelitian ini dilaksanakan dengan tujuan mengetahui aktivitas antimikroba N. sativa terhadap flora normal saluran pencernakan manusia agar diketahui dosis terapi yang tepat dan tidak membahayakan keseimbangan flora normal saluran pencernakan. Penelitian ini dilaksanakan secara in vitro dan in vivo. Pengujian in vitro aktivitas antimikroba ekstrak biji N. sativa terhadap strain bakteri probiotik usus dilakukan dengan uji difusi cakram dengan perlakuan pemberian ekstrak N. sativa fraksi n-heksan, etil asetat dan etanol konsentrasi 500 mg/ml; 250 mg/ml; 125 mg/ml; 62,5 mg/ml; 31,25 mg/ml; 15,625 mg/ml. Pengujian secara in vivo dilakukan dengan mencit dengan pemberian kombinasi antibiotik sefadroxil 1,3mg/20g BB mencit dan adjuvan berupa ekstrak N. sativa 1,3mg/20g BB, 2,6 mg/20g BB, dan 3,9mg/20g BB mencit. Hasil pengujian in vitro ekstrak biji N. sativa fraksi n-heksan memiliki potensi antimikroba tertinggi disusul kemudian fraksi etil asetat dan etanol. Daya antimikroba ekstrak biji N. sativa fraksi n-heksan muncul pada konsentrasi 15,63 mg/ml dan semakin kuat pada konsentrasi yang lebih tinggi. Secara in vivo didapatkan hasil dosis adjuvant ekstrak biji N. sativa 3,9 mg/20g BB memunculkan aktivitas antimikroba pada bakteri probiotik sebagai bagian flora normal di saluran cerna mencit. Kata kunci : N. sativa, antimikroba, bakteri probiotik, in vivo, in vitro ABSTRACT Nigella sativa has a lot of pharmacological activities. One of those is the broad range antimicrobial activity. The aims of this research are to find out the antimicrobial activity against probiotic bacteria as normal flora in gastrointestinal track (GIT), in order to determine the apropriate dose which not harm for GIT normal flora. This research was conducted by in vitro and invivo assay. In vitro assay for antimicrobial acitivity using disc diffusion method with n-hexane, ethyl acetate and ethanol fraction of N. sativa extract. The concentration of each fraction were varied into 500 mg/ml; 250 mg/ml; 125 mg/ml; 62.5 mg/ml, 31.25 mg/ml, 15.625 mg/ml, respectively. In vivo assay using mice (Mus musculus) to analyze the difference of the probiotic bacteria viability in the GIT of treated mice and in that of control mice. N. sativa extract at dose of 1.3 mg/20gBW, 2.6 mg/20gBW, and 3.9 mg/20gBW were used as adjuvant with sefadroxil 1,3 mg/20 g BB. The result showed that the n-hexane extract possessing the highest potency as antimicrobial against probiotic bacteria, wich the minimum inhibitory concentration was 15,625 mg/ml. The in vivo assay shows that the dose of 1.3 mg/20gBW of mice has antimicrobial activity and so do the higher doses. Key words: antimicrobial, probiotic bacteria, N. sativa, in vivo, in vitro

Abstract

Typhoid fever is an infection caused by the bacterium Salmonella typhi, spread through contaminated food or water. Typhoid fever can be treated with antibiotics, but at this time some pathogen microbes have been resistant to the available antibiotics. Antimicrobials derived from natural product might be the alternative to overcome antibiotic resistance. Eleutherine palmifolia (L.) Merr is empirically used to treat infectious diseases. E. palmifolia has chemical compounds of alkaloids, glycosides, flavonoids, phenolics, steroids, and tannins. This study aims to evaluate the antibacterial activity of ethyl acetate fraction against S. typhi. E. palmifolia bulbs were subsequently extracted using n-hexane solvents and followed by ethyl acetate solvents. The ethyl acetate fraction was tested for antibacterial activity using the disc diffusion method. The ethyl acetate fraction of E. palmifolia at the concentrations of 8, 6, and 8 mg/paper disc showed the diameters of the inhibition zone of 15.1±3.6, 15.3±3.3, and 16.9±1.9 mm, respectively. The conclusion of this study was the ethyl acetate fraction of E. palmifolia has a strong antibacterial activity against S. typhi.

Abstract

Indonesia has a high and diverse biodiversity, particularly in plant species. There are numerous advantages to using various plants that grow in Indonesia. Indonesia is also known for its abundance of spices and other natural resources. Rapid research is required in the use of this plant in order for bio-based products to be widely accepted. Using in-silico predictions by utilizing meta data provided by several credible sites is one of the important rapid methods in analyzing the benefits of the chemical content of Curcuma zedoaria (Temu putih). The goal of this in-silico analysis-based study is to gain an understanding of the pharmacology of a plant known as potency simplicia Curcuma zedoaria. Analyzing metadata from various sources is the research method. Prediction of absorption, distribution, metabolism, and excretion (ADME) was obtained from http://www.swissadme.ch/. The prediction of target proteins for phytochemical compounds of Curcuma zedoaria is available at http://www.swisstargetprediction.ch/, while the construction of active protein networks and interactions after induction of compounds contained in Curcuma zedoaria rhizome is available at https://string-db.org. According to the in-silico analysis performed with some of the software mentioned above, the rhizome of Curcuma zedoaria (Temu Putih) contains 71 active compounds, 64 of which are highly bioavailable. According to in-silico research, Curcuma zedoaria (Temu Putih) contains curcumin compounds (diarylheptanoid) and its derivatives have antioxidant activity, which functions to prevent stress from physiological stimulation that can increase the number of leukocytes.

Abstract

At the end of 2019, a new virus emerged from Wuhan, China named Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2). The key to viral infection to host cells is the use of the ACE2 receptor. This study aimed to explore the potential of drugs in Annona muricata plant using the Insilico method. Compounds obtained from KNApSAcK Based on the analysis using SwissADME (Lipinski creteria)criteria. 22 compounds passed the selection. Eight ligands were identified as being able to change the conformation of the initial form of RBD and ACE2 attachment. Eight molecules are able to change the initial conformation, namely Asimilobine, Coreximine, (+)-Stepharine, Coclaurine, Norcorydine, N- Nornuciferinhe, methoxyphenyl)methyl]-1,2,3,4-tetrahydroisoquinolin-5-ol, and Atherosperminine.

Abstract

Muhammadiyah memang sudah banyak menggunakan teknologi modern, termasuk media sosial, untuk melakukan komunikasi dan menyampaikan informasi dakwah, namun intensitas dan kapasitasnya masih terbilang rendah dibandingkan dengan komunitas yang harus dijangkau. Pemuda Muhammadiyah sebagai penerus gerakan Muhammadiyah tentu harus merumuskan dan segera mengeksekusi langkah-langkah konkrit dalam mendigitalisasi program dakwahnya. Platform digital http://dakwah.pwpmjatim.id adalah salah satu upaya Pemuda Muhammadiyah Jawa Timur bersama dengan Tim Pengabdian Masyarakat Universitas Muhammadiyah Malang dalam membangun ekosistem digital dakwah yang simpel dan mudah.  Fitur web disusun sebaran fasilitas layanan ibadah dan kanal media sosial masjid binaan Muhammadiyah Jawa Timur. Pin titik lokasi tunggal (berwarna biru putih merah), akan menunjukkan menu tautan media sosial instagram, facebook dan youtube sehingga dapat dimanfaatkan pengguna untuk lebih jauh menikmati layanan online masjid. Namun fitur ini masih terus dikembangkan karena mayoritas masjid belum memiliki kanal media sosial. Platform ini dibangun untuk membantu Muhammadiyah di Jawa Timur menampilkan peta lokasi, fasilitas, layanan dan kanal-kanal media sosial masjid dalam binaannya. Kedepan, platform ini dapat diintegrasikan dengan berbagai platform digital dalam binaan Muhammadiyah sehingga manfaatnya dapat lebih luas meliputi bidang pendidikan, sosial, budaya dan tentunya bidang dakwah Muhammadiyah. Platform ini juga direncanakan akan diperluas hingga dapat memetakan masjid binaan Muhammadiyah dalam skala nasional bahkan internasional.

Abstract

Covid-19 merupakan wabah penyakit yang menginfeksi sistem pernafasan pada manusia. Kasusnya terjadi hampir di seluruh negara di dunia dan keberadaannya sangat meresahkan karena penyebarannya terjadi dengan sangat cepat. Salah satu upaya yang dapat dilakukan untuk mencegah penyebaran penyakit Covid-19 adalah dengan meningkatkan sistem imun. Sistem imun melindungi tubuh dari infeksi dengan memproduksi molekul protein yang disebut antibodi yang mengikat antigen agen infeksi. Berdasarkan studi literatur yang telah dilakukan, beberapa tanaman lokal Indonesia memiliki potensi sebagai imunomodulator. Fokus penelitian ini ditujukan untuk mengetahui jejaring protein yang terkait dengan sistem imun tubuh yang teraktivasi karena pemberian ekstrak tanaman jati belanda (Guazuma ulmifolia Lamk.). Metode penelitian yang digunakan adalah deskriptif eksploratif berdasarkan online database dan web server (KNApSAck Family, Dr. Duke’s Phytochemical and Ethnobotanical Databases, SwissTargetPrediction, GeneCards, SwissADME, Venny, dan StringDB). Berdasarkan hasil penelitian, tanaman G. ulmifolia mengandung 17 senyawa metabolit sekunder, lima diantaranya memiliki bioavailabilitas yang tinggi meliputi ent-catechin, (-)-epigallocatechin, caffeine, kaempferol, dan quercetin. Senyawa (-)-epigallocatechin diprediksi dapat berinteraksi dengan MAPK14 yang memiliki koneksi dengan lima jalur penting dalam imunomodulator yaitu Fc epsilon RI signaling pathway, PD-L1 expression and PD-1 checkpoint pathway in cancer, THF17 cell differentiation, TNF signaling pathway, dan IL-17 signaling pathway.

Abstract

Hipertensi adalah kondisi kronis yang dapat menyebabkan komplikasi kardiovaskular serius, dan kawista (L. acidissima) telah digunakan dalam pengobatan tradisional untuk berbagai penyakit, termasuk hipertensi. Namun mekanisme molekuler L. acidissima dalam mengatur tekanan darah belum sepenuhnya dipahami. Penelitian ini bertujuan untuk menyelidiki potensi kawista L. acidissima dalam pengelolaan hipertensi melalui pendekatan jejaring farmakologi. Senyawa metabolit sekunder L. acidissima diambil dari literatur, protein yang dapat berinteraksi dengan metabolit sekunder diprediksi dengan menggunakan SwissTargetPrediction, protein yang berhubungan dengan hipertensi diperoleh dari GeneCards. Hasil irisan protein dari SwissTargetPrediction dan GeneCards dianalisis lebih lanjut menggunakan STRING dengan metode GO (Gene Ontology) dan KEGG (Kyoto Encyclopedia of Genes and Genomes) enrichment. Protein yang paling memiliki peranan dalam hipertensi dianalisis dengan menggunakan algoritma MCC (Maximal Clique Centralit) dari CytoHubba. Analisis GO enrichment menunjukkan keterlibatan protein-protein dalam regulasi tekanan darah sistemik dan respons biologis lainnya yang relevan dengan hipertensi. Analisis KEGG enrichment mengungkap keterlibatan jalur-jalur sinyal utama seperti cGMP-PKG dan HIF-1, serta jalur regulasi hormonal penting dalam regulasi tekanan darah. Analisis MCC top 10 dari CytoHubba menyoroti peran sentral gen-gen seperti ACE, NOS3, dan EDNRA dalam jaringan biologis, menunjukkan potensi mereka sebagai target terapeutik dalam pengelolaan hipertensi. Senyawa metabolit sekunder L. acidissima memiliki potensi digunakan sebagai terapi untuk hipertensi.

Abstract

Cancer remains a formidable health challenge worldwide, with complex molecular mechanisms driving its initiation, progression, and therapeutic resistance. In this study, we employed bioinformatics analyses to elucidate the molecular underpinnings of cancer biology, focusing on Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis. Our GO analysis revealed the enrichment of key biological processes such as protein phosphorylation, regulation of programmed cell death, and transmembrane receptor signaling pathways, underscoring the critical roles of signaling cascades and regulatory mechanisms in tumorigenesis. Similarly, molecular functions such as protein kinase activity and ATP binding were identified as significantly enriched, highlighting the importance of protein kinases and molecular interactions in cancer development and progression. The KEGG pathway analysis further delineated dysregulated signaling pathways associated with cancer, including the MAPK and PI3K-Akt signaling pathways, implicating these pathways as central regulators of cancer progression. These findings deepen our understanding of cancer biology and offer potential targets for therapeutic intervention. Integrating multi-omics data and systems biology approaches may provide deeper insights into the intricate networks underlying cancer pathogenesis, paving the way for developing more effective treatments for cancer patients.

Abstract

Background:  Diabetes mellitus prevalence in Indonesia has surged. In 2021, an estimated 19.5 million people had diabetes, with a 10.6% age-adjusted prevalence. Projections indicate around 9.5 million cases by 2024. Diabetes medications, such as metformin, are commonly used, although these medications have adverse effects. A common choice for chronic diseases like DM is the use of natural medications. A plant known as Clerodendrum inerme has the potential to alleviate diabetes, but little is known about its molecular mechanismsObjective: The purpose of this study was to explore the content of Clerodendrum inerme plant compounds and their potential for cases of Diabetes Mellitus.Methods: The KNApSAcK was used to conduct an analysis of plant parts of Clerodendrum inerme to seek out chemicals present in plants. A screening was done to find compounds by estimating Absorption, Distribution, Metabolism, and Excretion (ADME) parameters using the canonical Simplified molecular-input line-entry system (SMILES) on the SwissADME. On the SwissTargetPrediction tool, predictions of target proteins from compounds that pass screening are connected to various probable proteins. utilising the String-db  to show the network between target proteins and associated diseasesResults: The Clerodendrum inerme consist of 24 different compound. The 24 compounds were screened, and the results showed that 4 of them, specifically (Z)-3-Hexenyl beta-D-glucopyranoside, Rhodioloside, Sammangaoside B, and Clerodermic acid, had the potential to be developed into therapeutic agent. The compound are then analysed in order to find the protein target associated with diabetes mellitus and predict its networks. The findings indicate that multiple target proteins, including GSK3B, PPARG, DPP4, and STAT3, are connected to diabetes mellitus.Conclusion: It has been shown that (Z)-3-Hexenyl beta-D-glucopyranoside, or clerodermic acid, is able to attach to the proteins GSK3B, PPARG, DPP4, and STAT3, which are all linked to diabetes mellitus.
