Abstract

ABSTRACT South Kalimantan is rich with medicinal herbs, one of it is Argyreia nervosa. Ethanol extract of A. nervosa leaves has potential as antioxidant with IC50 value of 9,46 ppm. This study aims to isolate antioxidant compounds from ethyl acetate fraction of A. nervosa leaves. Extraction method used by maceration. Separation and purification of isolates was done by TLC, VLC, and CC. Qualitative and quantitative test of antioxidant activity using diphenyl picrylhydrazyl (DPPH) method. Identification of compounds uses UV-Vis and FTIR spectrophotometers. Argyreia nervosa leaf powder was extracted with 96% ethanol and obtained a yield of 16,916%. Rendemen of ethyl acetate fraction is 16,33%. The ethyl acetate fraction is fractionated again using VLC with eluen n-hexane:ethyl acetate (20:1, 15:1, 10:1,9:1, 8:2, 7:3, 6:4; 5:5; 4:6; 3:7) v/v, and methanol:ethyl acetate (1:4) v/v obtained 5 fraction. The fraction 4 is used for isolation using the gravity CC with eluen n-hexane:ethyl acetate (20:1, 15:1, 10:1, 9:1, 7:3, and 5:5) v/v. Qualitative TLC test showed isolates containing antioxidant compounds with DPPH reagent. Analysis with UV-Vis spectrophotometer yielded 344 nm and 299 nm λmax. Abstract

The Callicarpa longifolia Lam. known as ‘kareho’ is one of the plants used by several Dayak tribes for traditional medicine such as antiacne, swelling, wounds, diarrhea, diabetes, and lowering cholesterol levels. This study has obtained scientific data on the characteristics of dried leaves and determined the flavonoids levels of methanol extract of C. longifolia leaves. Characterization includes both qualitative and quantitative examination on the dried leaves of C. longifolia leaves, while the determination of total flavonoid content was determined by the aluminium chloride method and calculated as quercetin equivalent (QE). The results of characteristics observations obtained organoleptic in the form of powder, dark green, bitter taste and have a distinctive smell. Microscopic test of leaves had an actinositic stomata type, ethanol soluble extract content of 17.50 ± 0.10% and water soluble extract content of 16.53 ± 0.37%. Drying loss 8.16 ± 0.25%, total ash content of 5.52 ± 0.06%, and acid insoluble ash content of 0.07 ± 0.01%. The results of phytochemical screening confirmed the presence of saponins, flavonoids, steroids, tannins, glycosides, and phenols. TLC observasions using 254 nm UV light obtained 6 spots and 7 spots appearance on sulphuric acid. It has total flavonoid levels obtained at 7.995 ± 0.050% w/w QE.

Abstract

Nymphaea pubescens Willd telah diketahui mempunyai efek antibakteri, terutama pada biji dan daunnya. Tetapi sampai saat ini bagian bunga dari tanaman tersebut belum dilakukan pengujian aktivitas antibakteri. Tujuan dari penelitian ini untuk melakukan skrining fitokimia, uji aktivitas antibakteri dan penentuan konsentrasi hambat minimum (KHM) dari ekstrak metanol bunga N. pubescens terhadap S. aureus dan E. coli. Proses ekstraksi bunga N. pubescens dimaserasi menggunakan pelarut metanol dengan perbandingan 1: 4 b/v. Metode pengujian yang digunakan ada dua yaitu metode difusi untuk pengujian aktivitas antibakteri dan metode dilusi untuk pengukuran konsentrasi hambat minimum (KHM). Hasil skrining fitokimia didapatkan bahwa ekstrak metanol bunga N. pubescens mengandung senyawa golongan fenolik, saponin dan flavonoid. Hasil pengujian aktivitas antibakteri menunjukkan bahwa ekstrak metanol bunga N.pubescens mampu menghambat S. aureus (diameter hambat 10 ± 0,29 mm) dan E.coli (diameter hambat 10,2 ± 0,50 mm). Konsentrasi hambat minimum dari ekstrak metanol bunga N. pubescens terhadap S.aureus sebesar 12,5% dan terhadap E.coli sebesar 25%. Dapat disimpulkan bahwa ekstrak metanol bunga N. pubescens memiliki aktivitas sebagai antibakteri. Nymphaea pubescens Willd has known to have antibacterial effects, especially on the seeds and leaves. However, until now the flower of the plant has not been tested for antibacterial activity. The purpose of this study was to perform phytochemical screening, antibacterial activity test and determine the minimum inhibitory concentration (MIC) of the methanol extract of N. pubescens flowers against S. aureus and E. coli. The process of extracting N. pubescens flowers is macerated using methanol as a solvent with a ratio of 1: 4 w / v. There are two test methods used, namely the diffusion method for testing antibacterial activity and the dilution method for measuring the minimum inhibitory concentration (MIC). The results of phytochemical screening showed that the methanol extract of N. pubescens flowers contained phenolic compounds, saponins, and flavonoids. The results of the antibacterial activity test showed that the methanol extract of N.pubescens flowers was able to inhibit S. aureus (inhibition diameter 10 ± 0.29 mm) and E. coli (inhibitory diameter 10.2 ± 0.50 mm). The minimum inhibitory concentration of the methanol extract of N. pubescens flowers against S.aureus was 12.5% and against E. coli was 25%. Abstract

ABSTRAK Antioksidan berfungsi untuk menetralisir radikal bebas, sehingga dapat mencegah dan melindungi terjadinya kerusakan tubuh yang dapat menyebabkan timbulnya penyakit degeneratif. Salah satu tanaman endemik Kalimantan Selatan yang teridentifikasi mengandung antioksidan alami yaitu kasturi (Mangifera casturi Kosterm.). Penelitian ini bertujuan untuk mengisolasi dan mengidentifikasi senyawa antioksidan dari buah M. casturi. Ekstraksi dilakukan dengan metode maserasi. Pemisahan dan pemurnian dilakukan dengan KLT , KCV, dan kromatografi kolom gravitasi. Identifikasi senyawa dengan spektrofotometri UV-Vis dan FTIR serta uji kualitatif antioksidan dan aktifitas kuantitatif antioksidan isolat. Ekstraksi 2500 gram serbuk buah M. casturi dengan metanol diperoleh 840,69 gram ekstrak kental berwarna coklat. Fraksinasi 60 gram ekstrak metanol dengan fraksi etil asetat menghasilkan 8,3 gram ekstrak kering. Fraksi etil asetat difraksinasi kembali menggunakan KCV dengan eluen n-heksana : etil asetat (9:1; 8:2; 7:3; 6:4; 5:5; 4:6; 3:7) v/v, etil asetat : metanol (5:5 v/v), 100% metanol diperoleh fraksi A, B, C, D, E, F, G, dan H. Fraksi H dipilih untuk di isolasi menggunakan kromatografi kolom gravitasi dengan eluen n-heksana : etil asetat : metanol (5:3:0,5; 5:4:1; 5:4:2; 5:6:2; 5:6:2,5) v/v dan 100% metanol. Uji kualitatif KLT menunjukkan isolat H-7 mengandung senyawa antioksidan dengan pereaksi DPPH. Aktivitas antioksidan isolat H-7 memiliki nilai IC50¬ sebesar 4,61±0,57 ppm. Analisis isolat H-7 dengan UV-Vis menghasilkan puncak pada λ 274,8 nm (pita I) dan λ 218 nm (Pita II). Analisis FTIR menunjukkan gugus fungsi dari isolat H-7 yaitu -OH, C-H aromatik, C-H alifatik, C=O, C=C aromatik, dan C-O. Kata Kunci : Antioksidan, buah M. casturi, DPPH, fraksi etil asetat, Isolasi

Abstract

Kerusakan oksidatif dalam tubuh manusia dapat disebabkan oleh radikal bebas, sehingga menimbulkan berbagai permasalahan. Daun P. canescens memiliki kandungan senyawa fenolik, tanin, alkaloid, steroid, saponin dan flavonoid yang potensial sebagai antioksidan. Tujuan penelitian ini yaitu untuk melakukan uji aktivitas antioksidan dan antitirosinase fraksi n-butanol daun sungkai secara kualittaif menggunakan kromatografi lapis tipis. Metode yang digunakan yaitu preparasi sampel dengan pembuatan simplisia yang kemudian diekstraksi secara maserasi menggunakan etanol 96%. Hasil ekstrak kental kemudian difraksinasi bertingkat dengan pelarut n-heksan, etil asetat, dan n-butanol. Selanjutnya dilakukan skrining fitokimia, uji aktivitas antioksidan, dan antitirosinase secara kualitatif menggunakan kromatografi lapis tipis. Hasil rendemen ekstrak etanol daun P. canescens yaitu sebesar 7,28% sedangkan rendemen fraksi yaitu 24,8%. Skrining fitokimia ekstrak etanol P. canescens mengandung alkaloid, flavonoid, terpenoid, steroid, tanin, saponin, dan fenol. Kesimpulan yang didapatkan dari penelitian ini yaitu fraksi n-butanol daun P. canscens berpotensi memiliki aktivitas antioksidan yang ditandai adanya noda kuning pucat setelah penyemprotan dengan reagen DPPH, serta berpotensi memiliki aktivitas antitirosinase secara kualitatif yang ditandai dengan adanya bercak putih pada plat KLT. Kata Kunci: DPPH, L-DOPA, Asam askorbat, Enzim  Oxidative damage in the human body can be caused by free radicals, causing various problems. P. canescens leaves contain phenolic compounds, tannins, alkaloids, steroids, saponins and flavonoids that have potential as antioxidants. The purpose of this study was to test the antioxidant and antityrosinase activity of the n-butanol fraction of sungkai leaves qualitatively using thin layer chromatography. The method used is sample preparation by making simplicia which is then extracted by maceration using 96% ethanol. The viscous extract was then fractionated with n-hexane, ethyl acetate, and n-butanol as solvents. Furthermore, phytochemical screening, antioxidant activity, and antityrosinase tests were carried out qualitatively using thin layer chromatography. The yield of P. canescens leaf ethanol extract was 7.28%, while the fraction yield was 24.8%. Phytochemical screening of P. canescens ethanol extract containing alkaloids, flavonoids, terpenoids, steroids, tannins, saponins, and phenols. The conclusion obtained from this study is that the n-butanol fraction of P. canscens leaves has the potential to have antioxidant activity which is indicated by a pale yellow stain after spraying with DPPH reagent, and has the potential to have qualitative antityrosinase activity which is indicated by the presence of white spots on the TLC plate.

Abstract

Tumbuhan kapur naga (Calophyllum soulattri Burm F) merupakan salah satu spesies dari  keluarga Calophyllum yang terdapat di hutan Kalimantan khususnya daerah lahan basah. Secara empiris dimasyarakat memanfaatkan kulit batang C. soulattri untuk mengobati penyakit kulit yang salah satu mekanisme proses penyembuhan melawati tahap inflamasi. Penelitian ini bertujuan untuk melakukan penulusuran kandungan fitokimia dari fraksi n-heksana kulit batang C. soulattri dan melakukan uji aktivitas antiinflamasi in vitro dengan metode uji penghambatan denaturasi protein menggunakan spektrofotometer uv-vis. Metode penelitian diawali dengan tahapan pembuatan simplisia, ekstraksi dengan etanol 96%, dan difraksinasi menggunakan n-heksana.  Setelah itu dilakukan  uji penelusuran kandungan fitokimia menggunakan uji tabung dan pengujian antiinflamasi menggunakan spektrofotometri uv-vis menggunakan metode penghambatan denaturasi protein secara kuantitatif dengan natrium diklofenak sebagai kontrol positif. Hasil dari identifikasi fitokimia diperoleh bahwa fraksi n-heksana mengandung alkaloid, fenol, tanin, flavonoid, saponin, dan triterpenoid. Uji aktivitas antiinflamasi fraksi n-heksana kulit batang C. soulattri menghasilkan nilai IC50 sebesar 27,43±0,74 ppm dan natrium diklofenak dengan nilai  IC50 sebesar 39,17±0,86 ppm. Berdasaran uji t-test diperoleh hasil yang menunjukkan berbeda bermakna  dengan IC50 Natrium diklofenak (sig = 0,00). Kesimpulan dari penelitian ini yaitu fraksi n-heksana kulit batang C. soulattri memiliki aktivitas antiinflamasi yang lebih baik dari natrium diklofenak. Kata Kunci: Bovine Serum Albumin, IC50, Natrium Diklofenak, Kuantitatif Kapur naga (Calophyllum soulattri Burm F) is a species of the Calophyllum family that found in the forests of Kalimantan, especially in wetland areas. Empirically, the community uses the bark of C. soulattri to treat skin diseases, which is one of the mechanisms of the healing process through the inflammatory stage. The purpose of this study was to investigate the phytochemical content of the n-hexane fraction of the stem bark of C. soulattri and to test its anti-inflammatory activity in vitro with the protein denaturation inhibition test method using uv-vis spectrophotometer. The research method begins with the steps of making simplicia, extraction with 96% ethanol, and fractionation using n-hexane. After that, a test for tracing the phytochemical content was carried out using a tube test and an anti-inflammatory test using uv-vis spectrophotometry using a quantitative protein denaturation inhibition method with diclofenac sodium as a positive control. The results of the phytochemical identification showed that the n-hexane fraction contained alkaloids, triterpenoids, phenols, tannins, flavonoids, and saponins. The anti-inflammatory activity test of the n-hexane fraction of C. soulattri stem bark produced an IC50 value of 27.43±0.74 ppm and diclofenac sodium with an IC50 value of 39.17±0.86 ppm. Based on the t-test, the results showed that it was significantly different from the IC50 of diclofenac sodium (sig = 0.00). The conclusion of this study is that the n-hexane fraction of the stem bark of C. soulattri has better anti-inflammatory activity than diclofenac sodium.

Abstract

Kratom (Mitragyna speciosa Korth.) merupakan tanaman tropis yang memiliki kemampuan melindungi kulit dari paparan sinar matahari. Tujuan dari penelitian ini adalah untuk mengetahui aktivitas tabir surya ekstrak etanol daun M. speciosa kuantitatif menggunakan spektrometri UV-Vis. Ekstraksi dilakukan dengan metode maserasi menggunakan pelarut etanol 96%. Kandungan metabolit sekunder secara kualitatif dilakukan menggunakan skrining fitokimia. Aktivitas tabir surya diuji menggunakan lambda 290-320 nm dengan interval 5 nm. Ekstrak etanol daun M. speciosa diperoleh sebesar 86,09 gram (17,218%) dan pada konsentrasi 250 ppm menunjukkan adanya aktivitas tabir surya dengan nilai SPF sebesar 8,433. Daun kratom asal Kalimantan Selatan berpotensi sebagai produk kosmetik tabir surya. Kata Kunci: Kratom, Spektrofotometri UV-Vis, Tabir Surya, Flavonoid Kratom (Mitragyna speciosa Korth.) is a tropical plant that has the ability to protect the skin from sun exposure due to the presence of flavonoids. The purpose of this study was to determine the sunscreen activity of the M. speciosa leaves ethanolic extract, quantitatively by UV-Vis spectrometry. Extraction was carried out by maceration method in 96% ethanol. The content of secondary metabolites were qualitatively determined using the phytochemistry screening study. The sunscreen activity was tested using 290-320 nm (5 nm intervals). The ethanol extract of M. speciosa leaves was obtained by 86.09 grams (17.218%) and with a concentration of 250 ppm showed sunscreen activity with an SPF value of 8.433. Kratom leaves from South Kalimantan have potential as a sunscreen.

Abstract

Daun Sungkai (Peronema canescens Jack) mengandung metabolit sekunder antara lain tannin, alkaloid, terpenoid, steroid, dan flavonoid. P. canescens merupakan tanaman asli Kalimantan Selatan yang berpotensi sebagai perawatan kulit alami berdasarkarkan kandungan tannin yang memiliki aktivitas antioksidan sehingga dapat dikembangkan sebagai produk tabir surya. Penelitian ini bertujuan untuk menentukan nilai SPF dan tannin total fraksi etil asetat daun Sungkai (P. canescens) menggunakan spektrofotometri UV-Vis. Metode ekstraksi dilakukan dengan maserasi menggunakan etanol 96% dan penggunaan fraksi n-heksana dan etil asetat. Penentuan tanin total menggunakan standar katekin dengan konsentrasi 40,60, 80, 100 dan 120 ppm. Hasil kadar tannin total sebesar 0,5586% ±0.01516 ekivalen katekin dan nilai SPF sebesar 5.703±0.021. Dengan demikian dapat disimpulkan daun Sungkai (Peronema canescens Jack) dapat dikembangkan sebagai tabir surya alami.  Kata Kunci: Daun Sungkai, Tabir Surya, Tannin Total, UV-Vis, SPF  Sungkai leaves (Peronema canescens Jack). contains secondary metabolites i.e tannins, alkaloids, terpenoids, steroids, and flavonoids. P. canescens, natural plant from South Kalimantan, has potential benefits as natural skincare based on tannin which contains antioxidant activity that can be developed as a sunscreen product. This study aimed to determine the sun protection factor (SPF) and total tannin ethyl acetate fraction of sungkai leaves (P. canescens) using UV-vis spectrophotometry. The extraction method was maceration with 96% ethanol and the fraction used n-hexane and ethyl acetate. The determination of total tannin with catechin standards concentration 40,60,80, 100 and 120 ppm. The result of total tannin was 0.5586% ±0.01516 catechin equivalents and the SPF value was 5.703±0.021. The conclusion of the research is that Peronema canescens Jack can be developed into natural sunscreen.
