Abstract

The three-dimensional (3D) structure of proteins is necessary to understand the properties and functions of proteins. Determining protein structure by laboratory equipment is quite complicated and expensive. An alternative method to predict the 3D structure of proteins in the in silico method. One of the in silico methods is homology modeling. Homology modeling is done using the SWISS-MODEL server. Proteins that will be modeled in the 3D structure are proteins that do not yet have a structure in the RCSB PDB database. Protein sequences were obtained from the UniProt database with code A0A0B6VWS2. The results showed that there were two models selected, namely model-1 with the PDB code template 1q0e and model-2 with the PDB code template 3gtv. The results of sequence alignment and model visualization show that model-1 and model-2 are identical. The evaluation and assessment of model-1 on the Ramachandran Plot have a Favored area of ??97.36%, a MolProbity score of 0.79, and a QMEAN value is 1.13. Model-1 is a good 3D protein structure model.

Abstract

The ORF3a protein from SARS-CoV has functions in terms of ion channel activity, modulates the trafficking properties of SARS-CoV spike (S) protein, increases fibrinogen expression in pulmonary epithelial cells, and induces apoptosis. So that research is needed to overcome the ORF3a experiment. The method in this research uses the computational screening method with autodok4 software. The results of this study resulted in free binding energy between Hassk Rhodomyrtus tomentosa (Aiton) and ORF3a compounds, namely: α-tocopherol-quinone (-5.86); blumeatin (-4.98); methyl cinnamate (-4.44); myricetin (-4.49); naringenin (-4.93); quercetin (-4,9); rhodomyrtone (-6); rhodomyrtosone B (-7.11); rhodomyrtosone C (-6.77); tetrahydroxyflavanone (-4.91); α-tocopherol A (-6.72); verimol K (-4.89); watsonianone A (-7.55). Based on the data obtained, the ligand with the most potential due to the stability of the bond is watsonianone A.

Abstract

Many biscuits circulating in the community are not labeled and do not include the value of water and protein content in accordance with SNI. Therefore, it is necessary to determine the water content and protein content in the biscuits. Determination of water content and protein content in biscuits was carried out on 2 biscuit samples taken at the Banjarbaru Market. The water content test on the sample was carried out by the gravimetric method and the protein content test was carried out by the Kjeldahl method. The results obtained were the test of water content of 2 biscuit samples respectively at 4.43% and 3.84%. While the results of the protein content test from 2 samples are 5.31% and 6.89%, respectively. According to SNI 01-2973-2011 the water content limit does not exceed 5% while the protein content limit is a minimum of 5%. The results of 2 samples showed the water content and protein content in accordance with SNI.

Abstract

Nuclear factor kappa B (NF-κB) is a transcription factor that regulates the expression of genes important for innate and adaptive immune responses. NF-κB overactivation is associated with inflammatory diseases such as autoimmune, systemic lupus erythematosus (SLE) and rheumatoid arthritis (RA). This study aims to screen the chemical content of C.indicum against NF-κB-inducing kinase inhibitors. There are two methods used in this study, namely through PASSonline and docking. The docking method used was PLANTS. The results of PASSonline compounds that have P.a values above 0.7 weree arachidonic acid, linoleic acid, rutin, methyl urasolate, while the docking scores obtained were asquisqualic acid: -60.0382; arachidonic acid : -100,9410; linoleic acid: -93.8730; routine : -102,9320; methyl urasolate: -47,9166. This negative docking score indicates that the reaction between the compound and the target occurs easily. In conclusion, the compound predicted to have a potential as an inhibitor of NF-κB is Rutin.

Abstract

Abstract: Gluthathione s-transferases (GSTs) play an important role against carcinogens, therapeutic drugs, and various types of cellular oxidative damage. Judging from its role in overcoming cellular oxidative damage, the activity of GSTs is one of the antioxidant enzymes. One of the plants that has been tested for its antioxidant ability is M. paniculatus. The content of M. paniculatus has been found so that it can be used to test the stability of the interaction between ligands and receptors. The aim of this research is to find compounds from M. paniculatus that have the potential to stabilize the interaction with the receptors. This docking method begins with ligand preparation and protein preparation followed by docking, visualization of interactions and analysis of the results. The research resulted in various docking scores with the various proteins involved. The stability of the interaction is obtained when the ligand binds to Glutathione S-transferase type A3 is hesperetin, sitosterol and sitosterol, to Glutathione S-transferase type theta-2 is β-daucostero, to Glutathione S-transferase type P is quercetin. Keywords: GSTs, M. paniculatus, docking

Abstract

Not many researches on a fruit of limpasu (Baccaurea lanceolata Muell. Arg ) were found, while the use by tribal hamlets in Kalimantan already became a daily routine before they went into the field with the way made powder and smeared on the face. Therefore, the research on active substances needs to be done. For this study, we prepared two isolates which coded as K2 and CE, K2 from qualitative test negative isolates phenolic groups was identified by spray FeCl3, whereas qualitative test positive isolates were coded as CE and detected by a nitrogen-containing reagents dragendorf spray. The K2 isolate from SPF test resulted an SPF value of 3.2 at a concentration of 0.2 mg/ml, while CE isolate was given a value of SPF 5.0 at a concentration of 0.2 mg/ml. According to sensitivity skin test on rabbit skin, both K2 and CE isolated were given the value below 1, indicated the isolates were found to not irritate. This study proposes the potential from Baccaurea lanceolata Muell.Arg fruit isolates to be developed as a sunscreen for UV-protection.Keywords : Baccaurea lanceolata Muell.Arg, SPF test, sensitivity skin test

Abstract

Digestive tract disorders, especially gastric disorders, are often experienced by people. One drug to treat this disorder has a mechanism of blocking the H2 receptor. This research was conducted to find compounds from C.verum which have the stability of bind to H2 receptors. The method used is protein modeling with swiss-model, docking with PLANTS (CHEMPLP) and activity prediction. The test results obtained that the docking score was ?- amorphene (-65,79), ?-bergamotene (-65,48), ?-copaene (-66,62), ?-cubebene (-66,46), Cadinene (-64 , 79), Camphor (-52.15), Caryophyllene (-62.61), Cinnamaldehyde (-68.17), Epicatechin (-80.43), Ergosterol (-85.24), Eugenol (-67.35), Hydrocinnamaldehyde (-65,53), Quercetin (-74,38), Protocatechuic acid (-71,49), Stigmasterol (-88,88), 4- (2,3-dihydro-3- (hydroxymethyl) - 5- (3-hydroxypropyl) -7- (methoxy) benzofuranyl] -2-methoxyphenyl (-85,29). Combined with the probability activity of compounds that have the potential to be further developed are Epicatechin and urolignoside.

Abstract

UV rays cause skin damage such as loss of skin elasticity, wrinkles, sagging, and skin cancer. South Kalimantan is a wetland area with more than 4,000 species of medicinal plants. Mundar (Garcinia forbesii) is one of the plants in South Kalimantan. Mundar pericarp is empirically used for cosmetics which is believed to protect the skin from ultraviolet rays. The purpose of the study was to determine the sunscreen potency of the extract and fraction from pericarp of mundar (Garcinia forbesii) based on the Sun Protection Factor (SPF) parameter. The research started from material drying, making simplicia powder, extraction, and fractions. The extracts and fractions were measured for SPF values ??using a UV-Vis Spectrophotometer. The results showed that the ethanol extract and ethyl acetate fraction had weak protection, while the n-hexane fraction had extreme protective power with SPF values ??in the range of 6.51 – 30.00

Abstract

Fruits of Baccaurealanceolata is commonly used by tribe banjar in south Borneo to protect the skin damages due to sun light. People apply the pulverized fruits to the skin that will be exposed to sunlight. Antioxidant bioassay method (DPPH) is used to prove the effectiveness of the fruits.Baccaurea lanceolata fruitsvwere extracted by 70% ethanol, followed by fractionation with n-hexane, ether, ethyl acetate and methanol to give n-hexane (fr-H), ether (fr. Et2O), ethyl acetate (fr. EtOAc) and methanol (fr. MeOH) fractions respectively. Preparative TLC (SiO2) of the fr. EtOAc that showed the the highest antioxidant activity among those fractions. Based on spectroscopic data and comparison with reported data, the isolated compounds were identified as β-Sitosterol.

Abstract

Tumor necrosis factor-alpha (TNF-α) merupakan sitokin proinflamasi yang terlibat dalam berbagai penyakit seperti radang selaput otak, rematik, asma, tuberkolosiS, periontitis kronis, crohn desease, hepatitis dan sinusitis hidung. Sehingga ekspresinya perlu dikontrol untuk mengatasi penyakit ini. Penelitian ini bertujuan melakukan skrining inhibitor reseptor TNF-alpha convertase (TACE) sehingga menurunkan ekspersi TNF-α. Dengan menurunnya produksi TNF-α maka dapat sebagai antiinflamasi atau imunosupresan. Salah satu tanaman yang dapat dipergunakan untuk skrining ini adalah A.excels. Metode yang dipergunakan untuk skrining melalui PASSonline dan Docking dengan PLANTS serta dilihat prediksi memasuki peredaran darah dengan SWISSADME. Berdasarkan PASSonline Senyawa yang memiliki skor probability to be active imunosupreasant tinggi adalah batulinic acid (0,724), alphitolic acid (0,763), cis-coumaroyl alphitolic acid (0,771). Sedangkan yang memiliki probability to be active antiinflamasi adalah batulinic acid (0,741) dan cis-coumaroyl alphitolic acid (0,783). Berdasarkan stabilitas interaksi, senyawa yang memiliki kemampuan minimal 80% terhadap ligand referensi adalah alphitonin (98,25%), alphitexolide (92,33%), cis-coumaroyl alphitolic acid (98,31%). Kesimpulannya senyawa yang berpotensi yang memiliki stabilitas interaksi yang bagus adalah cis-coumaroyl alphitolic acid dan alphitonin
