Abstract

Penelitian ini bertujuan untuk mengkaji potensi pemanfaatan teknologi Augmented Reality (AR) dalam pelestarian dan edukasi sejarah lokal di Kampung Bandar, Pekanbaru. Kampung Bandar menyimpan berbagai bangunan bersejarah seperti Rumah Singgah Tuan Kadi, Istana Hinggap, dan Rumah Tenun yang memiliki nilai penting sebagai sumber sejarah dan identitas budaya. Pembelajaran sejarah lokal seringkali dianggap membosankan dan kurang interaktif, serta menghadapi kendala seperti keterbatasan sumber dan media pembelajaran. Penelitian ini menggunakan pendekatan kualitatif deskriptif untuk memperoleh pemahaman mendalam mengenai pemanfaatan teknologi AR. Data dianalisis secara sistematis untuk menggambarkan keterkaitan antara teknologi AR dan nilai historis lokal. Hasil penelitian menunjukkan bahwa Implementasi AR dapat menghadirkan kembali bangunan bersejarah seperti Istana Hinggap, Rumah Singgah Tuan Kadi, dan Rumah Tenun secara virtual dengan detail yang memudahkan pemahaman peserta didik, mengatasi kendala sifat statis dan abstrak materi sejarah dengan menghadirkan media belajar yang lebih interaktif dan kontekstual, sehingga meningkatkan daya tarik dan motivasi belajar peserta didik, serta memfasilitasi gaya belajar visual, audio, dan kinestetik. Dengan demikian, pengembangan dan penerapan teknologi AR di Kampung Bandar merupakan langkah strategis untuk pelestarian warisan budaya, penguatan identitas lokal serta peningkatan kualitas pendidikan sejarah lokal yang lebih bermakna dan relevan.

Abstract

Status dari kewarganegaraan merupakan hak dari setiap orang bahkan seorang anak juga berhak untuk mendapatkannya, dengan adanya status dari kewarganegaraan maka anak dikarenakan menunjukkan adanya hubungan hukum antara perorangan dan juga warganegara. Anak merupakan seseorang yang masih dibawah umur dan berhak mendapatkan penegakan hukum akibat dari kewarganegaraan ganda, dimana perlindungan hukum tersebut akan pemenuhan kesejahteraan serta keadilan juga kepastian termasuk perlindungan anak sampai beranjak dewasa sehingga dalam kehidupannya mendapatkan jaminan akan pemenuhan terhadap hak-haknya. Penegakan hukum yang diberikan juga jika timbul sengketa atau konflik jika terjadi pembagian warisan dan juga harta gono gini manakala berkaitan dengan kewarganegaraan ganda yang dianutnya, sehingga anak mendapatkan perlakuan yang adil dan tidak memihak dalam penyelesaian akan konfliknya atau sengketanya.

Abstract

Penelitian ini bertujuan untuk: (1) mengetahui korelasi kompetensi kepala sekolah dengan prestasi sekolah dasar, (2) mengetahui  korelasi kinerja guru dengan prestasi sekolah dasar, dan (3) mengetahui korelasi kompetensi kepala sekolah dan kinerja guru dengan prestasi sekolah dasar se Kecamatan Rejotangan. Penelitian ini mengunakan desain penelitian korelasi yang berbentuk ex-post facto untuk mengumpulkan data yang berasal dari fenomena kehidupan nyata dengan tidak memberikan perlakuan terhadap kondisi tertentu. Pengambilan sampel dilakukan dengan teknik multi stage sampling dengan 2 tahap yakni cluster sampling dan random sampling. Sampel dalam penelitian ini berjumlah 42 responden yang terdiri dari 5 kepala sekolah dan 37 guru. Penelitian ini menggunakan teknik pengumpulan data secara nontes berupa angket (kuesioner) dan studi dokumen. Analisis data yang digunkan dalam penelitian menggunakan teknik analisis regresi linear berganda menggunakan SPSS untuk mengetahui korelasi yang signifikan antara kompetensi kepala sekolah dan kinerja guru dengan prestasi sekolah. Berdasarkan hasil penelitian yang telah dilakukan dapat disimpulkan bahwa: (1)Terdapat korelasi positif antara kompetensi kepala sekolah dengan prestasi sekolah dasar se Kecamatan Rejotangan, ditunjukkan dengan nilai signifikansi t sebesar 0,012, berarti ada korelasi yang signifikan dari variabel X1 terhadap variabel Y, (2) terdapat korelasi positif antara kinerja guru dengan prestasi sekolah se Kecamatan Rejotangan dibuktikan dengan nilai signifikansi t sebesar 0,002, berarti ada korelasi yang signifikan dari variabel X2 terhadap variabel Y, (3) terdapat korelasi positif antara kompetensi kepala sekolah dan kinerja guru dengan prestasi sekolah dasar se Kecamatan Rejotangan,  dibuktikan dengan nilai F hitung sebesar 22,963, menunjukkan bahwa variabel X1 dan X2 secara bersama-sama mempengaruhi Y. Adapun saran yang disampaikan oleh peneliti adalah: kepala sekolah diharapkan mampu menerapkan kebijakan-kebijakan yang positif kepada guru dan guru hendaknya meningkatkan profesionalitas dan kinerjanya.

Abstract

Juridical Analysis of a Specific Time Work Agreement (PKWT) to Realize the Protection of Workers' Rights and Welfare (Research Study at the PT. Bersama Gemilang Property Batam Office), has been carried out as it should, in accordance with Law Number 11 of 2020 concerning Job Creation. There are factors related to the protection of the rights and welfare of contract workers. The qualifications/types of writing in this journal use normative legal writing, and then integrate it with sociological/empirical legal writing, and to analyze some of the problems in this journal, Lawrence M. Friedmen's big theory, Jeremy Bentham's middle theory, and application theory are used. / applied theory Mochtar Kusumaatmadja. The results of the analysis are still experiencing several obstacles both internally and externally. The obstacles faced have not been the implementation of rights and welfare, which have been prioritized and carried out but not yet perfectly. It is very necessary and improved in terms of supervision in terms of quality and quantity in order to suppress and not give space to violators who do not comply with contract workers. There is also a need for wider socialization in a timely manner regarding the implementation of the new law on the rights and welfare of contract workers.

Abstract

Efektivitas Program Percepatan Peningkatan Tata Guna Air Irigasi (P3 -TGAI) di UPT Dinas Pekerjaan Umum, Tata Ruang, Perumahan dan Permukiman Wilayah Sukaratu Kabupaten Tasikmalaya ditemukan adanya permasalahan kurang efektifnya P3-TGAI antara lain masih adanya saluran irigasi yang belum mendapatkan P3-TGAI dan kurangnya sosialisasi P3-TGAI oleh pegawai UPTD. Dari permasalahan tersebut, penulis merumuskan masalah. Bagaimanakah Efektivitas Program Percepatan Peningkatan Tata Guna Air Irigasi (P3 -TGAI) di UPT Dinas Pekerjaan Umum, Tata Ruang, Perumahan dan Permukiman Wilayah Sukaratu Kabupaten Tasikmalaya. Tujuan dari penelitian ini untuk mengetahui Efektivitas Program Percepatan Peningkatan Tata Guna Air Irigasi (P3 -TGAI) di UPT Dinas Pekerjaan Umum, Tata Ruang, Perumahan dan Permukiman Wilayah Sukaratu Kabupaten Tasikmalaya. Penelitian ini menggunakan metode penelitian deskriptif dengan pendekatan kualitatif. Teknik pengumpulan data yang digunakan adalah wawancara, observasi serta studi dokumentasi. Wawancara dan triangulasi dilakukan terhadap informan penelitian yaitu Kepala UPTD, Kepala Sub Bagian Tata Usaha, Pelaksana UPTD, Ketua P3A, dua orang anggota P3A dan 2 orang petani. Teknik analisis data dengan, reduksi data, display data serta verifikasi dan penarikan kesimpulan. Berdasarkan hasil penelitian dan pembahasan, penulis menyimpulkan bahwa efektivitas Program Percepatan Peningkatan Tata Guna Air Irigasi (P3 -TGAI) di UPT Dinas Pekerjaan Umum, Tata Ruang, Perumahan dan Permukiman Wilayah Sukaratu Kabupaten Tasikmalaya belum tercapai. Dari lima dimensi efektivitas yang dianalisis, dua dimensi efektivitas belum dilaksanakan dengan baik yaitu dimensi keberhasilan program dan dimensi keberhasilan sasaran sedangkan dimensi efektivitas yang telah dilaksanakan dengan baik adalah dimensi kepuasan terhadap program, dimensi tingkat input dan output, dan dimensi pencapaian tujuan menyeluruh.

Abstract

Glycerol is a renewable resource found as the main by-product in the transesterification of triglycerides and fat saponification. Due to the increased production of plant oils, especially palm oil in developing countries, and their larger use by the oleochemical industry, glycerol surpluses are on the world market and this may result in a decrease in glycerol price. As a consequence, biotechnological processes have been developed to convert this substrate into value-added products, such as 1,3-propanediol (1,3-PD). The microbial production of 1,3-PD could be competitive to chemical routes assuming that it is based on cheap raw material and an optimised process. In the screening for 1,3 PD–producing bacteria, raw glycerol as by-product from rapeseed oil processing unit was used as a carbon source compared with commercial glycerol. By using increasing concentration of both glycerols from 50 to 150 g/l, two potential bacteria were obtained from soil samples. BMP-1 was obtained from an enrichment culture using 50 g/l commercial glycerol, while BMR-1 was obtained from an enrichment culture using 100 g/l raw glycerol. The highest conversion yield obtained using the isolate BMP-1 was around 0.62 g 1,3-PD formed per mol glycerol consumed, and 0.73 mol 1,3-PD formed per mol glycerol using the isolate BMR-1. No bacteria were obtained from cultures using 150 g/l commercial and raw glycerol, respectively, which indicated that higher concentration of glycerol has inhibition effect.

Abstract

Cellulase enzymes have attracted considerable attention in recent years due to their great biotechnological and industrial potential. Cellulase enzymes provide a key opportunity for achieving tremendous benefits of biomass utilization through the bioconversion of the most abundant cellulosic material into the simplest carbohydrate monomer, glucose. Nowadays, the sources of cellulase-producing bacteria have been broadened into the presence of symbiotic bacteria in herbivorous animal and also from marine. Takifugu rubripes or known as Puffer fish is a unique poisonous vertebrate but nevertheless is considered a delicacy in Korea. The diet of the puffer fish includes mostly algae. This dietary habit considers Puffer fish as host of cellulase-producing bacteria, especially on its gut. In the present study an attempt has been made to search for the cellulolytic bacteria in the gut of Takifugu rubripes. Fifty five microorganisms have been isolated using 1% (w/v) carboxymethyl cellulose (CMC) as substrate. Congo red dye test and DNS method were then used for screening the extracellular cellulase activity of the strains. Among them, TD6 strain has shown the highest performance in term of cellulase activity. In order to evaluate the optimum culture condition of the isolate TD6 for cellulase production, the strain was grown at various temperatures, pH, carbon sources, and nitrogen sources. Under optimum condition, the maximum specific activity of 2.13 U/mg protein was achieved after growth the strain with 1.5% CMC at 45ºC pH 6 for 3 days, respectively. Based on 16S rRNA gene analysis it is proposed that the strain was identified as Bacillus subtilis.

Abstract

Transferrin receptor (TfR) levels are elevated in various types of cancer cells and correlate with the aggressive or proliferative ability of tumor cells. Therefore, TfR levels are considered useful as a prognostic tumor marker, and TfR is a potential target for drug delivery in therapy of malignant cells. In such kind of targeted delivery system, antibody fragments are frequently used as targeting moiety. Here, we report the generation of an anti-TfR single-chain antibody variable (scFv). The cDNA encoding the variable heavy and light chain domains of the scFv antibody fragment was derived from the anti-TfR monoclonal antibody LUCA31. The gene encoding the anti-TfR scFv fragment was codon optimized for expression in Escherichia coli, subsequently synthesized, and cloned into the pJExpress-804 Rhamex vector. The expression vector utilizes the E. coli rhaB promoter and corresponding regulatory genes, and is tightly regulated by the presence of L-L-rhamnose, it is also tightly regulated in the absence of L-L-rhamnose by the addition of D-glucose The His6-tagged anti-TfR scFv fragment was expressed in E. coli NiCo21 and purified by means of immobilized metal chelate affinity chromatography on TALON™ matrix. In SDS-PAGE, a single band corresponding to a molecular mass of approximately 30 kDa was observed which corresponded to the predicted molecular mass based on the amino acid sequence.

Abstract

A facile generation of a recombinant antibody fragment with intrinsic fluorescent properties of the monomeric mCherry fluorescent protein is described. The so-called mCherryBody was designed based on the structure model of the variable fragment of anti-transferrin receptor antibody LUCA31 and the X-ray crystallographic structure of the mCherry protein. mCherryBody was constructed to retain optimal spatial geometry between the C- and N-termini of the antibody light-chain (VL) and heavy-chain (VH) by mimicking the domains interface pairing in antibody Fab fragments and incorporation of the mCherry fluorescent protein as a bridging scaffold. The gene encoding the chimeric protein was cloned into the pJExpress414 expression vector, expressed and secreted into the periplasm of Escherichia coli NiCo21(DE3) for assembly and disulphide bond formation. Based on its amino acid sequence, mCherryBody was predicted to have a molecular weight of 51.46 kDa. The modular assembly used in the generation of mCherryBody may permit the interchange of binding sites and of fluorescent proteins to create robust panels of coloured antibody fragments. Thus, the mCherryBody platform facilitates rapid generation of colored single-chain variable fragment (scFv) chimeras that could be used for screening of antibodies against cell surface markers or receptors.
