Abstract

Flying fish (Exocoetidae) is a pelagic fishery commodity that holds economic value among the people of eastern Indonesia. Flying fish are advantageous as they are abundant in Indonesian sea waters, making them an affordable source of omega-3, which is beneficial for human health. However, the high demand for flying fish has resulted in overfishing, which poses a risk of reducing their natural population. Proper management of this commodity is necessary to minimize these risks. Effective management requires adequate information, including genetic diversity data. This study aims to determine the genetic diversity of flying fish in Southeast Sulawesi, specifically in the Banda Sea and Bone Bay populations. Tissue samples were collected from flying fish in the two populations during the study in March 2022. The results show that the genetic diversity values of flying fish in Bone Bay and the Banda Sea were 0.984 and 1.00, respectively, while the nucleotide diversity values were 0.021 and 0.018. High genetic diversity values and nucleotide diversity values were found between the two populations, with values of 0.990 and 0.020, respectively. Based on phylogenetic analysis, the two populations in Southeast Sulawesi are genetically similar.

Abstract

This community service activity is a training activity on the manufacture of health and food products that have the opportunity to improve the community's economy as well as providing education on the use of moringa leaves as face masks and papaya fruit as an analog tomato sauce to mothers in Tondonggeu Village, Nambo District, Kendari City. Tomato sauce is generally made from tomato raw materials, but it can also be made from other ingredients so it is called an analog tomato sauce. Papaya as a substitute for tomatoes in making analog tomato sauce can be done because papaya has characteristics like tomatoes, namely color, organic acid content, and viscosity, even papaya has a higher vitamin C level than tomatoes.  Moringa leaves contain many vitamins, including vitamin A, vitamin C, amino acids and antioxidants such as flavonoids, quercetin and zeatin which function to maintain dry skin hydration and prevent the appearance of wrinkles, because it helps fight free radicals, so it is very good to be used as a cosmetic ingredient in the form of a face mask. Participants in this training involved residents of Tondonggeu village, Thematic KKN students and a team of accompanying lecturers from the Biotechnology and Biology study program of FMIPA Halu Oleo University. The training methods used are lectures, demonstrations and discussions. The lecture method was carried out with the aim of providing information and knowledge about moringa masks and analog ketchup. The demonstration aims to provide skills to mothers through the practice of directly processing moringa leaves into masks and papaya fruits into analog tomato sauce while the discussion is intended to show the activeness and understanding of the community during the training process. The results of the training on making masks and analog tomato sauce carried out by mothers are in the good category with an average success of 85%, their response to the implementation of the training is very good as seen from the enthusiasm of the residents to participate in the activity from the beginning to the end. This training activity can increase knowledge and improve mothers' skills in producing moringa masks and papaya analogue tomato sauce.

Abstract

Several biotechnology industries are exploring the characteristics of lipase-dependent chaperones due to their distinctive biochemical traits. This study aimed to employ bioinformatics to analyze the molecular structure of Ralstonia picketii BK6's lipase-dependent chaperon (LipRM). The sequence mapping and amino acid distribution were examined using BioEdit (version 7.0.9.1). SignalP 5.0 and Interpro are employed for signal peptide detection, whereas Swiss-Model and VMD 1.9.2 are used for molecular dynamics modelling. The results showed that the Shine-Dalgarno sequence was discovered in the LipRM promoter, seven nucleotides upstream of the initiation codon (AUG) with the 5'-AGGAGA-3', and has a terminator region that facilitates the formation of a secondary structure. The protein's 3D structure prediction results indicate differences in the alpha helix chains (residues 166-174 and 254-271) between LipRM and the reference lipase. LipRM's molecular structure comprises a detachable signal peptide, and with variations in helix alpha chain conformation and ligand geometry.

Abstract

Madrasah Aliyah Negeri Insan Cendekia (MAN IC) in Kendari City is one of the OSN (National Science Olympiad) participants in the subject of biology, which is required to understand the idea and practice of PCR (Polymerase Chain Reaction). However, there are insufficient skilled PCR teachers at the school, and no PCR facilities are currently accessible. The goal of this mentoring is to provide an in-depth understanding of the concept of PCR as well as full training on how to use PCR effectively. The method consists of two components: community education and training. The introduction of the PCR concept in this community service includes genetic material, DNA concepts, genes, genomes, DNA isolation, and DNA replication, while the training material includes solution taking techniques, PCR solution preparation, PCR usage procedures, PCR result visualization with electrophoresis, and data interpretation. The PCR has three main steps consisting of denaturation, annealing and extension which are carried out for 30-35 cycles with different temperatures at each step. The evaluation results showed that this mentoring helped the student understand PCR and was beneficial for the OSN competition due to its relevance to the competition syllabus. Moreover, the student also received a bronze medal in the biology competition.

Abstract

ABSTRACT Coconut fruit processing produces by-products in the form of coconut pulp. Coconut pulp organic waste has a high fiber content that can benefit the body when consumed. So far, coconut pulp is only used as animal feed with very low product prices. The high amount of fiber content and the potential for protein and fat, provide opportunities for waste dregs to be developed into processed products. One of the efforts that can be done is to optimize the use of organic waste of coconut pulp into flour as raw material for making bread through community service activities. The problem faced by partners is the lack of information obtained by the community about making coconut pulp flour which can be used as a substitute for wheat flour for other food products.   The solution to this problem is to provide training and guidance on making coconut pulp flour and its processed products. This service aims to improve the skills of mothers by utilizing their potential and the potential of available natural resources optimally, in an effort to increase economic income. This training is the use of coconut pulp into flour for mothers in Talia Village, Abeli District, Kendari City. This method of service uses lectures, discussions and demonstrations. This training was attended by mothers in Talia Village, Abeli District. The results of the training obtained by the mothers of Talia Village can make bread from coconut pulp with various flavors. The training greatly supports the improvement of participants' skills and knowledge. During the training, all mothers enthusiastically participated in the course of activities and actively discussed both ways. Participants understand and can practice the technique of processing coconut pulp into raw materials for making bread and can be a business opportunity. Limbah organik ampas kelapa memiliki kandungan serat yang tinggi yang dapat bermanfaat bagi tubuh apabila dikonsumsi. Selama ini ampas kelapa hanya dimanfaatkan sebagai pakan ternak dengan harga produk yang sangat rendah. Kandungan jumlah serat yang tinggi serta potensi protein dan lemak, memberi peluang limbah ampas untuk dikembangkan menjadi produk olahan. Salah satu upaya yang dapat dilakukan yaitu dengan mengoptimalkan pemanfaatan limbah organik ampas kelapa menjadi tepung sebagai bahan baku pembuatan roti melalui kegiatan pengabdian masyarakat. Permasalahan  yang  dihadapi  oleh  mitra  adalah  kurangnya  informasi  yang  didapatkan  oleh masyarakat tentang   pembuatan   tepung   ampas   kelapa   yang   dapat dimanfaatkan   sebagai   pengganti   tepung   terigu   untuk   produk   pangan   lainnya.   Solusi dari permasalahan tersebut adalah memberikan pelatihan dan bimbingan tentang pembuatan tepung ampas kelapa beserta produk olahannya. Pengabdian ini bertujuan meningkatan keterampilan ibu-ibu dengan memanfaatkan potensi yang dimiliki dan potensi SDA yang tersedia secara optimal, dalam upaya meningkatkan pendapatan ekonomi. Pelatihan ini merupakan pemanfaatan ampas kelapa menjadi tepung pada ibu ibu Kelurahan Talia Kecamatan Abeli Kota Kendari. Metode pengabdian ini menggunakan metode ceramah, diskusi dan demonstrasi. Pelatihan ini diikuti oleh ibu ibu di Kelurahan Talia Kecamatan Abeli. Hasil pelatihan diperoleh ibu-ibu Kelurahan Talia dapat membuat roti dari ampas kelapa dengan berbagai rasa. Pelatihan tersebut sangat mendukung peningkatan keterampilan dan pengetahuan peserta. Selama pelatihan, semua ibu-ibu antusias mengikuti jalannya kegiatan serta aktif berdiskusi dua arah. Peserta memahami dan dapat mempraktikkan teknik pengolahan ampas kelapa menjadi bahan baku pembuatan roti dan dapat menjadi peluang usaha. Abstract

ABSTRACT Coconut plants can be utilized to make various products that have their own economic value. Almost every part of the coconut plant can be utilized to meet human needs, including VCO made from coconut meat and Nata de Coco from coconut water. However, until now, the processing of coconut meat and water into commercially valuable products is still very limited and not optimal. The limited knowledge and skills of the community in processing coconut fruit are the main problems. Therefore, this community service program is carried out to solve these problems. This community service aims to provide education and improve the knowledge and skills of the community in utilizing coconut meat into VCO and coconut water into Nata de Coco so that products with economic value are produced. This community service is carried out using lecture, demonstration and discussion methods. The lecture aims to provide insight to participants regarding the benefits and methods of making VCO and Nata de Coco. The demonstration was carried out by giving a demo to the participants and then making VCO and nata de coco together while holding a discussion. The participants involved in the activity were the people of Tondonggeu Village, Nambo District, as many as 20 participants. The evaluation results showed that this training activity could increase knowledge and improve the skills of the community in making VCO and Nata de Coco. In addition, the results of the activities carried out are processed coconut products in the form of VCO and Nata de Coco. Key words: Coconut Fruit, Virgin Coconut Oil (VCO), Nata De Coco, Fermentation   ABSTRAK Tanaman kelapa dapat dimanfaatkan menjadi berbagai macam produk yang memiliki nilai ekonomi tersendiri. Hampir setiap bagian tanaman kelapa dapat dimanfaatkan untuk memenuhi kebutuhan hidup manusia diantaranya adalah VCO yang dibuat dari daging kelapa dan Nata de Coco dari air kelapa. Namun hingga saat ini, pengolahan daging dan air  kelapa menjadi produk yang bernilai komersil masih sangat terbatas dan belum optimal. Keterbatasan pengetahuan dan keterampilan masyarakat dalam mengolah buah kelapa menjadi permasalahan utama. Oleh karena itu, program pengabdian ini dilakukan untuk menyelesaikan permasalahan tersebut. Pengabdian ini bertujuan untuk memberikan edukasi dan meningkatkan pengetahuan serta keterampilan masyarakat dalam memanfaatkan daging kelapa menjadi VCO dan air kelapa menjadi Nata de Coco sehingga dihasilkan produk bernilai ekonomi.  Pengabdian ini dilaksanakan menggunakan metode ceramah, demonstrasi dan diskusi. Ceramah bertujuan untuk memberikan wawasan kepada peserta terkait manfaat dan cara dalam membuat VCO dan Nata de Coco.  Demonstrasi dilaksanakan dengan memberikan demo kepada peserta kemudian sama sama membuat VCO dan nata de coco sambil melaksanakan diskusi. Peserta kegiatan yang terlibat adalah masyarakat Kelurahan Tondonggeu Kecamatan Nambo sebanyak 20 peserta. hasil evaluasi menunjukkan kegiatan pelatihan ini dapat menambah pengetahuan dan meningkatkan keterampilan masyarakat dalam membuat VCO dan Nata de Coco. Selain itu, hasil dari kegiatan yang dilakukan adalah produk olahan buah kelapa berupa VCO dan Nata de Coco. Kata Kunci: Buah Kelapa, Virgin Coconut Oil (VCO), Nata De Coco, Fermentasi.
