Abstract

Taman Nasional Lore Lindu Sulawesi Tengah memiliki produktivitas serasah yang tinggi, sehingga sangat bergantung pada keberadaan mikroorganisme tanah untuk mendekomposisinya menjadi unsur hara yang dimanfaatkan oleh tumbuhan. Kondisi hutan primer yang masih alami dapat mendukung pertumbuhan dan keanekaragaman mikroorganisme tanah terutama bakteri. Penelitian ini bertujuan untuk mengidentifikasi jenis-jenis bakteri tanah di hutan sekitar Danau Kalimpa?a kawasan Taman Nasional Lore Lindu. Penelitian ini dilakukan pada bulan Oktober sampai Desember 2016 di Laboratorium Bioteknologi Jurusan Biologi FMIPA Universitas Tadulako. Bakteri diisolasi dari sampel tanah rizosfir yang diambil secara random di beberapa area kecil, menggunakan metode enrichment dan spread plate untuk memperoleh isolat. Isolat dimurnikan dengan metode streak plate kemudian diindentifikasi berdasarkan uji mikrobiologis dan uji biokimiawi. Dari sampel tanah hutan di sekitar Danau Kalimpa?a Kawasan Taman Nasional Lore Lindu berhasil diisolasi 5 bakteri isolat terpilih dengan kode isolat DKL 1, DKL 2, DKL 3, DKL 4 dan DKL 5. Berdasarkan hasil identifikasi secara fenotipik, isolat terpilih merupakan anggota kelompok bakteri Gram negatif dengan bentuk sel coccus dan bacil, ada yang bersifat motil dan non motil, serta mampu menggunakan karbohidrat sebagai sumber energinya melalui proses fermentasi

Abstract

Hyptis capitata Jacq. plant has been using for traditional medicine. Utilization of medicinal plant must always consider to given dose because of toxic effect when too much medicine is taken. The aim of this study was to measure the liver damage in rats caused by H. capitata Jacq. leave extract. Either 300 (P1), 600 (P2) or 900 (P3) µg/kgBW H. capitata Jacq. leave extract was given orally to rats every 24 hours during 14 days. Zero point five ml ethanol 96% was given daily (K-) and without given anything (K+) was also conducted as a control. Both macroscopic and microscopic of liver damage were assessed. The result showed that rats given P3 treatment had the highest liver damage. The liver damage in rats was not statistically significant difference between P3 and K- treatments. The lowest liver damage was in rats given K+ treatment. There was no significant difference between P1 and P2 treatments on rats liver damage. Utilization of medicinal plant as traditional medicine should always be consider to doses.

Abstract

Gen HBB merupakan salah satu gen yang berperan dalam sintesis hemoglobin, khususnya protein β-globin. Berbagai metode analisis molekuler untuk mendeteksi kelainan atau varian pada gen penyandi hemoglobin membutuhkan produk amplifikasi DNA yang berkualitas. Penelitian ini bertujuan menentukan kondisi PCR paling optimal untuk mengamplifikasi sekuen gen HBB dari sampel darah. Kami melakukan uji kualitatif dan kuantitatif pada DNA sampel yang telah diisolasi, lalu mengamplifikasi sekuen gen HBB menggunakan 2 pasang primer dengan 6 variasi komposisi master mix dan suhu annealing. Hasil penelitian menunjukkan bahwa DNA sampel berhasil diisolasi dengan rasio kemurnian 1,925 dan konsentrasi 48,9 µg/ml. Produk amplifikasi menggunakan primer HBE01 sangat tipis dan tidak spesifik, sedangkan primer HBE02 berhasil mengamplifikasi sekuen gen HBB dengan spesifik dan tampak cukup tebal. Amplifikasi sekuen gen HBB lebih baik menggunakan primer HBE02 dengan komposisi master mix dan suhu annealing sesuai Mix 6.

Abstract

Corn, rice, and sugarcane are included in the Poaceae family which have high economic value as staple foods. Nitrate reductase plays a role in reducing nitrate to ammonia. The nitrate reductase activity testing can help biotechnology studies, in this case, fertilizer production or organic waste treatment, because the nitrate reductase enzyme can be used to convert nitrate into more useful nitrogen compounds. The purpose of this study was to analyze the activity of nitrate reductase (ANR) from the leaves of the Poaceae family, namely corn, rice, and sugarcane. In vivo ANR measurements were carried out using the spectrophotometric method with two types of spectrophotometers, namely the Vis/UV Spectrophotometer, Genesys10UV, and Visible Spectrophotometer, with a wavelength of 540 nm. This study concludes that rice had the highest ANR of 0.1503 µmol/gram leaf weight/hour of incubation. The increase in nitrate reductase activity indicates that the energy for nitrate reduction also increases and provides a large capacity for synthesizing amino acids, proteins, or total N assimilation.

Abstract

The hot spring of Pentadio Resort is one of the geothermal areas likely to be the place of bacteria or microorganisms that can produce amylolytic enzymes. This research aims to analyze bacteria in the Pentadio Resort hot springs that possess the potential to produce amylase enzymes, analyze the characteristics of bacterial isolates that produce amylase enzymes in Pentadio Resort hot springs and determine the types of bacteria that produce amylase enzymes in Pentadio Resort hot springs. The research employs a qualitative and descriptive method. The research involved several methods, including sample collection, media preparation, isolation and selection of thermophilic bacterial isolates, purification of thermophilic bacterial isolates, selection of isolates producing amylase enzymes, identification of selected isolates (motility test, biochemical test, gram staining), and molecular identification. The research yielded seven isolates, and isolate code A6 demonstrated the potential to produce amylase enzymes with a clear zone size of 23,26 mm, bacillus, gram positive, and positive motility. Subsequently, molecular identifidation was carried out in which, based on the results of phylogenetic tree reconstractiopisalate A6 was related to the bacteria Bacillus cereus strain MD 152, which belongs to the Bacillus cereus bacteria

Abstract

Pemphis acidula is a wild plant in rocky or sandy coastal areas and mangrove ecosystems. Different geographic characteristics may affect plant adaptability and have an impact on the emergence of various genotypes. This study was performed to reveal the phenetic relationship and genetic variation of P. acidula in 3 different areas in Tomini Bay, Gorontalo Province, Indonesia. We took 3 samples from each location and analysed them using 14 morphological characters and molecular approaches based on ISSR markers and ITS gene. The results showed that P. acidula on Olele had bigger sizes in some morphological features compared to the plants in other study areas. The phenetic analysis showed that P. acidula at Biluhu and Dulanga were more closely related, although P. acidula at the 3 locations had 100% similarity. Genetic variation analysis showed the highest genetic similarity based on ISSR markers was found in Dulanga and Biluhu samples (76.8%). Phylogenetic based on ITS gene revealed that Olele samples were in the same clade with P. acidula accession from GenBank (genetic distance 0-0.19%), while Biluhu samples were a sister group (genetic distance 24.97-25.03%) even though their percentage identity corresponds to P. acidula (81.34%). Plant adaptation to different habitat conditions may affect the genetic diversity of P. acidula.

Abstract

Fish as an important source of animal protein for humans is very easily damaged, so it must be processed immediately for consumption. People generally sell fish in fresh form or in smoked form. This community service activity aims to socialize the potential of developing fish-processed culinary products to improve society's economy. The activity was carried out in Bilungala Village, Bonepantai District, Bone Bolango Regency, Gorontalo Province. This activity uses the counseling method, including the delivery of material by the speakers, followed by discussion sessions and fish processing demonstration. Participants who were PKK women and Posyandu cadres in Bilungala Village were very enthusiastic and grateful for the materials and training provided because they broadened their horizons and knowledge, and increased their motivation to try to make culinary processed fish products independently at their respective homes.

Abstract

Gorontalo Province is one of the provinces that has a wealth of natural resources, especially fishery and marine products. One area that has the highest fish production is Bilungala Village, Bone Bolango Regency. Fish is a food rich in protein, minerals, vitamins, and fatty acids. In addition, fish also contain omega-3 fatty acids and vitamin K which the body needs. All the nutritional content in fish is very influential on the growth and development of children. But in fact, the level of fish consumption in Indonesia is still very low compared to other largest fish-producing countries. Therefore, socialization is carried out through the use of marine fish as a source of nutrient-rich food. The purpose of this service is to provide information to the public about the nutritional content of fish, increase fish consumption in the community, and increase the hobby of eating fish. The location of service is carried out in Bulungala Village, Bonepantai District, Bone Bolano Regency, Gorontalo Province. The target of this service is the people of Bilungala Village, especially PKK women with activities carried out in November 2022. The activities consist of the preparation, implementation, and closing stages. In the preparatory stage, a survey was carried out in the village, then in the implementation stage, socialization was carried out to the community through the provision of material and ended with a discussion/question and answer regarding the material presented.

Abstract

Usaha teknik ecoprint dianggap memiliki peluang besar untuk berkembang, terutama di wilayah pedesaan, karena memiliki potensi alam yang melimpah dengan pepohonan yang lebat, tumbuhan yang subur, dan beragam jenis dedaunan yang bisa dimanfaatkan untuk membuat produk ecoprint. Kegiatan  pembuatan ecoprint di  Indonesia  sudah  banyak  dilakukan oleh pecinta ecoprint, namun belum banyak dikenal oleh masyarakat umum. Di Provinsi Gorontalo sendiri, telah ada beberapa  usaha fashion lokal yang memproduksi produk ecoprint melalui pemesanan oleh konsumen terlebih dahulu, namun hal ini masih tidak familiar bagi masyarakat padahal bisa menjadi salah satu sumber penghasilan dalam bidang ekonomi kreatif, tidak terkecuali masyarakat di Dusun Pahu Desa Bontula, Kecamatan Asparaga Kabupaten Gorontalo. Kegiatan Pendampingan Pembuatan Ecoprint pada Kelompok Perempuan untuk Pengembangan Produk Fashion  bagi masyarakat Dusun Pahu, Desa Bontula, Kecamatan Asparaga Kabupaten Gorontalo ini penting untuk dilakukan sebagai usaha dalam menambah pengetahuan serta keterampilan masyarakat untuk mengembangkan hobi dan memperoleh tambahan penghasilan. Pada pendampingan ini, dipilih teknik pounding karena teknik ini relatif mudah dilakukan, tidak memerlukan alat dan bahan yang mahal. Kegiatan pengabdian ini dilaksanakan secara tatap muka. Peserta yang mengikuti kegiatan berjumlah 14 orang yang merupakan kelompok wanita Dusun Pahu. Kegiatan dilaksanakan pada tanggal 25 Mei 2024 bertempat di Dusun Pahu, Desa Bontula, Kecamatan Asparaga, Kabupaten Gorontalo. Kegiatan ini melalui beberapa tahapan pelaksanaan, mulai dari persiapan, perencanaan, pelaksanaan, dan evaluasi yang melibatkan narasumber yang sudah expert di bidang pembuatan ecoprint.

Abstract

Kecamatan Bonepantai merupakan salah satu kawasan pengembangan perikanan (KPP 4). Kecamatan Bonepantai memiliki 8 desa nelayan, salah satunya Desa Bilungala. Secara keseluruhan Kecamatan Bonepantai memiliki jumlah nelayan ataupun RTP tertinggi kedua setelah Kecamatan Kabila Bone. Melihat potensi Kelautan dan Perikanan yang cukup besar di Kecamatan Bonepantai, perlu dilakukan usaha strategis untuk pengembangan produk perikanan sebagai upaya pemanfaatan hasil tangkapan laut. Pengolahan ikan dikalangan masyarakat desa bilungala masih sebatas olahan bakar, abon, dan acar ikan. Kegiatan ini bertujuan memberdayakan masyarakat khususnya Ibu-Ibu dengan melakukan pelatihan pembuatan nugget berbahan dasar Ikan Cakalang. Metode yang digunakan adalah metode partisipatif dimana masyarakat ditempatkan sebagai subjek dalam setiap tahapan kegiatan. Kegiatan pelatihan pembuatan nugget dilaksanakan dengan jumlah peserta 15 orang Ibu-ibu Kader Posyandu Desa Bilungala. Kegiatan ini diawali dengan presentasi mengenai bahan baku yang dapat digunakan dalam pembuatan nugget dan penjelasan singkat terkait cara pembuatan dan penyimpanan yang baik dan benar. Melalui kegiatan pelatihan pembuatan nugget ikan ini diharapkan masyarakat Desa Bilungala mampu mengolah hasil tangkapan menjadi produk olahan dengan maksimal untuk menunjang kesejahteraan dan memilih nugget ikan sebagai cemilan sehat bagi anak-anak di Desa Bilungala
