Abstract

Multicomponent crystal (MCC) formulation consists of drug and co-former that spectrophotometrically has an interference each other. The aim of this research was to evaluate and develop the analytical method to obtain a fast, cheap, efficient, and adequate method to estimate desloratadine (DES) in MCC formulation using three different methods. Single wavelength calibration model was constructed at 290 nm, multiple component equations was obtained from 273 nm and 283 nm, and multivariate analysis consists of PLS and PCA, a chemo-metric based, assisted the model selection and model calibration. Single wavelength method and multivariate analysis model had been fully validated i.e. range, specificity, accuracy, precision, limit of detection, and limit of quantification. A multivariate analysis had more adequate ability to estimate the DES and co-former in the MCC formulation.Keywords: Desloratadine, multicomponent crystal, multivariate, PLS

Abstract

This research purposed to optimize, investigate and evaluate the influence of xanthan gum, sodium bicarbonate-citric acid as effervescent components and hardness on floatation behavior and drug release of gastro-floating captopril tablet with floating system. A 23 full factorial design (8 runs) was applied to optimize the floating captopril tablet using xanthan gum (X1), ratio of effervescent components (X2) and hardness (X3) as independent variables. Optimum area was determined by superimposed contour plot of floating lag time (Y1), cumulative drug release at 60min (Q60) (Y2) and drug release constant rate (Y3) using Design Expert® software. Xanthan gum, effervescent components and hardness were affected the floatation behavior and drug release. Hardness was the most dominant factor affected the floatation behavior and drug release. Based on superimposed contour plot, the optimum area was in range of xanthan gum 58–100mg, sodium bicarbonate 45–63mg, citric acid 7–25mg and hardness at 70–98N.

Abstract

ABSTRAK Berbagai polimer alam telah digunakan sebagai kontrol pelepasan obat dalam sediaan lepas lambat. Karagenan, pektin, dan glukomanan berpotensi sebagai matriks untuk mengontrol pelepasan obat. Penelitian ini bertujuan untuk mengetahui dan membandingkan pola dan metode pelepasan obat dati matriks berbasis natural-gum (karagenan, pektin dan glukomanan) dengan berbagai model kinetika pelepasan obat pada tablet lepas lambat teofilin. Teofilin digunakan sebagai model obat dan tablet dibuat dengan metode granulasi basah. Matriks yang digunakan adalah pektin, karagenan, dan glukomanan dengan konsentrasi 20%, 30%, dan 40%. Pemeriksaan mutu fisik dilakukan pada granul dan tablet. Pelepasan obat dilakukan dengan alat tipe II USP dengan putaran 75 rpm, medium dapar fosfat pH 4,5 selama 6 jam. Hasil dibandingkan secara statistic dengan ANOVA taraf kepercayaan 95%, model dependent dan model independent. Hasil menunjukkan bahwa peningkatan konsentrasi matriks glukomanan dan pektin menurunkan waktu alir dan peningkatan konsentrasi matriks menurunkan kompaktibilitas. Matriks pektin dan karagenan menunjukkan pelepasan konstan. Kinetika pelepasan yang paling dominan mengikuti kinetika orde nol dengan koefisien determinasi paling tinggi dan perbedaan antara data yang teramati dan data prediksi yang rendah (AICc dan RMSE) dan mekanisme pelepasan dikontrol oleh erosi pada matriks. Kata kunci: pektin, karagenan, glukomanan, pelepasan obat ABSTRACT Various natural polymers had been used as a control drug release in sustained release dosage form. Carrageenan, pectin, and glucomannan were potential as a matrix to control the release of drugs. This study is aimed at determining and comparing the patterns and methods of drug release from matrix natural-gum based (carrageenan, pectin, and glucomannan) with different models of drug release kinetics in sustained release theophylline tablets. Theophylline was used as a drug model and tablets which was made by wet granulation method. The matrix used is pectin, carrageenan, and glucomannan with the concentration 20%, 30%, and 40%. Physical quality inspection was conducted on granules and tablets. Drug release was carried out with apparatus type II USP at 75 rpm rotation, medium phosphate buffer pH 4,5 for 6 hours. The results were statistically compared by ANOVA level of 95%, dependent model and independent model. The results showed that the increasing concentration of glucomannan and pectin matrix decreased the flow time and increased concentrations of matrix lowers compatibility. Matrix of pectin and carrageenan showed a constant release. The most dominant release kinetics followed zero order kinetics with the highest determination coefficient and the difference between the observed data and the data predicted low (AICC and RMSE) and the release mechanism was controlled by the erosion of the matrix. Abstract

Sistem penghantaran sediaan patch transdermal mampu meningkatkan kepatuhan pasien, diabetes melitus dengan menghindari regimen dosis yang rumit serta penggunaan injeksi insulin. Senyawa 11β,13-dihydrovernolide yang diisolasi dari tanaman daun afrika (Vernonia amygdalina) terbukti secara efektif mampu menurunkan kadar gula darah. Microneedle merupakan alat untuk menghantarkan obat secara perkutan sehingga mampu mencapai sirkulasi sistemik. Penelitian ini bertujuan untuk memformulasikan nanoemulsi isolat ekstrak terpurifikasi daun afrika terinkorporasi dalam dissolved microneedle patch. Senyawa diisolasi dan diekstraksi menggunakan maserasi dengan pelarut metanol, dilanjutkan fraksinasi cair-cair, dan kromatografi kolom. Purifikat yang diperoleh dikarakterisasi profil kromatogram lapis tipis dan spektra vibrasional. Pengembangan formula nanoemulsi menggunakan teknik mixture design untuk memperoleh formula optimum yang diinkorporasikan ke dalam campuran polimer dan microneedle dicetak menggunakan master template. Pengujian in vivo dilakukan menggunakan tikus sebagai model hewan uji dan dikaji penurunan kadar gula darah tikus. Hasil fraksi semipolar diperoleh rendemen 11,99 % dan isolat daun afrika diperoleh rendemen 0,075%. Profil kromatogram lapis tipis menunjukkan spot tunggal pada sampel isolat dan spektra vibrasional mengkonfirmasi gugus-gugus senyawa 11β,13-dihydrovernolide. Karakterisasi ukuran droplet nanoemulsi menunjukkan hasil 73,54±3,92 nm dengan distribusi ukuran droplet yang seragam 0,594±0,058. Jarum yang dihasilkan berukuran pada rentang 100-250 µm dengan nilai folding endurance patch sebesar >300 kali. Hasil pengujian kadar glukosa menunjukkan bahwa kelompok patch transdermal nanoemulsi mampu menurunkan kadar gula darah sebear 60% dan tidak berbeda bermakna dengan control positif. Formula patch isolat tanpa nanoemulsi mampu menurunkan kadar gula darah tikus sebesar 20%. Formulasi nanoemulsi isolat daun afrika terinkorporasi dalam dissolving microneedle menunjukkan peningkatan efektivitas antidiabetes dibandingkan tanpa formulasi nanoemulsi.

Abstract

The process of making herbal powder needs to be optimized because it is not efficient, and the filling process is still manually into teabags. The purpose of empowerment is to increase the effectiveness and efficiency of the herbal medicine-making process through a pharmaceutical approach with granulation techniques. This service is based on data obtained quantitatively in the laboratory, including the characteristics of the size distribution of Simplicia powder and granules and the flowability of herbal medicine powders. The data obtained were analyzed statistically with a 95% confidence level (p=0.05). The results showed that the granulation technique increased the flowability of the herbal powder with a flowability of 0 to 11.94±0.54 g/second. The granulation technique is able to improve the process of extracting herbs during brewing. The success of this activity is measured through the technique of increasing the flowability of herbal powders and the use of automatic teabag fillers. The results of the program showed that the granulation technique was able to increase the efficiency of the herbal powder manufacturing process at the Suti Sehati Herbal Medicine Factory, Sukoharjo.

Abstract

The demand for traditional medicine increased during the pandemic, including instant herbal medicine for children. Children who consume herbal medicine are more interested in colorful products according to the advertised taste, while adult consumers are more interested in herbal compositions that are slightly sweeter (less sugar). This service aims to provide alternative natural dyes and non-sugar fillers for instant herbal products. Accordingly, the service was carried out in two stages. The service program was carried out using the tutorial method and focus group discussions (FGD). Tutorials were carried out online with natural materials as a source of alternative natural dyes and innovations in the use of non-sugar fillers. Group discussions focused on innovations in using non-sugar fillers with low production costs. The results showed that Rosella flowers (Hibiscus sabdariffa L.) are recommended as natural dyes for instant herbal medicine for children for red, yellow, and purple colors, while Suji leaves (Dracaena Angustifolia) for green colors. Additionally, Maltodextrin Fluidized Bed Dryer (FBD) ) is recommended as a non-sugar filler.

Abstract

Herein, this work aimed to develop an ultraviolet spectrophotometric assay for metformin HCl in comparative dissolution testing along with the acid stage medium due to low molar absorptivity. Metformin HCl was incorporated into the immediate tablet formulation to mimic the analytical matrix system. The dissolution validation procedure was carried out by linearity, accuracy, precision, the limit of quantification, filter compatibility, and aliquot stability that fulfil the USP method for validation of dissolution procedure, particularly assay of the analyte.The results revealed that metformin HCl had difficulty quantifying in acid pH due to the absence UV-band peak. The addition of pH-shifting agents promoted better performance for metformin quantification. Therefore, the method was successfully developed along with R2, predicted R2, accuracy, and precision of 0.9999, 0,9998, 100.57%, and 1.27%, respectively. In addition, it had the minimum placebo interference (0.47%) and was stable for long-term storage under determined conditions.

Abstract

Tamanu oil is a promising natural wound healer due to its chemical compounds, particularly fatty acids. Cold-pressed or hot-pressed methods are commonly used to obtain this oil from Calophyllum seed. However, there needs to be research documenting the impact of these two extraction methods on the fatty acid profile and wound healing activity. Therefore, the study aims to characterize the fatty acid profiles of cold-pressed and hot-pressed tamanu oils using Gas Chromatography and evaluate their wound healing activity in vivo. The fatty acid profiles were analyzed using GC, and the wound healing tests were conducted on animal subjects divided into four groups: negative control, positive control (Bioplacenton®), cold-pressed tamanu oil, and hot-pressed tamanu oil. Cold-pressed tamanu oil showed superior fatty acid characteristics with an acid value of 38,71 Mg KOH/g fat and a peroxide value of 3,0095 mEq O2/kg, indicating that oil is stable against oxidation.  The length of the wound was observed daily for up to 8 days to assess its effect. The parameters observed were the percentage of wound healing and the total AUC based on the average length of the wound. Cold-pressed tamanu oil demonstrated the highest wound healing efficacy compared to both the positive control and hot-pressed tamanu oils. Both cold (11,67± 0,78) and hot-pressed tamanu oil (11,87 ± 0,61) exhibited significant differences in AUC value compared to the negative control group (13,07 ± 0,38), highlighting the potential of tamanu oil as a wound healing agent.

Abstract

Indonesia memiliki keanekaragaman hayati yang melimpah. Banyak tanaman di negara ini memiliki potensi besar untuk dimanfaatkan sebagai bahan baku produk bernilai ekonomi tinggi. Salah satunya adalah tanaman lerak (Sapindus rarak). Namun, tidak semua masyarakat mampu mengoptimalkan potensi tersebut karena kurangnya pengetahuan dan keterampilan mengenai pengolahan dan pemanfaatan bahan alam. Oleh karena itu, pengabdian ini ditujukan untuk memanfaatkan sumber daya alam lokal dalam meningkatkan produktivitas industri rumah tangga. Pengabdian ini dilakukan dengan metode sosialisasi, pendampingan, dan pelatihan untuk masyarakat Dusun Ingasrejo dalam mengoptimalkan buah lerak sebagai sabun cuci antiiritasi dan ramah lingkungan serta memilki nilai komersial. Dari hasil pengabdian didapatkan masyarakat memiliki pengetahuan lebih dan mendalam mengenai bahan alam khususnya buah lerak. Hal tersebut dibuktikan dengan adanya peningkatan pemahaman warga setelah dilakukan evaluasi, serta mengasah keterampilan baru yaitu pembuatan produk sabun cuci buah lerak. Selain itu juga memberikan manfaat ekonomi serta pelestarian lingkungan.

Abstract

Jamu merupakan warisan budaya Indonesia yang telah digunakan secara turun menurun oleh nenek moyang. Penggunaan jamu dalam pengobatan hanya didasarkan pada data empiris yang diturunakan terus menerus antar generasi. Pembuktian khasiat dan kemanan jamu secara ilmiah telah diatur dalam Peraturan Menteri Kesehatan Republik Indonesia Nomor : 003/MENKES/PER/I/2010 dalam bentuk saintifikasi jamu. Saintifikasi produk jamu industri perlu dilakukan untuk mempertahankan eksistensi produk jamu di masyarakat. Tujuan pengabdian ini yaitu untuk meningkatkan penyediaan jamu berkhasiat melalui pembuktian khasiat jamu secara ilmiah. Standarisasi proses manufaktur granul jamu instan dilakukan sebagai bentuk saintifikasi jamu di industri. Standarisasi proses dilakukan berdasarkan penetapan kadar fenolik total dan nilai aktivitas antioksidan. Kadar fenolik total dianalisis dengan teknik Folin-Ciocalteu menggunakan asam galat sebagai senyawa pembanding. Aktivitas antioksidan dianalisis berdasarkan reaksi transfer electron senyawa antioksidan dengan •ABTS. Hasil penetapan menunjukkan adanya penurunan kadar fenolik total dari rimpang (0.1% GAE/g), hasil penyarian (0.065% GAE/g), dan hasil ruahan (0.057% GAE/g). Hasil penetapan aktivitas antioksidan menunjukkan adanya penurunan aktivitas antioksidan dari rimpang (29.79 IC50 mg/mL) ke hasil penyarian (34.62 IC50 mg/mL) serta terdapat peningkatan pada hasil ruahan (17.18 IC50 mg/mL) karena adanya bahan tambahan.
