Abstract

In an effort to improve public health by reducing the use of hazardous food additives in the city of Medan, community service has been carried out at the Perwiritan Batak Islam (PWBI) in Kwala Bekala, Medan Johor, Medan city on December 19-20, 2024. The training focused on learning how to test for formalin and borax in food using the natural color reaction of turmeric and KMnO4 (blue medicine) with 18 participants. This activity contributed to enhancing the technical skills of testing for formalin and borax to prevent and reduce the risk of using hazardous food additives. After this training, participants' knowledge about natural color tests on food containing formalin and borax improved. Pre-test and post-test evaluations of 20 participants showed a significant improvement in their understanding, with evaluation graphs indicating a shift from a low level of knowledge to a better level of knowledge.

Abstract

Background: Gayo tobacco (Nicotiana tabacum L.) is known for its distinctive quality and is highly sought after, particularly among young adults. This tobacco contains nicotine, a compound that plays a significant role in influencing human behavior, acting both as a stimulant and as a raw material for cigarette production. Understanding the nicotine content in Gayo tobacco is crucial for assessing its potential industrial applications and its impact on health. Objective: This study aimed to determine the nicotine content in Gayo tobacco leaves and analyze it using acidimetric titration and GC-MS methods to obtain more accurate results. Methods: This research employed two analytical methods: acidimetric titration for quantitative nicotine determination and Gas Chromatography-Mass Spectrometry (GC-MS) for qualitative analysis of other chemical compounds in the Gayo tobacco leaf extract. The tobacco leaf samples were obtained from the Gayo region of Aceh and extracted using 96% ethanol. Results: The study revealed that the nicotine content in Gayo tobacco leaves was 1,3% based on acidimetric titration, while the ethanol extract showed a higher nicotine concentration of 2.5%. GC-MS analysis identified various other compounds, including piperidine derivatives, terpenoids, and aromatic compounds, which have potential pharmaceutical and industrial applications. Conclusion: This study successfully determined the nicotine content in Gayo tobacco leaves using acidimetric titration and GC-MS. The findings provide a comprehensive overview of the chemical composition of Gayo tobacco, serving as a foundation for further research into its applications in pharmaceuticals and other industries.

Abstract

Staphyococcus aureus bacteria is one of the bacteria that contaminates crispy chicken meat and can cause Extraordinary Events or Kejadian Luar Biasa (KLB) in Indonesia as a cause of diarrhea. This is due to lack of hygiene in processing, storing and serving crispy chicken. Poisoning can occur due to food contamination by S. aureus enterotoxin. The aim of the research was to analyze the prevalence and identify the presence of S. aureus bacteria found in crispy chicken meat in the Amplas area of Medan city using Rabbit Coagulase Plasma with EDTA. Method for identifying S. aureus using BPA (Baird Parker Agar Base) and Egg Yolk media. Specific identification testing using Gram staining, catalase test, and coagulase test using Rabbit Coagulase Plasma with EDTA. The value of S. aureus bacterial contamination using the TPC (Total Plate Count) method and analysis of the prevalence of S. aureus contamination. The results of research on roadside cart krispy chickens were found to be positive for S. aureus with characteristics of Gram positive, positive coagulase and positive catalase as well as contamination values above 1X102 CFU/ml exceeding the threshold so they do not meet SNI standards. Meanwhile, the local fast food crispy chicken value for the presence of S. aureus bacteria does not exceed the threshold of 1X102 CFU/ml so it meets SNI standards. The prevalence value of S. aureus contamination in roadside cart krispy chicken was 75.0% and local fast food was 8.3%.

Abstract

Tobacco plants are considered more destructive plants than medicinal plants. This research aims to analyze using GCMS (gas chromatography mass spectrometry) the leaves extract of Bakong Gayo or Gayo tobacco (Nicotiana tabacum L) and its pharmaceutical uses. Extraction of Gayo tobacco leaves with chloroform solvent obtained 14 phytochemical compounds in the form of alkaloids, terpenoids and fatty acids. The phytochemical compounds in Gayo tobacco can be used as antibacterial, anticancer, anti-depressant, antioxidant, sedative, aromatherapy based on references and other potential to be developed as a medicinal plant.

Abstract

Gayo-origin tobacco leaves (Nicotiana tabacum L.) are a locally cultivated tobacco variety widely grown in Central Aceh, Indonesia. Tobacco leaves are known to contain nicotine, a bioactive compound with various pharmacological and economic potentials. This study aimed to identify the nicotine content and phytochemical profile of Gayo tobacco leaf extract. The research procedures included sample collection, macroscopic and microscopic examinations, simplicia preparation, characterization of the simplicia and extract, phytochemical screening, quantitative nicotine determination using acidimetry titration, and qualitative analysis using Gas Chromatography–Mass Spectrometry (GC–MS). Macroscopic examination showed that the leaves were oval to round in shape, broad-surfaced, green in color, with a characteristic odor and a bitter taste. Microscopic observation revealed the presence of vascular bundles, covering trichomes, and glandular hairs in the leaf fragments. The titrimetric test showed nicotine levels of 0.95 ± 0.3% in the simplicia powder and 1.87 ± 0.45% in the ethanol extract. GC–MS analysis identified several major compounds, including nicotine (20.99%) and Labda-8(20),13-dien-15-oic acid (29.85%) as dominant components, along with minor compounds such as Quinolin, 8-amino-6-hydroxy-Quinazoline (2.39%). These findings indicate that the Gayo tobacco leaf extract contains a relatively high level of nicotine and is dominated by alkaloid and diterpenoid acid derivatives, suggesting its potential for further phytochemical and pharmacological exploration.
