Abstract

Pleurotus ostreatus is a common mushroom cultivated in Indonesia, and potential properties of bioactive compounds for medicinal mushroom. This study was aimed at obtaining P.ostreatus extract bioactive compounds potential in inhibiting the proliferation of cervical cancer cells (HeLa) and evaluating the HeLa cell proliferation kinetics and HeLa cell death mechanisms. The research was beneficial in making this product can be easily applied in a more controlled industrial scale. Anticancer activity test through a cytotoxic test using the MTT [3- (4,5-dimetiltiazol-2-yl) -2.5-diphenyl tertrazolium bromide], the kinetics proliferation of HeLa cells and HeLa cell death mechanism was performed. Linear regression analysis was used to analyze the data. Ethyl acetate extract of P. ostreatus isolated from Madiun showed the best results with IC 50 = 107.59 g / ml. HeLa cell proliferation kinetics analysis showed that the application of bioactive compounds 100 g / ml resulted in an increase of in death of HeLa cells along with length of incubation time. An important finding was that HeLa cells death by apoptosis was greater than by necrosis. In conclusion, the extracts of P. ostreatus has the potential to inhibit the growth of HeLa cells.

Abstract

Myrmecophytes or locally known as simbagh utak are common medicinal plants used by locals in Bengkulu, especially in South Bengkulu, for treating various diseases. Despite their potential as medicines, there is no report on biotic factors can be used to indicate of Myrmecophytes species’ richness and habitat suitability. The objectives of this research were to analyze the Myrmecophytes’ species richness and habitat suitability. This study used the purposive sampling method by exploring the area where Myrmecophytes were commonly found. The biotic factors of Myrmecophytes were analyzed by identifying the tree host, the other epiphyte plant grew around them, and animal occupants on the tuber of the Myrmecophytes. The Myrmecophytes distribution was analyzed by using ArcGIS10.1. The results showed that there were two species of Myrmecophytes, namely Hydnophytum formicarum and Myrmecodia tuberosa in the study area. The Myrmecophytes attached and hung in 9 species of host trees. The characteristics of host trees are high trees with large diameter, mostly rough-barked; some were cracked and mossy. The highest frequency of host trees included Hevea brasiliensis and Durio zibethinus. Myrmecophytes coexisted with 12 species of epiphytes. Epiphyte plants like Dendrobium sp. and Drymoglossum piloselloides can be used to indicate the presence of Myrmecophytes. Ants made up the most predominantly animal found living inside the tubers of Myrmecophytes, with some cockroaches and termites found at the tuber of Hydnophytum. Information on species richness and habitat suitability of Myrmecophytes can serve as supporting data for conservation efforts in Bengkulu to prevent the extinction of this species.

Abstract

Abstract

Butterflies play an important role in the ecosystem of Suranadi Natural Park in West Lombok. Butterflies help preserving the existence and diversity of flora by facilitating the process of pollination so it is crucial for flowering plants. The present study aimed at analyzing the diversity and determining the priority of butterfly conservation at Suranadi Natural Park of West Lombok. This exploration is a descriptive study. Data were observed in four-time repetition in the morning and in the afternoon for two months. The sweeping net technique following observation path was employed in this research (line left, line right,  line central and line waterway). The data analysis used the Shannon-Wiener diversity index and priority for conservation determined by Ministry of Forestry Regulation Number: P.57 / Menhut-II / 2008. Fourty (40) species of butterflies belonging to 5 families identified. The diversity index (H ) of butterflies ranges from 2.63 to 3.43 (medium-high). The two species of the Papilionidae family found at TWA Suranadi, namely, Troides helena and Papilio memnon were the priority of conservation. This research would be helpful to consider conservation strategy and ecotourism.

Abstract

Aquaculture can be improved by a diversified approach in the form of domestication research of a wild fish. The research has been done by cultivating Javaen Barb/Brek (Puntius orphoides) from Serayu river, Banyumas at a natural pond and laboratory scale. Brek fish is relative slow in growth and development of gonad. This research is aimed to determine: (1) the rate of general growth of Brek in the provision of some kinds of additional food; (2) Relative Growth Rate (RGR); (3) Specific Growth Rate (SGR) and Feed Conversion (FC). The research used experimental methods. The method was a completely randomized design (CRD) of 4 treatments and 3 replicationsof: A = 100% pellet; B = 70% soybean sprout and 30% pellet; C = 70% Ipomoea aquatica leaf and 30% pellet; D = 35% soybean sprouts, 35% I. aquatica leaf and 30% pellet. Quantitative data were analysed by analysis of variance (ANOVA) of the fishes growth.The results show influence of feed types to the Relative Growth Rate (RGR) of Javaean Barb. The best RGR with the value of 72.40 was given by feed types in the D combination. Meanwhile, Specific Growth Rate (SGR) and Feed Conversion (FC) were not affected by the feed types. Abstract

Botryococcus sp. is one of microalgae species that has a high lipid content as much as 75% of their dry weight. But, lipid production by microalgae is regulated by their environmental condition (pH, light, temperature, nutrition, etc). Mutagenesis induced by Gamma 60Co irradiation can be utilized to alter the Botryococcus sp. genetic to get microalgae mutant strain that can produce a higher lipid content than the wild strain. Botryococcus sp. was irradiated with different doses of gamma ray of 60Co (0, 2, 4, 6, and 10 Gy), and the effect on the growth, lipid content, and fatty acid composition of microalgae were observed. Research design used is random complete (RAL) with 95 % confident level for quantitive analysis based on the biomass and lipid contents. More over fatty acid composition was analyzed by Gas Cromatography-Mass Spectrometry (GC-MS). Results showed that Gamma irradiated gave an effect on growth and lipid content of Botryococcus sp. But between the control treatment (0 Gy) with microalgae irradiated dose of 2 Gy, 4 Gy and 6 Gy were not significantly different. Whereas between the control with 10 Gy irradiated was significantly different. The highest biomassa and lipid content are found in 10 Gy irradiated microalgae with 0.833 gram biomass and 41% lipid content. Fatty acid profile of Botryococcus sp. control has 6 fatty acids while 10 Gy irradiated microalgae has 12 fatty acids, with the long-chain fatty acids increased, whereas short-chain fatty acids decreased.

Abstract

Actinomycetes SAE4034 isolates was isolated from Rhizophora apiculata rhizosphere mud showed some antibacterial properties. The antibacterial ability of this isolate has not been tested on antibiotic resistant bacterial pathogens. However, there was no research has been reported regarding actinomycetes from Segara Anakan mangrove area resulting compounds inhibit the growth of antibiotics-resistant bacteria. Therefore, it is important to investigate its capability against antibiotics resistant bacteria or multi drug resistant bacteria (MDR bacteria). The research aimed to know the ability of actinomycetes SAE4034 in inhibit MDR bacteria and to identify the species profiles. The research methods included isolate characterization involving morphology, physiology/enzymatic and molecular properties. MDR bacterial inhibition assay, antibacterial compound extraction and antibacterial compound test using thin layer chromatography (TLC) method, observation of morphological and biochemical properties, DNA isolation, amplification and analysis of 16SrRNA sequence, and phylogeny tree analysis. The methods of this study included MDR anti-bacterial assay and antibacterial compound test. Subsequent step was isolate characterization including observation of morphological and physiological / enzymatic properties, and 16S rRNA gene sequence. The results showed that culture extract was able to inhibit the growth of MDR bacteria i.e. Escherichia coli, Staphylococcus aureus, Klebsiella pneumoniae, Pseudomonas aeruginosa, Enterococcus sp., but no inhibition to Enterobacter cloacae. The bioactive compound showed 4 spots with Rf values of 0.36; 0.45; 0.54; and 0.6. Based on morphology, physiology / enzymatic and 16S rRNA gene sequences characteristics, actinomycetes SAE4034 isolate is Streptomyces sp. This research showed new Streptomyces strain that serves as a source of MDR antibacterial compounds and useful in development of antibiotic for combating infectious diseases caused by MDR bacteria

Abstract

Tempe content of aglikon isoflavone which have potency to as anticancer compound. During processing of tempe, beside tempe mold (Rhizopus oligosporus) share other microbial namely bacterium. One of the bacterium residing at tempe ecosystem is Micrococcus luteus, having the character of gram positive and can conduct genistein biotransformation and daidzein become factor II compound (6,7,4-tri hydroksi isoflavone). Its problems how amount of R.oligosporus cell and M. luteus at processing of tempe associated is full scale of yielded tempe isoflavone. The study aimed to improve full scalely tempe aglikon isoflavone through inoculation of M.luteus bacteria. Used complete random designd, with M.luteus bacteria, as independent variable and variable depended is the amount of mold cell (R..oligosporus), amount of full scale M.luteus cell and total of isoflavone. Result of one way Anava indicate that inoculation of M.luteus bacteria do not degrade the amount of R.oligosporus cell with F call (53,28) > F tab (3,10) at significancy level 5%. Amount of M.luteus cell from 1,3 x 109 cell / gr [at] fermentation first becomes 3,4 x 106 cell/gr and decrease after day fermentation second becoming 1,75 x 106 cell/gr. Proportion of increasing isoflavone content at day fermentation 1-2 from 19,1 becoming 52,7 mg/100gr. Inoculation of M. luteus bacteria increased to count isoflavone aglicone and no trouble at growth of tempe mold and it relate to the increasing of is full scale of yielded tempe isoflavone.

Abstract

This research supports the sustainable environmental development, especially at Perhutani area. The objective of this ethno-ecological study was to know relationship between daily life of local people related to their agroforestry practices in edge of the forest belong to Perhutani at the landscape of Bromo Tengger Semeru-East Java. The Non-Timber Forest Products (NTFPs) showed the importance of the forest for the local people. This study covered the perception and conception traditional management system of environment by the local society and also impact of their activities. Data were collected by applying ethno-ecology research methods. Quality of data were measured by means of participatory ethno-botanical appraisal (PEA) and some research methods included semi-structural and open discussion, in-deep interview, direct observation and plants identification. The results showed that development and management of the natural resources, in Komplangan agroforestry, were highly related to the plant conservation policy. The land use system was adapted from indigenous knowledge which consisted of holly sites (Pedanyangan), worship sites (Sanggar Pamujan), cemetery area and terrace. The conservation model and traditional knowledge in agricultural practices could be used as a model of komplangan area which should be taken into account as the key of biodiversity conservation. Traditional knowledge from these integrative studies will support the sustainable development of NTFPs.

Abstract

The present study aims at expressing and partially purifying PtpB in active form. To achieve this, Mtb PtpB gene has been cloned into pET30a vector and overexpressed in Escherichia coli BL 21(DE3) under IPTG induction in the form of an inclusion body. Following resolubilization by urea and dialysis, the resulted PtpB has been shown to be active against para-Nitrophenyl phosphate. It is concluded that the resulted PtpB has had been recovered from inclusion body to give the active form of the enzyme, and thus the success in overexpressing PtpB provides the required material to investigate the biochemical properties of the pathogen virulence factor further.
