Abstract

The present study aims at expressing and partially purifying PtpB in active form. To achieve this, Mtb PtpB gene has been cloned into pET30a vector and overexpressed in Escherichia coli BL 21(DE3) under IPTG induction in the form of an inclusion body. Following resolubilization by urea and dialysis, the resulted PtpB has been shown to be active against para-Nitrophenyl phosphate. It is concluded that the resulted PtpB has had been recovered from inclusion body to give the active form of the enzyme, and thus the success in overexpressing PtpB provides the required material to investigate the biochemical properties of the pathogen virulence factor further.

Abstract

Indonesia merupakan masyarakat yang memiliki beragam budaya lokal yang dapat dikembangkan dalam kehidupan. Keragaman budaya ini dapat dioptimalkan sebagai salah satu sumber pembelajaran di sekolah. Pada pembelajaran IPA di sekolah kemampuan mengaitkan pengetahuan sains dengan fenomena di sekitar secara kontekstual dapat diperoleh melalui etnosains. Salah satu tradisi yang dapat diintegrasikan dalam pembelajaran adalah tradisi mamaq/menyirih. Sehingga tujuan penelitian adalah untuk mengidentifikasi dan mendeskripsikan budaya lokal masyarakat yaitu mamaq/menyirih untuk diintegrasikan dalam pembelajaran. Metode yang digunakan dalam penelitian ini yaitu dengan cara observasi, wawancara, dokumentasi dan studi pustaka. Hasil penelitian menunjukkan bahwa pengintegrasian tradisi mamaq/menyirih materi IPA pada kelas VII dan kelas VIII. Diantara materi yang dapat diintegrasikan ke dalam pembelajaran IPA di SMP adalah morfologi tumbuhan, taksonomi tumbuhan, jaringan tumbuhan, sistem pencernaan pada manusia, serta zat adiktif dan aditif.

Abstract

Mengajar berkaitan dengan kemampuan guru dalam menguasai materi dan keterampilan dasar mengajar. Tujuan dari kegiatan ini untuk melatih mahasiswa calon guru melakukan praktek mengajar sebagai persiapan praktek lapangan persekolahan. Sasaran kegiatan ini adalah mahasiswa Pendidikan Biologi FKIP Unram, semester 6 kelas A sebanyak 23 orang yang mengambil matakuliah Pengajaran Mikro pada tahun ajaran 2023/2024. Pada waktu pelaksanaan praktek, mahasiswa dalam kelas dibagi menjadi tiga peran yaitu satu orang sebagai guru, dua orang sebagai pengamat dan sisanya sebagai siswa. Mahasiswa yang berperan sebagai pengamat, mengamati pembelajaran dari awal hingga akhir menggunakan lembar pengamatan pelaksanaan pembelajaran. Hasil pengamatan menunjukkan mahasiswa sudah melaksanakan pembelajaran dengan tahapan pendahuluan, inti dan penutup, semua keterampilan dasar mengajar sudah terlaksana, walaupun indikator pada beberapa aspek keterampilan masih harus ditingkatkan keterlaksanaannya dengan terus berlatih. Dapat disimpulkan, mahasiswa sudah siap untuk melaksanakan praktek persekolahan di sekolah yang sebenarnya.

Abstract

Kegiatan pengabdian kepada masyarakat ini bertujuan untuk memperkenalkan konsep dasar mikrobiom dan perannya bagi kesehatan kepada mahasiswa PG-PAUD semester 2 FKIP Universitas mataram. Kegiatan dilakukan dalam bentuk penyuluhan interaktif yang mencakup pretest, ceramah, diskusi, dan posttest. Instrumen yang digunakan adalah angket ya/tidak sebanyak 15 butir untuk mengukur tingkat pengetahuan awal dan pasca-penyuluhan. Hasil pretest menunjukkan bahwa seluruh peserta belum memiliki pengetahuan dasar tentang mikrobiom. Setelah kegiatan, seluruh peserta menyatakan telah mengetahui semua item yang ditanyakan dalam angket. Ini menunjukkan adanya peningkatan tingkat kesadaran pengetahuan dari tidak tahu menjadi tahu secara menyeluruh. Meskipun bukan merupakan pengukuran pemahaman konseptual mendalam, hasil ini mengindikasikan bahwa penyuluhan berhasil menyampaikan informasi baru secara efektif dan menarik. Kegiatan ini menjadi langkah awal dalam menyebarkan informasi ilmiah mengenai mikrobiom yang masih relatif baru dikenal masyarakat umum, terutama bagi calon guru PAUD yang akan berperan penting dalam membentuk pola makan dan kesehatan anak usia dini. Diharapkan kegiatan serupa dapat diperluas kepada kelompok masyarakat lainnya.

Abstract

Pendidikan memegang peranan yang penting dalam kehidupan karena pendidikan merupakan wahana dalam proses peningkatan kualitas sumber daya manusia secara menyeluruh. Pendidikan adalah suatu faktor penting bagi kemajuan suatu bangsa. Penelitian ini bertujuan untuk mengetahui efektivitas penerapan pembelajaran quantum dengan peta konsep untuk meningkatkan kecakapan akademik dan kecakapan sosial siswa. Penelitian ini merupakan penelitian eksperimen semu dengan desain pre-test dan post-test group design. Populasi penelitian sebanyak 63 siswa dengan sampel ditentukan dengan simple random sampling dan diperoleh 2 kelas sampel yaitu kelas VIII A dengan 31 siswa, VIII B dengan 32 siswa. Instrumen penelitian terdiri dari tes kecakapan akademik dan lembar observasi kecakapan sosial. Hasil penelitian menunjukkan bahwa nilai rata-rata kecakapan akademik pada kelas eksperimen lebih tinggi dari kelas kontrol (85,65 > 62,97) dan persentase kecakapan sosial kelas eksperimen 92,4% lebih tinggi dari kelas kontrol yang hanya sebesar 65,3%. Hasil uji hipotesis kecakapan akademik siswa diperoleh nilai thitung 14,610 > ttabel  1.696, sedangkan kecakapan sosial siswa diperoleh nilai thitung 10,148 >ttabel 1.696. Kesimpulannya bahwa pembelajaran quantum dengan peta konsep efektif meningkatkan kecakapan akademik dan kecakapan sosial siswa.

Abstract

PGPR are beneficial bacteria that play a role in increasing plant growth and productivity through hormone synthesis and provision of dissolved nutrients that are readily absorbed by plant roots. This study aims to determine the effect of IAA-producing and phosphate-solubilizing bacteria from corn plants (Zea mays L.) on the germination of legume plants (Vigna sinensis L.) in vitro. Rhizosphere bacteria from corn plants were isolated from Tetebatu Village, Sikur District, East Lombok Regency. IAA production and phosphate solubilization tests were carried out qualitatively and quantitatively. Qualitative phosphate solubilization was carried out by measuring the clear zone formed on Pikovskaya solid media and quantitatively using a spectrophotometer method with a wavelength of 690 nm. The test of the effect of bacteria on the germination of Wulung variety long bean plants planted on modified Murphy media with parameters of plant height, root length, fresh weight and dry weight of plants for 5 days was then analyzed using One Way Anova. The results showed that 7 isolates were able to produce IAA and dissolve phosphate and 3 other isolates were only able to dissolve phosphate. The highest IAA production (29.17 ppm) was produced by isolate RJ5 on the 5th day of incubation. The highest phosphate solubilization (18.46 ppm) was produced by isolate RJ1 on the 8th day of incubation. The results of the analysis showed that the effect of IAA-producing bacteria and phosphate-solubilizing bacteria on the germination of long bean plants was significantly different for plant height parameters, but not significantly different for root length, fresh weight and dry weight of plants. It can be concluded that bacteria isolated from the rhizosphere of corn (Zea mays) with the codes RJI and RJ5 are able to increase plant growth so that they have the potential as biofertilizer candidates that can be developed in the future.

Abstract

Chili plants have a high selling value when the supply of goods decreases due to late harvests in red chili centers in various regions. This can trigger Indonesian farmers to cultivate more quality chili plants because they have high economic value. This research aims to determine the effect of NPK fertilizer and banana peel liquid organic fertilizer on the growth of red chili (Capsicum annum L) and to assess the impact of the combination of NPK fertilizer and banana peel liquid organic fertilizer on red chili. This study employed an experimental approach, with data collected by assessing plant growth. NPK fertilizer had no discernible effect on the height or quantity of leaves on red chili peppers, but it did have an impact on leaf area, fresh weight, and dry weight. Red chili pepper plants' height, leaf count, leaf area, fresh weight, and dried weight were all unaffected by the application of a liquid organic fertilizer made from banana peels. Red chili pepper plant height and leaf count were not significantly impacted by the combination of NPK fertilizer and liquid organic fertilizer generated from banana peels; but, leaf area, fresh weight, and dry weight were.

Abstract

Gold mining activities can cause environmental degradation and have negative impacts on human health. This study aimed to analyze mercury concentrations in blood cockles (Anadara granosa) in the Essential Ecosystem Area (EEA) of the Bagek Kembar Mangrove, Sekotong, West Lombok. Sampling was conducted using 10 x 10 meter quadrants, and the data were analyzed using a Cold Atomic Absorption Analyzer of Mercury (CAAAM). Based on the analysis results, mercury concentrations in the flesh of Anadara granosa were in the range of 0.0057 to 0.0394 mg/kg, with the highest average concentration recorded at the station closest to the tourist area (0.0357 mg/kg). Despite this, the mercury levels remain below the safety threshold established by National Agency of Drug and Food Control (BPOM) Regulation No. 9 of 2022, which is 1.0 mg/kg. Therefore, regular environmental monitoring and increased public awareness about the dangers of mercury use are strongly recommended tu support the long-term preservation of marine ecological system and safeguard the safety of seafood consumption. Future research is suggested to assess the safe daily intake limit of Anadara granosa for local communities based on the detected mercury levels, in order to evaluate potential health risks to consumers.

Abstract

Fruit flies (Bactrocera sp.) are important pests of horticultural crops because they can cause serious damage and reduce the selling value of the fruit. This study aims to determine the effect of lemongrass (Cymbopogon nardus) plant extract on the number of fruit fly catches and to determine the most effective concentration as a natural attractant. The study was conducted experimentally in a crystal guava plantation in Dasan Tapen Village, West Lombok, using a Randomized Block Design (RBD) with five treatments of lemongrass extract concentrations (0% - 60%) and three replications. Data were analyzed using one-way ANOVA and continued with the LSD test. The results showed that there were four species of fruit flies caught, namely Bactrocera dorsalis, B. carambolae, B. cucurbitae, and B. papayae, with a total of 671 individuals. The 60% concentration produced the highest number of catches (an average of 69), while the control without attractant did not produce any catches. Statistical analysis showed a significant effect between treatments (p < 0.05). It can be concluded that lemongrass ethanol extract has a significant effect on the number of fruit fly catches, and the highest concentration in this study (60%) is more effective than the concentration below it. The concentration of 60% has the highest capture compared to the concentration below it. The use of lemongrass as a natural attractant has the potential as an alternative to environmentally friendly pest control in an integrated agricultural system.

Abstract

This study aims to determine the levels of heavy metal mercury (Hg) in the bodies of Tilapia (Oreochromis niloticus) fish taken from Lake Rawa Taliwang to enrich the material in Ecotoxicology. This study is particularly useful in protecting consumers from heavy metal contamination when consuming fish. This study was conducted at Lake Rawa Taliwang, located in West Sumbawa. There were two separate stations in the study, namely on the east and west sides of the lake. Tilapia (Oreochromis niloticus) fish were caught using gill nets. Three to four fish samples were taken from each station. Each fish sample was then placed in a plastic bag and stored in a sample box. The research samples were then analyzed at the West Nusa Tenggara Health Testing and Calibration Laboratory. Data analysis was carried out by taking fish muscle tissue and analyzing the heavy metal content of Hg using an Atomic Absorption Spectrophotometer (AAS). The wet digestion stage of sample digestion is the process of degrading organic material in the sample using a strong acid, namely HNO3, as a digesting agent. The data obtained is the heavy metal Hg content in ppm (parts per million). The next stage is to process the obtained Hg concentration descriptively and then display it in a table according to the AAS results. The conclusion of this study was that the heavy metal Hg content in Tilapia (Oreochromis niloticus) originating from Lake Rawa Taliwang to enrich the Ecotoxicology material ranged from 0.044 to 0.086 ppm.
