Abstract

This study used catfish Clarias gariepinus as an experimental animal, which given supplementation of curcumin and thyroxine hormone. The results showed that during the reproductive period there was an increase in body weight of female catfish in line with the development of the gonads. Through the histology of the gonads, it appeared that oocyte development occurs over time.  The results of liver histology showed a high activity of hepatocyte cells as indicated by the formation of vacuoles, as the storage site for vitellogenin, in their cytoplasm. Supplementation of curcumin and thyroxine was able to increase the specific growth rate of catfish, and the catfish group that given the combination treatment of curcumin supplementation (5 g.kg-1 of feed) with thyroxine hormone (0.1 mg.kg-1 of feed) showed the more optimal results.

Abstract

Cellulolytic microbes produce cellulase enzymes that can degrade cellulose. Cellulolytic microbes are found in many habitats, including in cattle waste (bio-slurry), which is result of hydrolysis of cattle feed ingredients sourced from cellulose. Cellulase enzymes play a role in the process of hydrolyzing cellulose into glucose. This study aims to isolate cellulolytic microbes from bio-slurry and measure activity of cellulase enzymes in cellulolytic microbes isolated from bio-slurry. We are taking bio-slurry samples with 5 sample points and isolating the microbes in Carboxyl Methyl Cellulose (CMC) medium, cellulase enzyme activity qualitative test with Congo Red Assay, and perform quantitative test with the DNS Assay. Microbial isolates which shows positive results for cellulolytic activity in the qualitative test called SIIC1, SIIIC5, SIIC3. In the quantitative test can be seen based on Optical Density (OD) for cell lysate, SIIC1 0.298; SIIC3 0.186; SIIIC5 0.247; and a 0.332 for blank solution. Whereas supernatant obtain SIIC1 value of 0.237; SIIC3 0.212; SIIIC5 0.198; and 0.195 for blank solution. Based on the results, it can be concluded that the cellulolytic activity of microbes in bio-slurry based on OD values shows good results in supernatant, but it’s not significant enough in the cell lysate.

Abstract

Cocoa (Theobroma cacao L) production in Indonesian has increased every year, therefore more cocoa pod husks are produced. Currently, the use of cocoa pod husks is still very limited, even most farmers only dump the pods and turn them into plantation waste which will later become new problems in the plantation environment, such as causing bad smell and spreading diseases to healthy cocoa. Even if there is use of cocoa pods, at this point it is only used for animal feed and compost. In fact, cacao pod husks contain flavonoids and phenolic compounds which have high levels of antioxidant activity which are very beneficial fot the health. Based on the results of the sample analysis test using the FTIR and XRF instrument, there are functional groups that contain antioxidants and based on the XRF test, the pods of cocoa also contain several minerals, including 60,69% potassium and 24,56 calcium. Antioxidants can prevent the activity of free radicals which are the main causes of degenerative diseases. Therefore, cacao pod husks are very useful for making herbal drinks by drying and pounding them into a homogeneous powder product.

Abstract

Cardiovascular disease cases increase due to consumption cholesterol dietary habit. It is well-known that squalence synthase (SQS) is the first committed enzyme for cholesterol synthesis. Therefore, SQS become target of anti-cholesterol. This paper aims to determine the potential binding pocket of SQS (PDB ID: 1EZF). Dogsitescorer, siteFinder, and DEPTH were used for binding pocket prediction and MOE 2009.10 was performed for molecular docking. We found that there are five out of 37 pockets which have druggability score above 0.8. Pocket_5 is the highest drugability and favorable for hydrophobic interaction, yet lower number of hydrogen bond with the ligand. However, Pocket_2, and Pocket_3 are suitable for hydrogen bond formation of ligand-protein. Molecular docking study showed that TAK-475, D99, and Cynarin inhibitors were embedded on the P_2 and P_3 of SQS, showing that P2_and P3 are promising binding pocket for ligand interactions. These results show a promising alternative to design anti-cholesterol using these potential pocket in silico.
